A composite skin nutritional composition comprising the following components: 5-80% of saturated fatty acid grease, 0.1-20% of conjugated linoleic acid, 1-40% of olive oil, 1-40% of walnut oil, 5-40% of rice bran oil and/or mink oil, 1-30% of wax fat, 0.1-30% of squalene, 0.1-20% of lecithin, 5-40% of egg yolk oil and 0.1-20% of vitamins; the percentage is mass percentage.

and/or the lecithin is present in an amount of 1% to 15%, for example 5% or 10%;

and/or the egg oil content is 5% to 30%, such as 10%, 20% or 25%.

The composite skin nutritional composition of claim 1, wherein the vitamins include one or more of vitamin a, vitamin D, and vitamin E; the vitamins preferably comprise 0.01-10% of vitamin A, 0.0001-1% of vitamin D and 0.1-99% of vitamin E; more preferably the vitamins consist of 2% vitamin a, 1% vitamin D and 97% vitamin E; the percentage is mass percentage.

or, the composite skin nutrition composition consists of the following components: 40% of saturated fatty acid grease, 1% of conjugated linoleic acid, 9% of olive oil, 2% of walnut oil, 8% of rice bran oil, 10% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 5% of vitamins;

or, the composite skin nutrition composition consists of the following components: 20% of saturated fatty acid grease, 5% of conjugated linoleic acid, 5% of olive oil, 5% of walnut oil, 10% of mink oil, 20% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 10% of vitamins.

9. A method for preparing the composite skin nutrition composition as claimed in any one of claims 1 to 8, which comprises mixing the components uniformly.

Use of a composite skin nutritional composition according to any one of claims 1 to 8 in skin care; wherein, said skin care preferably comprises treating a skin condition or improving healthy skin; the skin disorder is preferably a skin disorder caused by abnormal sebum metabolism or a skin disorder caused by damage to the skin barrier, such as seborrheic dermatitis, acne, eczema, psoriasis, follicular keratosis or keratosis; the healthy skin is preferably healthy oily skin or healthy dry skin.

Technical Field

Background

Common skin disorders are generally caused by abnormal sebum metabolism or damage to the skin barrier. Can cause various skin diseases, including acne, seborrheic dermatitis and the like, and brings various physical discomfort and psychological disorders to patients. The sebaceous membrane is a main structural unit of a skin barrier, is a layer of transparent thin film covered on the surface of the skin, is also called as a water-lipid membrane, is mainly formed by emulsifying sebum secreted by sebaceous glands, lipid produced by cells of horny layer and sweat secreted by sweat glands, is weakly acidic, mainly comprises ceramide, squalene, linoleic acid, linolenic acid and lipid components, and has the functions of locking water and resisting inflammation to a certain extent. Sebum and horny substances present in the horny layer form a stratified sheet membrane structure, which is the main structure of barrier function, and a lipid mixture in an appropriate ratio is the basis for forming a normal stratified sheet membrane structure. When the proportion of the lipid mixture is disordered, the abnormal change of the structure of the stratified lamina membrane can be caused, and further a series of skin diseases can be caused, including diseases such as eczema, psoriasis, parakeratosis and the like.

At present, many single medicines are applied to clinical treatment of skin diseases, but the curative effect is not ideal. The method mainly adopted by the acne is as follows: 1. tetracycline and macrolide medicines are adopted for antibacterial treatment, but the medicines have high liver and kidney toxicity; 2. tretinoin and adapalene gel are adopted to inhibit sebaceous gland secretion, but the medicines have large skin irritation and have teratogenic and mutagenic risks; 3. salicylic acid and tartaric acid are adopted to dissolve cutin and improve acne symptoms, but the skin is greatly damaged. Seborrheic dermatitis also lacks a specific safe treatment scheme, and ketoconazole and tetracycline with great side effects are mainly adopted at present. A 'regenerant-IL 4 antibody' which is marketed in the preparation of Xenoffy in 2017 is used for treating moderate and severe eczema, is combined with hormone for application, has the effective rate of only 48%, and has 2 deaths caused by adverse reactions in the use period. It can be seen that the single-drug therapy has low effective rate, and needs half a year of oral and external treatment, and the side effects are more beneficial than the treatment. Therefore, the development of a safe and effective preparation for caring skin diseases is of great significance.

