1. A microorganism having a function of degrading ethanol and acetaldehyde, which is selected from the group consisting of Lactobacillus brevis LMT1-73 (accession No. KCTC-13412BP) and Lactobacillus fermentum LMT2-75 (accession No. KCTC-13413 BP).

2. A composition comprising a microorganism of claim 1 and a diluent or carrier.

3. The composition of claim 2, for use in removing at least one of ethanol and acetaldehyde from a sample.

4. The composition of claim 3, wherein the sample is an intestinal fluid or blood comprising gastric fluid, duodenal fluid, intestinal fluid or large intestine fluid.

5. The composition of claim 1, which is a food product.

6. The composition according to claim 5, wherein the food is a dairy product, a food for preventing alcoholic liver disease or alleviating hangover, or a food additive.

7. The composition of claim 6, wherein the dairy product is fermented milk, cream, cheese, or milk powder.

8. The composition of claim 2, in the form of a liquid, powder, granule, tablet, or capsule.

9. A kit for removing at least one of ethanol and acetaldehyde from a sample comprising the microorganism according to claim 1 and a diluent or carrier.

FIELD

The present disclosure relates to a microorganism having a degradation function on ethanol and acetaldehyde, a composition and a kit comprising the same.

Background

Lactobacillus (Lactobacillus) belongs to the group of gram-positive, facultative anaerobic (Facultative anaerobic) or microaerophilic (microaerophilic), rod-shaped, or spore-free bacteria. Lactobacillus accounts for a major part of the Lactic Acid Bacteria (LAB) population.

Alcohol ingested by humans is readily absorbed by diffusion in the stomach and upper part of the small intestine. Alcohol metabolism usually occurs in liver tissue, alcohol is oxidized to acetaldehyde by yeast dehydrogenase (ADH), and acetaldehyde is converted to ethanol by acetaldehyde dehydrogenase (ALDH).

Acetaldehyde is a metabolite produced by alcohol oxidation and is a toxic substance with high reactivity. Acetaldehyde is covalently bound to various proteins within the liver, thereby altering liver function and structure. By binding to tubulin, polymerization of microtubules is reduced, impairing protein secretion and causing swelling of stem cells (hepatocyte). The formation of acetaldehyde adducts (adducts) impairs a portion of the enzymatic activity. Acetaldehyde promotes lipid peroxidation (lipid peroxidation), either directly or by binding to Glutathione (GSH). Acetaldehyde alters various mitochondrial functions, especially after chronic ethanol consumption, which sensitizes mitochondria to the toxic effects of acetaldehyde. In cultured myofibroblasts, acetaldehyde stimulates collagen production. It has been reported that this is the cause of liver fibrosis in chronic alcohol ingestors. Also, chronic alcohol intake may cause fatty liver, alcoholic hepatitis, and cirrhosis of the liver, etc. The acetaldehyde-protein adduct stimulates the production of antibodies directed against acetaldehyde epitopes. Such immune responses may exacerbate or perpetuate alcohol-induced liver damage. Also, acetaldehyde causes hangover (hangover).

However, even according to the prior art, there is still a need for lactobacillus bacteria having activity against ethanol and acetaldehyde degradation.

Disclosure of Invention

Technical problem

In one aspect, there is provided a microorganism having a function of degrading ethanol and acetaldehyde, selected from the group consisting of Lactobacillus brevis (Lactobacillus brevis) LMT1-73 (accession No. KCTC-13412BP) and Lactobacillus fermentum (Lactobacillus fermentum) LMT2-75 (accession No. KCTC-13413 BP).

Another aspect provides a composition comprising said microorganism or a lysate (lysate) thereof.

A kit for removing at least one of ethanol and acetaldehyde from a sample comprising the microorganism and a diluent or carrier is provided.

Technical scheme

In one aspect, there is provided a microorganism having a function of degrading ethanol and acetaldehyde, selected from the group consisting of Lactobacillus brevis (Lactobacillus brevis) LMT1-73 (accession No. KCTC-13412BP) and Lactobacillus fermentum (Lactobacillus fermentum) LMT2-75 (accession No. KCTC-13413 BP).

