Technical Field

Technical Field

The present invention relates to a composition containing ailanthus altissima (ailanthus altissima) extract as an active ingredient, and more particularly, to a composition for improving acne skin, suppressing sebum secretion or improving seborrheic skin, which contains an ailanthus altissima extract as an active ingredient.

Background

In general, acne is a common skin disease experienced by about 80% of korean citizens. Recently, as adolescence advances, a case has occurred in which a counseling dermatology is performed for acne on 5 or 6 grades of primary schools. Presumably, girls are in the second half to the first half of their teens, while boys are in the second half to the whole twenties. Most of them disappear in several years without any special treatment, but there are many cases where they do not disappear, and therefore, it is necessary to develop a technique for treating acne.

Acne, a disease that causes chronic inflammation of sebaceous glands, occurs mainly in the face, neck, chest where sebum is secreted much. Acne is classified into common acne (acne vulgaris), acne fulminans, premenstrual acne, prepubertal acne, neonatal acne, infantile acne, pediatric acne, occupational acne, and cicatricial acne.

Acne Vulgaris (Acne Vulgaris) is a complex effect of many causes, of which sex hormones are representative. The increase in sex hormones is increased due to various causes including puberty, and thus, sebum (sebum) is excessively secreted, and pores are clogged due to abnormal peeling of keratin.

The excessive secretion of sebum is caused by various reasons, and most importantly, excessive secretion of sebum is caused by activation of sebaceous gland cells by the amount of Dihydrotestosterone (DHT), which is one of hormones involved in the secretion of sebum in the activity of sebaceous glands. That is, in the skin or sebaceous glands, testosterone is converted into dihydrotestosterone by 5 α -reductase (5 α -reductase), cells of the sebaceous glands become active, differentiation is promoted, and sebum (s ebum) within the sebaceous glands is excessively secreted, causing acne (j. invest der skin 105:209-214 diabetes etc.).

Acne and the like, which are skin problems caused by the simple exuberance of sebum secretion, are further worsened by the subtle inflammatory reaction of the skin. When the production process of acne is observed, excessive sebum is accumulated in hair follicles, so that acne bacteria become active and inflammation is caused.

In addition, excessive secretion of sebum causes greasy (greasy) and bloody crust (scales) on the skin, and seborrheic dermatitis (seborrhoeic dermatitis) called seborrhoeic eczema (seborrhoeic eczema), as a disease of sebaceous glands or a skin problem.

As a method for treating acne, there are a method of applying an external preparation, a pimple removal technique, a chemical peeling, etc. together with the administration of an oral antibiotic and vitamin a, and antibiotics such as tetracycline (tetracycline), clindamycin (clindamycin), erythromycin (erythromycin) are used for the treatment of acne. However, long-term use of the antibody causes side effects or produces antibodies, which may reduce the therapeutic effect.

Recently, in order to solve the above problems, some cosmetic compositions for improving acne skin have been proposed which contain natural sesame oil such as tea tree oil or rosemary oil as an active ingredient. However, none of them can achieve satisfactory acne improvement effect.

On the one hand, Ailanthus altissima is bark of Ailanthus altissima and is known as a medicinal material with the effects of removing heat, drying dampness, astringing intestines, reducing urination, killing ascaris, excreting dampness and stopping leukorrhagia, and securing intestines and stopping diarrhea.

Disclosure of Invention

Technical problem

The present invention aims to provide a composition containing a ailanthus altissima extract as an active ingredient, which has an effect of improving acne skin, suppressing sebum secretion, or improving seborrheic skin.

Technical scheme

In order to achieve the above object, according to one aspect, the present invention provides a composition for improving acne skin, comprising ailanthus altissima extract as an active ingredient.

According to another aspect, the present invention provides a composition for inhibiting sebum secretion, which contains ailanthus altissima extract as an active ingredient.

According to still another aspect, the present invention provides a composition for improving seborrheic skin comprising a ailanthus altissima extract as an active ingredient.

Effects of the invention

The acne-improving composition according to the present invention contains ailanthus altissima extract as an active ingredient, and thus exhibits an excellent sebum secretion-inhibiting effect, whereby an excellent acne skin-improving or seborrheic skin-improving effect can be exhibited.

The present invention will be described in detail below.

According to one aspect, the present invention may relate to a composition for improving acne skin containing ailanthus altissima extract as an active ingredient.

