The method for preparing a composition for improving and/or treating depression with erectile dysfunction according to claim 5, which comprises the steps of: extracting the raw materials with water for 2-4 times, adding 8-12 times of water each time, decocting for 1-3 hr, mixing decoctions, filtering, and concentrating the filtrate to obtain extract with relative density of 1.10-1.50.

7. A medicament for improving and/or treating depression accompanied by erectile dysfunction, which is characterized by comprising the traditional Chinese medicine composition of any one of claims 1 to 3 and pharmaceutically acceptable auxiliary materials.

8. The medicament of claim 7, wherein the dosage form of the medicament is a solid oral dosage form.

9. The pharmaceutical composition of claim 8, wherein the pharmaceutical composition is in the form of granules, tablets or capsules.

10. Use of a Chinese medicinal composition according to any one of claims 1 to 3 for the preparation of a medicament for the amelioration and/or treatment of depression associated with erectile dysfunction.

Technical Field

The invention belongs to the field of traditional Chinese medicines, and particularly relates to a traditional Chinese medicine composition for improving depression accompanied by erectile dysfunction, and a preparation method and application thereof.

Background

Depression (depression) is a type of mood disorder disease characterized by high prevalence, high recurrence rate, high disability rate, and suicidal ideation. Penile Erectile Dysfunction (ED) is one of the major clinical symptoms in male patients and is associated with depressive severity and is also prone to prolonged disease progression and recurrence. ED means that the penis continues to fail to attain and/or maintain an adequate erection to achieve satisfactory sexual life. Investigations have shown that up to 52% of 40-70 year old men have varying degrees of ED, with an estimated 3 hundred million people worldwide by 2025; the tendency of patients suffering from ED in China and even in the world is gradually increased year by year.

ED is prevalent in depressed male patients, and studies have found that ED occurs 1.82 times more frequently in depressed patients than in patients without depression, and the more severe the depressive symptoms, the higher the incidence of ED. Aniah et al, who studied depression patients who were not given antidepressant therapy, found that 65% of the patients were accompanied by sexual dysfunction. Both depression itself and the use of antidepressants can cause sexual function problems, which are not favorable for the control of patients' illness to a certain extent, and many patients aggravate self-inferior feeling due to privacy problems, further aggravate depression symptoms, and finally cause self-mutilation and suicide. Therefore, attention should be paid to such diseases and prognosis is active.

The treatment methods of ED are various and comprise oral medicines, intracavernosal injection medicines, psychological treatment, prosthesis implantation surgery and the like, but the methods have more potential adverse reactions, such as penile pain, emission delay, complexion redness, headache, deafness, abnormal vision and the like, and can not fundamentally solve the problem, and simultaneously, the cost is high, so that a certain part of depression patients are difficult to bear. Therefore, the discovery of effective, safe, inexpensive alternative therapies to intervene in depression with erectile dysfunction is key to improving or ameliorating the quality of life and social satisfaction in patients with depression.

The traditional Chinese medicine has relative advantages in treating male erectile dysfunction (impotence), comprehensively evaluates the abnormality of the organism by applying the whole treatment thought, provides an effective treatment means for patients by carrying out syndrome differentiation treatment on the traditional Chinese medicine, and further improves the life quality of the patients. The depression accompanied with erectile dysfunction is called as stasis impotence in the traditional Chinese medicine, the traditional Chinese medicine considers that the key of the pathogenesis of the stasis is qi stagnation, liver qi loss and diarrhea, spleen dysfunction, heart failure and viscera disorder, and the impotence is mostly caused by decline of vital gate fire and deficiency and cold of vital essence and qi; liver and kidney are homologous, the mother and the son grow mutually, essence and blood are mutually transformed, and the storage and the release are mutually combined, so that the normal physiological functions of the pectoral muscles are maintained. At present, no patent and paper about improving the symptoms of depression and erectile dysfunction accompanied by liver depression and kidney yang deficiency is seen, but at present, clinically, impotence caused by single treatment or antipsychotic drugs is concentrated, and corresponding measures are not taken for treating erectile dysfunction accompanied by depression.

In one embodiment, the raw materials are taken according to the proportion and extracted with water for 2-4 times, 8-12 times of water is added for each time, the raw materials are decocted for 1-3h, then decoction liquids are combined, filtered, and finally the filtrate is concentrated to extract with the relative density of 1.10-1.50.

The invention also provides application of the traditional Chinese medicine composition for improving and/or treating depression accompanied by erectile dysfunction.

