The present invention relates to a syringe filled with a hydrogen peroxide solution in advance using an oily composition containing silicone oil.

[ background of the invention ]

Hydrogen peroxide solutions are used industrially as bleaching agents and in the food sector as fungicides. A hydrogen peroxide solution (also referred to as an oxidized alcohol in Japanese pharmacopoeia) containing 2.5 to 3.5 w/v% of hydrogen peroxide is medically used as a disinfectant.

The hydrogen peroxide solution may be mixed with hyaluronic acid (or a salt thereof (e.g., sodium hyaluronate)) at a predetermined ratio immediately before irradiation, and the mixture is applied to a tumor to be used as a radiation sensitizer (patent document 1). The liquid temperature of hydrogen peroxide is increased by the decomposition heat, and the decomposition is accelerated. Non-patent document 1 discloses a material suitable for hydrogen peroxide.

Disclosure of Invention

Problems to be solved by the invention

As disclosed in non-patent document 1, materials of a storage container usable for storing a hydrogen peroxide solution are limited, and the materials of the storage container cannot be freely selected.

As described in patent document 1, when a hydrogen peroxide solution is used as a radiation sensitizer, the hydrogen peroxide is rapidly decomposed when taken out from a light-shielded special container, and therefore, it is necessary to take out an appropriate amount or mass before injection and mix the hydrogen peroxide solution with a sodium hyaluronate solution, and a hospital pharmacy needs to take out the hydrogen peroxide solution and mix the hydrogen peroxide solution with the sodium hyaluronate solution, or a doctor needs to perform an operation at the bedside of a patient, which causes an excessive burden on medical staff who treat the patient. In the former case, the pharmacy staff is heavily burdened and there is a risk that the speed of transfer of the injection mixture from the pharmacy to the patient's bedside is delayed. In the latter case, the burden is borne by the medical staff at the patient's bedside. In either case, the complex process of weighing and mixing solutions increases the risk of errors that can compromise medical quality and endanger the patient.

In addition, if the hydrogen peroxide solution is prefilled using a conventional syringe of glass (the material is borosilicate glass or the like), problems such as swelling during storage and the gasket being pushed back occur, so that long-term storage is not possible.

Means for solving the problems

The present inventors have conducted extensive studies and found that an oily composition containing a silicone oil has an effect of stabilizing the storage of hydrogen peroxide, thereby completing the present invention. The present invention relates to an oily composition for stable storage of hydrogen peroxide, and a syringe prefilled with a hydrogen peroxide solution using the oily composition.

The present invention has an object to provide a syringe in which a hydrogen peroxide solution is prefilled, in which at least a syringe barrel is made of a material having a high decomposition capability of hydrogen peroxide, and an inner wall of the syringe barrel is coated with an oily composition containing silicone oil.

By using the oil composition, hydrogen peroxide can be stably stored in a hydrogen peroxide solution, and by applying the oil composition to the inner wall of a syringe, even a syringe filled with a hydrogen peroxide solution in advance can be stored for a long period of time.

Another object of the present invention is to provide an oily composition containing a silicone oil for the purpose of stably storing hydrogen peroxide, and use of the oily composition.

By using the oil composition, hydrogen peroxide can be stably stored in a hydrogen peroxide solution regardless of the material of a container for storing the hydrogen peroxide solution, and the hydrogen peroxide solution can be stored for a long period of time.

ADVANTAGEOUS EFFECTS OF INVENTION

According to the present invention, a prefilled syringe capable of storing a hydrogen peroxide solution for a long period of time until use as a radiosensitizer can be provided.

For convenience, certain terms used in the present application have been grouped together. Unless defined otherwise, all technical and scientific terms used herein have the same meaning as commonly understood by one of ordinary skill in the art to which this invention belongs. The singular forms "a", "an" and "the" include plural referents unless the context clearly dictates otherwise.

The numerical ranges and parameters set forth in this disclosure are approximations, but the numerical values set forth in the specific examples are reported as precisely as possible. Any numerical value, however, inherently contains certain errors necessarily resulting from the standard deviation found in each testing measurement. Additionally, the term "about" as used herein generally refers to within 10%, 5%, 1%, or 0.5% of the stated value or range. Alternatively, the term "about" means within an acceptable standard error as considered by one of ordinary skill in the art.

