1. A preparation method of a traditional Chinese medicine extract with an effect of treating myocarditis is characterized by comprising the following steps:

(1) mixing Ginseng radix and Tibetan herba Artemisiae Scopariae, extracting with ethanol under reflux, and concentrating the extractive solution until there is no ethanol smell to obtain extractive solution;

(2) adding ethyl acetate into the extracting solution for extraction, and concentrating and drying the extracting solution to obtain an ethyl acetate extract;

(3) and (3) applying the ethyl acetate extract to a silica gel column, and eluting by using a mixed organic solvent to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

2. The method for preparing the extract of Chinese traditional medicine for treating myocarditis according to claim 1, wherein the ethanol in step (1) is an aqueous ethanol solution with a volume fraction of 10% -30%; preferably, the ethanol in the step (1) is ethanol water solution with 15 percent of volume fraction.

3. The preparation method of the traditional Chinese medicine extract with the effect of treating myocarditis according to claim 1, wherein in the step (1), the weight ratio of ginseng to Tibetan capillaris is 1-3: 1-3; preferably, the weight ratio of ginseng to Tibetan capillaris is 2: 1.

4. The method for preparing a Chinese medicinal extract for treating myocarditis according to claim 1, wherein the volume of ethyl acetate used in step (2) is 0.5-2 times of the volume of the extractive solution.

eluting and removing impurities by using a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 85-88: 15-12; then eluting with a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 80-83: 20-17, collecting the eluted part eluted by the mixed organic solvent consisting of chloroform and methanol in a volume ratio of 80-83: 20-17, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Technical Field

The invention relates to the technical field of traditional Chinese medicines, and in particular relates to a traditional Chinese medicine extract with an effect of treating myocarditis as well as a preparation method and application thereof.

Background

Myocarditis refers to a disease that is mainly manifested by a localized or diffuse inflammatory disease of the myocardium, and is clinically manifested in various forms ranging from asymptomatic to severe arrhythmia, acute cardiac insufficiency, cardiogenic shock and even death. Common viral infection or immune response following viral infection is one of the common modes leading to myocarditis, with viruses that cause infections of the intestinal and upper respiratory tract being the most prevalent; such as echoviruses, polioviruses, coxsackie virus group a and coxsackie virus group B; the coxsackie B group virus is the most main pathogenic virus. Therefore, the further development of a medicament for treating myocarditis, in particular to the development of a medicament for treating viral myocarditis, has important significance.

The development of a drug having an effect of treating myocarditis from traditional Chinese medicines is one of the current research directions. The Chinese medicinal Ginseng radix is root of Panax ginseng C.A. Meyer of Araliaceae; it is used for invigorating primordial qi, restoring pulse, relieving depletion, invigorating spleen, benefiting lung, promoting fluid production, and tranquilizing; can be used for treating asthenia, collapse, cold limbs, slight pulse, spleen deficiency, anorexia, cough, asthma, and thirst due to body fluid consumption. The Tibetan capillary wormwood is a perennial herb of Artemisia of Compositae, and has the effects of clearing heat and eliminating dampness, and protecting liver and benefiting gallbladder. At present, no research report is available for preparing a traditional Chinese medicine extract with an effect of treating myocarditis from ginseng and Tibetan capillaris serving as raw materials.

Disclosure of Invention

In view of the above, the invention firstly provides a preparation method of a traditional Chinese medicine extract with an effect of treating myocarditis, and researches show that the traditional Chinese medicine extract prepared by the method has excellent activity of treating myocarditis.

A preparation method of a traditional Chinese medicine extract with an effect of treating myocarditis is characterized by comprising the following steps:

(1) mixing Ginseng radix and Tibetan herba Artemisiae Scopariae, extracting with ethanol under reflux, and concentrating the extractive solution until there is no ethanol smell to obtain extractive solution;

(2) adding ethyl acetate into the extracting solution for extraction, and concentrating and drying the extracting solution to obtain an ethyl acetate extract;

(3) and (3) applying the ethyl acetate extract to a silica gel column, and eluting by using a mixed organic solvent to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Preferably, the ethanol in the step (1) is ethanol water solution with the volume fraction of 10-30%; preferably, the ethanol in the step (1) is ethanol water solution with 15 percent of volume fraction.