Liver depression and kidney yang deficiency are the basic pathogenesis of depression type impotence, and the liver soothing and yang warming method is the therapeutic principle. Wherein, epimedium and morinda root enter liver and kidney channel, tonify kidney yang, strengthen tendons and bones, bupleurum enters liver and gallbladder channel, soothes liver and relieves stagnation, and raises yang qi to invigorate yang, all are monarch drugs, white peony root astringes yin, nourishes blood and softens liver, nourishes liver blood, regulates liver qi, can make bupleurum ascend and disperse without the disadvantage of damaging yin and blood, and is assisted with trifoliate orange fruit to regulate qi to relieve stagnation, discharge heat to break knot, and lowers with bupleurum to strengthen the function of smoothing qi movement, and is assisted with raw fossil fragments, raw oysters to relieve convulsion and calm nerves, subdue yang and strengthen kidney, and liquorice, harmonizing various drugs, benefiting spleen and regulating stomach. The whole formula is prepared by adding flavor to the Sini powder, has the effects of soothing the liver, strengthening the spleen, warming yang, tonifying the kidney and calming the nerves and is beneficial to improving the impotence symptom of a depression patient. The traditional Chinese medicine composition is prepared into a medicine for effectively treating depression accompanied by erectile dysfunction by a modern pharmaceutical technology according to the formula of traditional Chinese medicine by differentiation and treatment methods, and particularly has a good improving effect on depression accompanied by erectile dysfunction due to liver depression and kidney yang deficiency. Animal experiments and random control clinical experiments prove that the erectile dysfunction of rats or patients with depression can be effectively improved. In one embodiment of the invention, the invention provides a traditional Chinese medicine composition for improving and/or treating depression accompanied by erectile dysfunction, which comprises the following active ingredients in percentage by weight: 10-20 parts of radix bupleuri, 10-20 parts of radix paeoniae alba, 10-20 parts of immature bitter orange, 10-20 parts of liquorice, 8-20 parts of epimedium, 8-20 parts of radix morindae officinalis, 10-30 parts of raw keel and 10-30 parts of raw oyster.

Preferably, the effective components of the traditional Chinese medicine composition are prepared from the following raw materials in percentage by weight: 15 parts of radix bupleuri, 15 parts of radix paeoniae alba, 15 parts of immature bitter orange, 15 parts of liquorice, 10 parts of herba epimedii, 10 parts of morinda officinalis, 30 parts of raw keel and 30 parts of raw oyster.

Wherein, the licorice is preferably prepared licorice, namely, the licorice baked with honey is prepared according to a conventional method, for example, the method 1: diluting Mel with appropriate amount of boiling water, adding into Glycyrrhrizae radix, stirring, sealing thoroughly, parching in parching container to yellow to deep yellow, taking out when it is not sticky, and cooling. For example, the method 2 is: firstly, putting honey into a pot, refining the honey into medium honey, adding liquorice slices with slow fire, uniformly stirring and frying for 3-5 min, taking out the honey from the pot, putting the honey into a baking room or an oven, drying at 60 ℃ until the honey is not sticky, taking out the honey, and cooling.

The traditional Chinese medicine composition for improving and/or treating depression accompanied by erectile dysfunction can be prepared according to a conventional method in the field of traditional Chinese medicines.

Preferably, the present invention provides a method for preparing the above-mentioned composition for improving and/or treating depression accompanied by erectile dysfunction, comprising the steps of:

extracting the raw materials at a certain ratio with solvent, mixing extractive solutions, and filtering to obtain filtrate.

Further preferably, the solvent is water. The preparation method is preferably as follows: extracting the raw materials with water for 2-4 times, adding 8-12 times of water each time, decocting for 1-3 hr, mixing decoctions, filtering, and concentrating the filtrate to obtain extract with relative density of 1.10-1.50.

Still further, the preparation method preferably comprises: extracting the raw materials with water for 3 times according to a ratio, adding 10 times of water for each time, decocting for 3h, then combining the decoctions, filtering, and finally concentrating the filtrate to obtain an extract with a relative density of 1.50.

The embodiment of the invention also provides application of the traditional Chinese medicine composition in preparing a medicine for improving and/or treating depression accompanied with erectile dysfunction. Particularly, the medicinal composition is preferably applied to the treatment and improvement of patients with depression accompanied by liver-depression and kidney-yang deficiency in erectile dysfunction.

A series of animal experiments and clinical experiments show that the medicine for improving and/or treating depression accompanied by erectile dysfunction has a good curative effect on depression accompanied by erectile dysfunction, can relieve depression symptoms, improves sexual functions, and is beneficial to improvement of life quality and social satisfaction of patients.
