a scented tea with an efficacy of preventing and treating depression comprises the following components in parts by weight: 15-25 parts of saffron petals, 5-15 parts of rose and 1-10 parts of orange peels.

Preferably, the scented tea is prepared from the following raw materials in parts by weight: 15-20 parts of saffron petals, 10-15 parts of rose and 5-10 parts of orange peels.

More preferably, the scented tea is prepared from the following raw materials in parts by weight: 20 parts of saffron petals, 10 parts of rose and 5 parts of orange peels.

The scented tea with the effect of preventing and treating depression can be prepared by adopting the conventional method, and is preferably prepared by the following method: removing impurities from the raw materials, cleaning, drying, and mixing at a certain proportion.

Compared with the prior art, the method has the following advantages:

Embodiments of the present invention will be described in detail below with reference to examples, but those skilled in the art will appreciate that the following examples are only illustrative of the present invention and should not be construed as limiting the scope of the present invention.

Example 1

The preparation method of the safflower petal depression relieving scented tea comprises the steps of microwave drying the safflower petals and the rose, slicing the orange peel, feeding 3000g in total according to the formula proportion, mixing, packaging by adopting heat-sealing type tea filter paper, 3 g/bag, packaging in an outer packaging bag, and packaging by 10 bags and 1 bag to obtain the finished product.

(1) Experimental Material

Weighing saffron petals, roses and orange peels according to the table 1, adding 50mL of 90 ℃ pure water, fully soaking for 30min, filtering, collecting filtrate, and preparing scented tea samples with different ratios.

(2) Experimental methods

Comprehensively establishing a sensory evaluation table (table 1) of the saffron petal depression relieving scented tea according to GB/T23776-2009 tea sensory evaluation method and the characteristics of the scented tea, randomly selecting 10 laymen as examiners, observing soup color, smelling fragrance in a cup, tasting taste, scoring according to table 2, and taking the average score of the scores as a statistical result.

(3) Results of the experiment

The 10 panelists scored different combinations of saffron petal melancholy teas according to the scoring criteria of table 2, and the average score was multiplied by a weighting factor, with the results shown in table 3. In the aspect of liquor color, the crocus petal depression relieving flower tea soup obtained by combining and proportioning is light yellow to light orange, and has higher brightness, so the obtained scores are higher. In terms of fragrance, the crocus petal depression relieving flower tea soup prepared according to the ratio 6 is weak in fragrance, the rest fragrance is rich and pure, and the orange peels with fragrance and qi circulation promoting effects contained in the ratio 6 are possibly less, so that the whole fragrance is not strong enough. In terms of taste, the mouth feel scores of the saffron petal depression relieving flower tea soup prepared according to the proportion 5 and the proportion 7 are slightly lower, probably because the proportion of the rose flower with sweet and slightly bitter taste in the proportion 5 is higher, the proportion of the orange peel with slightly astringent mouth feel in the proportion 7 is higher, and the sample tea soup prepared according to the rest proportions is mellow and fresh, so that higher scores are obtained. Experimental example 1 study on in vitro antidepressant activity of different combinations of saffron petal depression-relieving scented tea

(1) Experimental Material

The scented tea extract with single component and the scented tea with different components are prepared by laboratories. The process is as follows: weighing 20g of saffron petals, 10g of rose and 5g of orange peel, adding 50mL of 90 ℃ pure water, fully soaking for 30min, filtering, and collecting filtrate to obtain a scented tea group 1; weighing 20g of saffron petals, and preparing into a scented tea group 2 by the same method; weighing 10g of rose, and preparing by the same method to obtain a scented tea group 3; weighing 5g of orange peel, and preparing into a scented tea group 4 by the same method; weighing 20g of saffron petals and 10g of roses, and preparing into a scented tea group 5 by the same method; weighing 20g of saffron petals and 5g of orange peel, and preparing into a scented tea group 6 by the same method; 10g of rose and 5g of orange peel are weighed and prepared according to the same method to obtain the scented tea group 7. Fluoxetine hydrochloride tablets (10 mg/tablet, febuxostat, four drugs pharmaceuticals, ltd.).

(2) Experimental methods

PC12 cells were diluted to 2X 10 cells per ml suspension in DMEM medium containing 10% fetal bovine serum5Inoculating the cells into a 96-well culture plate, culturing in a 5% CO2 incubator at 37 ℃ with 100 mu L of each well, removing cell liquid after the cells grow to the bottom of the well, adding a culture solution containing 400 mu M corticosterone (Cort), incubating for 24h, removing the culture solution, adding serum-free DMEM containing 0.5 mg/mL-1 MTT into each well, culturing for 4h, removing DMEM,after the blue particles were completely dissolved, 100. mu.L of dimethyl sulfoxide (DMSO) was added to each well, and the absorbance (A) of the sample at a wavelength of 570nm was measured by a microplate reader to calculate the cell viability.