Disclosure of Invention

The invention provides a composite skin nutrition composition and a preparation method and application thereof, aiming at solving the defects of low effective rate, long treatment period and large side effect of a single-medicament treatment scheme of skin diseases in the prior art. The composite skin nutrition composition has good treatment effect on skin diseases, short treatment period and high safety.

In order to achieve the purpose, the inventor of the invention utilizes a new concept of 'nature returning and recovery treatment', does not use medicines to carry out 'killing treatment' on diseases infected by non-pathogens, but utilizes the existing substances or natural substances in the body to recover the healthy substance composition under the normal physiological state, and replaces the disordered substance composition under the disease state with the healthy substance composition, so as to avoid the current situation of 'killing one thousand of enemies and eight hundred of self-injury' of modern medicine treatment. Based on the idea, ten years of research shows that the inventor successfully utilizes all natural products (without chemical synthetic substances) to prepare the composite skin nutrition composition which can highly simulate healthy skin lipid.

The invention provides a composite skin nutrition composition, which comprises the following components: 5-80% of saturated fatty acid grease, 0.1-20% of conjugated linoleic acid, 1-40% of olive oil, 1-40% of walnut oil, 5-40% of rice bran oil and/or mink oil, 1-30% of wax fat, 0.1-30% of squalene, 0.1-20% of lecithin, 5-40% of egg yolk oil and 0.1-20% of vitamins; the percentage is mass percentage.

In the invention, the saturated fatty acid oil refers to oil containing more than 90% of saturated fatty acid, and the main function of the saturated fatty acid oil is to maintain the stability of lipid and reduce the risk of lipid oxidation.

In the invention, the saturated fatty acid grease is preferably one or more of lard, goose oil, beef tallow, ostrich oil and coconut oil. Wherein, the lard can be extracted from the subcutaneous fat of the pig by adopting the conventional method in the field without any additional chemical substance. The goose oil can be extracted from goose subcutaneous fat by conventional method in the art, and no additional chemical substance is added for extraction. The beef tallow can be extracted from the lower fat of cowhide by conventional methods in the art without any additional chemical extraction. The ostrich oil can be extracted from the subcutaneous fat of the ostrich by adopting the conventional method in the field without adding any additional chemical substance for extraction. The source of coconut oil can be obtained by cold pressing coconut pulp using methods conventional in the art.

In the invention, the saturated fatty acid grease is preferably the combination of coconut oil and one or more of lard, goose oil, beef tallow and ostrich oil; even more preferably coconut oil in combination with lard or goose oil; most preferably a combination of coconut oil and lard.

When the saturated fatty acid oil is a combination of coconut oil and one or more of lard, goose oil, beef tallow, and ostrich oil, the mass ratio of the coconut oil to the one or more of lard, goose oil, beef tallow, and ostrich oil is preferably 1: (1-10). For example, the saturated fatty acid oil is a combination of coconut oil and lard in a mass ratio of 1: 1.

In the present invention, the content of the saturated fatty acid oil is preferably 20% to 60%, for example, 25%, 30%, 35%, 40%, 45%, 50%, or 55%.

In the invention, the conjugated linoleic acid, the olive oil, the walnut oil, the rice bran oil and/or the mink oil belong to unsaturated fatty acids. Wherein the source of the conjugated linoleic acid can be obtained by refining safflower by adopting a conventional method in the field. The olive oil can be obtained by olive pressing by conventional methods in the art. The walnut oil can be obtained by squeezing walnut by the conventional method in the field. The rice bran oil can be obtained from rice bran obtained in the rice processing process by pressing method by adopting the conventional method in the field. The mink oil can be extracted from the subcutaneous fat of minks by adopting a conventional method in the field, and no additional chemical substance is added for extraction.

In the present invention, the content of the conjugated linoleic acid is preferably 0.5% to 15%, for example, 0.8%, 1%, 5%, or 10%.

In the present invention, the content of the olive oil is preferably 5% to 30%, for example 9%, 10%, 15%, 20% or 25%.

In the present invention, the content of the walnut oil is preferably 2% to 30%, for example 5%, 10% or 20%.

In the present invention, the content of the rice bran oil and/or mink oil is preferably 7% to 30%, for example 8%, 10%, 15%, 20% or 25%.

