技术领域

本发明属于动物基因工程领域,具体地说,涉及一种乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体及应用。

背景技术

仔猪腹泻在养猪业危害中居首位,严重威胁着养猪业的健康发展,是集约化养猪生产条件下的一种典型的多因素性疾病。而哺乳仔猪以传染性腹泻较为常见,尤其以携带k88ac菌毛蛋白的大肠杆菌引起的仔猪腹泻最为常见。

大肠杆菌k88ac菌毛蛋白属于大肠杆菌纤毛抗原的一种,是蛋白质,不耐热,位于菌体表面,与菌株的肠病原性有着密切的关系。大肠杆菌k88ac是引起仔猪腹泻的主要病原,大肠杆菌k88ac菌毛蛋白则作为一种粘着因素在猪体内发挥作用,使大肠杆菌固着于前部肠道的粘膜绒毛上,产生肠毒素,引起常见的仔猪腹泻病。

然而,目前对于k88ac大肠杆菌并没有很好的单克隆抗体用以抵御k88ac大肠杆菌对仔猪危害,导致仔猪腹泻病仍然是威胁养猪业的一大原因。迄今为止,国内尚未见有特异性结合大肠杆菌k88ac,并可有效发挥免疫抗性的单链抗体的报道。因此,本发明人通过生物技术手段获得了抗k88ac大肠杆菌的单链抗体,同时具有显著的抑菌效果,提示可以通过转基因技术制备抗腹泻转基因猪、降低我国仔猪腹泻发病率的目的。

发明内容

本发明的目的是提供一种乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体及应用。

本发明构思如下:通过基因工程手段(通过构建乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体)将抗k88ac抗原抗体基因转入猪成纤维细胞内,并使其在乳腺特异表达,从而增加乳汁中抗k88ac抗原抗体的含量,针对乳猪达到降低仔猪腹泻发病率的目的。

为了实现本发明目的,第一方面,本发明提供乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体,所述同源重组载体是以真核表达载体为骨架载体,所述同源重组载体中至少包括正筛选元件、负筛选标记基因、抗k88ac抗原抗体基因表达盒以及rosa26基因的同源左臂和同源右臂。

优选地,所述同源重组载体是以载体BAC作为骨架载体。

其中,所述正筛选元件包括顺次连接的重组酶识别序列-正筛选标记基因-重组酶基因-重组酶识别序列。

所述正筛选元件的一端与所述同源左臂连接,另一端与所述抗k88ac抗原抗体基因表达盒连接;所述抗k88ac抗原抗体基因表达盒的另一端与所述同源右臂连接,同源右臂的外侧连接有负筛选标记基因。

优选地,所述正筛选标记基因包括但不限于编码新霉素磷酸转移酶(Neo)、潮霉素B磷酸转移酶、次黄嘌呤磷酸核糖转移酶或嘌呤霉素乙酰转移酶的基因。

优选地,所述负筛选标记基因包括但不限于编码白喉毒素或胸腺去氧核苷的基因(dTA)。

本发明所述重组酶基因包括编码Cre酶或Flp酶的基因。

优选地,所述同源重组载体中的正筛选元件为Loxp-OCT4启动子 -Cre-PGK-Neo-Loxp。其中,Cre表达框由胚胎期特异性启动子OCT4调控。所述新霉素Neo作为正筛选标记,dTA为负筛选标记。LoxP位点位于Cre表达框上游和Neo表达框下游。

优选地,所述抗k88ac抗原抗体基因表达盒为人乳铁蛋白hLF启动子-抗k88ac抗原抗体基因-hLF终止子。

更优选地,所述同源重组载体包括如下顺次连接的元件:同源左臂-Loxp-OCT4 启动子-Cre-PGK-Neo-Loxp-人乳铁蛋白hLF启动子-抗k88ac抗原抗体基因-hLF终止子 -同源右臂-PGK-dTA。

其中,所述同源左臂为来自于猪rosa26基因的同源左臂,优选来自于猪rosa26基因内含子1的一段至少1.1kb序列,核苷酸序列如SEQ ID NO:1所示。所述同源右臂为来自于猪rosa26基因的同源右臂,优选来自于猪rosa26基因内含子1的一段至少6.0kb 序列,核苷酸序列如SEQ ID NO:2所示。

