Background

The dental pulp periapical disease is a bacterial infectious disease occurring in dental pulp and periapical tissues, has a failure rate of 10 to 15 percent after the complete and standardized root canal treatment, and can develop into persistent periapical inflammation. There are two major factors that contribute to root canal treatment failure. 1. The infection control is not good, and the bacterial biofilm in the root canal is not removed in place. For example, the common bacterium in root canals, enterococcus faecalis, can invade dentinal tubules to grow, and complicated bacterial biofilms can be detected in the periapical part to cause periapical tissue damage. 2. Complicated root canal system anatomical structure exists complicated structures such as vice root canal, apical bifurcation, root canal isthmus, C type root canal, and traditional root canal preparation is difficult to thoroughly clear away the pathogenic biomembrane in deepening complicated root canal system, still has remaining bacterium at root canal narrow and small position and dentinal tubule depths.

Root canal filling is an important step in root canal treatment, and plays a role in sealing the root canal and preventing bacterial reinfection. In the root canal filling, a core filling material and a root canal sealer are commonly used for filling the root canal, and the root canal sealer, the wall of the root canal and the dentin tubules form micro leakage to cause bacterial reinfection, so that the failure of root canal treatment is caused.

The root canal sealer which is commonly used clinically at present comprises zinc oxide clove root canal sealer, calcium hydroxide root canal sealer, resin root canal sealer, biological ceramic sealer, polymethyl silane root canal sealer and glass ion root canal sealer.

The resin root canal sealer mainly includes AH-26, AH-plus, Endorez, Realseal, 2Seal, Dialet, Epiphany and the like.

DMADDM is a novel antibacterial monomer of quaternary ammonium salt, and a certain amount of DMADDM is added into a dental material, so that the antibacterial effect on bacteria and biological films can be shown on the premise of not damaging the performance of the material.

The inventor researches and discovers that DMADDM is added into the resin root canal sealing agent according to the mass fraction of about 2.5%, and a certain antibacterial effect is generated on the multi-strain biological membrane on the premise of not influencing the performance of the material. However, clinical use has found that the root canal sealer containing DMADDM has yet to be improved in therapeutic activity in dentinal tubules.

Disclosure of Invention

Based on the problems of the DMADDM-modified resin-based root canal sealer in the treatment of dentinal tubules, the inventors have conducted further studies and finally found that, since the root canal system itself is very complicated, structures such as apical bifurcation, secondary root canal, dentinal tubules and the like exist in addition to the primary root canal, the sealer to which DMADDM is added has antibacterial properties but cannot penetrate deep into the dentinal tubules.

The problem to be solved by the invention is how to promote the DMADDM modified resin root canal sealer to enter a finer structure in the root canal.

Magnetic Nanoparticles (MNPs), namely ferroferric oxide nanoparticles, can move directionally under the action of a Magnetic field due to small particle size and good Magnetic responsiveness. At present, MNPs have been added to dental adhesives in dental materials and have been shown to enhance the adhesion of composite resin materials to dentin. However, it has not been reported yet whether or not it can improve the penetration of the DMADDM-modified resin-based root canal sealer into dentinal tubules.

According to the invention, the coordination relationship between MNP and DMADDM modified resin root canal sealant is investigated by taking a plurality of performances of modified composite material, such as cytotoxicity, solubility, dentin tubule permeability, antibacterial property and the like as indexes, and finally, the MNP modified DMADDM modified resin root canal sealant is found to be capable of improving the dentin tubule permeability and the antibacterial property of the root canal sealant while maintaining the material performance of the sealant.

Specifically, the invention provides a dental material which comprises a resin root canal sealer, DMADDM and magnetic nano-particles MNP.

According to experimental results, the mass fraction of the magnetic nanoparticles MNP can be 1-2%. At the dosage, the biological safety, permeability and antibacterial property are good.

In the present invention, the mass fraction of MNP is calculated as the mass ratio of MNP to EndoREZ.

The mass fraction of DMADDM may be specified according to clinical practice reports, and may be, for example, 2.5%. In the present invention, the mass fraction of DMADDM is calculated as the mass ratio of DMADDM/EndoREZ.

In the present invention, the resin root canal sealer is selected from methacrylate resin type root canal sealers.

The selection of the resin material is based on that the resin can be crosslinked with DMADDM and the like, so that the problem of biological safety caused by separation of the raw materials from the resin in the long-term use process is avoided. EndorEZ is a methacrylate resin type root canal sealing agent commonly used in clinic, has the characteristics of chemical and light dual curing, X-ray radiation resistance, hydrophilicity and the like, and can enter a lateral branch root canal or a dentinal tubule to a certain degree after a stain layer is removed to form resin to increase the retention and sealing functions.

In the dental material of the invention, besides the three materials, other auxiliary agents commonly used in dental materials can be added; alternatively, the dental material of the present invention may be used in combination with other known therapeutic materials or in combination therapy.

The invention also provides a preparation method of the dental material, which is prepared by mixing the raw materials. In the mixing process, the conditions such as temperature and the like can be referred to the specification of the raw materials such as resin and the like. In the present invention, the mixing may be performed at room temperature.

The invention also provides application of the dental material in preparing antibacterial products.

The invention also provides the application of the dental material in preparing products for resisting bacterial biofilms.

Further, the bacteria include one or more of enterococcus faecalis, streptococcus gordonii, actinomyces naeslundii, or lactobacillus acidophilus.

The invention also provides the application of the dental material in preparing a product for treating decayed teeth.

The invention has the beneficial effects that:

on the basis of the research of the antibacterial resin root canal filling sealing agent, the invention researches the optimum dosage relation of MNP and EndorEZ by adding and screening magnetic nano particles and optimizing the cytotoxicity, the solubility and the permeability of the modified root canal sealing agent so as to prepare the novel resin root canal sealing agent material with effective antibacterial performance and rooting performance, thereby improving the performance of the original root canal sealing agent, inhibiting the growth of residual bacteria in the root canal, plugging the root canal and improving the success rate of root canal treatment.