In the invention, the wax fat can be conventional in the field, and can be prepared by compounding beeswax, cocoa butter and saturated fatty acid grease by adopting a conventional method in the field generally; the mass ratio of the beeswax, the cocoa butter and the saturated fatty acid grease is preferably 1: (1-2): (1-5), for example, 1:1:4, 1:1:1, 1:1:5 or 1:2: 2. Wherein the beeswax is fatty substance (wax) secreted by eight wax glands in the abdomen of the bee; the cocoa butter may be extracted from cocoa beans using methods conventional in the art; the saturated fatty acid grease is preferably lard, goose oil or beef tallow. Both high and low proportions of the wax lipid in the composite skin nutritional composition affect the fluidity and viscosity of the skin lipids, thereby inducing acne.

In the present invention, the content of the wax ester is preferably 5% to 25%, for example, 10%, 18%, or 20%.

In the present invention, the squalene can be extracted from shark liver oil by a method conventional in the art. The squalene has antioxidant effect, and can be used for treating acne with increased proportion in sebaceous gland. The squalene content is preferably 5% to 25%, for example 10%, 15% or 20%.

In the present invention, the lecithin may be extracted from egg yolk or soybean using a method conventional in the art. The lecithin is preferably present in an amount of 1% to 15%, for example 5% or 10%.

In the present invention, the egg oil may be extracted from the yolk of eggs by a method conventional in the art. The egg yolk oil can provide vitamins, lecithin, cholesterol, unsaturated fatty acid and the like, and also plays an important role in balancing fat composition. The egg oil content is preferably 5% to 30%, for example 10%, 20% or 25%.

In the present invention, the vitamins have an antioxidant effect and play a central role in skin cell differentiation. The vitamins are active substances necessary for skin nutrition, and can nourish the skin and improve the disease resistance of the skin through strengthening treatment. In the present invention, the vitamin may be a vitamin conventionally used in the art, and preferably includes one or more of vitamin a, vitamin D and vitamin E. The vitamins preferably comprise 0.01-10% of vitamin A, 0.0001-1% of vitamin D and 0.1-99% of vitamin E; more preferably, the vitamins consist of 2% vitamin a, 1% vitamin D and 97% vitamin E; the percentage is mass percentage.

In the present invention, the composite skin nutritional composition preferably consists of: 40% of saturated fatty acid grease, 1% of conjugated linoleic acid, 9% of olive oil, 2% of walnut oil, 8% of rice bran oil, 10% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 5% of vitamins, wherein the percentages are mass percentages.

In the present invention, the composite skin nutritional composition preferably consists of: 20% of saturated fatty acid grease, 5% of conjugated linoleic acid, 5% of olive oil, 5% of walnut oil, 10% of mink oil, 20% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 10% of vitamins, wherein the percentages are mass percentages.

The invention provides a preparation method of the composite skin nutrition composition, which comprises the step of uniformly mixing all the components.

The invention also provides application of the composite skin nutrition composition in skin care.

Wherein the skin care may include treating a skin condition or improving healthy skin. The skin disorder is preferably a skin disorder caused by abnormal sebum metabolism or a skin disorder caused by damage to the skin barrier, such as seborrheic dermatitis, acne, eczema, psoriasis, follicular keratosis or keratosis. The healthy skin may be healthy oily skin or healthy dry skin.

On the basis of the common knowledge in the field, the above preferred conditions can be combined randomly to obtain the preferred embodiments of the invention.

The reagents and starting materials used in the present invention are commercially available.

The positive progress effects of the invention are as follows:

the composite skin nutrition composition provided by the invention is based on the pathogenesis of skin diseases, adopts a treatment principle completely different from the current treatment strategy, starts from the construction of a healthy skin barrier, accurately performs the renaturation intervention (the restoration of the lipid composition of the normal physiological state of the skin), and intervenes in the pathogenesis of the skin diseases from the source. The components in the composite skin nutrition composition are cooperated to play roles in nourishing skin, regulating sebum secretion balance and reconstructing healthy skin barrier, and the composite skin nutrition composition has good treatment effect on skin diseases and short treatment period.

In addition, the composite skin nutrition composition is composed of safe edible oil and high-bionic healthy skin lipid, does not contain medicinal components and hormone, does not contain any additive (such as an emulsifier, a surfactant, a preservative and the like), and is high in safety.

Detailed Description

The invention is further illustrated by the following examples, which are not intended to limit the scope of the invention. The experimental methods without specifying specific conditions in the following examples were selected according to the conventional methods and conditions, or according to the commercial instructions.

