1 Material

The test articles comprise NDV standard antigen and positive serum, and are used for determining the NDVHI antibody titer.

Test animals: hailan brown chicken

Test methods and groups: 240 helan brown chicks are randomly divided into 12 groups, 4 replicates are arranged in each group, 5 chicks are replicated in each group, the chicks are flatly raised on the net according to a conventional raising method, and the chicks are fed with complete formula feed without any medicine, are fed freely and are drunk with water. When the chicks are raised to 14 days old, ND vaccines are used for carrying out primary immunization through eye drop and nose drop; the second immunization was carried out by drinking water at the age of 28 days. After the first and second immunizations, the drugs were administered continuously for 7 days, with free water, at doses as given in table 1 below, 2 chickens were randomly selected from each replicate group at 21 and 35 days of age, and 8 chickens per group were collected intravenously at 0.2ml, and placed in 1.5ml of serum for centrifugal separation, and the ND antibody levels of the chickens were determined by HI method, and the specific grouping and treatment methods are as given in table 2 below.

In 7 months of 2020, a chicken farm in Shandong with a history of Newcastle disease was selected for the test. 6000 avium broilers of 5 days old in the chicken farm were randomly divided into 4 groups of 1500, test 1, test 2, test 3 and test 4. Test results show that the traditional Chinese medicine composition prepared in the embodiment 5 and the embodiment 1 and the medicine in the comparative example 1 can effectively prevent the occurrence of the newcastle disease, the traditional Chinese medicine composition prepared in the embodiment 5 has the best effect, the morbidity is 0, the effective rate can reach 100 percent, and the traditional Chinese medicine composition is the optimal prescription of the invention, and the effect of preventing the newcastle disease of the invention is far better than that of the medicine group in the comparative example 1, and the difference of the traditional Chinese medicine composition and the medicine group is obvious compared with that of a blank control group. The medicine has obvious effect of preventing the newcastle disease.

Test example 3 Observation of the preventive Effect of the Chinese medicinal composition of the present invention on avian influenza

A chicken farm of Jiangsu Nanjing with a history of avian influenza is selected to carry out an avian influenza prevention test, and 2000 chicks are randomly divided into 4 groups of 500 chickens each. Group 1, 1 kg of the traditional Chinese medicine composition prepared in the embodiment 5 of the invention is added into each ton of feed for mixed feeding administration, and the feed is freely taken for 7 days; group 2, adding 1 kg of the traditional Chinese medicine composition prepared in the embodiment 3 of the invention into each ton of feed, and carrying out mixed feeding administration for 7 days after free feeding; group 3, 1 kg of the traditional Chinese medicine composition prepared in the comparative example 4 is added into each ton of feed for mixed feeding administration, and the feed is freely eaten for 7 days; in the first group, no drug was added to the feed. After the administration, the observation was carried out for 40 days. The test results are shown in Table 4.

Test results show that the traditional Chinese medicine composition of the embodiment 5 and the embodiment 3 and the medicine of the comparative example 4 have obvious effect on preventing avian influenza, the protection rates of the traditional Chinese medicine composition of the invention can reach 100 percent and 99.80 percent respectively, and are better than 95 percent of the comparative example 4, and the effect of synergy between the mannooligosaccharide and the traditional Chinese medicine composition of the invention is demonstrated. The test 1, 2 and 3 groups are significantly different from the blank control group. The Chinese medicinal composition has very good effect of preventing avian influenza.
