5. The F1 hybrid or a part thereof according to any one of claims 1 to 4, wherein the ratio (%) of the height of the flower at the lowermost part to the plant height is increased as compared to Nicotiana tabacum.

6. The F1 hybrid or a portion thereof according to any one of claims 1-5, having fewer flowers per 1 strain than Nicotiana tabacum.

7. The F1 hybrid or a portion thereof according to any one of claims 1 to 6, wherein the leaf length, leaf width, or both the leaf length and leaf width of the largest leaf is greater than that of Nicotiana tabacum.

8. The F1 hybrid or a portion thereof according to any one of claims 1-7, having reduced branching compared to Nicotiana tabacum.

9. The F1 hybrid or a portion thereof according to any one of claims 1-8, having an increased aerial yield as compared to Nicotiana tabacum.

10. The F1 hybrid or a portion thereof according to any one of claims 1-9, having an increased yield of dried leaves compared to nicotiana tabacum.

11. The F1 hybrid or a portion thereof according to any one of claims 1-10, having improved insect tolerance to foliar-feeding pests as compared to Chuanzhong tobacco.

12. The F1 hybrid or a part thereof according to any one of claims 1 to 11, which has improved insect resistance against leaf-eating pests as compared to both Sichuan tobacco and shade tobacco.

13. The F1 hybrid or a portion thereof according to any one of claims 1-12, having improved insect resistance to tobacco storage pests compared to Chuanzhong tobacco.

14. The F1 hybrid or a portion thereof according to any one of claims 1-13, having improved insect resistance against tobacco storage pests as compared to both Chuanzhong tobacco and shade tobacco.

15. The F1 hybrid or a portion thereof according to claim 11 or 12, wherein the leaf-eating pest is selected from the group consisting of oriental tobacco budworm (helicobacto budworm, Helicoverpa assulata), Heliothis virescens (helicobacto budworm, helicothios virescens), cotton bollworm (cotton bollworm, Helicoverpa armigera), tobacco hornworm (Helicoverpa hornworm, Manducasexa), Spodoptera litura (Spodoptera litura), and Spodoptera exigua (Spodoptera exigua).

16. A method of preparing an F1 hybrid, the method comprising:

mating the shade tobacco with Chuanshang tobacco.

17. The method of claim 16 wherein the seed parent is nicotiana tabacum and the pollen parent is nicotiana tabacum.

18. The method of claim 16 wherein the pollen parent is nicotiana tabacum and the seed parent is nicotiana tabacum.

19. The method of any of claims 16-18, further comprising:

and (3) mating the shade tobacco and Chuanwu tobacco, and then selecting an F1 hybrid in which the ratio (%) of the height of the lowermost flower to the plant height is increased as compared with the shade tobacco.

20. The method of any of claims 16-18, further comprising:

mating the shade tobacco and Chuanwu tobacco, and then selecting F1 hybrid seeds having less flower number per 1 plant than the shade tobacco.

21. The method of any of claims 16-18, further comprising:

a step of mating the shade tobacco and Chuanshang tobacco, and then selecting an F1 hybrid having a larger leaf length, leaf width, or both of the leaf length and leaf width than the shade tobacco.

22. The method of any of claims 16-18, further comprising:

and (3) mating the shade tobacco and Chuanwu tobacco, and then selecting an F1 hybrid having a reduced branching compared with the shade tobacco.

23. The method of any of claims 16-18, further comprising:

and (3) mating the shade tobacco and Chuanshang tobacco, and then selecting an F1 hybrid having an increased aerial yield as compared with the shade tobacco.

24. The method of any of claims 16-18, further comprising:

Technical Field

The present invention relates to a hybrid of Nicotiana F1 and use thereof.

Background

1. Plants of the genus nicotiana

Plants of the genus Nicotiana (also known as "tobaco genus") are widely distributed in the south/north continental americas, the australian continental and the african continental, and more than 70 species are now reported. The most notable are 2 cultivars of tobacco (Nicotiana tabacum) and yellow tobacco (Nicotiana rustica) that have been used as raw materials for tobacco products.

In many cases, genetic analysis and analysis of disease resistance have been reported for wild species of nicotiana other than the 2 species, including plants obtained by crossing the wild species with each other as material producers. For example, Takenaka, Jap.Jour.Genet.31.155-161,1956; MULKH RAJ AHUJA, Genetica 47,865-880, 1962. Although there are reports of using the wild species themselves as a raw material for tobacco products, the number is small. For example, U.S. patent No. 7,798,153. However, there has been no report of using F1 hybrid plants obtained by mating wild species with each other for tobacco products.

