1. A serum composition comprising docosahexaenoic acid, wherein the composition comprises:

docosahexaenoic acid 0.001-1 wt%;

0.1-5 wt% of emulsifier; and

1-45 wt% of humectant.

2. The serum composition according to claim 1, wherein the composition comprises:

docosahexaenoic acid 0.005-0.5 wt%;

0.2-4 wt% of emulsifier; and

1-40 wt% of humectant.

3. The serum composition according to claim 2, wherein the composition comprises:

docosahexaenoic acid 0.01-0.1 wt%;

0.3-3 wt% of emulsifier; and

1-30 wt% of humectant.

4. The serum composition according to any one of claims 1 to 3, wherein the emulsifier is selected from at least one of sucrose laurate, lecithin, PEG-40 hydrogenated castor oil, polysorbate-80 and sorbitan sesquioleate; and the humectant is selected from at least one of glycerin, glycol, sodium hyaluronate and hydrolyzed sodium hyaluronate.

5. The serum composition according to claim 4, wherein the glycol is selected from propylene glycol and butylene glycol.

6. The serum composition according to claim 5, wherein the propylene glycol is 1, 3-propylene glycol.

7. The essence composition according to any one of claims 1 to 3, wherein the composition further comprises 0 to 30% by weight of vegetable oil and fat, 0 to 1% by weight of an antioxidant, 0 to 1% by weight of a pH regulator, and 0 to 1% by weight of an anti-inflammatory soothing agent.

The essence composition according to any one of claims 1 to 3, wherein the vegetable oil is selected from squalane, sunflower seed oil or jojoba seed oil; the antioxidant is selected from tocopherol or tocopherol acetate; the pH regulator is selected from citric acid, sodium citrate, lactic acid or sodium lactate; and said anti-inflammatory soothing agent is selected from dipotassium glycyrrhizinate or mussel extract.

9. The serum composition according to any one of claims 1 to 3, wherein the lecithin is hydrogenated phosphatidylcholine.

14. A method of preparing the serum composition according to any one of claims 1 to 13, comprising the steps of:

1) adding an emulsifier and a humectant into water, and stirring until the emulsifier and the humectant are dissolved to obtain an aqueous solution;

2) heating a mixture of docosahexaenoic acid, optional vegetable oil and an antioxidant, and uniformly stirring to obtain an oil phase;

3) adding the oil phase into the aqueous solution, and emulsifying by using an emulsifying homogenizer to obtain an emulsion;

4) adding optional pH regulator and anti-inflammatory and soothing agent into the emulsion, and mixing to obtain the essence composition.

15. The process according to claim 14, wherein in step 2) the mixture of docosahexaenoic acid, optionally vegetable fat and oil and an antioxidant is heated to 75-80 ℃.

16. The serum composition obtained by the method according to claim 14 or 15.

Technical Field

The invention relates to an essence composition containing docosahexaenoic acid and a preparation method thereof.

Background

The normal healthy stratum corneum contains 10% -20% water to maintain the skin's moisture and elasticity. When the moisture content in the stratum corneum is reduced to below 10% due to changes in climatic conditions such as age, cold and dryness, the skin will appear dry, wrinkled and gradually age. Modern cosmetic development has focused on protecting skin and delaying skin aging. Therefore, it is of great significance to study the reason why the water in the stratum corneum of the skin can be maintained.

Fatty acid as one of PPAR agonists can stimulate the metabolism of cell lipid, and filaggrin as one of PPAR agonists can be activated and express up-regulated protein and plays a significant role in the integrity of skin barrier, the regulation of skin pH, UV resistance and the onset of atopic dermatitis. It is reported in the literature that the disruption of skin barrier function caused by abnormal expression of Filaggrin (FLG) protein may be one of pathogenesis of infant skin disease, and fatty acid can promote the expression of barrier protein from gene level, thereby possibly playing an important role in improving abnormal skin barrier function. The Martina S study showed that loss of FLG in the absence of inflammation is sufficient to alter the expression levels of proteins associated with the pathogenesis of atopic dermatitis. These include proteins that regulate inflammation, proteolysis, and cytoskeletal function.

Docosahexaenoic acid, DHA, is an unsaturated fatty acid that is very important to the human body and is often used in food as a nutritional additive. Docosahexaenoic acid as a highly unsaturated fatty acid has very important effects on the aspect of skin care, can achieve the effects of moisturizing, moistening, resisting inflammation, repairing and the like, and enhances the skin barrier, but the current market lacks a skin care product containing the docosahexaenoic acid.

Disclosure of Invention

The invention provides a docosahexaenoic acid-containing essence composition which has better tolerance to skin. Meanwhile, the essence composition containing docosahexaenoic acid can be stably stored for a long time, and has the effects of moisturizing, moistening, resisting inflammation, repairing, improving skin barrier and the like.

Accordingly, a first aspect of the present invention provides a docosahexaenoic acid-containing essence composition, wherein the composition comprises:

docosahexaenoic acid 0.001-1 wt%;

0.1-5 wt% of emulsifier; and

1-45 wt% of humectant.

According to a preferred embodiment of the invention, the composition comprises:

docosahexaenoic acid 0.005-0.5 wt%;

0.2-4 wt% of emulsifier; and

1-40 wt% of humectant.

According to another preferred embodiment of the invention, the composition comprises:

docosahexaenoic acid 0.01-0.1 wt%;

0.3-3 wt% of emulsifier; and

1-30 wt% of humectant.