In the following examples and comparative examples, the saturated fatty acid oil is a combination of coconut oil and lard, wherein the mass ratio of the coconut oil to the lard is 1: 1. The vitamins include 2% vitamin a, 1% vitamin D and 97% vitamin E. The wax fat is prepared by compounding beeswax, cocoa butter and lard in a mass ratio of 1:1: 1.

Example 1

The following components are mixed evenly to obtain the composite skin nutrition composition 1: 5% of saturated fatty acid grease, 15% of conjugated linoleic acid, 10% of olive oil, 2% of walnut oil, 8% of rice bran oil, 18% of wax fat, 10% of squalene, 1% of lecithin, 30% of egg oil and 1% of vitamins, wherein the percentages are mass percentages.

Example 2

Uniformly mixing the following components to obtain the composite skin nutrition composition 2: 60% of saturated fatty acid grease, 0.8% of conjugated linoleic acid, 1% of olive oil, 1% of walnut oil, 7% of mink oil, 5% of wax fat, 0.1% of squalene, 20% of lecithin, 5% of egg oil and 0.1% of vitamins, wherein the percentages are mass percentages.

Example 3

Uniformly mixing the following components to obtain the composite skin nutrition composition 3: 40% of saturated fatty acid grease, 1% of conjugated linoleic acid, 9% of olive oil, 2% of walnut oil, 8% of rice bran oil, 10% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 5% of vitamins, wherein the percentages are mass percentages.

Example 4

Uniformly mixing the following components to obtain the composite skin nutrition composition 4: 20% of saturated fatty acid grease, 5% of conjugated linoleic acid, 5% of olive oil, 5% of walnut oil, 10% of mink oil, 20% of wax fat, 10% of squalene, 5% of lecithin, 10% of egg oil and 10% of vitamins, wherein the percentages are mass percentages.

Comparative example 1

Uniformly mixing the following components to obtain the composite skin nutrition composition 5: 81% of saturated fatty acid grease, 0.05% of conjugated linoleic acid, 0.95% of olive oil, 3% of walnut oil, 5% of rice bran oil, 0.95% of wax fat, 4% of squalene, 2% of lecithin, 3% of egg yolk oil and 0.05% of vitamin, wherein the percentages are mass percentages.

Comparative example 2

Uniformly mixing the following components to obtain the composite skin nutrition composition 6: 5% of saturated fatty acid grease, 5% of olive oil, 5% of walnut oil, 10% of rice bran oil, 1% of wax grease, 35% of squalene, 2% of lecithin, 12% of egg oil and 25% of vitamins, wherein the percentages are mass percentages.

Effect example 1

KM mice, 90 females, weighing 25. + -.2 g, were used, as provided by the Wuhan university A3 animal center, and the certification numbers: SCXK (jaw 2016-. Mice were randomly divided into 9 groups of 10 mice each. The 9 groups are respectively: (1) a normal control group; (2) inflammatory acne model (M) group; (3) m + complex skin nutritional composition group 1; (4) m + complex skin nutritional composition group 2; (5) m + complex skin nutritional composition group 3; (6) m + complex skin nutritional composition group 4; (7) m + complex skin nutritional composition group 5; (8) m + complex skin nutritional composition 6 group; (9) the group of M + adapalene gels.

0.03mL of xylene was injected into the anterior and posterior lobes of the right ear of mice in groups (2) to (9) to induce acute ear edema in the right ear of the mice, and a mouse inflammatory acne model was established. The results are shown in table 1, wherein the ear swelling degrees of the groups (3) to (6) are respectively 13.73%, 18.91%, 21.22% and 10.55%, and the ear swelling degree of the group (9) is 32.22%, so that the composite skin nutritional compositions 1 to 4 of the embodiments 1 to 4 of the invention have good effect on resisting inflammatory acne, and are superior to the common medicine adapalene gel on the market. In addition, the ear swelling degrees of the groups (7) and (8) were 41.36% and 46.87%, respectively, illustrating that the composite skin nutritional compositions 5 and 6 of comparative examples 1 and 2 had no antagonistic effect on inflammatory acne, and it was found that the therapeutic effect could be lost when the composition component ratio was out of the range of the present invention, which is crucial for the treatment of inflammatory acne.

