Potentilla chinensis is dry whole plant of Potentilla chinensis of Rosaceae, is bitter and cold in taste, and enters liver and large intestine channels. Has effects of clearing away heat and toxic materials, cooling blood, stopping bleeding, relieving swelling, removing ulcer, and improving gastrointestinal health, and can be used for treating dysentery with bloody stool, abdominal pain, chronic dysentery, hemorrhoid hemorrhage, carbuncle, swelling, and sore.

A preparation method of the composition with the functions of reducing blood fat and improving gastrointestinal tract is characterized by comprising the following steps: comprises the following steps

(1) The corn protein powder is sequentially subjected to size mixing, enzymolysis, centrifugal filtration, reduced pressure concentration and spray drying to obtain the required corn oligopeptide, wherein the oligopeptide component with the molecular weight of less than 1000D in the corn oligopeptide accounts for more than 95 percent;

(2) respectively extracting flos Celosiae Cristatae and herba Potentillae chinensis with ethanol under reflux, concentrating under reduced pressure, vacuum freeze drying, and pulverizing to obtain flos Celosiae Cristatae and herba Potentillae chinensis extract;

(5) vacuum freeze drying and low-temperature superfine grinding are carried out on hawthorn fruits to obtain hawthorn fruit powder, and 100 percent of the hawthorn fruit powder can pass through a 300-mesh sieve;

(a) Size mixing: sieving corn protein powder with a 40-80-mesh sieve, preparing a corn protein solution by using purified water, and uniformly mixing the corn protein solution; adding purified water into the uniformly mixed zein solution to obtain a dispersion liquid; adjusting the pH of the dispersion liquid of the corn protein powder to 6-8;

(b) enzymolysis: heating the dispersion liquid of the corn protein powder to 50 ℃, keeping the temperature at 50 +/-5 ℃, adding neutral protease and papain into the dispersion liquid of the corn protein powder, uniformly stirring and performing enzymolysis for 1-3 hours, wherein the addition amount of the neutral protease is 0.3-0.5% of the content of the substrate, the addition amount of the papain is 0.1-0.4% of the content of the substrate, and performing enzyme deactivation by using a high-temperature instant enzyme deactivation method after the enzymolysis is finished;

(c) centrifugal filtration: carrying out centrifugal separation on the enzyme-inactivated enzymolysis liquid by a centrifugal machine at 13000-16000 rpm, removing impurities, collecting clear liquid, starting membrane filtration equipment, filtering the clear liquid by a microporous filter membrane, wherein the thickness of the filter membrane is 60-120 mu m, the aperture of the filter membrane is 3-6 mu m, the operating pressure is 0.01-0.25 MPa, ensuring that the filtrate is clear and transparent, and removing filter residues; passing the filtered filtrate through an ion exchange column and an active carbon column, performing desalination and decolorization treatment, and removing free amino acid by nanofiltration;

(d) and (3) concentrating under reduced pressure: carrying out vacuum reduced pressure concentration on the solution after ion exchange, wherein the concentration temperature is 60-80 ℃, and the vacuum degree is-0.045-0.085 Mpa, so as to obtain an oligopeptide concentrated solution, and the relative density is controlled to be 1.05-1.09;

(e) spray drying: and (3) carrying out spray drying on the oligopeptide concentrated solution, wherein the process parameters are that the nozzle temperature is 165-185 ℃, the outlet temperature is 80-92 ℃, and the feeding speed is 1.8-2.8L/min, so that the corn oligopeptide meeting the quality requirement is obtained.

Preferably, the cockscomb extract and potentilla chinensis extract in the step (2) can be prepared by the following steps

(a) Alcohol reflux extraction: taking the crushed cockscomb or potentilla chinensis, adding 50-70% ethanol (v/v) which is 6-8 times of the weight of the materials, carrying out reflux extraction for 2-4 times at 80-92 ℃ for 1 hour each time, filtering and combining the extracting solution;

(b) and (3) concentrating under reduced pressure: concentrating the extracting solution under reduced pressure, and recovering the solvent, wherein the concentration temperature is controlled to be 45-65 ℃, the vacuum degree is controlled to be-0.06-0.09 Mpa, and the relative density is controlled to be 1.12-1.20, so as to obtain an extract;

(c) vacuum freeze drying: putting the extract into vacuum freeze drying equipment for drying, and setting parameters as follows: the pre-freezing initial temperature is-35 to-45 ℃, the pre-freezing speed is 0.2 to 0.5 ℃/min, the pre-freezing end point temperature is-65 ℃, the pressure of a drying chamber is 40 to 90pa, the temperature of a heating plate is 35 to 42 ℃, and the drying time is 16 to 24 hours;

(d) crushing: pulverizing the dried extract, and sieving with 100 mesh sieve.

