5. The automatic nucleic acid extracting apparatus according to claim 1, wherein the bottom of the reaction chamber is provided with a control switch and a silica gel membrane positioned below the control switch.

The full-automatic nucleic acid extraction device according to claim 1, wherein the reaction chamber comprises a first reaction chamber and a second reaction chamber; the first reaction chamber is connected with the second reaction chamber through a channel; the first reaction chamber and the second reaction chamber are positioned below the storage chamber and are communicated with the storage chamber; the first reaction chamber and the second reaction chamber are both connected with the waste liquid chamber through a channel; the second reaction chamber is connected with the nucleic acid chamber through a channel.

7. The apparatus for extracting nucleic acid according to claim 6, wherein a filter membrane is disposed at the bottom of the first reaction chamber; and a silica gel film is arranged at the bottom of the second reaction chamber.

8. The fully automatic nucleic acid extraction apparatus according to claim 1, further comprising a heating device; the heating devices are positioned at two sides of the reagent card box; the heating device is connected with the circuit control system.

9. The automatic nucleic acid extracting apparatus according to claim 1, further comprising an upper cover; the upper cover is provided with a liquid pushing assembly; the liquid pushing assembly corresponds to the position of the storage chamber.

The invention belongs to the field of nucleic acid extraction, and particularly relates to a full-automatic nucleic acid extraction device.

Technical Field

Nucleic acid is a biological macromolecular compound formed by polymerizing many nucleotides, and the nucleic acid is often combined with other components (such as protein) in the organism to play a life role. At present, nucleic acid-based molecular diagnostic and detection techniques play an increasingly important role in many fields, and in the biological world, nucleic acids are used for detection of pathogenic microorganisms, species identification, species origin, diversity assessment, and research on genetic relationship, system evolution, genetic engineering operations, and the like. These studies require the isolation and purification of nucleic acids prior to detection.

At present, the existing nucleic acid extraction methods comprise a manual nucleic acid extraction method and an automatic nucleic acid extraction method, the manual nucleic acid extraction method has high labor intensity, low efficiency and long time consumption, and only one sample can be extracted each time; the automatic nucleic acid extraction method uses an automatic nucleic acid extraction device, and the existing automatic nucleic acid extraction device is fixed in form, so that only one sample can be extracted, or a fixed number of samples must be extracted at the same time, otherwise, consumable materials and reagents are wasted. Chinese patent application No. CN201410070377.4, full-automatic nucleic acid extraction instrument, discloses a full-automatic nucleic acid extraction instrument, which comprises a working platform, and a sample adding system, a deep-hole plate moving device and a nucleic acid extraction device which are arranged on the working platform. This full-automatic nucleic acid extractor draws the sample according to the number in the hole of deep-hole board mobile device, if the number of drawing the sample does not reach the number in hole, has partial hole by vacant, but still has consumptive material and reagent to be used to cause the waste of consumptive material and reagent.

Disclosure of Invention

In order to solve the technical problems, the invention provides a full-automatic nucleic acid extraction device, when the full-automatic nucleic acid extraction device is used for extracting nucleic acid, only manual sample adding is needed, other manual operations are not needed, the labor intensity is reduced, the full-automatic integrated extraction of nucleic acid is realized, errors caused by manual operations are avoided, the extraction result is stable, the efficiency is high, and the purity is good; in the nucleic acid extraction process, a reagent does not need to be added manually in each step of operation, so that the extraction time is saved; the number of samples extracted each time is variable, 1 sample can be extracted simultaneously, 100 samples can be extracted simultaneously, more samples can be extracted, and the used consumables and reagents can be determined according to the number of the extracted samples, so that the wastes of the consumables and the reagents cannot be caused.

Preferably, the upper part of the nucleic acid chamber is also provided with a buckle which detachably connects the nucleic acid chamber with the channel.

Preferably, the full-automatic nucleic acid extraction device further comprises a heating device; the heating devices are positioned at two sides of the reagent card box; the heating device is connected with the circuit control system.

Preferably, the full-automatic nucleic acid extraction device further comprises an upper cover; the upper cover is also provided with a liquid pushing assembly; the liquid pushing assembly corresponds to the position of the storage chamber. More preferably, the liquid pushing assembly is located above the storage chamber, corresponding to the position of the reagent tube.

Preferably, the reagent driving system comprises an air pump and a pipeline system connected with the air pump.

Preferably, the air pump is a positive pressure pump and/or a negative pressure pump.

Preferably, the pipeline system comprises a plurality of pipelines and a pipeline control switch for controlling whether the pipelines are unobstructed or not; and the pipeline control switch is connected with the circuit control system.

