s1, taking out viscera of fresh sea cucumbers, cleaning, putting the fresh sea cucumbers into a normal-temperature stirring pot, adding a proper amount of glacial acetic acid, starch and water into the stirring pot, soaking for 30min, then stirring at a low speed for 7min, taking out the sea cucumbers, and washing the sea cucumbers clean again for later use;

s2, grinding the sea cucumbers processed in the step S1 at a high speed to obtain sea cucumber slurry, putting the sea cucumber slurry into a sterile anti-reaction kettle, adding soda water and anthocyanin, stirring and uniformly mixing, transferring into a 0 ℃ refrigerating chamber, standing and refrigerating for 2.5 hours, and transferring out of the refrigerating chamber for later use;

s3, adding xylanase and protease into the sea cucumber slurry mixture treated in the step S2, stirring, slowly heating to 42 ℃, preserving heat, continuously and slowly stirring, and carrying out enzymolysis reaction for 4.5 hours under the condition that the pressure is 80 KPa;

s4, respectively grinding barley, peach gum, pummelo peel, angelica, lily, medlar, pawpaw and dried longan into powder for later use; parching brown rice with slow fire until the surface is yellowish, cooling, adding small amount of purified water, and grinding into brown rice pulp;

s5, adding the barley, peach gum, pummelo peel powder and brown rice pulp for later use into a reaction kettle while stirring, keeping the temperature at 42 ℃, continuously and slowly stirring for reaction for 1 hour, adding the peach gum, pummelo peel, angelica, lily, medlar, pawpaw and dried longan which are ground into powder, and purified water, stopping stirring, cooling to room temperature, homogenizing for 3-5min, discharging, adsorbing by activated carbon, filtering, and filling to obtain a finished product 3.

Wherein the adding ratio of the protease to the xylanase is 5: 1; the protease comprises mixed enzyme of subtilisin, trypsin and papain, and the addition ratio of the subtilisin, the trypsin and the papain is 2:1: 1.

Comparative example 1: the formula of the health product of example 3 and the corresponding preparation method were used to prepare a comparative product 1 without adding soda water.

Comparative example 2: the anthocyanin was not added, and the other ingredients were prepared using the formula of the health product of example 3 and the corresponding preparation method to give comparative product 2.

Comparative example 3: no xylanase is added, in order to keep the enzyme content consistent, the xylanase is replaced by protease, and the formula of the health care product in the embodiment 3 and the corresponding preparation method are adopted for preparation, so that a comparative finished product 3 is obtained.

Comparative example 4: the formula of the health product of example 3 and the corresponding preparation method were used to prepare a comparative product 4 without peach gum.

Comparative example 5: no exocarpium Citri Grandis was added, and the formula of the health product of example 3 and the corresponding preparation method were used to obtain comparative product 5.

Comparative example 6: no barley was added, the brown rice was used without being roasted, and the comparative product 5 was obtained by using the formula of the health food of example 3 and the corresponding preparation method.

Control group 1: the well-sold sea cucumber health care product purchased in the market is used as the product of the control group 1.

1. Long-term toxic and side effects tests were performed on the health products obtained in examples 1 to 3 and comparative examples 1 to 6 of the present invention.

Selecting 45 healthy rats with the same batch and the weight of 90 +/-10 g, randomly grouping the rats, wherein each group comprises 5 rats and 9 groups, feeding the same feed and water to all the rats tested in the whole experimental process, respectively carrying out tail vein injection administration on the rats of each group by using the finished products prepared in the examples 1-3 and the comparative examples 1-6, wherein the administration dose is 10ml/Kg, carrying out regular injection once a day, continuously administering for 3 months, measuring the weight of the rats every day, adjusting the administration dose according to the weight, and recording the health and development conditions of the rats.

After 2 months, the administration is stopped, the condition of the rats is recorded for 28 days, then 3 rats in each group are randomly selected and killed, blood is taken for measuring blood routine and blood biochemical indexes, internal tissues are planed, and the pathological histology of visceral organs of the planed rats is examined.