Preferably, the preparation method of the purslane microcapsules in the step (3) is as follows

(a) Pulverizing herba Portulacae into coarse powder, adding CO2Extracting by supercritical extraction at 32-42 deg.C under 28-35 Mpa of CO2The flow rate is 8-14 kg/h, and the extraction time is 120-180 min, so that a purslane extract is obtained;

(b) weighing maltodextrin and gamma-cyclodextrin, stirring and dissolving the maltodextrin and the gamma-cyclodextrin in hot water at 85 ℃ to obtain a wall material solution, wherein the total mass fraction of the maltodextrin and the gamma-cyclodextrin in the wall material solution is 5-10%, and the mass ratio of the maltodextrin to the gamma-cyclodextrin is 1: 1-3;

(c) adding the purslane extract into a 10% ethanol solution containing sucrose ester and phospholipid, wherein the mass ratio of the sucrose ester to the phospholipid is 1: 1-4, and the total mass fraction of the sucrose ester and the phospholipid in the ethanol solution is 1-3% to prepare a purslane emulsion with the mass fraction of 6-8%;

(d) pouring the purslane emulsion prepared in the step (c) into the wall material solution prepared in the step (b), wherein the volume ratio of the purslane emulsion to the wall material solution is 0.8-1: 1, and carrying out emulsification and homogenization treatment at 50 ℃ to prepare an initial product;

(e) and (3) carrying out spray drying on the prepared initial product, wherein the inlet temperature of the spray drying is 150-175 ℃, the outlet temperature of the spray drying is 80-90 ℃, and the feeding speed is 0.5-1.5L/min, so as to prepare the required purslane microcapsule finished product.

Preferably, the plantain seed husk dietary fiber and chia seed dietary fiber in the step (4) can be prepared by the following preparation method

(a) And (3) low-temperature superfine grinding: putting Plantago ovata seed husk or chia seed into low-temperature superfine grinding equipment for superfine grinding, and sieving with a 500-mesh sieve to obtain Plantago ovata seed husk superfine powder or chia seed superfine powder;

(b) enzymolysis: putting Plantago ovata husk superfine powder or chia seed superfine powder into purified water of 8-10 times, uniformly stirring to prepare a dispersion, heating to raise the temperature and keep the temperature at 50 +/-2 ℃, adding alpha-amylase into the dispersion, uniformly stirring for enzymolysis for 0.5-1.5 h, wherein the addition amount of the alpha-amylase is 0.4-0.6% of the content of a substrate, adjusting the pH value to 2.5-3.5, adding acidic protease and pectinase into the dispersion, uniformly stirring for enzymolysis for 2-4 h, wherein the addition amount of the acidic protease is 0.5-0.8% of the content of the substrate, the addition amount of the pectinase is 0.3-0.6% of the content of the substrate, and inactivating the enzyme by using a high-temperature instant enzyme inactivating method after the enzymolysis is finished;

(d) membrane separation: concentrating the enzymolysis liquid under vacuum and reduced pressure to recover the solvent, concentrating until the relative density is 1.01-1.02, filtering by a membrane separation technology to obtain another part of dietary fiber solution with the molecular weight obviously different from other nutrients, wherein the thickness of a filter membrane is 80-150 mu m, the pore diameter of the filter membrane is 2-8 mu m, and the operating pressure is 0.12-0.25 MPa; concentrating the dietary fiber solution under vacuum and reduced pressure to obtain a concentrated solution with the relative density of 1.15-1.25;

(e) vacuum microwave drying: collecting the dietary fiber concentrated solution obtained by membrane separation and the dietary fiber obtained by centrifugation, and placing the dietary fiber concentrated solution and the dietary fiber obtained by centrifugation in a vacuum microwave drying device for drying treatment, wherein the drying condition is a microwave vacuum drying stage, the vacuum pressure is 3.0-5.5kpa, the microwave power density is 0.75-0.85W/g, and the water content is controlled to be 2-4%, so that the psyllium husk dietary fiber and chia seed dietary fiber are finally obtained.

