Background

Obesity is a health problem facing the world, and the world health organization has listed obesity as the second leading cause of death second only to smoking. With the changes of dietary structure, environment and life style, the incidence of obesity in China in recent years is rapidly increasing. Obesity can cause metabolic syndromes such as hypertension, hyperlipidemia, insulin resistance, etc., and lead to type ii diabetes, cardiovascular disease and cancer, ultimately increasing mortality. In addition, overweight and obesity of pets and pregnant animals can also cause great threat to the health of the body, and need further research and solution.

Raffinose is a trisaccharide composed of galactose, glucose and fructose, and is a pure natural oligosaccharide found in vegetables such as beans, cabbage, brussels sprouts and some grains. Since the small intestine of mammals such as humans and pigs cannot secrete α -galactosidase required for the digestion of raffinose, raffinose cannot be directly utilized by these animals, but reaches the intestine and is fermented by microorganisms. Previous researches find that the raffinose has certain functions and functions, for example, the raffinose can be used as a prebiotic for proliferating bifidobacteria, and establishes a healthy intestinal flora environment by promoting the proliferation and growth of beneficial bacteria and effectively inhibiting the proliferation of harmful bacteria in the intestinal tract; in addition, raffinose has also been used in humans as a sugar substitute and sweetener due to its lower energy value than sucrose, coupled with its property of not being digested and absorbed into the blood in the stomach and small intestine.

In recent years, along with the development of functional oligosaccharides, the research and application of raffinose in animal production and human food are increasingly paid more attention, but no research on animal obesity by raffinose is found at present, and no report on the influence of raffinose on weight gain and obesity effect by taking a growing pig as a model is found.

Detailed Description

The technical solutions of the present invention will be further described with reference to the following examples, which are included in the following embodiments, but are not limited to the following examples, and the following examples are used to illustrate the present invention, but are not intended to limit the scope of the present invention. Unless otherwise specified, the technical means used in the examples are conventional means well known to those skilled in the art. The test methods in the following examples are conventional methods unless otherwise specified.

Raffinose is the best known trisaccharide in nature, is formed by combining galactose, fructose and glucose, is also called raffinose and melizitose, and can be widely applied to the industries of food, health-care food, medicine, cosmetics, feed and the like because raffinose has unique physicochemical and physiological effects, is used as a prebiotic for proliferating bifidobacteria, can also be used as a main component of a protective delivery liquid for transplanting living organs of human bodies and animals, and can also be used as a synergist for prolonging the survival period of living bacteria at normal temperature.

In experiments, the invention unexpectedly discovers that the addition of 0.5 percent of raffinose in the diet can unexpectedly reduce the feed intake and fat anabolism of animals, thereby achieving the effect of reducing weight and losing weight of the animals.

It should be noted that the pig is taken as the experimental subject in the embodiment of the invention, and the influence of the raffinose added to the diet on the weight of the pig is researched. The pig is an excellent model animal, has been widely applied to various aspects of life science research, and is very suitable for the research of obesity.

The addition of the raffinose can achieve the effect of reducing daily gain by reducing the daily feed intake of the growing pigs, can obviously reduce the digestibility of the growing pigs to fat and energy, reduce the contents of growth hormone and feeding promoting hormone neuropeptide Y in the serum of the growing pigs, obviously improve the content of the feeding inhibiting hormone casein peptide, and show that the raffinose can effectively reduce the appetite of animals and growth promoting hormone, and in addition, the addition of the raffinose can also obviously reduce the content of triglyceride in the serum of the growing pigs, thereby achieving the effect of relieving the obesity harm.

In the present embodiment, the "basal group", that is, the group using the basal diet, that is, the group using the non-high-fat diet, refers to the diet formulated with reference to NRC2012, and the "high-fat group", that is, the group using the high-fat diet, refers to the diet obtained by adding 10% of soybean oil or the like to the basal diet in place of corn starch.