2. Shade tobacco (Nicotiana umbellata)

"shade tobacco" (Nicotiana umbellata) is a species of the genus Nicotiana distributed in the northwest of Western Australia. The plant height is about 50-60 cm, the plant is a large number of annual herbaceous plants which branch into thin stems and are luxuriant, glandular hairs grow integrally, and the plant is adhered to emit strong fragrance. The leaf body is in an egg shape with a width of about 8cm, and a large number of flowers are arranged on the branch tip with the leaf and the axillary branch tip in a general inflorescence shape. The primary alkaloid is nicotine. Resistance to powdery mildew and downy mildew (common type) is observed (Nicotiana plant atlas, Nicotiana japonica Nippon Nicotiana Kabushiki Kaisha (editors), pp. 180-183, 3.1994, Chengtang Newcastle). The shade tobacco is considered to be preferable as a raw material for tobacco products because of its characteristic strong flavor. However, currently there is no report of the use of shade tobacco as a raw material for tobacco products.

On the other hand, a large number of literatures report genetic analysis and the like using nicotiana tabacum as a test material. For example, U.S. Subhashini et al have reported mating of nicotiana tabacum with other wild species of nicotiana (U.S. Subushini, Euphytoica, 23,289-293,1974; U.S. Subushini at., Cytologica 39,403-409,1974; U.S. Subushini at., Cytologica 40,409-413,1975).

3. Chuanshang tobacco (Nicotiana kawakamii)

"Chuanshana kawakamii" is a species of Nicotiana found in Gloyura in 1968 by Katong, Chuanshana et al. In the native place, half-woody shrubs with a height of about 4m have viscous stems and purple top ends. The leaves are wide ovals with a length of about 60 cm. The conifer has thick and wide branches, and the spiral branch tip has buds and flowers, and is lined with a deep red-purple capsule. The leaves are dense. The number of flowers is small, and the flowers are concentrated on the upper part of the plant body. The major alkaloid is nornicotine. It is resistant to powdery mildew and PVY (Nicotiana plant graphic, Nippon tobacco industry Co., Ltd. (edited), pp. 180-183, 3.1994, Chengtang Shuangshi, Kao corporation). The use in Tobacco products is reported in US patent No. 7,798,153 to US Smokeless tobaco co, but there is currently no report of the F1 hybrid, obtained by using choma Tobacco as a single parent, for Tobacco products.

Disclosure of Invention

Problems to be solved by the invention

In the case of using the shade tobacco as a raw material of tobacco products, there are the following significant problems: (i) the plant body is small, and the yield is low; (ii) flower buds are formed at each axillary bud/branch, and the topping operation (cutting of flower buds) for filling up leaves in general tobacco cultivation is complicated; and so on. The information obtained from the above documents is not clear as to a method for solving these problems without impairing the characteristics of the shade tobacco.

As one solution, it is assumed that F1 hybrid obtained by mating the shade tobacco with another nicotiana plant is used, but there is a problem that mating seeds cannot be obtained by a combination of mating and do not germinate even if the mating seeds are obtained. In addition, a lot of labor is required to select mating subjects satisfying the conditions for obtaining F1 hybrid seeds improved in yield and complexity of topping operation without impairing the characteristic strong flavor of the shade tobacco.

The present invention addresses the problem of providing a hybrid plant of F1, which has improved yield and improved top-cutting operation complexity, without impairing the characteristic strong aroma of sun tobaccos, and a method for producing the same.

The present inventors have made a detailed study on the characteristics of the obtained F1 hybrid by mating the shade tobacco with a plurality of nicotiana wild species, and have found that the F1 hybrid produced by combining the shade tobacco and the chuanqi tobacco has excellent properties as a raw material for obtaining a tobacco product.

ADVANTAGEOUS EFFECTS OF INVENTION

The F1 hybrid obtained by crossing Yingpo tobacco and Chuanzhong tobacco is an F1 hybrid having improved easiness of flower part removal without impairing the characteristic flavor of Yingpo tobacco and having an increased yield as compared with Yingpo tobacco. The cultivation and management of the F1 hybrid does not require special techniques or expertise, and can be easily carried out by cultivation to harvest in substantially the same manner as in the case of the tobacco of the usual cultivar.