Preferably, the preparation method of the hawthorn fruit powder in the step (5) is as follows

(a) Vacuum freeze drying: slicing hawthorn fruits, controlling the average thickness of the hawthorn fruit slices to be 2-4 mm, putting the hawthorn fruit slices into vacuum freeze drying equipment for drying, setting the parameters to be 0.4-0.6 ℃/min of pre-freezing rate, the pre-freezing initial temperature of-30-45 ℃, the pre-freezing final temperature of-70 ℃, the pressure of a drying chamber of 55-85 pa, the temperature of a heating plate of 38-45 ℃ and the drying time of 20-28 h;

(b) and (3) low-temperature superfine grinding: and (3) putting the dried hawthorn freeze-dried slices into a low-temperature ultrafine pulverizer for low-temperature ultrafine grinding, wherein 100% of the obtained hawthorn fruit powder is required to pass through a 300-mesh sieve.

In the invention, the Plantago ovata forsk shell dietary fiber and chia seed dietary fiber are obtained by carrying out low-temperature superfine grinding, membrane separation and vacuum microwave drying on Plantago ovata forsk shells and chia seeds, wherein the content of soluble dietary fiber and insoluble dietary fiber reaches more than 93%, and the Plantago ovata forsk shell dietary fiber and chia seed dietary fiber have the advantages of high purity, less impurities, good effect, high stability and greatly improved bioavailability; the hawthorn fruit powder is obtained by carrying out vacuum freeze drying and low-temperature superfine grinding on hawthorn fruits, the retention rate of various effective components such as vitamins, flavones and the like in the fruits is greatly improved, and 100% of the hawthorn fruit powder can pass through a 300-mesh sieve; the cockscomb extract and potentilla chinensis extract are obtained by carrying out alcohol reflux extraction, reduced pressure concentration, vacuum freeze drying and low-temperature superfine grinding on cockscomb and potentilla chinensis medicinal materials; the purslane microcapsule is obtained by sequentially carrying out supercritical extraction, emulsification and homogenization microcapsule embedding and spray drying on a purslane medicinal material, wherein the embedding rate of active ingredients in the purslane microcapsule is more than 90%, and the particle size is 50-260 mu m. The obtained purslane microcapsule has good solubility and good stability to light and heat. The corn oligopeptide is obtained by performing size mixing, enzymolysis, centrifugal filtration, concentration and spray drying on corn protein powder, wherein oligopeptide components with the molecular weight of less than 1000D in the corn oligopeptide account for more than 95 percent, so that the absorption and utilization rate of the corn oligopeptide by an organism is effectively improved, and the effect of absorption without digestion or with digestion slightly is achieved.

Preferably, the neutral protease is selected from bacillus subtilis neutral protease, and the enzyme activity is 20 ten thousand U/g.

The composition can be prepared into various pharmaceutically acceptable dosage forms such as hard capsules, granules, powder or tablets by a conventional preparation process so as to store and control the content of active ingredients.

Compared with the prior art, the invention has the advantages that:

Aiming at the efficacy of reducing blood fat, the purslane is mainly prepared into the microcapsule to be matched with hawthorn fruit powder, red yeast rice powder and corn oligopeptide which also have the functions of reducing blood fat and regulating blood pressure, so that the functions of regulating blood fat balance and reducing the contents of cholesterol and triglyceride in blood are obviously improved. Red yeast rice is rich in natural lovastatin, and has effects of invigorating spleen, resolving food stagnation, promoting blood circulation, removing blood stasis, reducing blood lipid, and treating and preventing cardiovascular disease and cerebrovascular disease. Red rice not only can strongly reduce Total Cholesterol (TC) and low-density lipoprotein (LDL), but also can reduce Triacylglycerol (TG) to a certain extent and can also increase high-density lipoprotein (HDL). The purslane has the functions of clearing away heat and toxic materials, cooling blood and stopping bleeding, treating diarrhea and enteritis of dysentery, carbuncle sore and pyogenic infections, diminishing inflammation and easing pain, improving the health of gastrointestinal tracts, protecting liver and reducing blood fat. Omega-3 fatty acids rich in purslane can inhibit the absorption of human body to cholesterol acid, inhibit the synthesis of lipid and lipoprotein in liver, reduce the concentration of blood cholesterol, promote the discharge of cholesterol from excrement, improve the elasticity of vessel wall, expand coronary artery, reduce thrombosis, delay the process of atherosclerosis, and is very beneficial to preventing and treating cardiovascular diseases. The hawthorn flavone rich in hawthorn fruits can obviously reduce the concentration of serum Total Cholesterol (TC), low-density lipoprotein cholesterol and apolipoprotein of hyperlipaemia people, obviously increase the concentration of high-density lipoprotein cholesterol (HDL _ C) and apolipoprotein, inhibit the synthesis of liver cholesterol, and promote the intake of plasma cholesterol by liver to play a role in reducing blood fat. The corn oligopeptide can protect liver, relieve alcoholism and sober up, and has auxiliary protection effect on chemical liver injury. A large number of experiments prove that the purslane microcapsule, hawthorn fruit powder, red yeast rice powder and corn oligopeptide are compatible for use according to a specific proportion, so that the synergistic effect can be achieved, the functions of regulating blood fat balance and reducing the contents of cholesterol and triglyceride in blood can be obviously improved.