Preferably, the dosage ratio of the ethanol water solution to the total weight of the ginseng and the Tibetan capillary artemisia is 8-15 mL: 1g of a compound;

researches show that the volume fraction of ethanol plays an important role in whether the traditional Chinese medicine extract with excellent effect of treating myocarditis can be prepared from ginseng and Tibetan capillaris, and the ginseng and Tibetan capillaris are extracted by adopting ethanol aqueous solution with the volume fraction of 10-30%, so that the traditional Chinese medicine extract with excellent effect of treating myocarditis can be prepared in the subsequent steps of the invention.

Preferably, the weight ratio of the ginseng to the Tibetan capillary artemisia in the step (1) is 1-3: 1-3; preferably, the weight ratio of ginseng to Tibetan capillaris is 2: 1.

Preferably, the volume of the ethyl acetate in the step (2) is 0.5-2 times of the volume of the extracting solution.

Preferably, the specific method for eluting with the mixed organic solvent in the step (3) is as follows:

eluting and removing impurities by using a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 85-88: 15-12; then eluting with a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 80-83: 20-27, collecting the eluted part eluted by the mixed organic solvent consisting of chloroform and methanol in a volume ratio of 80-83: 20-27, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Further preferably, the specific method for eluting with the mixed organic solvent in the step (3) is as follows:

The specific method for eluting by mixing organic solvents plays a decisive role in preparing the traditional Chinese medicine extract with the effect of treating myocarditis. The composition and elution conditions of the mixed organic solvent are not proper, and even the traditional Chinese medicine extract with the effect of treating myocarditis cannot be prepared. Research shows that the mixed organic solvent consisting of chloroform and methanol is selected, and the traditional Chinese medicine extract prepared under the elution condition has excellent effect of treating myocarditis.

The invention also provides a traditional Chinese medicine extract with the effect of treating myocarditis, which is prepared by the preparation method.

The invention also provides application of the traditional Chinese medicine extract in preparing a medicine with the effect of treating myocarditis.

Preferably, the myocarditis is viral myocarditis.

Most preferably, the viral myocarditis is myocarditis caused by a coxsackievirus.

Has the advantages that: the invention provides a brand-new traditional Chinese medicine extract, which has an excellent effect of treating myocarditis. Research shows that the traditional Chinese medicine extract prepared from ginseng and Tibetan capillaris by adopting the method of the invention has more excellent effect of treating myocarditis than the traditional Chinese medicine extract prepared from any one of ginseng and Tibetan capillaris by adopting the method of the invention. The traditional Chinese medicine extract has excellent effect of treating myocarditis; therefore, the traditional Chinese medicine extract can be used as an active ingredient for preparing a medicament with the effect of treating myocarditis.

The present invention is further explained below with reference to specific examples, which are not intended to limit the present invention in any way.

Example 1 preparation of extract of Chinese medicine having effect of treating myocarditis

(1) Mixing dried Ginseng radix and Tibetan herba Artemisiae Scopariae at a weight ratio of 2:1, heating and reflux-extracting with 15% ethanol water solution for 2 hr, and concentrating the extractive solution until there is no ethanol smell to obtain extractive solution; the dosage ratio of the ethanol water solution to the total weight of the ginseng and the Tibetan capillary artemisia is 10 mL: 1g of a compound;

(2) adding ethyl acetate with the same volume as the extracting solution into the extracting solution for extraction, and concentrating and drying the extract to obtain an ethyl acetate extract;

(3) passing the ethyl acetate extract through a silica gel column (the silica gel in the silica gel column is 200-300 mesh silica gel; the weight and the dosage of the silica gel are 30 times of the weight of the ethyl acetate extract), eluting and removing impurities by using a mixed organic solvent (the dosage is 6 times of the column volume) composed of chloroform and methanol with the volume ratio of 86:14, and then eluting by using a mixed organic solvent (the dosage is 4 times of the column volume) composed of chloroform and methanol with the volume ratio of 82: 18; collecting the eluted part eluted by a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 82:18, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Example 2 preparation of extract of Chinese traditional medicine for treating myocarditis

(1) Mixing dried Ginseng radix and Tibetan herba Artemisiae Scopariae at a weight ratio of 1:1, extracting with 30% ethanol water solution under reflux for 2 hr, and concentrating the extractive solution until there is no ethanol smell to obtain extractive solution; the dosage ratio of the ethanol water solution to the total weight of the ginseng and the Tibetan capillary artemisia is 10 mL: 1g of a compound;

