Technical Field

The present invention relates to quaternary ammonium-type antimicrobial compositions. More particularly, the present invention relates to compositions having mycobacterial and tuberculocidal (tuberculocidal) efficacy. The composition comprises a biocidal quaternary ammonium compound in combination with a water-insoluble bis-substituted alkylamide solvent.

Background

Quaternary ammonium compounds have been used as disinfectants for many years and these compounds have a broad spectrum of antimicrobial activity. While formulations comprising higher concentrations of quaternary ammonium compounds are known to be effective against certain gram-positive and gram-negative bacteria, these formulations are typically less effective against tuberculosis-causing bacteria.

U.S. patent No.5,444,094 discloses an effective tuberculocide composition comprising a quaternary ammonium salt and a minimum concentration of at least 8% by weight of a glycol ether. In particular, the composition has a weight ratio of glycol ether to quaternary ammonium salt of at least 4:1, and preferably 40: 1. Although the composition has tuberculocidal efficacy, glycol ethers are Volatile Organic Compounds (VOCs) which are undesirable from an environmental standpoint. Moreover, stricter regulations restricting VOCs are continually being proposed, which makes it important to restrict the use of glycol ethers.

U.S. patent No.5,908,854 discloses an aqueous disinfecting composition effective against mycobacteria wherein the composition comprises a germicidal cationic quaternary ammonium compound and a phenoxy alcohol or glycol ether solvent.

PCT published application WO 2015/078496 discloses disinfecting detergent compositions as follows: it is said to have tuberculocidal and mycobacterial killing efficacy and/or efficacy against norovirus, adenovirus and polyoma virus. The compositions include a combination of a specific quaternary ammonium chloride (i.e., N-alkyldimethylbenzylammonium chloride) and a bis (3-aminopropyl) alkylamine in a specific weight ratio.

U.S. published application No.2006/0094638 discloses a composition having tuberculocidal efficacy, wherein the composition comprises a quaternary ammonium compound, a guanidine derivative, and an alkyl amine.

U.S. patent No.6,245,361 discloses that compositions comprising a combination of quaternary ammonium compounds and chlorine bleach can be effective disinfectants for mycobacteria.

There remains a need for improved antimicrobial compositions as follows: it has tuberculocidal and mycobacterial killing efficacy, and has low toxicity, low VOC and low impact (impact) on the environment.

Xcomprises the following steps: cl-、Br-、F-、I-CH2COO-

Exemplary quaternary ammonium compounds within the formula include alkyltrimethylammonium halides, dialkyldimethylammonium halides, alkyldimethylbenzylammonium halides, dialkylmethylbenzylammonium halides, alkyldimethylethylbenzylammonium halides, and dialkylmethylethylbenzylammonium halides. The quaternary ammonium salt comprises didecyl dimethyl ammonium chloride, dioctyl dimethyl ammonium chloride, octyl decyl dimethyl ammonium chloride, (C)12-C18) Alkyl dimethyl benzyl ammonium chloride, and (C)12-C18) Alkyl dimethyl ethyl benzyl ammonium chloride. The quaternary ammonium compound need not be a single entity (entity), but can be a blend of two or more quaternary ammonium compounds.

The disinfecting compositions of the present technology in the form of concentrates may comprise from about 0.05% to about 50% by weight, alternatively from about 1% to about 25% by weight, alternatively from about 8% to about 16% by weight, of the quaternary ammonium compound, based on the total weight of the composition. In some embodiments, the amount of the quaternary ammonium compound is 16% by weight of the total concentrate composition. Disinfecting compositions of the present technology in the form of ready-to-use (or diluted) compositions may include from about 100ppm to about 10,000ppm, alternatively from about 200ppm, about 300ppm, about 400ppm, about 500ppm, about 600ppm, alternatively from about 700ppm to about 5000ppm, about 6000ppm, about 7000ppm, or about 8000ppm of the quaternary ammonium compound, based on the total weight of the composition. In some embodiments, the quaternary ammonium compound is present in an amount of 5000ppm by weight of the ready-to-use disinfectant.

