1. The preparation method of the beautiful millettia root extract is characterized by comprising the following steps:

(1) 1g of beautiful millettia root and a solvent according to the mass-volume ratio of 1 g: mixing 9-11 ml of the raw materials, and soaking;

(2) extracting for 1-2 h, collecting the extracting solution, and filtering to obtain the liquid medicine containing the beautiful millettia root extract.

2. The preparation method according to claim 1, wherein when the solvent in the step (1) is water, the soaking time is 1-3 h, and the extraction in the step (2) is boiling extraction.

3. The preparation method according to claim 1, wherein when the solvent in step (1) is 70-80% ethanol water by volume, the soaking time is 10-14 h, and the extraction in step (2) is heating reflux extraction at 90-110 ℃.

4. The preparation method according to any one of claims 2 or 3, wherein the step (2) of obtaining the radix Millettiae Speciosae extract-containing liquid medicine further comprises concentrating and drying the radix Millettiae Speciosae extract-containing liquid medicine.

5. The preparation method according to claim 4, wherein the concentration is rotary evaporation concentration, the condensation temperature of the rotary evaporation concentration is-2 to 3 ℃, and the heating temperature is 35 to 60 ℃.

The method for preparing the beautiful millettia root extract according to claim 7, characterized in that the precipitate is vacuum-dried to obtain beautiful millettia root extract which is beautiful millettia root polysaccharide; and concentrating and drying the filtrate to obtain the beautiful millettia root extract which is the beautiful millettia root polysaccharide filtrate.

9. The Millettia speciosa champ extract prepared by the preparation method according to any one of claims 1 to 8.

10. Use of the millettia speciosa champ extract of claim 9 for the preparation of a medicament for lowering blood lipids and/or weight loss.

Background

Obesity and dyslipidemia are important risk factors for cardiovascular and cerebrovascular diseases. With the increase of living standard of people, the obesity and overweight people rapidly rise, the obesity becomes one of the great factors causing human death, and at least 280 ten thousand adults die every year in the world due to obesity-related diseases. Hyperlipidemia is one of the most typical complications of obesity, and 1200-1600 million people die of cardiovascular diseases caused by hyperlipidemia every year in the world. Therefore, the research and development of lipid-lowering and weight-losing medicines have great significance in both scientific and civil aspects, and the development potential is great. At present, weight-loss drugs such as sibutramine on the market have side effects such as abnormal liver function although they have some effects.

Radix Millettiae Speciosae is root of Millettia speciosa of Millettia of Leguminosae, and is named as radix Chimaphilae, radix Stephaniae Tetrandrae, rhizoma Nelumbinis, radix seu caulis Kadsurae Heteroclitae, and DALI. Produced in southern areas of China such as Fujian, Hunan, Guangdong, Guangxi, Hainan, Yunnan and Guizhou. The record of the materia medica records beautiful millettia root, strong bones and muscles, detoxication, internal injury regulation, traumatic injury treatment, wine infusion and kidney nourishing. The Chinese herbal medicine compilation records that the beautiful millettia root has sweet nature and flavor, is mild, tonifies deficiency and moistens lung, strengthens tendons and activates collaterals, and is used for lumbar muscle strain, rheumatoid arthritis, phthisis, chronic bronchitis, chronic hepatitis, spermatorrhea and leucorrhea. The Chinese plant record states that the beautiful millettia root has rich starch, can be used for brewing wine and can be used as a medicine, and has the functions of dredging the meridian passage, tonifying deficiency, moistening lung and strengthening spleen. Radix Millettiae Speciosae contains protein, polysaccharide, flavonoid, terpenoids, alkaloid, etc., and has effects of enhancing immunity, eliminating phlegm, relieving cough, resisting inflammation and relieving fatigue. The beautiful millettia root is commonly used for cooking soup, making medicated diet, medicinal liquor and the like in southern areas of China, such as Guangdong, Guangxi, Hainan and other areas, and has the remarkable effects of tonifying kidney and waist and strengthening tendons and bones. The health care and edible value of the beautiful millettia root is very high, and the beautiful millettia root is also a raw material medicine of Chinese patent medicines such as waist strengthening and kidney invigorating pills, strong body building capsules, and radix astragali tendon relaxing and collateral dredging tablets. However, the application of beautiful millettia root medicinal materials has not been developed completely, and the pharmacopoeia does not contain beautiful millettia root medicinal materials.

Disclosure of Invention

In view of the above, the present invention aims to provide a beautiful millettia root extract, a preparation method thereof and applications thereof in medicines for reducing blood fat and losing weight.

