Compared with the prior art, the invention has the following advantages and effects:

(1) loquat flower is flower of Eriobotrya japonica (Thunb.) lindl of Rosaceae, and has effects of dispelling pathogenic wind, relieving cough, and dredging nasal orifice. It is commonly used to treat wind syndrome of the head, nasal obstruction, nasal discharge, chronic cough due to asthenia, and bloody sputum. The invention innovatively discovers that the extract of the loquat flower also has the effects of strongly activating the growth of skin cells, inhibiting tyrosinase, removing free radicals in vivo and the like through research, and is a very good anti-aging active substance.

In order to obtain the best extraction effect, the invention firstly carries out scientific experiments on the extraction solvent, uses water, ethanol, 1, 3-butanediol, PEG400, petroleum ether and the like as the extraction solvent respectively, and finds that the ethanol aqueous solution has the best effect. The heating extraction method, the microwave extraction method, the ultrasonic extraction method, the supercritical extraction method and the high-pressure homogenization extraction method are respectively adopted, and the result shows that the high-pressure homogenization extraction method has the best effect. A great deal of scientific research is carried out on the extraction conditions.

In order to obtain the best separation and purification effect, membrane separation and macroporous resin separation are respectively adopted, and as a result, the activity of the extract is obviously improved after the membrane separation and the macroporous resin separation are found, and the activity of the extract obtained by the macroporous resin separation is better, and the conductivity of the extract is less than 300 mu S/cm and is much lower than that of the extract obtained by a membrane separation method (about 1200 mu S/cm). The scientific experiments of the separation of the extracts are respectively carried out by using the types of macroporous resins of AB-8, X-5, D101, LSA-20 and the like, and the results show that the effect is the best after the treatment by the AB-8 macroporous absorption resin. The eluents were examined for 10%, 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, 95% and 100% ethanol by volume fraction, respectively, and it was found that 70% ethanol elution gave the best results of the extract.

The extraction and separation processes are carried out at normal temperature to ensure that the active ingredients are not deactivated by heat treatment.

(2) The loquat flower extract obtained by the method can strongly promote the proliferation and growth of human embryonic lung fibroblasts, strongly remove free radicals, inhibit the activity of tyrosinase, reduce the generation of melanin, accelerate the renewal of skin surface cells and delay skin aging, and has obvious anti-aging effect when being added into cosmetics.

The present invention will be described in further detail with reference to examples and drawings, but the present invention is not limited thereto.

The test methods in the following examples, in which specific experimental conditions are not specified, are generally performed according to conventional experimental conditions or according to the experimental conditions recommended by the manufacturer. The materials, reagents and the like used are, unless otherwise specified, reagents and materials obtained from commercial sources.

EXAMPLE 1 preparation of loquat flower extract (I)

1kg of dried loquat flower is taken, the loquat flower is crushed, screened by a 60-mesh sieve and mixed to obtain loquat flower powder;

secondly, according to the mass ratio of liquid to material of 12: 1 adding an extraction solvent, soaking for 2.5h, and extracting at normal temperature by using a high-pressure homogenizing extractor under the extraction pressure of 5MPa to obtain an extracting solution; the extraction solvent is ethanol water solution with the volume fraction of 50 percent;

③ centrifuging the extract (centrifugation radius is 10cm, centrifugation condition is 3000rpm, normal temperature centrifugation is 30min), adsorbing the centrifugate by AB-8 macroporous adsorbent resin (purchased from Tianjin Praded Biotech limited) and eluting by ethanol water solution with volume fraction of 70%;

fourthly, concentrating the eluent under reduced pressure until the mass ratio of the concentrated solution to the loquat pollen is 1: 1, adding 1, 3-butanediol with the same mass as the concentrated solution to obtain the loquat flower extract (I).

Example 2 preparation of loquat flower extract (II)

1kg of dried loquat flower is taken, the loquat flower is crushed, screened by a 80-mesh sieve and mixed to obtain loquat flower powder;

secondly, according to the mass ratio of liquid to material of 15: 1, adding an extraction solvent, soaking for 2 hours, and then extracting at normal temperature by using a high-pressure homogenizing extractor under the extraction pressure of 4MPa to obtain an extracting solution; the extraction solvent is ethanol water solution with the volume fraction of 40 percent;

③ centrifuging the extract (centrifugation radius is 12cm, centrifugation condition is 3000rpm, normal temperature centrifugation is 20min), adsorbing the centrifugate by AB-8 macroporous adsorption resin, and eluting by ethanol water solution with volume fraction of 70%;

fourthly, concentrating the eluent under reduced pressure until the mass ratio of the concentrated solution to the loquat pollen is 1: 1, adding 1, 3-butanediol with the same mass as the concentrated solution to obtain the loquat flower extract (II).

Example 3 preparation of loquat flower extract (III)

1kg of dried loquat flower is taken, the loquat flower is crushed, screened by a 40-mesh sieve and mixed to obtain loquat flower powder;

secondly, according to the mass ratio of liquid to material of 10: 1 adding an extraction solvent, soaking for 3 hours, and then extracting at normal temperature by using a high-pressure homogenizing extractor under the extraction pressure of 6MPa to obtain an extracting solution; the extraction solvent is ethanol water solution with the volume fraction of 60%;

③ centrifuging the extract (centrifugation radius is 8cm, centrifugation condition is 3000rpm, normal temperature centrifugation is 40min), adsorbing the centrifugate by AB-8 macroporous adsorption resin, and eluting by ethanol water solution with volume fraction of 70%;

fourthly, concentrating the eluent under reduced pressure until the mass ratio of the concentrated solution to the loquat pollen is 1: 1, adding 1, 3-butanediol with the same mass as the concentrated solution to obtain the loquat flower extract (III).

