Background

Weeds are a large group of biological disasters that severely threaten crop production. The harm of weeds is manifested in direct competition with crops for nutrients, moisture, light and growing space, resulting in reduced crop yields. The use of the herbicide can not only effectively prevent and kill farmland weeds, improve the quality and the yield of crops, but also save a large amount of labor cost. However, the long-term use of chemical herbicides in large quantities has made the problem of weed resistance prominent. By 2016 (11) months, 476 biotypes of 251 weeds (146 dicots, 105 monocots) had developed resistance to 161 products of 26 classes of chemical herbicides worldwide. In addition, due to improper use, fog drop drifting, leaching, liquid medicine volatilization and the like, the chemical herbicide not only seriously affects the growth and development of crops, but also brings the problems of residual toxicity and phytotoxicity, yield reduction of next crops, land degradation and the like. Some highly toxic herbicides, such as paraquat, are life threatening when they are ingested by humans. Currently, over 100 chemical herbicides are banned or deregistered in over 30 countries around the world. With the need of agricultural sustainable development and the improvement of environmental awareness, the development and application of novel biopesticides to control the occurrence of diseases, pests and weeds is one of the key points of future pesticide research.

Eudesmol, also known as 4- (1-methylethyl) -phenol, has the following chemical structure:

the essential oil exists in essential oils of various plants, has simple chemical structure and is easy to synthesize. The method can provide a material guarantee for developing a novel botanical herbicide by fully utilizing the eudesmol in the follow-up process. Through retrieval, no report about the agricultural activity of the eudesmol exists at home and abroad at present, and no report about the herbicidal activity of the eudesmol exists.

Determination of inhibitory activity of eudesmol on agricultural weeds

1. Weed to be tested

Echinochloa crusgalli (Echinochloa crusgalli), Cyperus rotundus (Cyperus rotundus), Salvia miltiorrhiza (Sorghum sudanense), Abutili coronaria (Abutilon hybridum), Amaranthus mangostana (Amaranthus tricolor), Euphorbia lathyris (Leptochloa chinensis), Amaranthus retroflexus (Amaranthus retroflexus), Echinochloa crusgalli (Echinochloa phyllopogon), Rhizophora viridis (Chlorris virgata), Eleusines indica (Eleusines), Digitaria sanguinalis (Digitaria sanguinalis) and Borreria latifolia (Borreria latifolia), all of which are provided by the group of agricultural chemical biology and application topics of the environmental and plant protection institute of the Chinese institute of Tropical sciences.

2. Test method

2.1 in vitro Activity assay

In order to test the herbicidal activity of the eudesmol, the present invention evaluated the herbicidal activity of the compounds of the present invention against agricultural weeds by using a seed germination method. Dissolving the eudesmol with constant volume in ethanol, diluting to 1mg/mL for test, repeating the treatment for 3 times, and repeating for 20 weed seeds. The culture dish is put into a constant temperature incubator [ (28 +/-1) DEG C, the relative humidity is 80-85%, and the L/D is 12h/12h ] for culture, the root length and the stem length of the seedling are measured when the blank control grows to the proper length, and the inhibition rate is calculated according to the following formula.

Inhibition rate ═ control (root or stem length) -treatment (root or stem length) ]/control (root or stem length) × 100%

2.2 data analysis

The weeding activity results of the eudesmol on 12 weeds to be tested are shown in table 1, and experiments prove that the eudesmol provided by the invention has strong root length and stem length inhibition effects on barnyard grass, sudancao, garland hemp, amaranth, redroot amaranth, cockspur grass and crab grass when the test concentration is 1mg/mL, the inhibition rates are all more than 90%, and particularly, the root length and stem length of the eudesmol are all completely inhibited on the barnyard grass and the amaranth; the herbicide also has certain root length and stem length inhibiting effects on moleplant seed, tiger tail grass, eleusine indica and broadleaf fenghuacao, but the sensitivity of the herbicide is lower than that of the weeds; the herbicide activity to cyperus rotundus is lowest, and the inhibition rate of root length and stem length is only 74.72% and 57.26%.

2.3 potting test method

The reference method for the in vivo pot culture test of eudesmol on weeds is slightly improved: taking barnyard grass, garland hemp and amaranth as weeds to be tested, disinfecting and soaking seeds in warm water at 50 ℃ for 2h, wrapping the seeds with a wet towel, placing the seeds into a constant-temperature incubator [ (28 +/-1) DEG C, the relative humidity of 80-85% and the L/D of 12h/12h ] for accelerating germination for 24h, dibbling the seeds after the seeds are whitened in plastic pots filled with planting soil, uniformly dibbling 15-20 seeds in each pot, completely sprinkling the planting soil with the thickness of 1cm, properly watering to keep the humidity, placing the plants in a greenhouse with the temperatures of [ (26 +/-1) DEG C, the relative humidity of 70-90% and the L/D of 12h ], when 2-3 new leaves grow on the plants, uniformly spraying liquid medicines with the pipermide treatment concentrations of 10, 5 and 2.5mg/mL to the treated plants by using a small-sized spray pot, spraying 2-3mL each time, using clean water as blank control and ethanol as solvent control, the treatment concentration of the control medicament 5% eucalyptus oil soluble liquid is 1mg/mL, each treatment is repeated for 3 times, the symptoms of treated plants are regularly observed and recorded, the fresh weight of the overground part is weighed 7 days after treatment, and the prevention and treatment effect is calculated.

Control effect ═ control (fresh weight) -treatment (fresh weight) ]/control (fresh weight) × 100%

The in-vivo pot culture result shows that the control effect of 10mg/ml eudesmol treated cockspur grass, abutilon, amaranth is respectively 88.4%, 84.9% and 88.3% after 7 days, the weeding activity is higher, and the difference of the weeding activity and the control medicament 5% eucalyptus oil soluble liquid is not significant. And the control effect of the eudesmol treated by 5mg/ml on three kinds of weeds reaches more than 65% (tables 2 and 3). In addition, toxicity symptoms of the eudesmol on weeds are observed, the weeds show symptoms of lodging and withered leaves after being treated for 2 hours by 10mg/ml of the eudesmol and a control agent, and the treatment effects of 5mg/ml and 2.5mg/ml of the eudesmol are not obvious; after 24 hours, all the medicaments are subjected to treatment, and lodging, withered leaves and greenness generation are caused; after which the symptoms are more pronounced.

TABLE 3 potted plant test results of Audrecol vs. amaranth

The invention discloses application of eudesmol in preventing and controlling agricultural weeds. The test proves that the eudesmol has excellent herbicidal activity. When the tested concentration is 1000mg/L, the compound has stronger root length and stem length inhibition effects on barnyard grass, sudancao, crowndaisy hemp, amaranth, redroot pigweed, barnyard grass and crab grass, the inhibition rate is over 90 percent, and particularly, the root length and stem length of the compound are completely inhibited on the barnyard grass and the amaranth; pot experiments further verify that the compound has a strong contact poisoning effect on barnyard grass, crowndaisy hemp and amaranth, and the control effect of the compound is not significantly different from that of a control medicament 5% eucalyptus oil soluble liquid under the same concentration. The above results show that the eudesmol has a broad-spectrum herbicidal effect and can be deeply developed as a contact herbicide.