According to another aspect, the present invention may relate to a sebum secretion suppressing composition containing ailanthus altissima extract as an active ingredient.

According to still another aspect, the present invention may relate to a composition for improving seborrheic skin comprising a ailanthus altissima extract as an active ingredient.

The Ailanthus altissima bark is not particularly limited to Ailanthus altissima bark, and any Ailanthus altissima bark may be used as long as it is a commonly available Ailanthus altissima bark.

According to an embodiment of the present invention, the ailanthus altissima extract may be an extract extracted with an extraction solvent selected from water, a lower alcohol having 1 to 4 carbon atoms, or a mixed solvent thereof, and preferably, the extraction solvent may be absolute ethanol of 99.5% (v/v) or more.

In addition, according to an embodiment of the present invention, the ailanthus altissima extract may be included in an amount of 0.001 to 10 wt% based on the entire weight of the composition.

When the content of the ailanthus altissima extract is less than 0.001 wt% based on the total weight of the composition, the sebum secretion inhibiting effect is weak, and when it exceeds 10 wt%, problems in skin stability or formulation may occur.

According to one embodiment of the present invention, the composition can inhibit 5 α -reductase activity.

According to an embodiment of the invention, the composition may be a pharmaceutical composition.

The pharmaceutical composition may contain, in addition to the cortex ailanthi radicis extract, a preservative, a stabilizer, a wettable powder or an emulsion accelerator, a pharmaceutical adjuvant such as a salt and/or a buffer for adjusting osmotic pressure, and other therapeutically useful substances, and may be formulated into various oral administration preparations or non-oral administration preparations according to a conventional method.

Examples of the agent for oral administration include tablets, pills, hard and soft capsules, liquids, suspensions, emulsifiers, syrups, powders, fine granules, pellets and the like, and these dosage forms may contain, in addition to the active ingredient, a surfactant, a diluent (e.g., glucose, sucrose, mannitol, sorbitol, cellulose and glycine), a lubricant (e.g., silica, talc, stearic acid and magnesium or calcium salts thereof and polyethylene glycol). The tablet may further contain a binder such as magnesium aluminum silicate, starch paste, gelatin, tragacanth, methylcellulose, sodium carboxymethylcellulose, and polyvinylpyrrolidone, and may optionally contain pharmaceutical additives such as a disintegrant such as starch, agar, alginic acid or a sodium salt thereof, an absorbent, a coloring agent, a flavoring agent, and a sweetening agent. The tablets may be prepared by conventional mixing, granulating or coating methods.

The non-oral administration form may be a form of transdermal administration, and examples thereof include injections, drops, ointments, emulsions, gels, creams, sprays, suspensions, oils, suppositories, patches, and the like, but are not limited thereto.

The pharmaceutical compositions may be administered parenterally, rectally, topically, transdermally, subcutaneously, and the like. The amount of the effective ingredient to be administered is within the level of those skilled in the art, and the daily dose of the drug varies depending on various causes such as the degree of progression of the disease, the time of onset, the age, the health condition, complications and the like of the subject to be administered, however, on an adult basis, the composition is usually administered in an amount of 1. mu.g/kg to 200mg/kg, preferably 50. mu.g/kg to 50mg/kg, divided once to three times a day, and the amount to be administered does not limit the scope of the present invention in any way.

According to an embodiment of the present invention, the composition may be a cosmetic composition.

The cosmetic composition may contain a cosmetically or dermatologically acceptable medium or base. It can be provided in the form of, for example, a solution, a gel, a solid, an anhydrous paste, an emulsion obtained by dispersing an oil phase in an aqueous phase, a suspension, a microemulsion, a microcapsule, a fine particle sphere or an ionic (liposome) form, and a nonionic crop dispersing agent, or in the form of a cream, a lotion, an emulsion, a powder, an ointment, a spray, or a concealer stick, as all formulations suitable for topical application. The composition can be prepared by a method generally used in the art. The cosmetic composition according to an embodiment of the present invention may also be used in the form of an aerosol composition further containing a propellant compressed in the form of a foam (foam).