The oily composition according to the present embodiment contains a silicone oil, and is an oily composition for the purpose of stabilizing the storage of hydrogen peroxide. The oily composition can be used as a medicament for stably storing hydrogen peroxide. The above oily composition may contain a pharmaceutically acceptable ingredient (sterile water or the like) other than silicone oil. The above oil composition can be applied to the inner wall of a container or a syringe. The above oil composition can inhibit the decomposition of hydrogen peroxide by a material having a high decomposition ability of hydrogen peroxide (for example, glass, a plastic having a high decomposition ability of hydrogen peroxide (for example, polyethylene terephthalate), and stainless steel). Therefore, the material of the container or the syringe may be the above-mentioned material having a high decomposition capability of hydrogen oxide.

As the silicone oil, a silicone oil having a kinematic viscosity at 25 ℃ of 20 to 40,000cSt, preferably 500 to 30,000cSt, more preferably 800 to 20,000cSt, and still more preferably 1,000 to 15,000cSt, can be used.

The kinematic viscosity of the silicone oil at 25 ℃ can be measured according to JIS Z8803.

The silicone oil preferably uses a linear silicone represented by the following formula (1).

In the formula (1), R independently represents an organic functional group such as an alkenyl group and/or an SiH group which does not contain a functional group participating in a hydrosilylation addition reaction, a monovalent hydrocarbon group or a hydroxyl group, and each R may be the same or different. x represents an integer of 10 to 1200. R preferably represents an alkyl group or an aryl group, and more preferably represents a methyl group, an ethyl group or a phenyl group.

The linear silicone represented by the above formula (1) may be a polydiorganosiloxane terminated at both ends with a triorganosilyl group, preferably a polydialkylsiloxane, a polydiarylsiloxane, a polyalkylarylsiloxane or a copolymer of these, more preferably a polydimethylsiloxane or a polymethylphenylsiloxane, and particularly preferably a polydimethylsiloxane in which all R groups are all methyl groups.

Syringe with a needle

Fig. 1 is a schematic view showing a pre-filled syringe 1 filled with a hydrogen peroxide solution 50 according to the present embodiment. In the present embodiment, the syringe 10 (particularly, the syringe barrel 20) is generally cylindrical in shape, one end of the syringe 10 is provided with the needle mounting portion 30 for discharging the hydrogen peroxide solution 50, the other end of the syringe 10 is provided with the rod insertion portion 80 for inserting the plunger rod 70, and the periphery of the rod insertion portion 80 is provided with the flange 90. The pre-filled syringe 1 shown in fig. 1 was coated with silicone oil on the inner wall of the syringe 10. In order to seal the filled hydrogen peroxide solution 50, the prefilled syringe 1 shown in fig. 1 includes a cap 40 in the needle mounting portion 30, and a plunger rod 70 having a spacer 60 is inserted from a rod insertion portion 80.

In the present embodiment, the syringe for a hydrogen peroxide solution means a syringe having a low capability of decomposing hydrogen peroxide in a hydrogen peroxide solution. In the present embodiment, the hydrogen peroxide solution means a solution obtained by dissolving hydrogen peroxide in a solvent (e.g., water), and may contain additives (e.g., phosphoric acid and phenacetin) as necessary. In the present embodiment, the syringe may be made of a single material, or may be made of a plurality of materials (including a multilayer structure such as a plated film). When the syringe is made of a single material, the entire syringe may be made of a material having high decomposition ability of hydrogen peroxide (e.g., glass, plastic having high decomposition ability of hydrogen peroxide (e.g., polyethylene terephthalate), and stainless steel). When the syringe is made of a plurality of materials, the inner wall of the syringe is made of glass, and the other parts may be made of a material having a high decomposition ability of hydrogen peroxide. In addition, not all the portions that come into contact with the hydrogen peroxide solution are made of a material having a high decomposition capability of hydrogen peroxide, and it is sufficient that the main portion, including the inner surface of the syringe barrel of the syringe main body, is made of a material having a high decomposition capability of hydrogen peroxide. That is, other plunger rods, luer locks, caps, spacers, and the like that may come into contact with the hydrogen peroxide solution need not be made of the above-described material having a high decomposition capability of hydrogen peroxide. The syringe according to the present embodiment can be used for a radiosensitizer.