The invention has the beneficial effects that:

1. in the nucleic acid extraction process, only need artifical application of sample, then starting device need not artifical reagent of adding and shift, filter reaction liquid, has avoided the error that manual operation arouses, has not only reduced intensity of labour, and extraction result is stable moreover, efficient, purity is good, and the extraction time is short.

2. A reagent card box for reacting can draw a plurality of samples simultaneously for dismantling reagent card box, draws at every turn, and the figure of drawing the sample at every turn can change moreover, only need install the reagent card box of corresponding figure can, easy operation is swift, can not cause the waste of consumptive material and reagent yet.

The technical solutions of the embodiments of the present invention will be described clearly and completely with reference to the accompanying drawings in the embodiments of the present invention, and it should be apparent that the drawings described below are only some embodiments of the present invention, but not all embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.

As shown in FIGS. 1-4, the present invention provides a fully automatic nucleic acid extracting apparatus, comprising at least one card slot, at least one reagent cartridge, a reagent driving system, and a circuit control system connected to the reagent driving system; the reagent card box is a detachable reagent card box and is arranged in the clamping groove; the reagent card box comprises a storage chamber, a reaction chamber, a waste liquid chamber and a nucleic acid chamber; the storage chamber is connected with the reaction chamber; the reaction chamber is respectively connected with the waste liquid chamber and the nucleic acid chamber through a channel. Wherein, a control switch can be arranged in the channel and is connected with the circuit control system.

The reagent cartridges are used for nucleic acid extraction reactions, one reagent cartridge to extract one sample. In order to facilitate the addition of the sample, the reaction chamber is provided with a sample inlet and a sample inlet cover, the sample can be added into the reaction chamber through the sample inlet, and the storage chamber does not need to be opened.

In order to make the reaction liquid fully mixed and react more completely, a stirring device is arranged in the reaction chamber.

As an alternative embodiment, the bottom of the reaction chamber is provided with a control switch and a silica gel membrane positioned below the control switch. Wherein, the control switch is connected with the circuit control system.

In the reaction process, the control switch is in a closed state; after the reaction is finished, the control switch is in an open state, and the liquid in the reaction chamber passes through the silica gel membrane. The silica gel membrane is used for specifically combining nucleic acid, and can improve the purification quality of nucleic acid.

As an alternative embodiment, the reaction chamber comprises a first reaction chamber and a second reaction chamber; the first reaction chamber is connected with the second reaction chamber through a channel; the first reaction chamber and the second reaction chamber are positioned below the storage chamber and are communicated with the storage chamber; the first reaction chamber and the second reaction chamber are both connected with the waste liquid chamber through a channel; the second reaction chamber is connected to the nucleic acid chamber via a channel.

Wherein, a control switch can be arranged in the channel and is connected with the circuit control system.

Wherein, the bottom of the first reaction chamber is provided with a filter membrane; the bottom of the second reaction chamber is provided with a silica gel film.

The filter membrane can be used for collecting bacterial thalli and precipitating protein and genome compound; the silica gel membrane is used for specifically binding nucleic acid; the arrangement of the filter membrane or the silica gel membrane can improve the purification quality of nucleic acid.

The upper part of the nucleic acid chamber is also provided with a buckle which connects the nucleic acid chamber with the channel in a detachable way. The purified nucleic acid enters the nucleic acid chamber, which can be removed for direct use in subsequent operations.

The full-automatic nucleic acid extraction device also comprises a heating device; the heating devices are positioned at two sides of the reagent card box; the heating device is connected with the circuit control system.

The full-automatic nucleic acid extraction device also comprises an upper cover; the upper cover is also provided with a liquid pushing assembly; the liquid pushing component corresponds to the position of the storage chamber. The liquid pushing assembly is positioned above the storage chamber and corresponds to the position of the reagent tube.

The reagent driving system comprises an air pump and a pipeline system. The air pump is a positive pressure pump and/or a negative pressure pump. The pipeline system comprises a plurality of pipelines and a pipeline control switch for controlling whether the pipelines are unobstructed or not; the pipeline control switch is connected with the circuit control system.

The air pump causes a pressure difference to exist between the reaction chamber and the waste liquid chamber or the nucleic acid chamber (or between the first reaction chamber and the second reaction chamber) in the reagent cartridge through the piping system, and the pressure difference causes the liquid in the reaction chamber to enter the waste liquid chamber or the nucleic acid chamber (or causes the liquid in the first reaction chamber to enter the second reaction chamber).

The reagent driving system effectively reduces the residual liquid amount in the reaction chamber after transferring the liquid, thereby further improving the purification quality of the nucleic acid.