The cosmetic composition is not particularly limited in its formulation, and may be appropriately selected according to the purpose. For example, the skin lotion can be prepared into dosage forms such as a skin lotion (skin deposition), a skin softener (skin softener), a skin toner (skin toner), a milk lotion (deposition), a milk lotion (milk deposition), a moisturizing lotion, a nourishing lotion, a massage cream, a nourishing cream, a moisturizing cream, a hand cream, a foundation, a essence, a nourishing essence, a mask, a soap, a cleansing foam, a cleansing cream, a makeup remover, a powder, a body lotion, a body cream, a body oil, a body wash, a body lotion essence, and the like.

When the formulation of the present invention is a paste, cream or gel, animal fibers, vegetable fibers, waxes, paraffin, starch, tragacanth, cellulose derivatives, polyethylene glycol, silicone, bentonite, silica, talc, zinc oxide, or the like can be used as a carrier component.

When the dosage form of the present invention is a powder or a spray, lactose, talc, silica, aluminum hydroxide, calcium silicate or polyamide powder can be used as a carrier component, and particularly, when the dosage form is a spray, a propellant such as chlorofluorocarbon, propane/butane or dimethyl ether may be further contained.

In the case of the formulation of the invention as a solution or emulsion, as a carrier component, a solvent, solubilizer or demulsifier is used, such as water, ethanol, isopropanol, ethyl carbonate, ethyl acetate, benzyl alcohol, benzyl benzoate, propylene glycol, 1, 3-butyl glycol oil, glycerol fatty acid ester, polyethylene glycol or fatty acid ester of sorbitan.

In the case where the dosage form of the present invention is a suspension, as a carrier ingredient, a liquid diluent such as water, ethanol or propylene glycol; suspending agents such as ethoxylated isostearyl alcohols, polyoxyethylene sorbitol esters and polyoxyethylene sorbitan esters; microcrystalline cellulose, aluminum metahydroxide, bentonite, agar-agar or tragacanth, and the like.

When the formulation of the present invention is a surfactant-containing detergent, fatty alcohol sulfate, fatty alcohol ether sulfate, sulfosuccinic acid monoester, isethionate, imidazoline derivative, methyltaurate, sarcosinate, fatty acid amide ether sulfate, alkylamide betaine, fatty alcohol, fatty acid glyceride, fatty acid diethanolamide, vegetable oil, linolenic acid derivative, ethoxylated glycerol fatty acid ester, or the like can be used as the carrier component.

The cosmetic composition may include functional additives and ingredients included in general cosmetic compositions, in addition to the ailanthus altissima extract. The functional additive may include a component selected from the group consisting of water-soluble vitamins, oil-soluble vitamins, polymer peptides, polymer polysaccharides, sphingolipids, and seaweed juice.

In addition, the composition may contain, together with the functional additive, components contained in a general cosmetic composition as needed, and examples of the components contained in addition to the above include an oil and fat component, a moisturizer, an emollient (emollients), a surfactant, an organic and inorganic pigment, an organic powder, an ultraviolet absorber, a preservative, a bactericide, an antioxidant, a plant extract, a pH adjuster, ethanol, a pigment, a perfume, a blood circulation promoter, a cold feeling agent, an antiperspirant, purified water, and the like.

According to an embodiment of the invention, the composition may be a food composition.

The food composition may be in the form of a liquid or solid, and examples thereof include various foods, drinks, gums, teas, vitamin complexes, health foods, and the like, and may be in the form of powder, granules, tablets, capsules, or drinks. In addition to the ailanthus altissima extract, the food composition of each dosage form can be appropriately selected and matched by those skilled in the art without difficulty according to the dosage form or the purpose of use, and can obtain a synergistic effect when applied together with other raw materials.

The liquid component that may be contained in addition to the ailanthus altissima extract is not particularly limited, and as an additional component, there may be mentioned, for example, a common drink including various kinds of flavors or natural carbohydrates, etc. Examples of the natural carbohydrates include monosaccharides; disaccharides such as glucose and fructose; polysaccharides such as maltose and sucrose; common sugars such as dextrin and cyclodextrin; and sugar alcohols such as xylitol, sorbitol, erythritol and the like. As the flavoring agent, natural flavoring agents (thaumatin (tau marti n), stevia extracts (e.g., rebaudioside a, glycyrrhizin, etc.), and synthetic flavoring agents (e.g., saccharin, aspartame, etc.) can be advantageously used. The natural carbohydrate ratio may be from about 1 to 20 grams, in one aspect from about 5 to 12 grams, per 100 milliliters of the composition disclosed herein.

