Background

In the process of movement, the energy consumption of people is increased, and due to a large amount of sweating, water and electrolyte are lost along with the sweat, so that the internal environment of the body is disordered and the function is reduced; the oxidative free radicals generated by exercise attack muscle cells, which are seriously damaged skeletal muscle cells, and may cause damage to the cardiac muscle cells to some extent. If the injury of muscle cells occurs continuously and cannot be corrected in time, sports fatigue can occur, and energetic physical strength cannot be ensured; athletes exercising for a long time with great intensity often suffer from poor physical performance and even "reduced exercise immunity" due to poor recovery after training, which is particularly easy to happen in the middle and later periods of the training period or the competition period, and is mainly characterized in that: slow muscle growth, strength decline, muscle ache and stiffness for a long time, poor mental state, reduced body resistance, susceptibility to diseases such as cold, fever and the like, thereby seriously affecting the game achievement.

Disclosure of Invention

The invention aims to develop a preparation which can improve the metabolism ability of people, supplement elements required by human bodies, enable people to be vigorous, relieve fatigue and improve the immunity of athletes and a preparation method thereof.

The invention relates to a preparation capable of relieving fatigue, improving immunity and delaying senescence, which comprises the following components in percentage by weight:

l-carnitine 45-55 taurine 18-25 inositol 1.5-2.5

Vitamin PP0.25-0.35 vitamin E0.15-0.25 selenium-rich soybean 15-18

Microcrystalline cellulose 7-9 sodium carboxymethylcellulose 2.5-3.5

Magnesium stearate 0.45-0.55

The edible fungus extract: is prepared from more than one of Lentinus Edodes, Hericium Erinaceus, Agaricus Blazei Murr or Flammulina velutipes.

The preparation method of the preparation capable of relieving fatigue and improving the immunity of athletes comprises the following steps:

(1) weighing L-carnitine, taurine, an edible fungus extract, inositol, vitamin PP, vitamin E, microcrystalline cellulose and selenium-enriched soybean according to the weight percentage of the components, crushing and sieving by a sieve of 80 meshes;

1. l-carnitine: l-carnitine is a retinoid, is an essential nutrient for the human body, and has a main function of promoting the combustion of fatty acids to provide energy. Animal experiments at home and abroad show that the L-carnitine has the function of improving the swimming capability and anoxia endurance of normal mice. It was further observed in foreign trials that vigorous exercise accelerated carnitine metabolism and increased urinary acetyl-carnitine excretion. There are many researchers who reported that the maximum oxygen consumption and aerobic metabolic capacity of athletes were remarkably improved by supplementing 4 g of L-carnitine per day, and also reported that the maximum oxygen consumption and energy output during exercise were greatly improved by administering 2 g of L-carnitine one hour before the competition, and the generation of carbon dioxide and the concentration of lactic acid in plasma were remarkably reduced, so it was considered that L-carnitine could remarkably exert anti-fatigue effect and improve athletic performance of athletes.

2. Taurine: taurine is an essential nutrient for human body, and can eliminate fatigue for people with excessive use of brain, exercise and work; plays a certain role in regulating the functions of heart, liver, endocrine and the like; maintaining normal physiological functions of human brain; maintain normal visual function. In addition, taurine also has the effects of resisting oxidation, delaying aging, improving the immunity of the organism, protecting the liver and the like.

3. Inositol: inositol is a necessary substance for the growth of human, animals and microorganisms, and has important physiological, pharmacological and nutritional functions of promoting metabolism, improving cell nutrition, promoting cell growth, stimulating appetite, recovering physical strength, preventing fat accumulation in liver, and the like. Mutual compensation of inositol among tissues in the human body is quite slow, and the constant supply of exogenous inositol must be relied on to maintain normal physiological activities of the body.

4. Vitamin E: vitamin E is an antioxidant. In animal tissue cell membranes, it protects polyunsaturated fatty acids, the cytoskeleton, and sulfhydryl groups of other proteins in the membrane and nucleic acids of the cellular aeolian against attack by free radicals. This function is closely related to the normal functioning of many systems of the body, such as immunity, nerves, exercise, cardiovascular and reproduction. In animals with vitamin E deficiency, muscular dystrophy, dysfunction of the neuromotor system, and circulatory impairment are common symptoms. Therefore, proper vitamin E supplementation is very important for improving the physical fitness and the immune function.