The test results show that: all rats tested were in stock, none died, and gained normal weight and developed normally, after 2 months of dosing and 28 days of drug withdrawal; the detection results of the blood routine and the blood biochemical index of each group of rats are normal; pathological histology examination is carried out on visceral organs of the debrided rat, and no pathological changes and abnormalities are found in brain, heart, liver, thymus, kidney, spleen, intestines and stomach and the like of the rat. The preparation has the advantages of mild components, no toxic or side effect after long-term use and good safety.

2. The hydrolysis degree of the sea cucumber protein is measured and tested: the Degree of hydrolysis of the sea cucumber proteins of examples 1 to 3 and comparative examples 1 to 6 was determined by trinitrobenzene sulfonic acid (TNBS), which is a method for determining the Degree of reliable hydrolysis of proteins, wherein the Degree of Hydrolysis (DH) of proteins is a percentage of peptide bonds in proteins that are hydrolyzed, and is calculated by using the following calculation formula:

DH% -the number of peptide bonds hydrolyzed/total number of peptide bonds in the protein X100%

=h/htot×100%

Wherein h refers to the millimole number of peptide bonds (mmol/g protein) cleaved per gram protein after the hydrolysis of sea cucumber protein, and h refers tototThe number of millimoles of peptide bonds in each gram of sea cucumber protein is shown, and when all the peptide bonds in the protein are hydrolyzed to generate free amino acids, the hydrolysis degree is 100 percent; when peptide bonds in the protein are not hydrolyzed, the degree of hydrolysis is 0. The final calculation and statistics are shown in fig. 1.

As can be seen from the results shown in fig. 1, the hydrolysis degree of the holothurian protein in the preparation methods of the holothurian health care products of examples 1 to 3 and comparative examples 4 to 6 is much higher than that in the preparation methods of the holothurian health care products of comparative examples 1 to 3, so that the method is proper in the process of processing holothurian, and the addition of soda water, anthocyanin and xylanase in the preparation process plays a critical role in the hydrolysis of holothurian protein, and the three are deficient and need to supplement each other, so that the efficient hydrolysis of holothurian protein can be realized in a relatively mild environment.

In particular, the preparation method of example 3, which has a degree of proteolysis as high as 74.4%, is also significantly higher than the preparation method of examples 1-2, and it can be seen that when the ratio of protease to xylanase is 5: 1; when the addition ratio of the subtilisin, the trypsin and the papain contained in the protease is 2:1:1, the effect of remarkably improving the degree of proteolysis can be achieved, which is also the best collocation and proportion discovered by the team of the invention through long-term research.

3. Test for improving immunocompetence: 55 mice of 35-40 g size purchased from the same batch are randomly divided into 11 groups of 5 mice each, and the mice are fed with the same feed in the same environment; wherein the health care products of examples 1-3, comparative examples 1-6 and control group 1 are respectively added into the drinking purified water of the mice of the 1 st to 10 th groups, and the adding amount is 5 percent; group 11 mice were fed for 25 days with purified water alone as drinking water.

The mouse eyeball blood sampling is carried out on each group of mice, the serum lysozyme content is determined, the agar plate with micrococcus and standard lysozyme with different concentrations are adopted, the mouse serum is diluted into samples with different concentrations, the samples are placed in a thermostat for 24 hours, the diameter of a lysozyme ring is measured, a standard curve is drawn, so that the lysozyme content of each serum sample is obtained, and the results are summarized as shown in the following table 2. The anticoagulation of heparin is adopted for each group of mice, the phagocytic function of white blood cells in blood is measured, the inactivated staphylococcus albus suspension is adopted for testing, the phagocytic speed and the phagocytic rate of the anticoagulation of the heparin of each group of mice are calculated, and the results are summarized as shown in the following table 2.

