Technical Field

The invention belongs to the field of medicines, and particularly relates to application of spring mulberry leaves in preparation of an anti-aging medicine.

Background

Aging is a natural process of life, and with the increase of age, organisms have hypofunction, and the internal environment stability and the stress ability are reduced. Cells, tissues and organs in an aging body often show various degenerations in structure and function, and are accompanied by the generation of aging-related diseases such as diabetes, neurodegenerative diseases, cancer and the like, so that the quality of life is reduced, and a heavy burden is imposed on families and society (Collins et al, 2006; Muller et al, 2007).

The 21 st century is a century of aging of the population, which has brought about a series of very serious medical and social problems. In order to fundamentally improve the living quality of the elderly, a method for improving physical functions and delaying aging needs to be found to ensure that the elderly can be aged healthily. This goal has motivated modern scientists to screen for anti-aging drugs.

The application history of Chinese herbs in China has been 5000 years (Yu et al, 2010) so far. The traditional Chinese medicine also plays a role in improving the life quality of the old and promoting the healthy aging of the human. Folium Mori is derived from dried leaves of Morus albal. Generally, people divide mulberry leaves into spring mulberry, frost mulberry and mulberry leaves picked between the two. Spring mulberry is mostly used for eating, spring mulberry is mostly used for breeding silkworms after being picked, and frost mulberry is collected after being frosted for the first time and is used as a medicine. The frosted mulberry is obtained by removing impurities after picking and drying in the sun, is a wind-heat dispersing medicine and can be taken orally or applied externally. It is sweet and bitter in taste and cold in nature, enters lung and liver meridians, and has the functions of dispelling wind and heat, clearing lung-heat and moistening dryness, and clearing liver and improving vision. At present, effective components such as flavone, polyphenol, alkaloid, polysaccharide and the like are separated and extracted from downy mulberry, and the downy mulberry has various pharmacological activities such as oxidation resistance, inflammation resistance, blood sugar reduction, blood pressure reduction, blood fat reduction, immunity improvement, aging resistance, digestive system and excretory system health maintenance, tumor resistance and the like, and becomes a raw material for developing functional foods such as mulberry leaf tea, mulberry leaf juice beverage, mulberry leaf noodles, mulberry leaf powder and the like. Caenorhabditis elegans (Caenorhabditis elegans) has been used for life analysis abroad, and has been in progress for 30 years to date (Johnson and Wood, 1982). It is a good model organism for aging and anti-aging research. It is cheap and easy to culture; the generation period is short, the number of offspring is large, a large number of individuals with consistent backgrounds can be obtained, the experimental repeatability is ensured, and the experiment is carried out by adopting a large sample amount, so that the influence of individual difference is eliminated; conserved in gene and molecular pathways with higher organisms (Chua Seiko et al, 2008); nematodes live through the same aging process as humans, with progressive accumulation of lipofuscin in the gut, progressive loss of muscle motor capacity, and progressive loss of reproductive capacity until death (Vayndorf, 2009). Thus, nematodes became the first model organism for senescence studies (Johnson, 2008). Nematodes are also increasingly being used in the research and development of anti-aging traditional Chinese medicines in China (Chua Seijie et al, 2008; Konhongjie et al, 2011; Wang Xiaoyan et al, 2010; Kan et al; 2011).

The invention provides application of spring mulberry leaves in preparation of a medicine for resisting aging and treating diseases related to mitochondrial disorder. A large number of documents show that frosted mulberry leaves are the best to be used as medicines, although frosted mulberry leaves are frosted. Unexpectedly, compared with the mulberry leaves as frost, the mulberry leaves as picked in the beginning of spring have more obvious anti-aging effect, and are particularly suitable for preparing the medicament for delaying the premature senility.

Disclosure of Invention

The invention aims to provide application of spring mulberry leaves in preparation of anti-aging medicaments.

The folium mori is picked in the early spring.

The invention also aims to provide application of the mulberry leaves in spring in preparing the medicine for delaying the premature senility, wherein the mulberry leaves are picked in the early spring.

The invention also aims to provide application of the mulberry leaves in preparing the medicines for treating diseases related to mitochondrial dysfunction, wherein the mulberry leaves are picked in the early spring.

The dosage form of the mulberry leaf spring of the invention is one of tablets, capsules, powder, syrup, granules, medicinal granules, pills, mixture or dropping pills.

The experiment is carried out by taking caenorhabditis elegans as model organisms, the caenorhabditis elegans is a mitochondrial respiratory chain complex II large subunit mev-1 mutant, a mitochondrial respiratory chain complex III iron-sulfur protein isp-1 mutant and an N2 wild type nematode respectively, and water extracts of different mulberry products are adopted to treat the tested caenorhabditis elegans. The results show that: the mulberry leaf water extract has a remarkable treatment effect on mitochondrial dysfunction, namely senilism caenorhabditis elegans, and tends to prolong the life of the nematode with mitochondrial dysfunction. However, the longevity of the N2 wild-type nematode was not affected. Meanwhile, the mulberry leaf water extract obviously improves the movement capacity of the senilism nematodes without influencing the reproduction of the senilism nematodes. The results indicate that the mulberry leaf water extract not only has the potential of delaying premature senility, but also has the potential of treating diseases related to mitochondrial dysfunction. Surprisingly, the anti-aging effect of the mulberry leaves in spring is more remarkable compared with the conventionally used frosted mulberry leaves.

The technical solution of the present invention is further described in detail with reference to the following specific examples, but the scope of the present invention is not limited to the following.