In order to achieve the above purpose, the invention provides the following technical scheme:

the invention provides a preparation method of a beautiful millettia root extract, which comprises the following steps:

(1) 1g of beautiful millettia root and a solvent according to the mass-volume ratio of 1 g: mixing 9-11 ml of the raw materials, and soaking;

(2) extracting for 1-2 h, collecting the extracting solution, and filtering to obtain the liquid medicine containing the beautiful millettia root extract.

Preferably, when the solvent in the step (1) is water, the soaking time is 1-3 h, and the extraction in the step (2) is boiling extraction.

Preferably, when the solvent in the step (1) is ethanol water solution with the volume fraction of 70-80%, the soaking time is 10-14 h, and the extraction in the step (2) is heating reflux extraction at 90-110 ℃.

Preferably, when the solvent in the step (1) is water or an ethanol aqueous solution with a volume fraction of 70-80%, after the medicinal liquid containing the beautiful millettia root extract is obtained in the step (2), the method further comprises concentrating and drying the medicinal liquid containing the beautiful millettia root extract.

Preferably, the concentration is rotary evaporation concentration, the condensation temperature of the rotary evaporation concentration is-2 to 3 ℃, and the heating temperature is 35 to 60 ℃.

Preferably, the drying is vacuum drying, the vacuum degree of the vacuum drying is 0.1Mpa, the drying is carried out for 2 hours at 100 ℃, and then the drying is carried out for 3 hours at 80 ℃.

Preferably, when the solvent in the step (1) is water, after the medicinal liquid containing the beautiful millettia root extract is obtained in the step (2), the method further comprises the steps of mixing the medicinal liquid with absolute ethyl alcohol until the volume fraction of the absolute ethyl alcohol in the mixed liquid is 80-90%, standing for 46-50 h, and filtering to obtain filtrate and precipitate.

Further, vacuum drying the precipitate to obtain the beautiful millettia root extract which is beautiful millettia root polysaccharide; and concentrating and drying the filtrate to obtain the beautiful millettia root extract which is the beautiful millettia root polysaccharide filtrate.

The invention also provides the beautiful millettia root extract prepared by the preparation method.

The invention also provides application of the beautiful millettia root extract in preparing medicines for reducing blood fat and/or losing weight.

Compared with the prior art, the invention has the following beneficial effects:

the invention discovers for the first time that the beautiful millettia root extract comprises a beautiful millettia root water extract, beautiful millettia root polysaccharide, a beautiful millettia root polysaccharide filtrate and a beautiful millettia root alcohol extract, can inhibit the proliferation of precursor fat cells, inhibit the differentiation of fat cells and inhibit the formation of fat droplets, can reduce blood fat and lose weight, can be applied to blood fat reducing and/or weight losing medicines, provides scientific basis for reducing blood fat and losing weight of the beautiful millettia root, solves the blank of the beautiful millettia root in the field of reducing fat and losing weight, and expands the application range of the beautiful millettia root.

Detailed Description

The invention provides a preparation method of a beautiful millettia root extract, which comprises the following steps:

(1) 1g of beautiful millettia root and a solvent according to the mass-volume ratio of 1 g: mixing 9-11 ml of the raw materials, and soaking;

(2) extracting for 1-2 h, collecting the extracting solution, and filtering to obtain the liquid medicine containing the beautiful millettia root extract.

The source of the beautiful millettia root is not specially limited, the beautiful millettia root is prepared from conventional commercial products, and beautiful millettia root decoction pieces are preferably adopted; in the present invention, preferably, the beautiful millettia root is pulverized and mixed with the solvent, the pulverized particle size is preferably 2mm, and the pulverized product is preferably sieved by a 10-mesh sieve; the pulverization is preferably carried out by dry pulverization by a pulverizer.

In the present invention, when the solvent in step (1) is preferably water, the mass-to-volume ratio of beautiful millettia root to water is 1 g: mixing 9-11 ml of the raw materials, and soaking; extracting for 1-2 h, collecting an extracting solution, and filtering to obtain a liquid medicine containing the beautiful millettia root aqueous extract; concentrating and drying to obtain the beautiful millettia root aqueous extract.

