Toothpastes are complex mixtures typically made by mixing abrasives (e.g., calcium carbonate, dibasic calcium phosphate, calcium pyrophosphate, silica, aluminum hydroxide), humectants (e.g., glycerin, sorbitol, propylene glycol, polyethylene glycol, and water), surfactants (e.g., sodium lauryl sulfate, sodium 2-acyloxy sulfonate, sodium lauroyl sarcosinate), thickeners (e.g., carboxymethylcellulose, antler pectin, hydroxyethyl cellulose, xanthan gum, guar gum, carrageenan, etc.), sweeteners (e.g., glycerin, sodium cyclohexylamine sulfonate, sodium saccharin, etc.), preservatives (e.g., potassium sorbate and sodium benzoate), active additives (e.g., chlorophyll, fluoride), and pigments, flavors, and the like. The special toothpaste is toothpaste with special properties.

The fluorine-containing toothpaste contains active substances such as sodium fluoride, stannous fluoride, sodium monofluorophosphate and zinc fluoride, and is effective in preventing dental caries. The chlorophyll toothpaste is added with chlorophyll, and has special effects of preventing gingival bleeding and preventing halitosis. The toothpaste containing enzyme can decompose residual food, and has effects of cleaning oral cavity and preventing worm damage. The medicated toothpaste can be added into toothpaste for preventing and relieving oral diseases. The toothpaste of radix scutellariae, the toothpaste of calculus bovis and the like sold in the market have obvious functions of slowing down and treating gingival bleeding, gingival swelling, halitosis, dentin hypersensitivity and the like.

The functional toothpaste in the market at present mainly comprises two main types, one is used for preventing and assisting to treat oral diseases, such as periodontitis, gingivitis, oral ulcer, hemostasis and pain relief, tooth sensitivity and the like, and the other is used for helping to adjust and improve oral conditions, such as tooth whitening, oral odor removal, dental calculus removal, stain removal, dental plaque removal and the like. Some effective components of Chinese herbal medicines such as pseudo-ginseng, paeonol, radix zanthoxyli, glabrous sarcandra herb and the like are added into the toothpaste, some western medicine components such as tranexamic acid or other chemical components such as strontium chloride, sodium fluoride and the like are added into the toothpaste, some antibiotics are even added into the toothpaste, certain toxic and side effects are caused to a human body, and certain hidden danger exists in safety performance.

Disclosure of Invention

Aiming at the defects of the prior art, the invention provides the toothpaste with the efficacy of preventing and treating the oral diseases and the preparation method thereof, which have no toxic or side effect on a human body and have good sensitivity effect on teeth.

The invention is realized by the following technical scheme, and the toothpaste with the efficacy of preventing and treating oral diseases comprises the following components: fluoride, biological enzyme agent, bioactive glass, chitosan porous microsphere, glycerol and abrasive;

the abrasive is one of main components of toothpaste, is used for removing dirt on the surface of teeth and endowing the teeth with original luster, generally requires that the hardness of the abrasive is lower than the hardness of enamel on the surface of a dental crown by a proper degree, and takes the principle of not damaging the texture of the teeth as a principle, and the abrasive of the powder abrasive for the toothpaste generally has the hardness of 2-3.5, so that the aims of not only removing the dirt accumulated on the surface of the teeth but also not damaging the glaze surface of the teeth are fulfilled. The abrasives selected by the invention comprise calcium carbonate, calcium hydrophosphate, calcium pyrophosphate, aluminum hydroxide, silicon dioxide and the like;

stannous fluoride toothpastes are incompatible with dicalcium phosphate and compatible with insoluble calcium metaphosphate. Early stannous fluoride toothpastes were susceptible to hydrolysis and oxidation in solution due to the chemical reactivity and instability of stannous fluoride, had the disadvantages of staining and metallic taste, and had a short shelf life. Through research and improvement, the novel stannous fluoride toothpaste has good stability and biological activity, can prolong the storage time, and has the effects of preventing dental plaque, gingivitis and caries. Stannous fluoride has two antibacterial mechanisms: firstly, sterilization, because stannous fluoride is nonspecific, stannous fluoride can react with bacterial cell membranes to cause cell membrane rupture, so that bacteria can be dissolved and killed; secondly, the sugar metabolism of the bacteria is influenced by inhibiting the metabolic enzyme of the bacteria, the reproduction of the bacteria is reduced, the adhesion of the bacteria to the tooth surface is prevented, and the inflammation-causing metabolite generated by the bacteria is reduced; the toothpaste can release stannous ions and fluoride ions in the process of tooth brushing, play roles in resisting bacteria, neutralizing LPS and the like, and thus provide good anti-gingivitis effect; according to the preparation method of the toothpaste, sodium fluoride, bioactive glass and glycerol are added at first to construct a system which is not easy to oxidize, chitosan, a biological enzyme agent and stannous chloride are added, the sodium fluoride and the stannous chloride can be well loaded through the bioactive glass and the chitosan, and fluorine ions and tin ions are released when the toothpaste is in air bubbles in an oral cavity to generate stannous fluoride, so that the stability of the toothpaste (sterilization) is prolonged due to the construction of the loading system.