5. Vitamin PP: vitamin PP acts as a dehydrogenase mainly in the form of coenzyme i (nad) and coenzyme ii (nadp) in vivo. It is involved in the process of protein ribosylation, and is used as glucose tolerance factor to promote insulin response. The deficiency of vitamin PP can cause fatigue, listlessness, weight loss, anorexia, insomnia, headache, hypomnesis, etc. Therefore, proper vitamin E supplementation is also important for relieving physical fatigue and enhancing immune function.

6. The edible fungus extract: the edible fungus extract contains polysaccharide peptide, and can enhance activity of T cell, B cell, NK cell and mononuclear macrophage, improve immunity, and recover immunity reduced by strenuous exercise training.

T3 increased the number of macrophages in the abdominal cavity in the humoral circulation, and T3 had an anti-inflammatory effect. When T4 is normal, the T3 is too high, which indicates that the body may produce inflammation.

T3 normal range: 0.8ng/ml to 1.9 ng/ml;

t4 normal range: 5 mu g/dl to 13 mu g/dl;

performance: stabilizing body temperature, improving state, improving constitution of susceptible cold, and restoring mental state.

The preparation method of the edible fungus extract comprises the following steps: weighing at least one of Lentinus Edodes, Hericium Erinaceus, Agaricus Blazei Murr or Flammulina velutipes (Fr.) Sing as raw material, pulverizing, adding water, extracting at 100 deg.C (by soaking edible fungi in boiling water) for 2 hr, and filtering; adding water into the filter residue obtained by filtering, extracting for 1.5 hours at the temperature of 100 ℃, and then filtering again; combining the filtrates obtained by the two times of filtration, and carrying out vacuum concentration under the conditions that the temperature is 60-85 ℃ and the vacuum degree is-0.06-0.08 Mpa; and spray drying at inlet temperature of 180 deg.C and outlet temperature of 90 deg.C to obtain edible fungus extract.

The preparation capable of relieving fatigue and improving the immunity of athletes and the preparation method have the advantages that: the preparation can greatly improve the maximum oxygen consumption of a user and the energy output during exercise, and obviously reduce the generation of carbon dioxide and the concentration of lactic acid in blood plasma; fatigue is eliminated; resisting oxidation, delaying aging, improving immunity, and protecting liver; promoting metabolism, improving cell nutrition, promoting cell growth, stimulating appetite, and restoring physical strength; enhancing body constitution and immunity.

Animal experiment results of the preparation

The effect of the preparation on the weight bearing swimming time of the mice: weighing the weight of the mouse at the beginning, the middle and the end of the experiment respectively, and after the last gastric lavage at the end of the experiment for 30min, putting the mouse in a constant-temperature swimming box for heavy swimming. Body weight and weight swimming time are shown in table 1:

The body weight of each group of mice is increased during the test period, but no significant difference exists between the same group of mice, which indicates that the body weight of the mice is not obviously influenced by the tested substances. The result of the mouse weight swimming time shows that the weight swimming time of the three-dose groups is obviously prolonged, wherein the difference of the 10-time dose group is significant compared with that of the negative control group; the difference between the 20-fold and 30-fold different groups is very significant compared with the negative control group (F is 5.03, and P is 0.0052). The results show that the preparation has the function of prolonging the weight swimming time of mice.

The influence of the preparation on serum urea nitrogen after mouse movement is as follows: the results of the measurements of the mice body weight three times (initial, middle and final) and the serum urea nitrogen level after swimming during the test period are shown in table 2:

The body weight of each group of mice is increased during the test period, but no significant difference exists between the same group of mice, which indicates that the body weight of the mice is not obviously influenced by the tested substances, and the result is consistent with the 2.1 test result. Serum urea nitrogen measurement results after swimming of the mice show that the BUN of the serum of the three-dose group is obviously reduced, wherein the difference of the 20-fold and 30-fold dose groups is respectively significant and extremely significant compared with a negative control group (F ═ 4.70, and P ═ 0.0072). The results show that the preparation has the function of reducing the serum BUN level of the mice after exercise.

The preparation has the following effects on the accumulation and removal of blood lactic acid after the movement of mice: during the experiment, the mice were weighed three times (initial, intermediate and terminal) as in table 3:

The body weight of each group of mice is increased during the test period, but no significant difference exists between the same group of mice, which indicates that the body weight of the mice is not obviously influenced by the tested substances, and the result is consistent with the results of the previous two experiments.