The problems of the prior art can be significantly improved by preparing an F1 hybrid based on wild species of Nicotiana tabacum and Nicotiana chuanensis, and using the F1 hybrid which does not impair the characteristic flavor of Nicotiana tabacum and improves the yield and ease of flower removal. In addition, the F1 hybrid has the effect of greatly suppressing the extent to which the tobacco budworm is bitten by.

Detailed Description

The present invention includes the following embodiments, but is not limited thereto.

Hybrid F1

The present invention relates to F1 hybrid of Nicotiana tabacum and Nicotiana chuanxiong, or a portion thereof.

"shade tobacco" (Nicotiana umbellata) is a species of the genus Nicotiana distributed in the northwest of western australia. The number of chromosomes (2n) was 46. The plant height of the shade tobacco is about 50-60 cm, a large number of branches are flourishing, and the shade tobacco has a large number of small leaves about 5-15 cm. The shoot apex forms a large number of flowers in a general inflorescence shape, but is not limited thereto. The shade tobacco is not limited to this, but is sticky due to a large amount of secretion from the fuzz of the plant as a whole, and emits a strong fragrance as a whole. The primary alkaloid is nicotine.

"Chuanshana kawakamii" is a species found in Gliviya, Katong, Chuanshana et al, 1968. The number of chromosomes (2n) was 24. The average height of Sichuan tobacco is more than 200cm, and the leaves are large. The main alkaloid component is nornicotine, but is not limited thereto.

The "F1 hybrid" is a progeny derived from plants of different species and varieties. The term "F1 hybrid" as used herein and in the appended claims means the F1 hybrid of the first hybrid generation (F1). F1 is the progeny of the first generation resulting from the mating of two parents having a different allele in homozygous form in an organism. F1 has genes of both parents in heterozygous form, and the genotypes are uniform. The number of chromosomes (2n) was 35.

The part of the F1 hybrid in the present invention includes not only a part of the mature body of a plant but also all forms from the seed to the mature body of a plant.

The "part" is selected from the group consisting of, but not limited to, a leaf (including a leaf shaft and a petiole), a stem, a root, a seed, a flower, pollen, an anther, an ovule, a petiole, a meristem, a cotyledon, an hypocotyl, a pericycle, an embryo, an endosperm, an explant, a callus, a tissue culture, a bud, a cell, and a protoplast. Preferably a leaf or stem.

"petiole" refers to the petiole of a plant that joins a leaf to a stem. "petiole" refers to the short stalk of a seed plant that links a single petal to a stem that connects inflorescences. The "pericycle" is a plant tissue and is a layer of cells surrounding xylem and phloem present in the center of a root. The pericycle forms a lateral root by splitting. "explant" refers to a tissue of a plant obtained by culturing a part of an embryo cut from the plant under artificial conditions. "callus" is a plant cell mass which is cultured on a solid medium or the like in an undifferentiated state. The "tissue culture" is a culture of a plant tissue obtained by culturing an arbitrary tissue of a plant under artificial conditions.

In the F1 hybrid, either of the Nicotiana tabacum and Nicotiana kawakamii may be a seed parent or a pollen parent.

In another embodiment, the seed parent is Nicotiana tabacum and the pollen parent is Nicotiana armandii. In another embodiment, the pollen parent is Nicotiana tabacum and the seed parent is Nicotiana armandii. More preferably, the seed parent is Nicotiana tabacum and the pollen parent is Nicotiana tabacum.

Characterization of the F1 hybrid

(1) Ease of flower removal

In order to efficiently harvest the above-ground parts including leaves and stems useful for the manufacture of tobacco products from tobacco plants, flowers need to be removed from the tobacco plants. The F1 hybrid is easier to remove flowers than tobacco shade.

In one embodiment, the ratio (%) of the height of the flower at the lowermost part of the F1 hybrid to the plant height is increased as compared to the shade tobacco. The ratio (%) of the height of the flower at the lowermost part to the plant height is preferably 20% or more, 30% or more, 40% or more, 50% or more, 60% or more, and 65% or more. In one embodiment, the ratio (%) of the height of the lowermost flower to the plant height is 40% or more.

