Technical Field

The invention relates to the field of artificial feeding of insects, in particular to a prodenia litura artificial feed containing traditional Chinese medicine components and a preparation method thereof.

Background

Indoor breeding of insects is an important link in pesticide activity screening, insect biological characteristic research, insect function and insect gene research at present, however, in the indoor breeding process, a large amount of insects die due to the influence of environmental conditions, nutrient elements and microorganisms. Improving the survival rate of insects becomes a key technology for indoor feeding of insects.

Prodenia litura (Spodopteralitura (Fabricius)) is a lepidoptera, and insects of the Spodoptera family are one of agricultural pests which are ubiquitous in the world, seriously harmful and difficult to control, and meanwhile, the prodenia litura is also a relatively mature insect. There are a lot of literature reports on indoor breeding technology and artificial feed composition of prodenia litura, and these artificial feeds mainly comprise natural feeds and artificial feeds, wherein the artificial feeds are widely researched and applied, and contain nutritional ingredients such as proteins, amino acids, saccharides, vitamins, inorganic substances, lipids, sterols, and the like. However, when the artificial feed is used for breeding insects indoors, the nuclear polyhedrosis virus infects spodoptera litura larvae, or the larvae generate drug resistance, so that the insects die in a large amount, or the insects die in a large amount in a short time, or the insect population declines, and the survival rate of the artificially bred spodoptera litura is low. The spodoptera litura larvae are very small in size, 1-3 instar larvae are smaller in size, the immunity is low, and the spodoptera litura larvae are extremely easy to die due to factors such as feeding conditions and unbalanced nutrient elements in the indoor feeding process. The spodoptera litura is frequently subjected to phenomena of soft larva body, outflow of body fluid and the like in the indoor breeding process, and finally the spodoptera litura is killed in a large amount. The reason for this phenomenon has been studied mainly due to bacterial, fungal and viral infections, and to the decline of the insect population. The immunity is the ability of the insect body to resist external invasion and maintain the stability of the internal environment. When the immune system of the insect is low, the insect can not normally play a protective role, and is very easy to cause infection of bacteria, viruses, fungi and the like. The feed disclosed by the invention takes the improvement of the immunity of prodenia litura as a breakthrough, and traditional Chinese medicine components for improving the physique of the larvae, regulating the immunity of the larvae, enhancing the anti-infection capacity of the larvae, regulating the immunity of the larvae, improving the physique of the larvae and resisting the infection of germs and viruses are added into the feed. The invention takes corn flour, wheat germ and yeast powder as protein and amino acid sources. The wheat germ contains common amino acids, saccharides, fatty acids, minerals and B vitamins required by insects, but the amino acids are not balanced. The sorbic acid can effectively inhibit the activity of mould, microzyme and aerobic bacteria, thereby effectively prolonging the preservation time of the food and keeping the flavor of the original food. The methyl p-hydroxybenzoate and the sorbic acid are usually mixed and added into the artificial insect feed. Astragalus membranaceus is a labiate plant, is a commonly used qi tonifying medicine in China, has more than 2000 years of history so far, has the functions of enhancing the immunity of organisms, resisting stress and resisting wide bacteria, and can be used for symptoms such as spontaneous perspiration, night sweat due to yin deficiency, asthenia, yang-qi deficiency, lung-qi deficiency, sore ulcer and the like caused by weak body and weak surface in the medical field. Cortex Phellodendri is Rutaceae plant, and its medicinal part is cortex, and has heat clearing and detoxicating effects; heat-clearing and dampness-drying herbs mainly used for treating damp-heat dysentery and diarrhea; bone-steaming, fatigue-heat, night sweat; and sores in the mouth and tongue; conjunctival congestion with swelling and pain; carbuncle, deep-rooted carbuncle and sore; eczema of skin, etc. The radix et rhizoma Rhei is dried root and rhizome of Rheum palmatum L., Rheum tanguticum Maxim. and Rheum officinale Baill. of Polygonaceae, has effects of removing stagnation, clearing away dampness and heat, purging pathogenic fire, cooling blood, removing blood stasis, removing toxic substance, and inhibiting bacteria, and is mainly used for treating acute gastric ulcer and duodenal ulcer complicated with hemorrhage.