The water-insoluble bis-substituted alkylamides have the following general structure:

Figure BDA0002229545130000031

or

Figure BDA0002229545130000032

Wherein R is a linear or branched, saturated or unsaturated alkyl or alkene group having 4 to 16, preferably 8 to 10, carbon atoms, n ═ 2 or 3; and

r1 and R2 are independently alkyl groups having 1 to 4 carbon atoms. In one embodiment, the water-insoluble solvent is an N, N-dimethyl alkylamide having 8 to 10 carbon atoms in the alkyl group.

When the disinfecting compositions of the present technology are in the form of concentrates, the compositions may comprise from about 0.05% to about 50% by weight, alternatively from about 1%, about 2%, about 3%, about 4%, about 5%, about 6%, about 7%, or about 8% by weight to about 16%, about 17%, about 18%, about 19%, about 20%, about 25%, or about 29% by weight of the N, N-bis-substituted alkylamides, based on the weight of the composition. When the disinfecting composition is a ready-to-use (or diluted) composition, the composition may include from about 100ppm to about 10,000ppm, alternatively from about 200ppm, about 300ppm, about 400ppm, about 500ppm, about 600ppm, or about 700ppm to about 5000ppm, about 6000ppm, about 7000ppm, or about 8000ppm of the N, N-bis-substituted alkylamide, based on the total weight of the composition.

It has been unexpectedly found that the combination of one or more quaternary ammonium compounds and one or more N, N-bis-substituted alkylamides provides effective disinfecting agents for a variety of bacteria including the tuberculogenic bacteria pseudomonas aeruginosa (Pa), staphylococcus aureus (Sa) and salmonella enterica (Se). The disinfecting compositions of the present technology are also effective against other microorganisms including viruses, molds and fungi. In addition, the compositions of the present technology are efficacious without the need for any additional antimicrobial active ingredient (active). Without being limited by theory, it is believed that the combination of one or more quaternary ammonium compounds and one or more N, N-bis-substituted alkylamides forms a synergistic mixture that provides efficacy against a wide variety of bacteria, including tuberculosis-causing bacteria. Thus, the use of glycol ethers or alcohol solvents, chlorine bleach and guanidine derivatives can be avoided in the disinfecting compositions of the present technology.

The disinfecting compositions of the present technology are in liquid form and include a carrier in addition to the quaternary ammonium compound and the N, N-bis-substituted alkylamide. Water (deionized water or ordinary tap water) is a particularly suitable carrier for ready-to-use formulations. Water may be used as the carrier, either alone or in combination with other suitable carriers. In some embodiments, the disinfecting composition is a liquid concentrate formulation that does not include added water. In such concentrate formulations, as an environmentally friendly filler/carrier, glycerol may be used, which unexpectedly also provides phase stability to the concentrated formulation. These concentrate formulations can be diluted with water at a dilution ratio of, for example, 1:32 and still unexpectedly have efficacy against tuberculosis-causing bacteria as well as a wide variety of other biocidal targets (targets) such as other types of bacteria, viruses, fungi and yeasts.

Depending on the end use, one or more optional components may be included in the disinfecting composition of the present invention. For example, one or more surfactants may be added to the composition in order to improve cleaning, detergency and/or bactericidal efficacy. Surfactants are particularly useful in one-step sanitizing formulations because a pre-cleaning step to first remove soils and stains is not required. Surfactants also help to wet the surface and thus enable the combination of quaternary ammonium compounds and N, N-bis-substituted alkylamides to kill microorganisms more effectively. In the disinfecting compositions of the present invention, the surfactant also helps solubilize the water-insoluble N, N-bis-substituted alkylamide solvent and optionally the water-insoluble fragrance. The surfactant may be a nonionic or amphoteric/zwitterionic surfactant or a combination thereof. Useful nonionic surfactants useful in the disinfecting compositionsExamples of surfactants include fatty alcohols having from 5 to 18, preferably from 6 to 12, carbon atoms in the alkyl chain, further ethoxylated with from 5 to 15, preferably from 6 to 9, ethylene oxide units. Such nonionic surfactants may also include some propylene oxide units, provided that there are sufficient ethylene oxide units to maintain the water solubility of the surfactant. Although less preferred, ethoxylated alkylphenols may also be used as surfactants. Particularly useful nonionic surfactants include, for example, Bio-

Figure BDA0002229545130000051

N1-9 (ethoxylated C11 alkyl 9EO alcohol). Examples of useful amphoteric/zwitterionic surfactants include alkyl amine oxides, betaines, and sarcosinates. The surfactant is included in the composition in an amount such that the weight ratio of the quaternary ammonium compound and surfactant combined to the di-substituted alkylamide is from 1:10 to 10:1, preferably from 1:3 to 3: 1.