(2) adding ethyl acetate with the same volume as the extracting solution into the extracting solution for extraction, and concentrating and drying the extract to obtain an ethyl acetate extract;

(3) passing the ethyl acetate extract through a silica gel column (the silica gel in the silica gel column is 200-300 mesh silica gel; the weight and the dosage of the silica gel are 30 times of the weight of the ethyl acetate extract), eluting and removing impurities by using a mixed organic solvent (the dosage is 5 times of the column volume) composed of chloroform and methanol with the volume ratio of 85:15, and then eluting by using a mixed organic solvent (the dosage is 2 times of the column volume) composed of chloroform and methanol with the volume ratio of 80: 20; collecting the eluted part eluted by a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 80:20, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Example 3 preparation of extract of Chinese traditional medicine for treating myocarditis

(1) Mixing dried Ginseng radix and Tibetan herba Artemisiae Scopariae at a weight ratio of 1:2, extracting with 10% ethanol water solution under reflux for 2 hr, and concentrating the extractive solution until there is no ethanol smell to obtain extractive solution; the dosage ratio of the ethanol water solution to the total weight of the ginseng and the Tibetan capillary artemisia is 10 mL: 1g of a compound;

(2) adding ethyl acetate with the same volume as the extracting solution into the extracting solution for extraction, and concentrating and drying the extract to obtain an ethyl acetate extract;

(3) passing the ethyl acetate extract through a silica gel column (the silica gel in the silica gel column is 200-300 mesh silica gel; the weight and the dosage of the silica gel are 30 times of the weight of the ethyl acetate extract), eluting and removing impurities by using a mixed organic solvent (the dosage is 8 times of the column volume) composed of chloroform and methanol with the volume ratio of 88:12, and then eluting by using a mixed organic solvent (the dosage is 5 times of the column volume) composed of chloroform and methanol with the volume ratio of 83: 17; collecting the eluted part eluted by a mixed organic solvent consisting of chloroform and methanol with the volume ratio of 83:17, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Comparative example 1 preparation of extract of Chinese medicine having effect of treating myocarditis

(1) Taking dried ginseng, then heating and refluxing the dried ginseng by using an ethanol water solution with the volume fraction of 15% for 2 hours, and concentrating the extracting solution until no alcohol smell exists to obtain an extracting solution; the dosage ratio of the ethanol water solution to the ginseng is 10 mL: 1g of a compound;

(2) adding ethyl acetate with the same volume as the extracting solution into the extracting solution for extraction, and concentrating and drying the extract to obtain an ethyl acetate extract;

(3) passing the ethyl acetate extract through a silica gel column (the silica gel in the silica gel column is 200-300 mesh silica gel; the weight and the dosage of the silica gel are 30 times of the weight of the ethyl acetate extract), eluting and removing impurities by using a mixed organic solvent (the dosage is 6 times of the column volume) composed of chloroform and methanol with the volume ratio of 86:14, and then eluting by using a mixed organic solvent (the dosage is 4 times of the column volume) composed of chloroform and methanol with the volume ratio of 82: 18; collecting the eluted part eluted by a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 82:18, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

The difference between the comparative example 1 and the example 1 is that the comparative example 1 only takes ginseng as a raw material, and the traditional Chinese medicine extract with the effect of treating myocarditis is prepared by adopting the method disclosed by the invention. In the embodiment 1, the traditional Chinese medicine extract with the effect of treating myocarditis is prepared by using ginseng and Tibetan capillaris as raw materials by adopting the method disclosed by the invention.

Comparative example 2 preparation of extract of Chinese medicine having effect of treating myocarditis

(1) Taking dried Tibetan capillary, heating and refluxing with 15% ethanol water solution by volume fraction for 2h, and concentrating the extractive solution until no alcohol smell exists to obtain extractive solution; the dosage ratio of the ethanol water solution to the Tibetan capillary is 10 mL: 1g of a compound;

(2) adding ethyl acetate with the same volume as the extracting solution into the extracting solution for extraction, and concentrating and drying the extract to obtain an ethyl acetate extract;

(3) passing the ethyl acetate extract through a silica gel column (the silica gel in the silica gel column is 200-300 mesh silica gel; the weight and the dosage of the silica gel are 30 times of the weight of the ethyl acetate extract), eluting and removing impurities by using a mixed organic solvent (the dosage is 6 times of the column volume) composed of chloroform and methanol with the volume ratio of 86:14, and then eluting by using a mixed organic solvent (the dosage is 4 times of the column volume) composed of chloroform and methanol with the volume ratio of 82: 18; collecting the eluted part eluted by a mixed organic solvent consisting of chloroform and methanol in a volume ratio of 82:18, concentrating and drying to obtain the traditional Chinese medicine extract with the effect of treating myocarditis.