所述抗k88ac抗原抗体基因的核苷酸序列如SEQ ID NO:3所示。

具体实施方式

中,所述乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体为pHS-AVC-YW079 HLF-K88(图1),其中,O代表猪早期胚胎发育转录因子OCT4的特异性启动子;C代表编码重组酶Cre基因;N代表真核细胞常用的药物筛选标记基因Neo;k88代表抗k88ac抗原抗体基因;R代表***位点rosa26基因。

所述同源重组载体pHS-AVC-YW079 HLF-K88的核苷酸序列全长如SEQ ID NO:4所示。

第二方面,本发明提供乳腺特异性表达抗k88ac抗原抗体基因的细胞系,利用上述同源重组载体转染哺乳动物体细胞,获得的中靶阳性细胞克隆,即为乳腺特异性表达抗k88ac抗原抗体基因的细胞系。

第三方面,本发明提供所述同源重组载体或所述细胞系在制备乳腺特异性表达抗k88ac抗原抗体转基因动物中的应用,其中,所述动物包括猪。

前述的应用,将所述同源重组载体转染到与其同源臂DNA来源相同的猪细胞系中。

借由上述技术方案,本发明至少具有下列优点及有益效果:

本发明首次构建得到乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体,并成功获得了乳腺特异性表达抗k88ac抗原抗体的转基因猪细胞系,为通过转基因技术制备抗腹泻转基因猪奠定基础。

具体实施方式

以下实施例用于说明本发明,但不用来限制本发明的范围。若未特别指明,实施例均按照常规实验条件,如Sambrook等分子克隆实验手册(Sambrook J&Russell DW,Molecular Cloning:a Laboratory Manual,2001),或按照制造厂商说明书建议的条件。

实施例1乳腺特异性表达抗k88ac抗原抗体基因的同源重组载体的构建

1、构建中间载体pHS-AVC-YW056

1)从本实验室前期构建的pBC1-hLZ-GFP-Neo(Jia et al.,2011)扩增同源左臂Left arm,扩增引物如下:

吹打混匀体系,并放入金属浴中反应,50℃保持恒温反应60min即可进行后续的转化实验。

2、构建中间载体pHS-AVC-YW059

1)从本实验室前期构建的pBC1-hLZ-GFP-Neo(Jia et al.,2011)载体为模板扩增OCT4-Cre-PGK-Neo(分两段扩增)

3)重组反应的建立

将PCR片段与载体酶切片段按照Easyclone拼装试剂进行组装反应。吹打混匀体系,并放入金属浴中反应,50℃保持恒温反应60min即可进行后续的转化实验。

3、构建中间载体pHS-AVC-YW063

1)从本实验室前期构建的pBC1-hLZ-GFP-Neo(Jia et al.,2011)载体为模板扩增Loxp-HLF(分两段扩增)

3)重组反应的建立

将PCR片段与载体酶切片段按照Easyclone拼装试剂进行组装反应。吹打混匀体系,并放入金属浴中反应,50℃保持恒温反应60min即可进行后续的转化实验。

4、构建中间载体pHS-AVC-YW068

将pHS-AVC-YW063,pENTR-L1-k88ac-L2(基因合成),

Seq3-ccdB-Seq4(骨架载体)分别稀释成20nM(10ul体系),各取0.5ul加入0.5ulLR 酶及0.5ul ddH2O,室温反应3h即可。

5、构建中间载体pHS-AVC-YW058

1)从本实验室前期提供的pBC1-hLZ-GFP-Neo(Jia et al.,2011)载体为模板扩增同源右臂Right arm,扩增引物如下:

3)重组反应的建立

将PCR片段与载体酶切片段按照Easyclone拼装试剂进行组装反应。吹打混匀体系,并放入金属浴中反应,50℃保持恒温反应60min即可进行后续的转化实验。

6、构建中间载体pHS-AVC-YW055

1)将pENTR-L4-mPGK-R(pHS-AVC-YW053)、 pENTR-L1-dTA-L2(pHS-AVC-YW054)、Seq5-GTW-Seq6(骨架载体)分别稀释成 20nM(10ul体系)。