On the basis of mastering the biology, ecological habits and growth and development rules of spodoptera litura larvae, combining theories and methods of modern biology and breeding science, selecting traditional Chinese medicines such as scutellaria baicalensis, golden cypress, raw rhubarb and radix scrophulariae to be added into the feed according to the principle of distinguishing and treating in traditional Chinese medicine and on the basis of the understanding and treatment principle of the traditional Chinese medicine on improving resistance and strengthening the body mechanism, and organically combining and synergizing a plurality of traditional Chinese medicines, so that the feed disclosed by the invention can improve the autoimmunity and disease resistance of insects while meeting the basic nutritional requirements of the spodoptera litura. The invention is to prepare the traditional Chinese medicine into alcohol extract and then mix the alcohol extract with the feed. The alcohol extract prepared from the traditional Chinese medicine has high content of effective components, is easy to be uniformly mixed with feed, and has simple and convenient operation and low preparation cost.

The feed provided by the invention can obviously improve the immunity of spodoptera litura larvae and improve the survival rate of the larvae, particularly the survival rate of 1-3 instar larvae. The invention focuses on improving the immunity of prodenia litura, has scientific and reasonable formula, ensures that the feed has comprehensive nutrition, high nutritional value and good taste, can improve the feeding rate of prodenia litura larvae, and can improve the physical quality of the prodenia litura larvae. The 1-3 instar larvae fed by the feed have high survival rate and regular growth and development, and are favorable for meeting the supply of insect sources for indoor bioassay. The effect of the spodoptera litura special feed containing the traditional Chinese medicine components is tested by experiments of feeding spodoptera litura larvae with the artificial feeds prepared in examples 1-5 respectively:

(1) the feeds prepared in examples 1 to 5 were prepared into small pieces of 5g each, and day-old larvae hatched on the same day were selected;

(2) taking 5 cleaned straight tubes, placing 20 larvae in each tube, and placing 1 small piece of feed with weight of 5g prepared in examples 1-5 into 5 tubes;

(3) when the larva grows to 3 years old, the larva is changed to eat the feed once;

(4) after 4 years of age, the animals are raised singly and changed once every 2 days, and 5g of the feed is added every time.

The growth of the 1-6 th-instar spodoptera litura larvae reared using the feeds prepared according to the formulations of examples 1-5 of the present invention and the above-described methods was observed as shown in table 3.

The artificial feed for prodenia litura prepared as shown in table 4 is comparative example 1 and comparative example 2, the raw materials of comparative example 1 and comparative example 2 are similar in variety and different in quantity, but are within the range of the raw material variety and quantity in the claims of the present invention, and comparative example 1 and comparative example 2 are different from the present invention in that the alcohol extract of the traditional Chinese medicine of the present invention is not added, but oxytetracycline which is commonly used in feed is added.

The preparation methods of comparative examples 1 and 2 are substantially the same as the method of the present invention. The method comprises the following specific steps:

(1) weighing the raw materials according to the table 4, uniformly mixing wheat germ, corn flour, tomato sauce, yeast powder, agar, sucrose and casein in the raw materials, adding distilled water, uniformly stirring to form paste, sterilizing at 121 ℃ for 30min, and cooling to 60-70 ℃ to obtain a material 1;

(2) pulverizing oxytetracycline in the raw materials, uniformly mixing with vitamin C, methylparaben, sorbic acid, compound vitamin, Vickers salt and choline chloride, adding distilled water, and stirring into paste to obtain a material 2;

(3) uniformly mixing the material 1 and the material 2, and cooling to 25 ℃, thus obtaining the spodoptera litura special feed in the comparative example block shape; storing at 4 ℃.

The effect of the feed containing the traditional Chinese medicine components special for prodenia litura of the invention is checked by contrast through experiments of feeding the prodenia litura larvae with the feeds prepared in example 1, comparative example 1 and comparative example 2 respectively:

(1) the feeds prepared in example 1, comparative example 1 and comparative example 2 were prepared into 5 g-weight small pieces, and day-old larvae hatched on the same day were selected;

(2) taking 3 cleaned straight tubes, placing 20 larvae in each tube, and respectively placing 1 small piece of feed with the weight of 5g prepared in example 1, comparative example 1 and comparative example 2 in the 3 straight tubes;

(3) when the larva grows to 3 years old, the larva is changed to eat the feed once;

(4) after 4 years of age, the animals are raised singly and changed once every 2 days, and 5g of the feed is added every time.

The growth and development of 1-6 th instar spodoptera litura larvae reared by the same method using the feeds prepared according to the formulations of example 1, comparative example 1 and comparative example 2 of the present invention were observed as shown in table 5.