As is apparent from the results in table 2 above, the sea cucumber health products prepared in examples 1 to 3 have much more significant effects on enhancing the immunity, including increasing the content of serum lysozyme and increasing the phagocytic capacity of leukocytes, than the top-market sea cucumber health products purchased in control group 1, while the sea cucumber health products prepared in comparative examples 1 to 3 have much less effect on enhancing the immunity than the top-market sea cucumber health products prepared in examples 1 to 3, and even have less effect than the top-market sea cucumber health products purchased in control group 1. Therefore, the health care effects of the sea cucumber health care products obtained in the embodiments 1 to 3 are more comprehensive and efficient, and the hydrolysis degree of the sea cucumber protein and the retention degree of the active substances can be remarkably improved in the preparation processes of the embodiments 1 to 3, so that the overall efficiency is improved, therefore, the soda water, the anthocyanin and the xylanase added in the preparation processes not only play a critical role in the hydrolysis of the sea cucumber protein, but also can effectively avoid the loss or the loss of the active substances in the sea cucumber, so that the nutrient substances in the sea cucumber can be efficiently and fully utilized, and the effects of the sea cucumber health care products are improved.

4. And (3) taking effect test: after long-term toxic and side effects are completed and the results show safety and no toxic and side effects, and the health detection standards of food and drug products are passed, volunteer effect comparison tests are performed on the products of the above examples 1-3, comparative examples 1-6 and control group 1.

In the same area, 50 volunteers aged 35-55 years were collected and randomly divided into 10 groups of 5 persons, each volunteer had wrinkles of different depths on the face and had occasional insomnia symptoms, general mental state and no other serious diseases. The 10 groups of volunteers respectively drunk the health products of examples 1 to 3, comparative examples 1 to 6 and control group 1 once a day, 15ml each time, immediately stopped taking if any physical discomfort occurs in the period, and immediately hospitalized, each volunteer fed back the physical condition and the feeling after taking to the following person every day for 28 days, and finally summarized the test data and analyzed to obtain the test results as shown in the following table 3.

As is apparent from the test results in table 3 above, compared with the commercially available sea cucumber health products in the control group 1, the sea cucumber health products prepared in the embodiments 1-3 have excellent anti-wrinkle effects, and can significantly reduce or slowly lighten deep wrinkles on the face, and slowly improve skin vitality, while the sea cucumber health products prepared in the comparative examples 4-5 have much less anti-wrinkle and skin-tendering effects than the sea cucumber health products prepared in the embodiments 1-3, and the anti-wrinkle effects of the health products prepared in the comparative examples 1-3 are also significantly reduced, so that the synergistic effect of peach gum and pummelo peel can be seen, which has a critical effect in exerting excellent anti-wrinkle effects, and simultaneously needs to interact with active substances in the sea cucumber, and if the loss of the active substances in the sea cucumber is large, the anti-wrinkle effect can be influenced, only the active substances in the peach gum, the pummelo peel and the sea cucumber interact with each other to exert obvious effects of resisting wrinkles and tendering skin, so that the skin becomes young and active.

The test results in table 4 show that the top-selling sea cucumber health care product purchased in the market of the control group 1 is sweet and greasy in taste and is not suitable for the elderly to eat, even the diabetics eat with caution, while the sea cucumber health care products prepared in the comparative examples 3 and 6 are far from the taste of the sea cucumber health care products prepared in the embodiments 1 to 3, so that the mouth feeling of the xylanase, barley and brown rice can be obviously improved in the preparation process, the health care products are sweet but not sweet and greasy in taste, and the appetite of the health care products can be improved. While the sea cucumber health care products obtained in the examples 1 to 3 are mild and friendly to the intestines and the stomach, and the problems of dyspepsia or discomfort of the intestines and the stomach cannot occur, while the health care products in the comparative examples 1 to 3 and 6 and the control group 1 have certain stimulation to the intestines and the stomach and are not mild enough, so that the barley and the roasted brown rice are added, the full hydrolysis of the sea cucumber protein is ensured, the intestines and the stomach can be effectively protected, and the digestion and the absorption of the intestines and the stomach are facilitated.

In conclusion, the sea cucumber health care product prepared by the invention can fully hydrolyze sea cucumber protein in a relatively mild environment, simultaneously retain more active ingredients, has better health care effect, has the effects of beautifying and resisting wrinkles, enables skin to recover young vitality, is mild to intestines and stomach, does not generate toxic or side effect after being eaten for a long time, and ensures the ingredient safety.