In one embodiment, the F1 hybrid has fewer flowers per 1 strain than nicotiana tabacum. In example 1 described later, the "number of flowers" of the F1 hybrid was reduced to about 1/5 per 1 strain at about 50 days after transplantation, as compared to nicotiana tabacum. The number of flowers per 1 strain of the F1 hybrid is preferably 2/3 or less, 1/2 or less, 1/3 or less, 1/4 or less, and 1/5 or less, as compared with nicotiana tabacum.

Since the F1 hybrid has a small number of flowers and is localized on the upper part, the flowers can be easily removed.

(2) Harvesting of the overground part

Preferably, the F1 hybrid can harvest more of the useful aerial parts including leaves, stems for the manufacture of tobacco products than the P.tabacum.

In one embodiment, the yield of aerial parts of the F1 hybrid is increased over that of nicotiana tabacum.

The yield per 1 strain of aerial parts is preferably 1.2 times or more, 1.5 times or more, 1.8 times or more, 2 times or more, 2.1 times or more as compared with tobacco of the shade.

In one mode, the yield of dried leaves of the F1 hybrid is increased over that of nicotiana tabacum. The "dried leaves" are leaves of tobacco plants which are dried by subjecting the leaves to drying treatment such as freeze drying, heat drying, sun drying, air drying, and the like.

In one embodiment, the leaf length, leaf width, or both the leaf length and leaf width of the largest leaf of the F1 hybrid is greater than that of the shade tobacco. In example 1, the leaf length and leaf width of the largest leaf of the F1 hybrid were about 3 times as compared with those of the shade tobacco 90 days after transplantation. Therefore, the leaf length and leaf width of the largest leaf, or both the leaf length and leaf width of the largest leaf are preferably 1.5 times, 2 times, 2.5 times, or 3 times as large as those of F1 hybrid.

In one embodiment, the F1 hybrid has reduced branching compared to nicotiana tabacum. In one embodiment, the "number of branches" is about the same as the number of leaves. Leaf harvesting is easier than in nicotiana tabacum. In one embodiment, the side shoots are extracted to a lesser extent than the shade tobacco. Therefore, the F1 hybrid is easy to harvest leaves.

In a preferred embodiment, the F1 hybrid has a higher aerial part yield than nicotiana tabacum and leaves can be harvested easily.

(3) Insect resistance to pests

The F1 hybrid preferably has strong tolerance to pests.

It is known that, in general, Sichuan tobacco, which is a wild species, has low insect resistance against many pests, and shadow tobacco has high insect resistance against pests.

In one embodiment, the F1 hybrid has improved insect tolerance to foliar-feeding pests over Sichuan tobacco. More preferably, the F1 hybrid has improved insect resistance against leaf-feeding pests as compared to both Sichuan tobacco and shade tobacco.

In one embodiment, the F1 hybrid has improved insect tolerance to storage pests compared to choma tobacco. More preferably, the F1 hybrid has improved insect resistance against storage pests compared to both Chuanshang tobacco and Yingshao tobacco.

In one embodiment, the F1 hybrid has improved insect tolerance to foliar-feeding pests over the cultivar tobacco.

In one embodiment, the F1 hybrid has improved insect tolerance to storage pests over cultivated tobacco.

In the present specification and claims, "insect pests" are used as a generic term for any insect that causes damage to tobacco plants. Examples of foliar-eating pests include: oriental tobacco budworm (helicobacto budworm, Helicoverpa assaulta), tobacco budworm (helicobacto budworm, helicothris virescens), cotton bollworm (heliconebacterium, Helicoverpa armigera), tobacco hornworm (helicobacto hornworm, Manduca sexta), Spodoptera litura (Spodoptera litura), and beet armyworm (Spodoptera exigua).

"tobacco budworms" are known as pests of plants of the solanaceae family, such as tobacco and pimento, water, flowers, and the like, which are bitten by their larvae (budworms) in particular. "greens" are green insects with no long hairs covering the body in the larvae of the lepidoptera (lepidoptera, butterflies, moths).

Examples of storage pests include tobacco beetles (Lasioderma sericorne), tobacco leaf rollers (Ephestia lutella), and insects which feed on dry leaves of tobacco.

(4) Ingredients contained in tobacco

The F1 hybrid is preferred to have no reduction in all or a part of the components contributing to the characteristic flavor (which is a preferred flavor of tobacco) as compared with the shade tobacco. Examples of the component contributing to the preferable flavor of tobacco include 1-isopropyl-5- (hydroxymethyl) -8-methyltricyclo [4.4.0.02,8] dec-4-ene, 3-methylpentanoic acid, 4-methylhexanoic acid, and the like.