Cell survival (%) × 100% (experimental group a value/normal group a value).

The cell experiment is divided into (1) normal control group, which comprises sucking off original cell culture fluid and changing to complete culture medium; (2) in the corticosterone group, the original cell culture solution is aspirated and replaced by a complete culture medium containing 400 mu M corticosterone; (3) in fluoxetine group, the original cell culture solution is aspirated and replaced by a complete culture medium containing 400 μ M of corticosterone, and fluoxetine (50 μ M) is added at the same time; test drug groups: the stock cell culture was aspirated and replaced with 400. mu.M complete medium containing corticosterone, while different drugs (200. mu.g/mL) were added. After adding the drug, the cell survival rate is detected by MTT after the culture is continued for 24 hours.

(3) Results of the experiment

After the PC12 cells are damaged by 400 mu M corticosterone for 24h, the cell survival rate is obviously lower than that of a normal control group, and the difference is statistically significant compared with the control group (P < 0.001). As can be seen from fig. 1, the scented tea combination groups 1, 3, 4, 5, 6, and 7 all have certain activities, and the differences have statistical significance (P <0.05) compared with the corticosterone group, wherein the scented tea combination groups 1, 6, and 7 are more obvious, and the three-component scented tea combination group 1 is the most preferable. Therefore, the efficacy of the single-component scented tea for resisting corticosterone-induced PC12 apoptosis is inferior to that of two scented tea combinations, and the effect of the three scented tea combinations for improving corticosterone-induced apoptosis is the best. The experimental results show that the three-component combined scented tea has better in-vitro antidepressant activity, and the three-component combined scented tea has better synergistic effect.

Experimental example 2 influence of saffron petal depression relieving scented tea on chronic unpredictable stress-induced depression mice

(1) Experimental Material

The experimental animal SPF-grade BALB/c mouse is male, has the weight of 18-22 g, and is purchased from model animal center of Nanjing university. Scented tea with different components of medicines and reagents is prepared by laboratories, and the specific process is as follows: weighing 20g of saffron petals, 10g of rose and 5g of orange peel, adding 50mL of 90 ℃ pure water, fully soaking for 30min, filtering, and collecting filtrate to obtain a scented tea group 1; weighing 20g of saffron petals, preparing according to the same method, weighing 2g of scented tea, weighing 10g of rose, preparing according to the same method, weighing 3g of scented tea, weighing 5g of orange peel, preparing according to the same method, weighing 4 g of scented tea, weighing 20g of saffron petals and 10g of rose, preparing according to the same method, weighing 5g of saffron petals and 5g of orange peel, preparing according to the same method, weighing 6 g of scented tea, weighing 10g of rose and 5g of orange peel, preparing according to the same method, and preparing 7 g of scented tea. Fluoxetine hydrochloride tablets (10 mg/tablet, febuxostat, four drugs pharmaceuticals, ltd.). Mouse IL-1 β, IL-6 and TNF- α ELISA kits were purchased from Wuhan Huamei.