According to a preferred embodiment of the present invention, the emulsifier is selected from at least one of sucrose laurate, lecithin, PEG-40 hydrogenated castor oil, polysorbate-80, sorbitan sesquioleate; the emulsifier in the composition of the present invention improves the surface tension between the various constituent phases of the composition to form a uniform and stable dispersion.

According to a preferred embodiment of the present invention, the humectant is selected from at least one of glycerin, glycol, sodium hyaluronate and hydrolyzed sodium hyaluronate. The moisturizer in the essence composition can form a film on the surface of the skin, seals water in the skin to prevent the water from evaporating, and does not interfere the absorption of the water in the air of the skin, so that the proper humidity of the skin is maintained.

According to another preferred embodiment of the invention, the diols are preferably propylene glycol and butylene glycol, more preferably 1, 3-propanediol.

According to another preferred embodiment of the invention, the composition further comprises 0-30 wt.% of vegetable fat and oil, 0-1 wt.% of an antioxidant, 0-1 wt.% of a pH adjusting agent and 0-1 wt.% of an anti-inflammatory soothing agent.

The vegetable oil and fat in the essence composition can be retained on the surface of the skin, plays a role in lubrication in the cutin layer, fills the space on the surface of the skin, replaces the lipid lost in the cutin layer, keeps the skin soft, flexible and smooth, reduces the scale-shaped skin and improves the appearance of the skin.

The antioxidant in the essence composition of the invention can prevent the adverse effect of oxygen, thereby helping to capture and neutralize free radicals, and removing the damage of the free radicals to human skin.

The pH regulator in the essence composition can interact with the acidic substances in the composition, so that the pH value of the composition is regulated.

The anti-inflammatory and soothing agent in the essence composition has the effects of resisting inflammation, sterilizing and soothing nerves.

According to a preferred embodiment of the invention, the vegetable oil is selected from squalane, sunflower seed oil, jojoba seed oil; the antioxidant is selected from tocopherol or tocopherol acetate; the pH regulator is selected from citric acid, sodium citrate, lactic acid or sodium lactate; and said anti-inflammatory soothing agent is selected from dipotassium glycyrrhizinate or mussel extract.

According to a particularly preferred embodiment of the invention, the lecithin is hydrogenated phosphatidylcholine.

according to a preferred embodiment of the invention, the docosahexaenoic acid has a purity greater than or equal to 80%.

The essence composition can be prepared into different formulations according to different requirements, and preferably skin external preparations including liquid agents, gels, emulsions, creams, foams, patches and the like are selected. However, it will be understood by those skilled in the art that the compositions of the present invention may be supplemented with various desired additives, including but not limited to, humectants, vegetable fats and oils, anti-inflammatory soothing agents, antioxidants, and pH adjusting agents, and the like, depending on the various dosage forms of the composition. The composition of the present invention may further comprise various excipients as required depending on various formulations of the composition. For example, when the composition of the present invention is prepared as a liquid, the excipient may be selected from water, physiological saline, etc.; when the composition of the present invention is prepared into a gel, the excipient may be selected from cellulose derivatives, starch, gelatin, agar, polysaccharides, and the like; when the composition of the present invention is prepared into a cream, the excipient may be selected from glycerin, vaseline, paraffin, etc.; when the composition of the present invention is prepared into a foaming agent, the excipient may be selected from hydroxypropyl methyl cellulose, sodium lauryl sulfate polyethylene glycol, sodium fatty alcohol-polyoxyethylene ether sulfonate, etc.; when the composition of the present invention is prepared as a patch, the excipient may be selected from cellulose derivatives, starch, gelatin, agar, polysaccharides, and the like.

A second aspect of the present invention provides a method for preparing the essence composition, comprising the steps of:

1) adding an emulsifier and a humectant into water, and stirring until the emulsifier and the humectant are dissolved to obtain an aqueous solution;

2) heating a mixture of docosahexaenoic acid, optional vegetable oil and an antioxidant, and uniformly stirring to obtain an oil phase;

3) adding the oil phase into the aqueous solution, and emulsifying by using an emulsifying homogenizer to obtain an emulsion;

4) adding optional pH regulator and anti-inflammatory and soothing agent into the emulsion, and mixing to obtain the essence composition.

According to a preferred embodiment of the invention, wherein in step 2) the mixture of docosahexaenoic acid, optionally vegetable fat, antioxidants is heated to 75-80 ℃.

The third aspect of the present invention also provides the essence composition obtained by the method according to the present invention, which comprises docosahexaenoic acid as an active ingredient, and the purity of docosahexaenoic acid is not less than 80%.

The various dosage forms of the present invention may be prepared according to conventional methods known to those skilled in the art.

The essence composition containing docosahexaenoic acid has the advantages that: the essence composition has better tolerance to the skin than essence compositions using other emulsifiers. Meanwhile, the composition can remarkably increase the relative immunofluorescence intensity and the content of long-chain fatty acid of filaggrin, loricrin and endointegin, so that the composition has the effects of moisturizing, moistening, resisting inflammation, repairing, improving skin barriers and the like.

FIG. 1 shows the effect of the essence compositions according to the present invention on the expression amounts of filaggrin, loricrin and incrustin, which were measured and analyzed by a 3D epidermis model experiment using the essence compositions prepared in examples 1 to 4 according to the present invention.

Fig. 2 shows that the essence compositions prepared in examples 1 to 4 of the present invention were measured and analyzed for the effect of long-chain fatty acids by a 3D skin model experiment.
