A method for extracting polyphenol components in acer tegmentosum includes the following steps:

taking dry stem skin of Acer tataricus, and weighing;

the ratio of the materials to the liquid is 1g: (20-40) mL, adding a 95% ethanol solution, and soaking for one week;

Influence of the feed-to-liquid ratio

Table 1 shows the results of a single-factor experiment of the feed-liquid ratio at an extraction temperature of 80 ℃ and an extraction time of 3h, wherein the results show that: the ratio of the feed to the liquid is 1g to 30 mL.

The results show that: the extraction temperature is preferably 80 deg.C.

Influence of extraction time

Table 3 shows the results of a single-factor experiment of extraction time at an extraction temperature of 80 ℃ with a feed-to-liquid ratio of 1g to 30 mL: the extraction time is preferably 3 h.

And (4) conclusion: through single factor experiment and orthogonal experiment, the extraction rate of polyphenol components under different extraction process conditions is obtained, and the optimal process conditions are that the material-liquid ratio is 1g:30mL, the extraction temperature is 80 ℃, and the extraction time is 3 hours.

Acer tegmentosum polyphenol extract reduction test

The reduction capability is an important expression of potential antioxidant activity of the plant extract, and the strength of the reduction capability shows the strength of the antioxidant activity of the plant extract to a certain extent. The antioxidant (reducing agent) is a compound which can scavenge free radicals by giving electrons through its own reducing action, and the stronger the reducing ability is, the stronger the oxidation resistance is. The reduction capability of the antioxidant substance is utilized to reduce the ferricyanate into ferricyanate, and the ferricyanate and ferric iron generate ferric ferrocyanide under an acidic medium. Therefore, the more ferrocyanide is produced, the greater the absorbance is, and the stronger the reducing ability is.

Vitamin c (Vc) is used as a positive control, and the antioxidant capacity of the Acer tegmentosum polyphenol extract is compared with that of Vc. From fig. 1, it can be seen that the acer tegmentosum phenolic extract and Vc both have certain antioxidant capacity, and the antioxidant activity is enhanced with the increase of concentration. When the mass concentration of the sample is more than 0.4 mg/mL, the antioxidant capacity of the Acer tegmentosum polyphenol extract is more than Vc, and the difference of the antioxidant activity increases along with the increase of the concentration.