Test example 1 Effect of loquat flower extract on cell proliferation

The MTT experiment detection principle is as follows: succinate dehydrogenase in mitochondria of living cells can reduce exogenous MTT (thiazole blue) into water-insoluble blue-violet crystalline formazan and deposit in the cells, but dead cells do not have the function; then, the formazan in the cells is dissolved by DMSO, and the measurement is carried out at 570nm by a microplate reader; since the production amount of formazan is generally proportional to the number of living cells, the number of living cells can be reflected by the OD number measured by the microplate reader.

Test example 2 Effect of loquat flower extract on cell growth Curve

To further verify the long-term effect of the loquat flower extract on cell viability, the long-term growth relationship of MRC5 cells was determined in terms of days and PD (population doubling) using the loquat flower extract of example 1, which accurately describes the growth of cells throughout the culture. The MRC5 cells were selected as normal human embryonic lung fibroblasts that had only limited ability to divide in vitro and then entered the senescence stage. Test example 3 Effect of loquat flower extract on tyrosinase and free radical

Adding samples according to the table 2, comparing without adding tyrosinase as a control, and placing the solution in a water bath at 37 ℃ for constant temperature for 10 min; tyrosinase (110U/mL) was added, and the solution was incubated in a water bath at 37 ℃ for 10min, and then the absorbance value was measured at 475 nm.

4.5mL of Tris-HCl buffer (0.05 mol/L, pH 8.2) was preheated in a water bath at 25 ℃ for 20 min. Then adding 1mL of sample and 0.4mL of 25mmol/L pyrogallol solution, mixing uniformly, reacting in a water bath at 25 ℃ for 5min, and adding 1.0mL of 8mol/L HCl to terminate the reaction. Experiments show that the loquat flower extract can effectively remove free radicals, has good tyrosinase activity inhibition effect and has obvious anti-aging function.

Application example 1 anti-aging nutrient cream

Table 4 shows the formula of the anti-aging cream containing the loquat flower extract of the present invention.

The preparation method comprises the following steps: dissolving phase A at 80 deg.C, mixing phase B at 80 deg.C, adding phase B before emulsifying cyclomethicone, adding phase B during emulsifying, homogenizing for 5min, cooling to 50 deg.C, adding phase C, homogenizing for 2 min, aging for 24 hr, and packaging.

Application example 2 anti-aging emulsion

Table 5 shows the formula of an anti-aging emulsion containing the loquat flower extract of the present invention.

The preparation method comprises the following steps:

dispersing the uniform phase C in advance for later use; putting the phase A component into an oil phase pot, heating to 80 ℃, and stirring until the phase A component is completely dissolved; putting the phase B component into an emulsifying pot, heating to 80 ℃, and stirring to dissolve until the phase B component is completely transparent; keeping the temperature of an emulsifying pot at 80 ℃, controlling the stirring speed to be in a vacuum foamless state of 300r/min, and slowly pumping the phase A; homogenizing at 3000r/min, and emulsifying at 80 deg.C for 10 min; after homogenizing, putting the C phase component into an emulsifying pot, keeping the C phase component in a vacuum state, stirring uniformly under the condition of 400r/min, and cooling; adding the heat sensitive component D phase component into an emulsifying pot at 50 ℃, and stirring until the components are uniformly mixed; cooling to 38 deg.C, discharging, filling and sealing.

Application example 3 anti-aging facial mask solution

Table 6 shows the formula of an anti-aging emulsion mask solution containing the loquat flower extract of the present invention.

The preparation method comprises the following steps: dispersing the phase B in advance for later use; putting the phase A component and the dispersed and uniform phase B into an emulsifying pot, heating to 80 ℃, stirring to dissolve until the components are completely transparent, and then cooling; cooling to 45 deg.C, adding the C phase active substance and D phase into emulsifying pot, stirring and dissolving to mix well; cooling to 38 deg.C, discharging, filling and sealing.

Application example 4 anti-aging gel

Table 7 shows an anti-aging gel formulation containing the loquat flower extract of the present invention.

The preparation method comprises the following steps: putting the raw material of the component A into a manufacturing pot, heating to 70 ℃, starting stirring and homogenizing at 3000rpm for 10min until the raw material is uniformly dispersed. Cooling to 45 deg.C, adding the pre-mixed and dissolved component B and the pre-mixed and dissolved component C, stirring for dissolving completely, vacuum degassing, filtering and discharging.

Application comparative example 1 ordinary emulsion

The results are in accordance with Table 5 except that loquat flower extract (I) was not contained.

Effect example 1 human body trial evaluation experiment

1. Experimental methods

Human body trial evaluation was performed using the emulsions prepared in example 2 and comparative example 1, respectively. 50 volunteers of 40-50 years old are selected and all female. The application method was to wash the face twice a day (once in the morning and evening), the emulsion prepared in application example 2 was applied to the left face, and the emulsion prepared in application comparative example 1 was applied to the right face. The experimental period was two months, and the skin color and wrinkles of the left and right faces were observed after two months of use.

2. Results of the experiment

After two months of use, the skin color of the left face using the lotion prepared according to application example 2 was significantly better than the skin color of the right face using the lotion prepared according to application comparative example 1 for 43 users, as determined by VISA skin detector and comparison with the skin color before use, and the wrinkles on the left face were significantly improved and the wrinkles on the right face were not improved, and the effective rate was 86%. Wherein, comprehensive evaluation is carried out from humidity retention, glossiness, compactness and anti-wrinkle effect, wherein the highest score is 5, the lowest score is 1, and the score higher than 3.5 is improved.

The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.