Thus, the F1 hybrid can be used for tobacco products, flavors, and the like while maintaining the preferred flavor of the shade tobacco.

3. Method for preparing F1 hybrid

In one embodiment, the present invention provides a method for preparing an F1 hybrid. The method of the invention comprises mating shade tobacco with Sichuan tobacco. The mating may be performed by a known method for mating tobacco plants.

In the method of producing the F1 hybrid of the present invention, either of the shade tobacco and the chuanzhong tobacco may be a seed parent or a pollen parent.

In one embodiment, the seed parent is nicotiana tabacum and the pollen parent is nicotiana tabacum. In another embodiment, the pollen parent is Nicotiana tabacum and the seed parent is Nicotiana armandii. More preferably, the seed parent is Nicotiana tabacum and the pollen parent is Nicotiana tabacum.

In one embodiment of the method for producing the F1 hybrid of the present invention, the method may further comprise, after mating the shade tobacco and the chuanzhong tobacco: and (3) selecting the F1 hybrid having the more preferable characteristics. The step of selecting the F1 hybrid can be performed based on the ratio (%) of the height of the lowest flower to the plant height, the number of flowers per 1 plant, the leaf length and leaf width of the largest leaf, or both the leaf length and leaf width of the largest leaf, the number of branches, the yield of overground parts, the yield of dried leaves, and the like, but is not limited thereto. These characteristics are as described in the section "characteristics of 2.F1 hybrid". In the step of selecting the F1 hybrid, a plurality of selection steps may be combined for a plurality of characteristics exemplified above. The order of performing the plurality of sorting steps is not particularly limited.

4. Composition comprising a metal oxide and a metal oxide

In one embodiment, the present invention provides a composition comprising the F1 hybrid described above or a portion thereof, or an extract comprising the same.

The composition may be used as it is or as a part of a tobacco plant, or may be used as it is, or as a material obtained by cutting, pulverizing or grinding the tobacco plant into a fine sheet, slurry or fine powder. In the case of the composition, the tobacco plant or a part thereof may be used as it is for harvesting tobacco from a field or the like, or tobacco from which moisture is partially released indoors or outdoors for a given period of time may be used, or tobacco from which moisture is substantially completely released by a dryer or the like may be used.

The "extract" is a substance obtained by extracting a material derived from the leaf, stem, or the like of a tobacco plant in order to improve the flavor of a tobacco product and to reduce the content of a specific component in the tobacco product. The extraction method may use a known method for extracting essential oils, specific components, and the like from plants.

The composition is in the form of cut filler, powder, tablet, or solution, but is not limited thereto.

5. Product(s)

The hybrid F1 or a part thereof, or an extract thereof, and the composition containing the hybrid F1 or a part thereof, or an extract thereof, have a characteristic flavor derived from shade tobacco. Therefore, in one embodiment, these materials can be used as a raw material for a tobacco product or the like to impart a characteristic flavor derived from shade tobacco to the product.

In one embodiment, the present invention provides a product comprising the F1 hybrid described above or a portion thereof, or an extract comprising the same.

In one embodiment, the present invention provides a product comprising a composition comprising the F1 hybrid described above or a portion thereof, or an extract comprising the same. The product includes, but is not limited to, tobacco products, flavors, agricultural chemicals, and pharmaceuticals.

The kind of "tobacco product" is not particularly limited. In addition to cigarettes (cigarettes), cigars, pipe tobacco, snuff, chewing tobacco, shredded tobacco, hookah, and the like are also included. In addition, a heating type flavor inhaler using an aerosol generated by heating tobacco as an aerosol source, a non-heating type flavor inhaler for extracting flavor without heating tobacco, and the like are also included.

The kind of the perfume or the pesticide is not limited. Examples of the agricultural chemicals include agricultural chemicals intended to prevent and control bacteria, nematodes, acarids, insects, rats, other animals and plants, viruses, to promote or inhibit physiological functions of agricultural crops, and to inhibit germination. The perfume, pesticide and medicine can be used alone or together with other effective components.

In one embodiment, the present invention provides the use of the F1 hybrid described above, or a portion thereof, or an extract thereof, in the manufacture of a composition or product.