The invention has no special limitation on water, and can be realized by adopting water which is conventional in the field; the mixing mode of the beautiful millettia root and the water is not specially limited, and the conventional mixing mode in the field is adopted; in the present invention, the mass-to-volume ratio of beautiful millettia root to water is preferably 1 g: 10 ml. In the invention, the soaking time is preferably 1-3 h, more preferably 2h, and the soaking temperature is preferably 17-23 ℃, more preferably 20 ℃; in the present invention, the number of times of extraction is preferably 1 to 3 times, more preferably 2 times, and the preferred mode of each extraction is 230 to 270 ℃ heating and boiling, then 100 to 130 ℃ extraction, more preferably 250 ℃ heating and boiling, then 115 ℃ extraction, and the time for each extraction is preferably 1.5 hours. In the invention, the extracting solutions obtained by several times of extraction are combined and filtered; the filtering mode is preferably double-layer 200-mesh silk cloth filtering; in the invention, the concentration is preferably rotary evaporation concentration, the condensation temperature of the rotary evaporation concentration is preferably-2-3 ℃, the heating temperature is preferably 35-60 ℃, and the volume ratio of the concentrate obtained by the rotary evaporation concentration to the original solution is preferably 1: 10-20; in the invention, the drying is preferably vacuum drying, and the conditions of the vacuum drying are preferably 0.1Mpa, 100 ℃ for 2h, and 80 ℃ for 3 h.

In the invention, when the solvent in the step (1) is preferably water, the method further comprises the steps of mixing the liquid medicine containing the beautiful millettia root aqueous extract with absolute ethyl alcohol until the volume fraction of the absolute ethyl alcohol in the mixed liquid is 80-90%, standing for 46-50 h, and filtering to obtain filtrate and precipitate; vacuum drying the precipitate to obtain radix millettiae speciosae extract which is radix millettiae speciosae polysaccharide; and concentrating and drying the filtrate to obtain the beautiful millettia root extract which is the beautiful millettia root polysaccharide filtrate.

The mixing mode of the liquid medicine containing the beautiful millettia root aqueous extract and the absolute ethyl alcohol is not specially limited, and the conventional mixing mode in the field is adopted; the invention has no special limitation on the absolute ethyl alcohol, and the absolute ethyl alcohol is conventionally used in the field; in the present invention, the volume fraction of the anhydrous ethanol in the mixed solution is preferably 85%; in the invention, the standing time is preferably 48h, and the standing temperature is preferably 17-23 ℃, more preferably 20 ℃; in the invention, the concentration is preferably rotary evaporation concentration, the condensation temperature of the rotary evaporation concentration is preferably-2-3 ℃, the heating temperature is preferably 35-60 ℃, and the volume ratio of the concentrate obtained by the rotary evaporation concentration to the original solution is preferably 1: 15-17, more preferably 1: 16; in the invention, the drying is preferably vacuum drying, and the conditions of the vacuum drying are preferably 0.1Mpa, 100 ℃ for 2h, and 80 ℃ for 3 h.

In the invention, when the solvent in the step (1) is preferably ethanol water with a volume fraction of 70-80%, the mass-volume ratio of the beautiful millettia root to the ethanol water with a volume fraction of 70-80% is 1 g: mixing 9-11 ml of the raw materials, and soaking; extracting for 1-2 h, collecting an extracting solution, and filtering to obtain a liquid medicine containing the millettia speciosa champ alcohol extract; concentrating and drying to obtain the beautiful millettia root alcohol extract.

In the present invention, the solvent is more preferably an aqueous ethanol solution with a volume fraction of 75%; the mixing mode of the invention is not particularly limited, and the conventional mixing mode in the field is adopted; in the present invention, the mass-to-volume ratio of beautiful millettia root to the ethanol aqueous solution is preferably 1 g: 10 ml; in the invention, the soaking time is preferably 10-14 h, more preferably 12h, and the soaking temperature is preferably 17-23 ℃, more preferably 20 ℃; in the invention, the extraction mode is preferably heating reflux extraction, the heating reflux extraction temperature is preferably 90-110 ℃, more preferably 100 ℃, and the extraction time is preferably 1.5 h; in the invention, the filtering mode is preferably filtering by using double-layer 200-mesh silk cloth; in the invention, the concentration is preferably rotary evaporation concentration, the condensation temperature of the rotary evaporation concentration is preferably-2-3 ℃, the heating temperature is preferably 35-60 ℃, and the volume ratio of the concentrate obtained by the rotary evaporation concentration to the original solution is preferably 1: 15-17, more preferably 1: 16; in the invention, the drying is preferably vacuum drying, and the conditions of the vacuum drying are preferably 0.1Mpa, 100 ℃ for 2h, and 80 ℃ for 3 h.

The invention also provides the beautiful millettia root extract prepared by the preparation method, and the beautiful millettia root extract is preferably a beautiful millettia root water extract, a beautiful millettia root alcohol extract, beautiful millettia root polysaccharide or beautiful millettia root polysaccharide filtrate.

The invention also provides application of the beautiful millettia root extract in preparing medicines for reducing blood fat and/or losing weight. In the present invention, the hypolipidemic and/or anti-obesity drug preferably comprises the beautiful millettia root extract and a pharmaceutically acceptable carrier. In the invention, the mass percentage content of the beautiful millettia root extract in the medicine is preferably 10-25%.
