Background

Functional Dyspepsia (FD) belongs to digestive system diseases and can be classified into subgroups of dyskinetic dyspepsia, ulcerative dyspepsia, and nonspecific dyspepsia. The main symptoms are various digestive tract symptoms such as nausea, distention after eating for a long time, persistent abdominal distension, belching, epigastric pain, abnormal defecation and the like, but the auxiliary examination of the internal medicine has no obvious organic lesions. In FD diseases, the secretion of digestive enzymes, the absorption of food, the functions of a secretion system, a digestive tract power system, immunity, psychology and the like in an organism are abnormal, the pathological mechanism is complex, symptoms can occur repeatedly, the life quality of a patient is seriously reduced, and the mood of the patient is influenced.

At present, no specific therapy aiming at functional dyspepsia is available, experience treatment is mainly used, and different treatment schemes such as gastric acid secretion inhibition, gastrointestinal motility promoting drugs, helicobacter pylori eradication, antidepressant drugs and other gastric mucosa protective agents are performed according to the condition of a patient.

Pregnant people have a series of reactions such as dizziness, hypodynamia, inappetence, preference for acid food, aversion to greasy food, nausea, morning vomiting and the like due to the fact that chorionic gonadotropin (HCG) is increased, gastric acid secretion is reduced and gastric emptying time is prolonged, and the reactions are collectively called pregnancy reactions. Symptoms are similar to functional dyspepsia symptoms, but the market for products that alleviate pregnancy reactions is rare.

Chinese patent application CN105212205A discloses a composition for relieving vomiting of pregnant women and a preparation method thereof, wherein the preparation method of the composition comprises the following steps: (1) decocting: 5-9 parts of decocted honey-fried licorice root, 8-12 parts of dried orange peel and 4-6 parts of shaddock peel for 10-20 minutes, and 15-25 parts of concentrated solution is extracted after cooling for later use; (2) juicing: respectively juicing 20-50 parts of ginger, 30-50 parts of carrot, 1-5 parts of dragon fruit and 10-12 parts of dark plum, mixing with 10-15 parts of perilla leaf extract and 25-30 parts of fructus amomi extract, and filtering through fine cotton cloth for later use; (3) and (5) preparing a product.

Chinese patent application CN104815275A discloses an oral medicine for treating pregnancy reaction and a preparation method thereof, wherein the medicine comprises the following components in parts by mass: radix pseudostellariae, polygala tenuifolia, fructus amomi, spina date seed, semen cuscutae, radix ophiopogonis, fried eucommia ulmoides, dark plum pulp, cornus officinalis and ginger bamboo shavings 1-3 parts respectively. The preparation method of the medicine comprises the following steps: A. selecting high-quality clean first-grade radix pseudostellariae, polygala tenuifolia, fructus amomi, spina date seed, semen cuscutae, radix ophiopogonis, fried eucommia ulmoides, dark plum pulp, cornus officinalis and ginger bamboo shavings according to the mass part ratio, and respectively removing residual impurities and dust for later use; B. soaking the raw materials in cold boiled water for about half an hour until the materials penetrate into the tissues of the medicines, then placing the soaked medicines into an earthen pot, adding cold boiled water which is 45mm above the medicines, boiling with strong fire, decocting with slow fire for 35 minutes, pouring out the liquid medicine, adding warm boiled water again, decocting for 30 minutes, mixing twice, and filtering to obtain 600 ml of the liquid medicine.

The invention verifies the digestion-aiding effect of the product from two aspects of the gastric emptying experiment and the digestive enzyme activity of the rat.

Gastric emptying experiment

Experimental animals: SPF grade SD rats, female, 180-. During the experiment, the rats are raised in a quiet environment and are raised in cages, and are fed with common feed (which accords with GB 14924.3-2001 experimental animal mouse and rat compound feed) and freely eat drinking water. The drinking water and the feed are changed every day, so that the ventilation, cleanness and sanitation of the living environment of the animals are kept. Day (12 +/-2) hours, indoor temperature (20 +/-2) DEG C and relative humidity of 40-60 percent. Rats were housed for one week at baseline and were randomized into groups.

Modeling functional dyspepsia: the rat was held at the distal end 1/3 of the tail of the rat with a sponge of long sea, and the rat was subjected to violent and aggressive tearing with other rats to irritate the whole cage rat for 0.5 h/time. With the aggravation of fighting, rats can be scratched, in order to avoid inflammation interference, the injured parts are smeared with 0.5% iodophor to prevent infection, stimulation is carried out for 1 time and 4 times per day every 3 hours, the food intake is reduced after continuous stimulation is carried out for 7 days and 7 days, the gastric emptying rate is reduced, and the success of model building can be judged.

And (3) determination of digestive enzyme activity: the digestive enzyme activity of each experimental rat is tested after the model is made and the experimental period is over, and the test method refers to the experimental method for the digestive enzyme activity determination in the technical Specification for health food inspection and evaluation (2003 edition).

Calculating the gastric emptying rate: the rats are forbidden to eat water for 24h, then the rats are filled with 1g/mL of semisolid nutrient paste at a ratio of 10mL/kg, after standing for 1h, the rats are killed by cutting off the head, the abdominal cavity is cut open, the cardia and the pylorus are tied by using threads, the esophagus and the duodenum are cut off by clinging to the tying threads, and the stomachs are taken out. The stomach mass is weighed and recorded by using the balance; then cutting the stomach wall, washing the stomach contents with distilled water, sucking the water with filter paper, weighing the mass of the empty stomach, and finally calculating the gastric emptying rate: the gastric emptying rate is [ gavage amount- (stomach mass-stomach net mass) ]/gavage amount x 100%.

The formula of the semi-solid nutrient paste comprises: 10g of sodium carboxymethylcellulose (CMC-Na) is dissolved in 200mL of distilled water, 16g of milk powder, 8g of sugar, 8g of starch, 4mL of carbon ink and distilled water are respectively added, the mixture is uniformly stirred once, 300mL of black semisolid paste is prepared, the mixture is refrigerated in a refrigerator, and the black semisolid paste is taken out 2h before use and is returned to the room temperature.

The administration time is as follows: and (3) administration is started the next day after the molding is finished, the intragastric administration is carried out once every day at a fixed time, the intragastric distilled water with the same volume is infused in an experimental group without administration, and the administration period is 10 days.

Grouping and administration dose:

the normal control group is not subjected to molding treatment, and is fed with normal diet without medicine;

after model building, eating normally without dosing;

example 3, example 4, comparative examples 1-3, the dosage was calculated based on crude drug amount, and the dosage was 500mg/kg based on crude drug amount.BW;

The number of rats in each experimental group was 5.

The results are shown in Table 1.

Table 1 data of experimental results of each group

Compared with a normal control group: p < 0.05, P < 0.01

The digestive powder can well recover the pepsin activity and the gastric emptying capacity of rats with functional dyspepsia, and can be used for well helping digestion in improving functional dyspepsia.

Finally, it should be noted that the above-mentioned contents are only used for illustrating the technical solutions of the present invention, and not for limiting the protection scope of the present invention, and that the simple modifications or equivalent substitutions of the technical solutions of the present invention by those of ordinary skill in the art can be made without departing from the spirit and scope of the technical solutions of the present invention.