Other components or additives may also be included in the disinfecting composition. Additional components may include pH adjusting agents, water soluble or other solubilizing agents for obtaining and maintaining a clear, single phase concentrated or ready-to-use composition, electrolytes for enhancing the detergency of surfactants, chelating agents for enhancing the surfactant detergency and cationic surfactant effectiveness, perfumes for different appealing odors, dyes for desirable color, and other functional ingredients.

In some embodiments, the final pH of an industrial product is more easily controlled within specification by including one or more water-soluble primary amines in the formulation as organic basic pH buffers. Suitable primary amine buffers include 2-aminoethanol and 2-amino-2-methyl-1-propanol. When the ready-to-use formulation is made with water, any pH buffering agent having a pKa of greater than 8.5 (preferably greater than 9.0) can be used in place of the water-soluble organic primary amine. The weight ratio of buffer to quaternary ammonium compound is in the range of 3:1 to 1:3, preferably 2:1 to 1: 2.

The compositions according to the present technology may be supplied in a variety of forms depending on the desired end use. In some embodiments, the composition is in a ready-to-use (RTU) form that can be used without dilution. In accordance with other embodiments of the present technology, the composition comprising the quaternary ammonium compound and the N, N-bis-substituted alkylamide is a dilutable concentrate product. Dilutable concentrate products are products that require dilution with a diluent (such as water) in a ratio such as, but not limited to, 1:256, 1:128, 1:64, or 1:32 before they can be applied to an article or surface to be disinfected. Suitable dilution ratios for the compositions of the invention in the form of concentrates are in the range of 1:1 to 1:256, alternatively 1:5 to 1:128, alternatively 1:8 to 1: 64. The concentration of the active ingredient in the dilutable concentrate product may vary depending on the desired dilution ratio.

In some embodiments of the present application, it is contemplated that the disinfecting composition is used as a spray. The disinfecting composition may be used as a spray in an RTU formulation, or the concentrate formulation may be used as a spray using a dilution of the disinfecting composition, e.g., 1:8, 1:10, 1:32, or 1: 64. In some embodiments, the composition may be used in a wipe. In other embodiments, the composition may be used, for example, in a wet wipe for use with an applicator pad. In some embodiments, it is contemplated that the compositions of the present application are concentrates that can be packaged, for example, in a bag or pod, that can be added to water at an appropriate dilution ratio.

In some embodiments, the present application provides methods of cleaning a hard surface by: the disinfecting composition of the present technology is provided in its end use concentration and at least one contaminated surface or substrate is contacted with the composition for a set period of time and then the composition or contaminant (soil) is removed from the surface or substrate.

Further, in other embodiments, the present application provides methods of disinfecting hard surfaces by: the disinfecting composition of the present technology is provided in its end-use concentration, and at least one surface to be disinfected is contacted with the composition. In some embodiments, the surface to be disinfected is contacted for a specified period of time, e.g., about 5 minutes or less, or about 3 minutes or less, and the composition is then removed from the surface.

It is contemplated that the method of contacting a surface with the disinfecting composition to clean and/or disinfect a hard surface is used by, for example, but not limited to, a mop, sponge, wipe, towel, wet wipe, automatic floor cleaning device, manual floor cleaning device, wet pad applicator, and the like.

The presently described technology and its advantages will be better understood by reference to the following examples. These examples are provided for the purpose of describing particular embodiments of the present technology. It is not intended to limit the scope or spirit of the present technology by providing these specific embodiments. It will be appreciated by those skilled in the art that the full scope of the presently described technology encompasses the subject matter defined by the claims appended hereto as well as any variations, modifications or equivalents of those claims.