The microorganism has excellent resistance to ethanol, degradation function of ethanol and/or acetaldehyde, acid resistance, bile acid resistance, and intestinal adhesion. The microorganism is isolated from kimchi.

Another aspect provides a composition comprising said microorganism or a lysate (lysate) thereof.

The composition may include a diluent or carrier acceptable in foodstuffs. The diluent may be a buffer such as water, medium or Phosphate Buffered Saline (PBS). The carrier may be a conventional excipient, disintegrant, binder, lubricant, thickener or filler. The diluent or carrier may be lactose, glucose, sucrose, sorbitol, mannitol, xylitol, erythritol, maltitol, starch, acacia, alginate, gelatin, calcium phosphate, calcium silicate, cellulose, methyl cellulose, amorphous cellulose, polyvinylpyrrolidone, water, parabens, hydroxybenzoate, talc, magnesium stearate, or a mineral fuel. Also, the composition may include a lubricant such as magnesium stearate, talc.

The composition can be used to remove at least one of ethanol and acetaldehyde from a sample. The term "removing" means reducing the concentration of at least one of ethanol and acetaldehyde in a sample, and also includes complete removal. The sample may be a body fluid. The sample may be an intestinal fluid or blood. The enteral fluid may be gastric fluid, duodenal fluid, small intestinal fluid or large intestinal fluid.

The composition may be in a form for oral administration. The composition can be granule, powder, liquid, tablet, capsule or dry slurry. The composition may be, for example, a culture product obtained by culturing the microorganism in a medium or a dried product thereof.

The composition may be a food product. The food may be a dairy product, a food for preventing alcoholic liver disease or relieving hangover, or a food additive. The dairy product may be fermented milk, cream, cheese, or milk powder. The food may be a health functional food. The health functional food may be a health functional food for preventing alcoholic liver disease or relieving hangover. The food can also be beverage, biscuit, diet biscuit, chocolate, pizza, stretched noodles, other noodles, chewing gum, and ice cream.

The food product may include ingredients that are typically added when the food product is manufactured, including, for example, proteins, carbohydrates, fats, nutrients, flavors and aromas.

Carbohydrates used in the preparation of food products may be monosaccharides such as glucose, fructose, etc.; disaccharides such as maltose, sucrose, oligosaccharides, and the like; and polysaccharides such as dextrin, cyclodextrin and other conventional sugars and sugar alcohols such as xylitol, sorbitol and erythritol. Also, natural flavors and synthetic flavors such as saccharin and aspartame can be used as the flavors. The natural flavoring agent may be stevioside extract, such as thaumatin, steviolbioside, glycyrrhizin, etc.

The health functional food is a food which has a specific health effect when ingested.

In the composition, the content of the microorganism may fall within a range of 0.01 wt% to 50 wt% or 0.1 wt% to 20 wt% based on the weight of the composition. Also, the composition can include 105 to 1x 109CFU/g or 1x 105 to 1x 108CFU/g of cells based on the weight of the composition.

Another aspect provides a method of removing at least one of ethanol and acetaldehyde from an individual, comprising administering to the individual a microorganism having a degrading function for ethanol and acetaldehyde selected from the group consisting of Lactobacillus brevis (Lactobacillus brevis) LMT1-73 (accession number KCTC-13412BP) and Lactobacillus fermentum (Lactobacillus fermentum) LMT2-75 (accession number KCTC-13413 BP). The method may be used for preventing or treating diseases associated with the in vivo accumulation of ethanol and/or acetaldehyde. The disease may be alcoholic liver disease or hangover. The subject may be a mammal. The mammal may be a human or another mammal other than a human.

On the other hand, a kit for removing at least one of ethanol and acetaldehyde from a sample containing the microorganism and a diluent or carrier is provided. The kit may be provided separately from the microorganism and diluent or carrier.

The microorganism, the composition including the microorganism, and the kit according to an aspect may be used for removing at least one of ethanol and acetaldehyde from a sample.

These specific embodiments may, therefore, be embodied in many different forms and should not be construed as limited to the description set forth herein. As used in this specification, the term "and/or" includes any and all combinations of at least one of the associated listed items.

Advantageous effects

The microorganism and the composition and the kit each including the microorganism according to an aspect may be suitable for removing at least one of ethanol and acetaldehyde from a sample.
