Technical Field

The invention belongs to the field of medicines, and particularly relates to a natural cinnamon essential oil disinfectant and a preparation method thereof.

Background

Along with the continuous progress of society, the living standard of people is increasingly improved, and the health consciousness is also continuously enhanced. Meanwhile, with the rapid development of industry, the environment in which human beings live is also affected, and the occurrence of various novel bacteria and viruses increases the probability of human beings suffering from diseases. The active ingredients of the disinfection solution sold in the market at present mainly comprise peroxyacetic acid, 75% ethanol, formaldehyde, benzalkonium bromide and the like, and the disinfection solution has irritation and toxic and side effects and is not suitable for being generally used in places such as families and the like and being used for a long time. Therefore, an efficient and safe disinfectant suitable for long-term use in daily life is urgently needed to meet the requirements of people on sanitation and safety. Cinnamon is also known as cinnamon, cinnamon bark, cinnamon of the Lauraceae family (Latin's name: Cinnamomum cassia Presl), tropical evergreen arbor. Mainly distributed in Fujian, Taiwan, Hainan, Guangdong, Guangxi, Yunnan and other places. The essential oil obtained by distilling bark, branch and leaf of cinnamon has the effects of dispelling wind-damp, warming and strengthening spleen and stomach. It is commonly used for treating rheumatism, skin pruritus, yang collapse of spleen and stomach, cold limbs, slight pulse, cold accumulation of stomach and abdomen, abdominal pain, diarrhea, cold hernia and dolphin. Modern pharmacological research shows that the cinnamon oil has a killing effect on escherichia coli, staphylococcus aureus, bacillus subtilis, candida albicans and aspergillus flavus, and has a strong inhibiting effect on typhoid bacillus, paratyphoid bacillus, cholera bacillus, shigella, pneumococcus, bacillus anthracis, salmonella enteritidis, aerobacter, proteus and the like.

However, because cinnamon essential oil is insoluble in water and easily oxidized in air, the application of cinnamon essential oil is greatly limited by the properties. Therefore, it is necessary to provide a disinfectant solution using cinnamon essential oil as a sterilization component, which has the advantages of simple components, uniform and stable properties, convenient preparation, low cost and no environmental pollution. 2. The quantitative sterilization test of the suspension of the disinfectant refers to the quantitative sterilization test of the suspension specified in 2008 edition 'sterilization technical code', and the sterilization effect of the disinfectant prepared in the steps on escherichia coli (ATCC 8099), staphylococcus aureus (ATCC 6538) and candida albicans (ATCC 10231) is detected, specifically as follows:

1) selecting three strains of staphylococcus aureus, escherichia coli and candida albicans to respectively prepare bacterial suspensions according to the sterilization technical specification, wherein the concentration of the bacterial suspension is 1 multiplied by 108cfu/mL~5×108cfu/mL for standby;

2) taking a sterile centrifuge tube, adding 0.5mL of bacterial suspension, adding 0.5mL of 0.3% calf serum albumin, uniformly mixing, sucking 4.0mL of the disinfectant prepared in each embodiment by using a pipette gun, injecting into the disinfectant, rapidly and uniformly mixing, and timing;

3) the test bacteria and the disinfectant are acted for each preset time, 0.5mL of mixed solution of the test bacteria and the disinfectant is respectively absorbed and added into 4.5mL of sterilized neutralizer, and the mixture is uniformly mixed;

4) adding neutralizer into the mixed solution of the test bacteria and the disinfectant for 10min, sucking 1.0mL of the mixed solution, measuring the number of viable bacteria according to a viable bacteria culture counting method, and inoculating 2 plates to each tube of test solution. If the number of colonies growing on the flat plate is more, serial 10-time dilution can be carried out, and viable bacteria culture counting is carried out;

5) all test samples are cultured in a thermostat at 37 ℃, and the results are observed for 24h of bacterial culture and 48h of fungal culture;

6) the test was repeated 3 times, and the viable bacteria concentration (cfu/mL) of each group was calculated and converted to a logarithmic value (N), and then the killing logarithmic value was calculated according to the following formula:

log Kill (KL) = log of mean active bacteria concentration of control group (N0) -log of mean active bacteria concentration of test group (Nx). If the killing log value of each time is more than or equal to 5.00, the disinfection is judged to be qualified.
