Technical Field

The invention relates to the field of cosmetics, in particular to application of lycium ruthenicum anthocyanin in cosmetics with whitening effects and a cosmetic composition containing the lycium ruthenicum anthocyanin.

Background

Various components such as the physiological surface state of the skin, blood flow, moisture, and pigment are intricately combined to determine the skin color of a human body. The color of the skin is mainly determined by melanin, which plays a role in protecting the body from uv rays, but excessive and uneven accumulation of melanin is a major cause of skin darkening and senile plaque. Melanin is produced from small organ melanosomes in pigment cells of the basal epidermis layer, then migrates to epidermal cells, decomposes in the epidermal cells while entering the cornified layer, and finally disappears from the skin together with exfoliated fragments of keratinocytes.

Tyrosinase, a key rate-limiting enzyme in the biosynthesis of melanin in the skin, acts on dopa to produce dopaquinone, which spontaneously undergoes a series of reactions to form melanin. Since tyrosinase plays a key role in the process of melanin synthesis, a large class of whitening agents achieve the effect of whitening skin by reducing melanin production by inhibiting tyrosinase activity.

In recent years, the development of skin whitening cosmetics has been actively promoted, and studies on the cause of skin melanin, the whitening mechanism, the evaluation of whitening effect, the selection of whitening agents, and the like have been intensively conducted in the skin whitening cosmetics. Emphasizes the use of natural active whitening agents in cosmetics, people hope to appear a series of skin care cosmetics prepared from natural raw materials which are harmless to human bodies and have the effects of beautifying and health care.

Lycium ruthenicum Murr is perennial shrub of Lycium L of Solanaceae, and is a special wild plant variety in northwest of China. According to records of Tibetan book Jingzhu materia Medica and Uygur medicine record, the lycium ruthenicum fruit has the functions of strengthening kidney, moistening liver, improving eyesight, invigorating stomach, tonifying brain, resisting aging and dredging channels, is used for treating heart heat disease, heart disease, irregular menstruation, menopause and the like, and has obvious drug effect. Recently, the lycium ruthenicum mill has whitening efficacy (1, white wave, etc., patent application number: 201310557610.7; 2, Cao Youlong, etc., patent application number: 201710351479.7), but the basis of the efficacy material is unclear, and the whitening efficacy of the lycium ruthenicum mill anthocyanin monomer is not reported at present.

The invention firstly provides that the lycium ruthenicum anthocyanin has the skin beautifying effect and is suitable for cosmetics with the skin whitening effect.

Disclosure of Invention

At present, the medlar is mostly applied to the field of food. The literature does not relate to the whitening effect of the anthocyanin of the lycium ruthenicum, and the anthocyanin is not mentioned to be added into cosmetics as a functional additive. That is, the invention firstly proposes that the lycium ruthenicum anthocyanin has the skin beautifying effect, can be added into cosmetics as a functional additive and is used for realizing the whitening effect.

The invention is further illustrated below with reference to specific examples. It is to be understood, however, that these examples are illustrative only and are not to be construed as limiting the scope of the present invention.

Lycium ruthenicum is produced in Ningxia as described in examples 1-4 below.

Example 1

Adding deionized water into the dried lycium ruthenicum mill fruits according to the solid-liquid ratio of 1:8, leaching for 4 hours at room temperature, adding 75% edible ethanol according to the solid-liquid ratio of 1:6, and extracting twice at 45 ℃ for 2 hours each time. Mixing extractive solutions, filtering, concentrating under reduced pressure, and spray drying to obtain Lycium ruthenicum Murr extract. Weighing about 30g of lycium ruthenicum extract, adding 100ml of 1% formic acid-water for dissolving to obtain a lycium ruthenicum extract solution, separating by using a macroporous resin column, taking ethanol-water (adding 1% formic acid) as an eluent, sequentially eluting by using 30% ethanol to remove impurities, obtaining an anthocyanin crude extract by using 50% ethanol, and finally washing the column by using 95% ethanol. The obtained crude anthocyanin extract is subjected to liquid phase preparation by Waters, C18 column, and isocratic elution by using 20% acetonitrile (added with 1% formic acid) as eluent to obtain a component rich in anthocyanin. And finally preparing a liquid phase by Shimadzu, separating and purifying by using a C18 column, carrying out isocratic elution by using 20% acetonitrile (added with 1% formic acid) as an eluent to obtain a purer anthocyanin component, continuously preparing the purer anthocyanin, and then preparing the petunidin-3-O- (6-O-p-coumaroyl) rutinoside-5-O-glucoside, wherein the preparation is shown in figure 1.
