Background

As is well known, the existing collagen process is: in the extraction process of the collagen, firstly, adding a proper amount of water into the raw materials, mixing, steaming at high temperature, adding a proper amount of enzymolysis agent under a proper pH condition, carrying out enzymolysis extraction, and carrying out fine filtration after completion; and (3) concentrating and dehydrating the filtrate in an evaporation concentration system, and after the concentrated liquid reaches a certain concentration, feeding the concentrated liquid into spray drying equipment to obtain powder, namely the collagen.

However, the conventional collagen preparation process has many defects, such as: (1) the volume of the extracting solution is large, and the production period of the product is long; (2) the enzymolysis liquid does not thoroughly remove impurities, so that the quality of the collagen is influenced; (3) the resulting finished product often has a bitter and fishy smell; (4) the filtration grade is coarse and low, and the water solubility of the product is poor; (5) the salinity is high; (6) the molecular weight distribution of the product is wide.

The technical problem to be solved by the invention is to provide a collagen extraction and purification system, which has high extraction efficiency, and the purity of the obtained product is high.

In order to solve the technical problem, the invention provides a collagen extraction and purification system, which comprises the following devices which are sequentially communicated with each other: the collagen production device comprises a degreasing tank and a leaching device for degreasing raw materials, a crusher for crushing the degreased raw materials, an enzymolysis tank for performing enzymolysis treatment on the crushed raw materials, a refrigerated centrifuge for performing refrigerated centrifugation treatment on enzymolysis liquid, an alkaline washing tank for performing alkaline washing treatment on precipitates obtained by refrigerated centrifugation, a salting-out unit for performing salting-out treatment on alkaline washing liquid, a filtering unit and a dialysis unit for purifying the salted-out crude collagen, and a freeze drying unit for solidifying the purified collagen.

In a preferred embodiment of the present invention, the dialysis unit comprises a dialysis tank and a dialysis bag disposed in the dialysis tank, the dialysis bag is communicated with the filtration unit, the dialysis tank is filled with distilled water, and the dialysis tank is provided with a water inlet and a water outlet.

In a preferred embodiment of the present invention, the filtering unit is a filtering barrel, and an ion exchange membrane is disposed in the filtering barrel.

In a preferred embodiment of the present invention, a stirring device is disposed in the enzymolysis tank.

According to the collagen extraction and purification system, the collagen is extracted by an enzymolysis method, so that the problems of collagen denaturation caused by acid-base hydrolysis and damage caused by an amino group molecular structure are solved, and a natural active triple-helix structure is preserved. Meanwhile, through two-stage purification of the filtering unit and the dialysis unit, the purity of the collagen is greatly improved, the purity, color, taste, clarity, ash content and other qualities of the prepared collagen are well characterized, the molecular weight uniformity of the obtained collagen is good, and the pharmaceutical activity of the collagen can be fully exerted when the collagen is used in the fields of medicines and cosmetics.