Effect of test substances on blood lactic acid levels before and after swimming of mice: the blood lactic acid levels and their differences before, after and after 20min rest of the mice before, after and after swimming are shown in table 4:

As can be seen from Table 4, the blood lactic acid level of each group of mice increased significantly after exercise and decreased after 20min rest. The results of the analysis of variance and the Q test show that: (1) pre-exercise blood lactate levels: no significant difference between groups (F ═ 0.08, P ═ 0.0244); (3) blood lactate levels after 20 minutes of rest: the 5-time, 20-time and 30-time three-dose groups are obviously lower than the negative control group, and the difference is very significant (F is 12.94, and P is less than 0.0001); (4) amplitude of increase in blood lactate level after exercise: the 5 times dosage group is lower than the negative control group, and the difference is significant; compared with the negative control group, the difference between the 20-time and 30-time two-dose groups is very significant (F is 5.07, and P is 0.0050); (5) extent of decrease in blood lactate level after rest: the 30-fold dosage group is higher than the negative control group, and the difference is significant (F is 3.48, and P is 0.0258). The above results show that: the preparation has functions of reducing accumulation of lactic acid and promoting lactic acid clearance after mouse movement.

The influence of the preparation on the content of mouse liver glycogen: the results of the three (initial, intermediate and final) body weights and liver glycogen measurements of the mice during the test are shown in table 5:

As can be seen from the above table, the body weight of each group of mice increased during the test period, but no significant difference was observed between the groups of the same period of body weight, which indicates that the test substance had no significant effect on the body weight of the mice, and the result is consistent with the previous test results. Liver glycogen measurements indicated that liver glycogen levels were elevated in the three-dose groups of mice after administration of the test substance, with a significant difference between the 20-and 30-fold two-dose groups as compared to the negative control group (F ═ 3.68, and P ═ 0.0207). The results show that the preparation has the function of increasing the content of glycogen of mouse liver.

The physical fatigue relieving experiment result of the preparation shows that: the weight-bearing swimming time of the mice with 10 times of dosage is longer than that of the negative control group, the difference is significant, and compared with the negative control group, the difference between the mice with 20 times of dosage and 30 times of dosage is very significant; 20. after 30 times of two dose groups of mice swim, the serum urea and nitrogen levels are lower than those of a negative control group, and the differences are respectively significant and extremely significant; after the mice in the 5-time dosage group move, the rise amplitude of the blood lactic acid is lower than that of the negative control group, the difference is significant, and compared with the negative control group, the difference between the two 20-time and 30-time dosage groups is significant; after the 30 times dosage group mice rest, the reduction amplitude of the blood lactic acid is higher than that of a negative control group, and the difference is significant; 20. the liver glycogen content of the 30-time two-dose group is obviously higher than that of the negative control group, and the difference is significant.

According to the 'evaluation procedure and inspection method specification of health food functionality', the preparation is judged to have the effect of relieving physical fatigue.

Detailed Description

Example one

Referring to fig. 1, a method for preparing a preparation for relieving fatigue and improving immunity of athletes comprises the following steps:

(1) weighing 44.05kg of L-carnitine, 25kg of taurine, 1.5kg of inositol, 0.02kg of edible fungus extract, 0.35kg of vitamin PP and 0.15kg of vitamin E, weighing 18kg of selenium-enriched soybean and 7kg of microcrystalline cellulose, and respectively crushing and sieving by a 80-mesh sieve;

(2) uniformly mixing the raw materials which are sieved by the 80-mesh sieve in a V-shaped mixer under the clean environment of 10 ten thousand grades, wherein the rotating speed of the mixer is 150 revolutions per minute, and the time is 6 minutes;

(3) weighing 3.5kg of sodium carboxymethylcellulose under a clean environment of 10 ten thousand grades, dissolving the sodium carboxymethylcellulose in purified water to prepare a sodium carboxymethylcellulose solution with the concentration of 2%, adding the sodium carboxymethylcellulose solution with the concentration of 2% into the uniformly mixed powder obtained in the step II, and uniformly mixing the powder with a groove type mixer to prepare a soft material, wherein the dryness and humidity of the soft material are proper;

(4) under a clean environment of 10 ten thousand grades, the prepared soft material is made into wet granules by a granulator through a 16-mesh screen;

(8) pouring the prepared uniformly mixed particles into a tablet machine for tabletting under a clean environment of 10 ten thousand grade to ensure that each tablet weighs 0.5 g;

(9) and adding the tablets into a cleaned and sterilized racking machine for subpackage under a clean environment of 10 ten thousand.