The ratio of the concentration of hydrogen peroxide in the hydrogen peroxide solution after a specific storage period under specific temperature conditions (residual rate of hydrogen peroxide) to the concentration of hydrogen peroxide in the hydrogen peroxide solution before the start of storage determines the decomposition ability of hydrogen peroxide. The storage is carried out in a closed state. The temperature conditions are not particularly new and may be 35 ℃, 37 ℃, 40 ℃ or 60 ℃. The storage period is not particularly limited, and may be 1 week, 2 weeks, 3 weeks, or 4 weeks, or may be 4 weeks or more. The concentration of hydrogen peroxide in the hydrogen peroxide solution before the start of storage may be measured at any concentration in the range of, for example, 0.01 to 40% (w/v). In an embodiment, the resin may have a lower hydrogen peroxide decomposition ability than glass. The resin may have a residual hydrogen peroxide content of 70% or more, preferably 75% or more, more preferably 78% or more, and still more preferably 80% or more, when the solution containing 2.5 to 3.5 w/v% hydrogen peroxide is stored under a closed condition at 60 ℃ for 4 weeks. The amount of hydrogen peroxide in the hydrogen peroxide solution was determined by titration with a potassium permanganate solution according to the hydrogen peroxide quantitation method of the japanese pharmacopoeia.

This embodiment provides a syringe that is pre-potted with a hydrogen peroxide solution. The syringe prefilled with the hydrogen peroxide solution is provided with a gasket slidably attached to the syringe. The needle mounting portion of the syringe prefilled with the hydrogen peroxide solution is sealed (for example, by a cap or the like).

In the present embodiment, the concentration of hydrogen peroxide in the hydrogen peroxide solution in the prefilled syringe is, for example, 0.01, 0.02, 0.03, 0.04, 0.05, 0.06, 0.07, 0.08, 0.09, 0.1, 0.5, 1, 5, 10, 15, 20, 25, 30, 35, or 40%, and may be in a range between any two of the values exemplified herein, for example, 0.01 to 40% (w/v), preferably 0.05 to 30% (w/v).

The oily composition according to the present embodiment can be applied using a sprayer or the like. The oily composition may be dissolved in an appropriate solvent at the time of coating. When the inner wall of the syringe is coated with the oil composition, the oil composition is applied at a concentration of 0.02 to 0.2mg/cm2The amount of application of (3) is sprayed to the syringeAn inner wall. After the coating of the inner wall of the syringe is completed, the thickness of the oil composition may be made uniform by using a device such as a water-absorbing wiper, or the thickness of the oil composition may be made uniform by softening the silicone oil in the oil composition by heat treatment (e.g., autoclaving treatment).

[ examples ] A method for producing a compound

Stability test of hydrogen peroxide solution was performed using a glass container coated with silicone oil and a glass container not coated with silicone oil. Silicone oils are commercially available from Dow Corning (product name: Dow Corning 360Medical Fluid (12,500 cSt)).

The containers were filled with hydrogen peroxide solution and sealed and stored at 60 ℃ for 4 weeks. The residual rate of hydrogen peroxide in the hydrogen peroxide solution after storage was measured. Hydrogen peroxide solution Kenei (containing 2.5-3.5 w/v% hydrogen peroxide, phosphoric acid and phenacetin) manufactured by Jianrong pharmaceutical company is used. The amount of hydrogen peroxide in the hydrogen peroxide solution was determined by titration with a potassium permanganate solution according to the hydrogen peroxide quantitation method of the japanese pharmacopoeia. FIG. 2 shows the results.

In the glass container not coated with the silicone oil, the residual hydrogen peroxide content was about 70%. The content of the silicone oil-coated glass container was 78% or more. From the above results, it is clear that the glass container coated with the silicone oil can suppress the decomposition of hydrogen peroxide as compared with the glass container not coated with the silicone oil.