In one aspect, the food composition may comprise various nutrients, vitamins, minerals (electrolytes), flavors such as synthetic flavors and natural flavors, colors and thickeners (cheese, chocolate, etc.), pectic acids and salts thereof, alginic acid and salts thereof, organic acids, protective colloid thickeners, pH adjusters, stabilizers, preservatives, glycerin, alcohols, carbonating agents used in carbonated beverages, and the like. In another aspect, pulp for use in producing natural fruit juices as well as vegetable drinks can be included. The components may be used independently or in combination. The proportions of the additives may vary, but are generally selected in the range of about 0.001 to 20 parts by weight per 100 parts by weight of the composition disclosed in this specification.

The present invention will be described in more detail below with reference to examples and test examples. However, the examples and test examples are disclosed for understanding the contents of the present invention, and the scope of the present invention is not limited to the examples and test examples, and modifications, substitutions, and insertions, which are generally known in the art, may be made, and are also included in the scope of the present invention.

[ example 1 ] preparation of Ailanthus altissima extract

1000ml of absolute ethanol was added to 100g of bark of Ailanthus altissima purchased from Korean Plant extraction Bank (Plant Extract Bank, Korean national field), and the mixture was heated at 80 ℃ for 2 hours using an extractor (HB 4basic, IKA) equipped with a reflux condenser to Extract. After filtering the extract with filter paper (Whatman No.2, Whatman co.), the remaining residue (residue) was extracted again 1 time by the same method as above, and the obtained extract was concentrated under reduced pressure at 40 ℃ using a reduced pressure concentrator (rotavap, Buchi co.), followed by freeze-drying to obtain a dried extract of ailanthus altissima bark.

[ test example 1 ] sebum secretion-inhibiting Effect

In order to evaluate the effect of inhibiting sebum secretion of the ailanthus altissima extract of the present invention, sebaceous gland cells and keratinocytes were used together, and an in vitro (in vitro) model of an environment similar to that of human sebaceous glands was used. Specifically, 24-well plates (wells) were seeded with 3.75 × 10 cells4Human HaCaT keratinocytes of (ATCC, VA, USA) and 6.0x104Human sebaceous gland cells (Cellprog en, CA, USA). After inoculation, the cells were allowed to stick to the bottom of the plate for 24 hours. At this time, as the medium, a DMEM medium (Invitrogen, Garlsbad, Calif.) supplemented with 10% fetal bovine serum (total bovine serum; JRH Bioscience, Tokyo (Tokyo), Japan) and 100mg/ml penicillin (penicillin) and 100mg/ml streptomycin (Streptomyces in) (all of the reagents are GIBCO, Milan (Milan), Italy) was used. In cells over one day, 50 μ M of linoleic acid (linoleic acid), 50 μ M of arachidonic acid (arachidonic acid), 10nM of dihydrotestosterone (dihydrotestosterone) were treated as a stimulus for causing seborrheic dermatitis. Meanwhile, in the experimental group (stimulus source + ailanthus altissima extract), the ailanthus altissima extract of example 1 was dissolved in dmso (dimethyl sulfoxide) to prepare a 1% solution, diluted into a DMEM medium to a final concentration of 50ppm, and after one day, the amount of neutral fat generated in the cells was measured using an oil Red o (oil Red o) staining method。

The oil Red O (oil Red O) staining method specifically comprises fixing the cells washed with PBS (phosphate buffered saline) with a 3.7% formaldehyde solution for 30 minutes, washing the fixed cells with PBS 3 times, washing with 70% (v/v) ethanol 1 time, and staining with 0.4% (v/v) oil Red O (oil Red O) solution for 30 minutes. The stained cells were washed 1 time with 70% (v/v) ethanol and 3 times with PBS, and then the oil Red o (oil Red o) solution was dissolved again with isopropanol, and absorbance was measured at 520nm using a spectrophotometer (spectrophotometer) and quantified. The results are shown in fig. 1.

From the results of fig. 1, it was confirmed that the amount of sebum increased more than 4 times (about 470%) compared to the control group when the stimulus was treated, and the amount of sebum decreased nearly 2 times (about 236%) when the ailanthus altissima extract was treated thereon.

That is, it was confirmed that the ailanthus altissima extract according to the present invention shows an excellent sebum secretion suppressing effect, and thus can show an excellent acne skin improving or seborrheic skin improving effect.