Example two

Referring to fig. 1, a method for preparing a preparation for relieving fatigue and improving immunity of athletes comprises the following steps:

(1) weighing 51.95kg of L-carnitine, 18kg of taurine, 2.5kg of inositol, 0.035kg of edible fungus extract, 0.25kg of vitamin PP and 0.25kg of vitamin E, weighing 15kg of selenium-enriched soybean and 9kg of microcrystalline cellulose, and respectively crushing and sieving by a sieve of 80 meshes;

(2) uniformly mixing the raw materials which are sieved by the 80-mesh sieve in a V-shaped mixer under the clean environment of 10 ten thousand grades, wherein the rotating speed of the mixer is 150 revolutions per minute, and the time is 6 minutes;

(3) weighing 2.5kg of sodium carboxymethylcellulose under a clean environment of 10 ten thousand grades, dissolving the sodium carboxymethylcellulose in purified water to prepare a sodium carboxymethylcellulose solution with the concentration of 2%, adding the sodium carboxymethylcellulose solution with the concentration of 2% into the uniformly mixed powder obtained in the step II, and uniformly mixing the powder with a groove type mixer to prepare a soft material, wherein the dryness and humidity of the soft material are proper;

(4) under a clean environment of 10 ten thousand grades, the prepared soft material is made into wet granules by a granulator through a 16-mesh screen;

(5) putting the prepared wet particles into a drying disc under a clean environment of 10 ten thousand grade, wherein the thickness of the prepared wet particles is not more than 1.0cm, putting the drying disc into an oven with the temperature of 75-80 ℃, drying and sterilizing, turning the dried material once every 15 minutes, and recording the drying and sterilizing temperature to prevent coking; controlling the moisture content of the granules within a specified range (less than or equal to 5%), and the whole drying and sterilizing process takes about 2.5-3 hours;

(8) pouring the prepared uniformly mixed particles into a tablet machine for tabletting under a clean environment of 10 ten thousand grade to ensure that each tablet weighs 0.5 g;

(9) and adding the tablets into a cleaned and sterilized racking machine for subpackage under a clean environment of 10 ten thousand.

EXAMPLE III

Referring to fig. 1, a method for preparing a preparation for relieving fatigue and improving immunity of athletes comprises the following steps:

(1) weighing 48kg of L-carnitine, 21.5kg of taurine, 2kg of inositol, 0.05kg of edible fungus extract, 0.3kg of vitamin PP and 0.2kg of vitamin E, weighing 16.5kg of selenium-enriched soybean and 8kg of microcrystalline cellulose, and respectively crushing and sieving by a 80-mesh sieve;

(2) uniformly mixing the raw materials which are sieved by the 80-mesh sieve in a V-shaped mixer under the clean environment of 10 ten thousand grades, wherein the rotating speed of the mixer is 150 revolutions per minute, and the time is 6 minutes;

(3) weighing 3kg of sodium carboxymethylcellulose under a clean environment of 10 ten thousand grades, dissolving the sodium carboxymethylcellulose in purified water to prepare a sodium carboxymethylcellulose solution with the concentration of 2%, adding the sodium carboxymethylcellulose solution with the concentration of 2% into the uniformly mixed powder obtained in the step II, and uniformly mixing the powder by using a groove type mixer to prepare a soft material, wherein the dryness and humidity of the soft material are proper;

(4) under a clean environment of 10 ten thousand grades, the prepared soft material is made into wet granules by a granulator through a 16-mesh screen;

(5) putting the prepared wet particles into a drying disc under a clean environment of 10 ten thousand grade, wherein the thickness of the prepared wet particles is not more than 1.0cm, putting the drying disc into an oven with the temperature of 75-80 ℃, drying and sterilizing, turning the dried material once every 15 minutes, and recording the drying and sterilizing temperature to prevent coking; controlling the moisture content of the granules within a specified range (less than or equal to 5%), and the whole drying and sterilizing process takes about 2.5-3 hours;

(6) cooling the dried particles and screening and finishing the particles in a 10 ten thousand grade clean environment;

(7) adding 0.5kg of magnesium stearate into the dry granules under a clean environment of 10 ten thousand grades, and uniformly mixing, wherein the rotating speed of a mixer is 150 revolutions per minute, and the time is 6 minutes;

(8) pouring the prepared uniformly mixed particles into a tablet machine for tabletting under a clean environment of 10 ten thousand grade to ensure that each tablet weighs 0.5 g;

(9) and adding the tablets into a cleaned and sterilized racking machine for subpackage under a clean environment of 10 ten thousand.