Comparative example 3 differs from example 1 in that the elution conditions of the silica gel column are different; comparative example 3 the impurities were removed by elution with a mixed organic solvent of chloroform and methanol at a volume ratio of 95:5, followed by elution with a mixed organic solvent of chloroform and methanol at a volume ratio of 90: 10; in example 1, the mixed organic solvent of chloroform and methanol was used in a volume ratio of 86:14 to remove impurities, and then the mixed organic solvent of chloroform and methanol was used in a volume ratio of 82:18 to elute.

Injecting Kunming white mouse into abdominal cavity with 0.2mL of 100TCID50Preparing an Eagle culture solution of Coxsackie virus B3(CVB3) to prepare a viral myocarditis mouse model; after modeling, selecting mice with equivalent serum cardiac troponin I (cTnI) concentration, and dividing the mice into an experimental group 1-7 and a control group, wherein each group comprises 20 mice; wherein, the experimental groups 1 to 7 are respectively perfused with the traditional Chinese medicine extract with the effect of treating myocarditis prepared in the examples 1 to 3 and the comparative examples 1 to 4 according to the dose of 10mg/kg/dTaking the object; the control group was administered with equal volume of saline daily; meanwhile, a normal group is arranged, and normal saline with the same volume as the stomach is infused every day; after 2 weeks of continuous administration and 2 hours after the last administration, serum was extracted from peripheral blood of each group of mice to measure the concentration of myocardial troponin I (cTnI) in serum of each group of mice, and the test results showed that the model group had significant difference (P) from the normal group<0.05), the experimental groups 1-3 had significant differences (P) compared with the model group<0.05); compared with a model group, the experimental groups 4-7 have no significant difference (P is more than 0.05); specific results are shown in table 1. Wherein, the concentration of the serum cardiac troponin I (cTnI) is obtained by testing a cTnI detection kit (ELISA).

Serum cardiac troponin i (ctni) concentration is currently the most sensitive and specific detection indicator for diagnostic myocarditis. As can be seen from the data in table 1, the cTnI concentration of the model group was significantly higher than that of the normal group; this demonstrates the success of modeling the mouse myocarditis model of the present invention.

As can be seen from the data in Table 1, the cTnI concentration of the experimental groups 1-3 is significantly lower than that of the model group; this shows that the traditional Chinese medicine extract prepared from ginseng and Tibetan capillaris by the method of the invention can significantly reduce the concentration of serum cardiac troponin I (cTnI) of mice; further, the traditional Chinese medicine extract prepared from ginseng and Tibetan capillaris by adopting the method disclosed by the invention has a good treatment effect on myocarditis.

As can also be seen from the data in table 1, the cTnI concentrations for both experimental groups 4 and 5 were significantly higher than experimental group 1; the effect of the traditional Chinese medicine extract prepared by taking any one of the ginseng or the Tibetan virgate wormwood herb as the raw material and adopting the method for treating myocarditis is far smaller than that of the traditional Chinese medicine extract prepared by taking the ginseng and the Tibetan virgate wormwood herb as the raw materials and adopting the method for treating myocarditis. The method also indicates that the traditional Chinese medicine extract with excellent effect of treating myocarditis can be prepared by using ginseng and Tibetan capillaris simultaneously as raw materials.

As can also be seen from the data in table 1, the cTnI concentrations for both experimental groups 6 and 7 were significantly higher than experimental group 1; this shows that in the method of the present invention, the elution condition of the mixed organic solvent plays a decisive role in whether the traditional Chinese medicine extract with the effect of treating myocarditis can be prepared; the traditional Chinese medicine extract with excellent effect of treating myocarditis can be prepared only under the elution condition of the mixed organic solvent (namely, the mixed organic solvent consisting of chloroform and methanol with the volume ratio of 85-88: 15-12 is used for elution and impurity removal, and then the mixed organic solvent consisting of chloroform and methanol with the volume ratio of 80-83: 20-17 is used for elution); under the elution condition of other mixed organic solvents, the traditional Chinese medicine extract with excellent effect of treating myocarditis cannot be prepared.