(2) Experimental methods

Grouping, dosing and modeling 120 Balbc mice were randomly divided into 10 groups (12/group) by body weight: a blank control group, a model control group, a positive drug (fluoxetine) control group, a scented tea 1 group, a scented tea 2 group, a scented tea 3 group, a scented tea 4 group, a scented tea 5 group, a scented tea 6 group and a scented tea 7 group. The scented tea is administrated by intragastric administration for 3 times every day from day 1, each time is 0.2 mL/flower, the blank group and the model group are administrated with purified water with the same volume, and the positive drug group is administrated with fluoxetine 10mg/kg 1 time every day. Chronic unpredictable stress stimulation started on day 31, following the following specific procedure: mice were subjected to different types of stress: restraint was 4 hours, cage tilt 24 hours, wet bedding 24 hours, swimming 5 minutes in cold water at 4 ℃, swimming 5 minutes in hot water at 45 ℃, shaking horizontally for 10 minutes, tail compression for 1 minute, light/dark cycle inversion for 24 hours, fasted and water deprived overnight. Mice received one of these pressures daily, these 9 pressures were randomly applied for 30 days, and the same pressure was not applied for two consecutive days, to minimize the predictability of each pressure. Three-day behavioural assessment from day 61 onwards, the procedure was as follows 1) sucrose preference test: firstly, carrying out sugar water adaptability exercise, and forbidding water for 24 h. The test was started after 24h, two bottles of 100mL water, one bottle of 1% sugar water and one bottle of pure water, were placed in each mouse cage, and the recording was started after 2h, and the consumption of sugar water and pure water was recorded, respectively. In the experimental process, the amount of each bottle of water needs to be carefully and accurately read, the horizontal placement position needs to be kept consistent, and the mice can be guaranteed to normally drink water. 2) Forced swimming test: the mice were placed individually in 40cm x 20cm glass jars (water 20cm, water temperature around 25 ℃), their activity was observed over 6 minutes using a small animal behavioral analysis system (Etho-Vision XT9), and the fixed time was recorded over the last 4 minutes over 6 minutes. Mice are judged to be standing still when they stop struggling and float on the water surface or only do some small movements to keep their heads on the water. After the experiment of each rat is finished, the water tank needs to be cleaned, water needs to be changed, and then the next rat is tested continuously to avoid interference. 3) And (3) tail suspension test: a small clamp is connected with the center of the top plate of the tail suspension box (25cm multiplied by 35 cm). The tail part of the mouse is stuck to the position 2cm away from the tail tip by using an adhesive tape, the adhesive tape is clamped by using a clamp, the mouse is suspended in a black tail suspension box, and the head part of the mouse is 5cm away from the box bottom. The animals were observed for activity within 6 minutes using the MedLab BioInformatics collection System (MedLab-U/4C501H, Napkinsonkyu technologies, Inc. China) and the time to immobility was recorded for the last 4 minutes. The mouse is fixed when the mouse stops struggling and is in a static vertical suspension state. Mouse hippocampal tissue was isolated starting at day 64 and assayed for levels of the relevant inflammatory factors IL-1 β, IL-6 and TNF- α.

(3) Results of the experiment

a) Influence of different scented tea groups on sucrose preference experiment

As shown in fig. 2, the sugar water preference index of the mice in the model control group was significantly decreased compared to the normal control group. Compared with a model control group, the fluoxetine hydrochloride group and the scented tea group have obviously improved sucrose preference index of mice. The scented tea group 1 has a particularly obvious effect. The results show that the scented tea group 1 consisting of 20g of saffron petals, 10g of roses and 5g of orange peels is obviously superior to a single component or the combination of two components.

b) Influence of different scented tea groups on forced swimming experiment

As shown in table 4, the activity resting time of the model group mice was significantly prolonged (P <0.01) compared to the normal control group. Compared with the model control group, the fluoxetine group and the scented tea group can obviously shorten the resting time. The immobility time of the scented tea 1 is obviously reduced. The results show that the scented tea group 1 consisting of 20g of saffron petals, 10g of roses and 5g of orange peels is obviously superior to a single component or the combination of two components.

c) Influence of different scented tea groups on tail suspension test experiment

As shown in table 4, the increased immobility time of the tail suspension test experiment in the model group mice was significantly prolonged compared to the normal control group. Compared with a model control group, the fluoxetine group and the scented tea group can obviously shorten the immobility time. Compared with other scented tea groups, scented tea 1 obviously reduces immobility time of mice. The results show that the scented tea group 1 consisting of 20g of saffron petals, 10g of roses and 5g of orange peels is obviously superior to a single component or the combination of two components.

d) Unpredictable weight change after chronic mild stress of different scented tea groups

As shown in FIG. 3, the body weight of the mice in the model group was significantly reduced, and the body weight of the mice in the fluoxetine (10mg/kg) and scented tea group was increased compared to the body weight of the mice in the model group. The mice in scented tea group 1 had insignificant weight loss. The results show that the scented tea group 1 consisting of 20g of saffron petals, 10g of roses and 5g of orange peels is obviously superior to a single component or the combination of two components.

e) Influence of different scented tea groups on inflammatory factor level in hippocampus

As shown in FIG. 4, the levels of IL-1 β, IL-6, TNF- α were significantly increased in the hippocampus of the model group (P <0.05) compared to the normal control group. Compared with the model group, the content of the hippocampus of the mouse in the scented tea group is obviously reduced (P is less than 0.05), wherein the scented tea group 1 is most obviously reduced. The results show that the scented tea group 1 consisting of 20g of saffron petals, 10g of roses and 5g of orange peels is obviously superior to a single component or the combination of two components.

The experiments fully confirm that the scented tea composed of the saffron petals, the roses and the orange peels has a synergistic effect and has a remarkable anti-depression effect.