Effect example 2

KM mice, 90 females, weighing 25. + -.2 g, were used, as provided by the Wuhan university A3 animal center, and the certification numbers: SCXK (jaw 2016-. Next, the mice of groups (3) to (9) were applied with the corresponding drugs (for example, the mice of group (3) were applied with the composite skin nutritional composition 1, and the mice of group (9) were applied with the positive control dexamethasone cream). 0.1g each, once a day for 2 weeks.

After 14 days, the skin condition of each group of mice is shown in fig. 1. The results of measuring the skin dehydration rate, the degree of dermatitis, the skin pH, the skin moisture content, the degree of skin edema, and the skin density of each group of mice are shown in table 2, and it can be seen from table 2 that:

the skin percutaneous water loss rate of the mice in the group (2) is 6.3 times higher than that of the mice in the group (1), and the percutaneous water loss rate of the mice in the groups (3) to (6) is obviously reduced, wherein the percutaneous water loss rate of the group (6) is close to that of the normal group. The dermatitis degree score of the mice in the group (2) is 9.85, and the dermatitis degree scores of the mice in the groups (3) to (6) are known to be remarkably improved by the composite skin nutrient compositions 1 to 4 in the embodiment of the invention, wherein the dermatitis degree score of the group (6) (composite skin nutrient composition 4) is reduced to 1.84. The skin pH of the mice in group (2) was significantly higher than that of the mice in group (1), and it was found from the skin pH of the mice in groups (3) to (6) that the skin pH of the composite skin nutritional compositions 1 to 4 according to the embodiments of the present invention was significantly reduced. The skin moisture content of the mice in the group (2) is remarkably reduced to only 48.8% of that of the mice in the group (1), and the skin moisture content of the mice in the groups (3) to (6) can be remarkably improved by the composite skin nutritional compositions 1 to 4 in the embodiment of the invention. The skin of the mice in the group (2) is obviously edematous, the skin edema degree is 73.3% of that of the mice in the group (1), and the skin edema degrees of the mice in the groups (3) to (6) show that the composite skin nutrient compositions 1 to 4 in the embodiment of the invention can obviously reduce the skin edema degree. The skin density of the mice in the group (2) is remarkably reduced to 26% of that of the mice in the group (1), and the skin density of the mice in the groups (3) to (6) can be remarkably improved by the composite skin nutrition compositions 1 to 4 in the embodiment of the invention, wherein the skin density of the mice in the group (2) is basically recovered to be normal.

From the test results of the mice of the groups (7) and (8), it was found that the composite skin nutrient compositions 5 and 6 of comparative examples 1 and 2 did not significantly improve each index of skin inflammation. It can be seen that the efficacy can be lost when the composition has component ratios outside the scope of the present invention, which are critical to the treatment of eczema. According to the test results of the mice in the group (9), although the dexamethasone cream can reduce the transdermal water loss rate, the skin inflammation degree, the pH value, the skin water content and the skin edema degree, the effect is weak, and the curative effect is inferior to that of the composite skin nutritional composition 1-4 in the embodiment of the invention; also, dexamethasone cream had no improving effect on skin density.

Effect example 3

40 patients with acne were treated with the composite skin nutritional composition 4 of example 4, with 5 cases of class II, 26 cases of class III, and 9 cases of class IV according to the pilsbury classification. The observation period is 3 months, the effective rate is 100 percent, the cure rate is 90 percent, and the cure time is from 1 month to 3 months. Fig. 2 is a recovery chart of 1 example of patients with grade IV facial acne. As can be seen from fig. 2, the acne on the face of the patient was completely cured after 1 month of care for zero 21 days.

Acne grade II, III, IV in the pilsbury classification: grade II (medium): acne exists, pimples and pustules with medium number, and the total number of lesions is 31-50; grade III (moderate): a large number of papules and pustules are occasionally large inflammatory lesions, the distribution is wide, the total number of lesions is 51-100, and the number of nodules is less than 3; grade IV (heavy): nodular, cystic or polymeric, with pain and formation of cysts, with more than 100 lesions and more than 3 nodules or cysts.

Effect example 4

The composite skin nutrition composition 4 of the embodiment 4 is adopted to nurse 10 cases of eczema patients, the treatment course is 6 months, the effective rate is 100 percent, and the cure rate is 40 percent. Fig. 3 is a rehabilitation chart of 1 example of foot eczema patients. As can be seen in fig. 3, the patient had a complete cure of eczema on the feet after about 5 months of care.