In order to better understand the technical scheme provided by the invention, the following specific examples respectively illustrate the application of raffinose provided by the above embodiments of the invention in preparing a diet for reducing the feed intake of mammals and relieving weight gain, and the effect detection.

Example 1 effects of Raffinose in pig diets

The growing pig test was performed with raffinose. The test selected 40 healthy pigs of similar body weight (45.03 + -0.92 kg) and divided into 4 treatment groups, each treatment group having 10 replicates, each replicate having 1 pig, and fed in a single cage. The treatment was basal group, raffinose group, high fat group, and high fat-supplemented raffinose group, respectively. The raffinose group contains raffinose 0.5%, the high fat group contains soybean oil 10.0%, and the high fat group contains raffinose 0.5% and soybean oil 10.0%. The addition amounts of raffinose and soybean oil were calculated based on the total amount of the diet, and the formulation of each group is shown in table 1. The test period was 28 days with the test diet fed directly three times a day. After the test is finished, the pigs are slaughtered, blood sampling is carried out, subsequent index measurement is carried out, and the results are expressed by mean values and Standard Error (SE). The results are shown in tables 2 to 5.

Table 1 diet formula for each treatment group

TABLE 2 Effect of Raffinose addition to feed on growth performance of growing pigs

TABLE 3 Effect of Raffinose addition to feed on nutrient digestibility of growing pigs

TABLE 4 Effect of Raffinose addition to diets on serum hormones in growing pigs

TABLE 5 influence of raffinose addition to diets on indexes related to lipid metabolism in serum of growing pigs

As can be seen from the data in table 2, the addition of raffinose to the basal diet can achieve the effect of reducing daily gain by reducing the daily feed intake of growing pigs. When the feed is fed by high-fat diet, the daily gain of the growing pigs can be obviously improved, the feed conversion ratio of the growing pigs is reduced, and the weight gain is too fast; the addition of the raffinose can obviously reduce the daily feed intake of the growing pigs to achieve the effect of reducing the daily gain.

As can be seen from table 3, the addition of raffinose to both the basal diet and the high fat diet reduced the fat and energy digestibility of the growing pigs. The high-fat diet can obviously improve the dry matter, fat, protein and energy digestibility of the growing pigs, and the effect of reducing the fat and energy digestibility of the growing pigs after the raffinose is added is obviously better than the effect of adding the raffinose into the basic feed.

As can be seen from Table 4, the addition of raffinose to both the basal diet and the high-fat diet can significantly reduce the content of growth hormone and the feeding-promoting hormone neuropeptide Y in the serum of the growing pigs, and significantly increase the content of the feeding-inhibiting hormone casein peptide, which indicates that raffinose can effectively reduce the appetite of animals and promote growth hormone.

As can be seen from table 5, the addition of raffinose to the basal diet and the high-fat diet significantly reduced the triglyceride content, thereby achieving the effect of alleviating the risk of obesity. Wherein, the effect of adding the raffinose in the high-fat diet is obviously better than that of adding the raffinose in the basic diet.

In conclusion, the influence of feed on weight gain and obesity of a growing pig model can be reduced by inhibiting the feed intake, fat and energy digestibility of animals by adding raffinose into non-high-fat feed and high-fat feed, and especially in the high-fat feed, the raffinose can obviously inhibit the over-fast growth of the growing pig caused by the high-fat feed and the increase of the blood fat content in serum, so that the obesity caused by the high-fat feed and the negative influence thereof can be relieved. The high-fat diet can cause the growth rate of the growing pigs to be too fast and the blood fat content to be increased, and the effect of the high-fat diet added with the raffinose is better than that of the basic diet added with the raffinose.

Finally, it should be noted that: the above embodiments are only used to illustrate the technical solution of the present invention, and not to limit the same; while the invention has been described in detail and with reference to the foregoing embodiments, it will be understood by those skilled in the art that: the technical solutions described in the foregoing embodiments may still be modified, or some or all of the technical features may be equivalently replaced; and the modifications or the substitutions do not make the essence of the corresponding technical solutions depart from the scope of the technical solutions of the embodiments of the present invention.
