Performance test method

1. Free radical scavenging ability test:

(1) DPPH method

Accurately weighing 6mg of DPPH (1, 1-diphenyl-2-trinitrophenylhydrazine), dissolving with 95% VOL ethanol water solution, dissolving with the aid of ultrasonic wave, then fixing the volume to a 50mL volumetric flask, and diluting with 95% VOL ethanol water solution until the DPPH concentration is 3mg/100mL to obtain DPPH mother liquor; the DPPH mother liquor is stored in a refrigerator at 0-4 ℃ in a dark place.

Dissolving 2g of the acanthopanax senticosus extract obtained in the example 1 in deionized water to obtain a sample to be detected, adding an equal volume of DPPH mother liquor into the sample to be detected, carrying out a light-shielding reaction at 26 ℃ for 30min, measuring the absorbance at 525nm, and calculating the IC50 (half inhibition concentration); see table 1.

At the same time, 2g of ascorbic acid was used as a control group, and the above-mentioned operation was repeated to determine the half inhibitory concentration of ascorbic acid against DPPH.

The lower the semi-inhibitory concentration to DPPH, the greater the free radical scavenging ability of the sample.

(2) ABTS method

Preparing ABTS (2, 2-diazo-bis (3-ethyl-benzothiazole-6-sulfonic acid) diammonium salt) aqueous solution with the concentration of 7mmol/L, mixing the ABTS aqueous solution with 2.45mmol/L potassium persulfate aqueous solution according to the weight ratio of 1:1, stirring the mixture for 16 hours in a dark place, and then adding PBS solution (phosphate buffer solution) to dilute the mixture until the light absorption value at 734nm is 0.70 +/-0.02 to obtain ABTS mother liquor.

Diluting the Acanthopanax senticosus extract of example 1 with 50% VOL petroleum ether ethanol solution to 3. mu.g/mL, 6. mu.g/mL, 13. mu.g/mL, 25. mu.g/mL, 50. mu.g/mL; mixing the samples with the concentration gradient with ABTS mother liquor respectively, wherein the volume ratio of the samples to the ABTS mother liquor is 1: 20; after mixing, the mixture was placed at 26 ℃ in the dark for 6min, and the absorbance at 734nm was measured to calculate the ABTS clearance D, see Table 2.

D ═ 100% (1-a1/a0) ×, where a1 is the absorbance of the ABTS mother liquor and a0 is the absorbance measured after mixing the ABTS mother liquor with the sample.

The higher the ABTS clearance rate D, the stronger the free radical scavenging ability and the oxidation resistance of the sample.

2. Skin care effect:

180 subjects were screened and divided into 3 groups of 60 persons each. Selecting the left and right cheekbones of the face of a subject, the left and right cheeks at the intersection of the nose tip and the pupil, and the left and right sides at the intersection of the temple and the mouth corner as test areas, firstly, measuring the skin texture of the test part before applying the cosmetics by a technician by using a visia facial image analyzer, measuring five times at each point, and taking an average value. The subjects continued to use the skin care products of examples 2-4 for four weeks (1 each day in the morning and evening), and again tested for skin texture. Counting the skin texture degree value measured at each time of the experimental part of the subject, and analyzing the change rule of the skin texture, thereby determining the effect of the skin care product of the embodiment 2-4 on improving the skin texture; see table 3.