The biological enzyme preparation, biological enzyme in oral cavity is secreted with saliva, it has functions of dissolving bacteria, resisting inflammation, repairing tissue, inhibiting hemorrhage and regulating oral cavity flora balance, and is an important component of human body natural defense system. However, in most cases, the content of the biological enzymes in human oral cavity is insufficient, such as when people sleep or rest, the body is weak, the medicine is taken for a long time, and the mental stress is too large, so that the content of the biological enzymes in saliva is reduced, the oral cavity balance is damaged, the natural defense ability is lost, and the dental disease is caused. After the biological enzyme is added into the toothpaste, the biological enzyme can be supplemented during tooth brushing, so that oral diseases caused by insufficient biological enzyme due to the reasons can be well solved;

the bioactive glass is prepared by reacting with body fluid in biological environment to form porous SiO2 gel layer at its interface, and then adding Ca in the bioactive glass2+And PO4 in body fluids3-The bioactive glass is diffused and deposited to an SiO2 gel layer, and is gradually mineralized, so that hydroxyapatite and HA covering layers are formed on the surface of the SiO2 gel layer, the chemical bond combination between the bioactive glass and host bones is realized, and the bond is very firm and is equivalent to the strength of a normally healed fracture interface. The bioactive glass is a new high-tech bioactive material capable of producing physical and chemical reaction with tooth tissue, and can quickly produce continuous ion exchange with water and saliva, raise oxygen partial pressure andthe pH value and the generation of stronger negative charges form a porous network bracket consisting of HA (hydroxyapatite) in the oral cavity instantly, and the net frame adsorbs a large amount of silicon ions, sodium ions, strontium ions, calcium ions and phosphorus ions, is easy to attach and permeate into dental tissues and HAs good biocompatibility with dental bone tissues and soft tissues.

The elastic particles formed by swelling chitosan through water are high-quality friction materials for replacing white carbon black in toothpaste; the chitosan has antibacterial and antiinflammatory effects, and can be used for treating oral diseases.

The toothpaste with the effect of preventing and treating oral diseases comprises the following components in parts by weight: 1-2 parts of fluoride, 1-2 parts of biological enzyme agent, 0.2-0.3 part of bioactive glass, 0.2-0.3 part of chitosan, 100 parts of glycerol and 10-15 parts of abrasive

The fluoride comprises the following raw materials in parts by weight: 0.07-0.15 part of sodium fluoride and 0.03-0.05 part of stannous chloride.

Wherein the biological enzyme agent is lysozyme;

wherein the abrasive is calcium carbonate;

moreover, the toothpaste of the invention also comprises 0.5-1 part of foaming agent;

wherein the foaming agent is one or more of sodium lauroyl amide, sodium dodecyl sulfate and betaine;

similarly, the invention also provides a preparation method of the toothpaste with the efficacy of preventing and treating oral diseases, which comprises the following steps:

a. putting sodium fluoride and a friction agent into water diluted by 20 times, uniformly stirring at the rotating speed of 800r/min at 500-;

Culturing at 37 deg.C for 48h, and calculating the diameter of the antibacterial ring around the paper to measure the antibacterial activity of the toothpaste.

The results are shown in Table 1

Therefore, the toothpaste disclosed by the invention has a large bacteriostatic circle and an excellent bacteriostatic effect.

2. Surface microhardness value test

The experiment adopts a method of a toothpaste-artificial saliva extracorporeal circulation model, a bovine enamel sample is polished and subjected to acid etching treatment, the daily use of toothpaste (examples 1-3 and comparative example 1) is simulated, the acid etched and demineralized enamel sample is treated, and the change of the Surface Microhardness (SMH) of the enamel in the comparative experiment process is measured, so that the repair of the acid etched and demineralized enamel by the toothpaste is inspected; the results are summarized in Table 2 below

TABLE 2

Thus, it can be seen that the toothpaste according to the present invention can effectively restore the hardness of the enamel of teeth to the level of the hardness before erosion after 2 days of use, while comparative example 1 cannot be more reduced in hardness with the increase of the use time; therefore, the toothpaste provided by the invention can improve the hardness of the enamel and help the remineralization of the enamel.

Currently, enamel demineralization is a process in which minerals slowly and continuously dissolve, which can lead to a decrease in enamel hardness and even loss of enamel. Hardness is one of the important physical properties of enamel, is the basis of enamel function, and is also a reflection of the degree of enamel mineralization and the content of minerals, and changes in hardness can reflect the acquisition and loss of enamel minerals. Factors causing enamel demineralization are many, such as microbial fermentation to produce acid, frequent drinking of acidic foods and beverages, frequent entry of gastric acid into the oral cavity to contact the teeth due to certain diseases, occupational exposure to acid mist, and the like. When the enamel is demineralized, elements such as calcium, phosphorus and the like are lost, the mineral density is reduced, the surface hardness of teeth is reduced, and the enamel robustness is reduced; on the contrary, when the dental enamel is remineralized, elements such as calcium, phosphorus and the like form crystals again, the mineral density is increased, and the dental enamel robustness is improved. Early demineralization of children dental caries usually presents as white spot clinically, surface demineralization continues to expand, obvious caries lesion can be formed, and mineral calcium and fluorine can help teeth remineralize and repair white spot.

Therefore, the invention ensures effective persistence of fluoride in the toothpaste and can effectively remove caries lesions by reasonably designing the loading mode of the fluoride.

Those skilled in the art will recognize that numerous variations are possible in light of the above description, and thus the examples are intended to describe one or more specific embodiments.

While there has been described and illustrated what are considered to be example embodiments of the present invention, it will be understood by those skilled in the art that various changes and substitutions may be made therein without departing from the spirit of the invention. In addition, many modifications may be made to adapt a particular situation to the teachings of the present invention without departing from the central concept described herein.