Aiming at the effect of improving the gastrointestinal function, the cockscomb extract and the potentilla chinensis extract are combined with the polydextrose, the konjac powder and the pueraria powder which are rich in dietary fibers, so that the stimulation to the gastrointestinal tract is small, and the special effects of mildness and nourishing, bowel relaxing, heat clearing and detoxifying, hemostasis and detumescence and ulcer removal, intestinal flora balance regulation and gastrointestinal function improvement can be achieved. The composition is rich in various soluble dietary fibers and insoluble dietary fibers, has low energy, is easy to generate satiety, can promote gastrointestinal peristalsis and accelerate gastrointestinal tract emptying, and simultaneously has strong water absorption of the dietary fibers, thereby playing the roles of softening stool, relaxing bowel and preventing constipation. The traditional Chinese medicines of the cockscomb and the potentilla chinensis in the formula are mainly used for clearing away heat and toxic materials, stopping bleeding, reducing swelling and removing ulcer, and a large number of experiments prove that the cockscomb and the potentilla chinensis are matched according to the formula proportion of the composition disclosed by the invention, so that the synergistic effect is achieved, and the effects of relaxing bowel, clearing away heat and toxic materials, stopping bleeding, reducing swelling and removing ulcer and regulating intestinal flora balance can be greatly improved.

The blood fat reducing composition comprises purslane microcapsules, hawthorn fruit powder, red yeast rice powder and corn oligopeptide, and has good functions of reducing blood fat and reducing cholesterol, and meanwhile, a large amount of dietary fibers in the hawthorn fruit powder and the red yeast rice powder cannot be directly absorbed by a human body, but can be effectively utilized by intestinal beneficial bacteria groups, so that colonization and multiplication of intestinal beneficial bacteria are promoted, intestinal microecological balance is improved, and intestinal peristalsis is stimulated, so that the effects of enhancing the bowel relaxing and gastrointestinal functions of the composition are achieved.

The preparation method of the invention is that purslane is processed by supercritical CO2Extracting and embedding to prepare microcapsules, extracting the traditional Chinese medicines such as cockscomb, potentilla chinensis and the like by alcohol reflux, concentrating under reduced pressure, freeze-drying in vacuum, and crushing to prepare extracts, so that the main functional components of the medicines are retained to the maximum extent, and the drug effect is remarkably improved; the Plantago ovata forsk husk and chia seed are subjected to ultramicro crushing, enzymolysis, alcohol dissolving, membrane separation and vacuum microwave drying treatment to extract dietary fiber with the purity of more than 93 percent, so that the dosage of the composition is obviously reduced; the hawthorn fruit is subjected to vacuum freeze drying and low-temperature superfine grinding to prepare hawthorn fruit powder, so that the nutrient substances in the hawthorn fruit are retained to the greatest extent; corn oligopeptide is prepared by mixing, carrying out enzymolysis, carrying out centrifugal filtration, carrying out reduced pressure concentration and spray drying on corn protein powder, wherein the oligopeptide component with the molecular weight of less than 1000D accounts for more than 95 percent, so that the absorption and utilization rate of the corn oligopeptide by an organism is effectively improved, and the effect of absorption without digestion or with digestion slightly is achieved; through the treatment of the preparation process, the edible amount of the composition is obviously reduced, the efficacy and the nutrient components are enriched, the efficacy is obviously improved, and the bioavailability of the raw materials is improved.