2)按照下表的反应体系进行LR反应,室温反应3h。

3)反应完成后,进行转化实验。

7、多个元件载体的拼装

拼装同源左臂-Loxp-OCT4启动子-Cre-PGK-Neo-Loxp-人乳铁蛋白hLF启动子-抗k88ac抗原抗体基因-hLF终止子-同源右臂-PGK-dTA,即同源重组载体 pHS-AVC-YW079HLF-K88。

构建方法如下:

1)线性化片段的制备

①将pHS-AVC-YW056/059/068/058/055每个载体计算42fmol的量,放在同一离心管中使用I-SceI酶切3小时(10ul体系),然后80℃热失活20min。

②将pZ306载体(42fmol)使用PacI酶切3小时(10ul体系),然后80℃热失活20min。

③将pKan-6-X载体(42fmol)使用XbaI/XhoI酶切3小时(10ul体系),然后80℃热失活20min。

2)重组反应体系的建立

①从-20℃冰箱中取出1.33×gibson assembly组装混合液置于冰上保存,如有结冰情况,请置于冰上融化。

②取出15μl 1.33×gibson assembly放入PCR管中,按照等摩尔的比例依次加入含有连接片段的溶液,总体积加至20μl,如果加入所有的片段溶液后,总体积不足20μl,用ddH2O补齐至20μl。按下表在0.2ml离心管中加入各试剂:

③吹打混匀体系,50℃保持恒温反应60min。

④反应完成后,如不能马上进行转化,应置于-20℃保存。

⑤测序验证其正确后,扩大培养,去内毒素大提。即得同源重组载体 pHS-AVC-YW079 HLF-K88(图1,SEQ ID NO:4)。

实施例2乳腺特异性表达抗k88ac抗原抗体转基因细胞系的制备

选用性别为母的50胎龄的大白猪胎儿成纤维细胞系,细胞复苏、培养和传代参照《精编细胞生物学实验指南》(J.S.博尼费斯农等著,章静波等译,2007年,科学出版社)。待细胞生长汇合至80%左右,消化收集细胞(约1×106个),加入实施例1 构建的载体pHS-AVC-YW079 HLF-K88 2μg和100μL Nucleofector试剂混匀,加入电击杯中,用A-024程序进行电击转染。然后按1∶20接种至10cm培养皿中,37.5℃,5%CO2培养箱中培养。24h后更换含500μg/mL G418的完全培养基(10%FBS+DMEM),每3~4 天换液一次,72h后G418浓度增加到800μg/mL。培养至第8天可见克隆点形成。镜下标记克隆点位置,用细胞克隆环将其消化,接种到24孔板。待汇合度达90%,消化细胞,一部分留在原孔继续培养用于鉴定其整合位点,另一部分接种到6孔板用于细胞冻存。

阳性细胞单克隆鉴定:

对获得的360个细胞克隆进行鉴定。短臂引物SF: 5’-TGGGTGCTTGAGGTGGTCT-3’,短臂引物SR: 5’-CAAGCATAAAAACGCACCAATTGTC-3’,PCR扩增产物大小为1300bp(图2A)。长臂引物LF:5’-GGAAGTGAGCAGGTAGAGGGTC-3’,长臂引物LR: 5’-ATCCAGTGTTGCCGTGAGC-3’,PCR扩增产物大小为7975bp(图2B)。消化12孔板单克隆细胞,提取基因组,以野生型猪成纤维细胞基因组为阴性对照,用KOD聚合酶对长、短臂进行扩增,0.8%琼脂糖电泳检测。结果如图2所示。其中阳性克隆为 7个,状态良好,适宜做核移植供体细胞。即,本发明中k88ac基因定点整合效率约为 2.5%。

虽然,上文中已经用一般性说明及具体实施方案对本发明作了详尽的描述,但在本发明基础上,可以对之做一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
