Technical Field

The invention belongs to the technical field of biological medicines, and particularly relates to a composition consisting of a hydrogenated cyclopentanoperhydrophenanthrene derivative and a cellulose derivative.

Background

Cellulose derivatives represented by hydroxyethylcellulose are commonly used auxiliary materials in oral and external solid preparations, however, a research result of Raffai T et al, published in Antimicob Agents Chemother, journal 2018, Vol.63, No. 1, proves that hydroxyethylcellulose can remarkably enhance the growth and infectivity of Chlamydia trachomatis D and E types. Although this study was conducted in vaginal simulants, it has been reported that Chlamydia trachomatis also causes infection of the digestive tract (e.g., Genitourin Med.1990 Feb; 66(1):26-7.), and that, for some special populations with some non-sexual activity, Chlamydia trachomatis growing in the reproductive tract is entirely likely to enter the digestive tract. Experiments carried out by adopting gastric acid simulation liquid as a medium show that cellulose acetate, low-substituted hydroxypropyl methylcellulose, low-substituted hydroxypropyl cellulose, methyl cellulose, croscarmellose sodium, microcrystalline cellulose, cellulose acetate, ethyl cellulose, hydroxyethyl cellulose, hydroxypropyl methylcellulose, hydroxypropyl cellulose, hydroxyethyl methylcellulose, hydroxyethyl cellulose, sodium carboxymethyl cellulose and other cellulose derivative auxiliary materials have similar effects. Therefore, the oral preparation containing the cellulose derivative as the auxiliary material can also promote the growth of chlamydia trachomatis in the alimentary canal after oral administration, and brings certain potential safety hazard to high-risk patients.

Disclosure of Invention

The invention aims to provide a composition consisting of a hydrogenated cyclopentanoperhydrophenanthrene derivative and a cellulose derivative, which can significantly reduce the growth-enhancing effect of the cellulose derivative on chlamydia trachomatis.

In order to achieve the above object, the present invention provides a composition comprising a hydrogenated cyclopentanophene derivative and a cellulose derivative, wherein the cellulose derivative is one selected from the group consisting of cellulose acetate (C1), low-substituted hypromellose (C2), low-substituted hypromellose (C3), methylcellulose (C4), croscarmellose sodium (C5), microcrystalline cellulose (C6), cellulose acetate (C7), ethylcellulose (C8), ethylcellulose (C9), hypromellose (C10), hypromellose (C11), hydroxyethylmethylcellulose (C12), hydroxyethylcellulose (C13), and carboxymethylcellulose sodium (C14); the hydrogenated cyclopentanoperhydrophenanthrene derivative is one of the following compounds 1-10:

the cellulose derivatives of the present invention are all Excipients found in Handbook of Pharmaceutical Excipients (Six Edition) written by Raymond C Rowe et al and meet the quality standards thereof.

Preferably, the mass ratio of the cellulose derivative to the hydrogenated cyclopentanophenanthrene derivative in the composition according to the invention is not higher than 100000: 1.

In vitro test results show that the composition can obviously reduce the growth effect of the cellulose derivative auxiliary materials on enhancing the chlamydia trachomatis.
