6. A preparation method of a dressing for promoting the healing of a skin wound surface is characterized by comprising the following steps:

s1, preparing a medical wound covering layer capable of completely covering a wound according to the size of the wound;

s2, soaking at least the part, in contact with the wound, of the medical wound covering layer in the step S1 by using 0.01-0.1 g/L sodium 3-hydroxybutyrate solution to prepare the dressing for promoting the healing of the skin wound.

The method of preparing a dressing for promoting healing of skin wounds according to claim 6, wherein in step S1, a medical absorbent cotton gauze capable of completely covering a wound and a medical transparent dressing capable of completely covering the medical absorbent cotton gauze and larger than the medical absorbent cotton gauze are prepared according to the size of the wound, and the medical wound covering layer is obtained by covering the medical transparent dressing on the outer side of the medical absorbent cotton gauze.

The preparation method of the dressing for promoting skin wound healing according to claim 6, wherein in step S2, sterile PBS or deionized water is used to dissolve the sodium 3-hydroxybutyrate powder with purity of more than 98%, and the sodium 3-hydroxybutyrate solution is obtained by fully shaking and centrifuging to remove impurities.

Technical Field

The invention relates to the technical field of biology, in particular to a dressing for promoting skin wound healing and a preparation method thereof.

Background

The skin is the first barrier between the body and the nature, and can prevent the invasion of environmental pathogenic microorganisms and the loss of body fluid. At present, the treatment of skin wounds which are difficult to heal is still a problem which needs to be solved clinically urgently, so that a plurality of dressings for promoting the healing of the skin wounds are produced at the same time, but the current clinical medical dressings cannot well meet the requirements. The normal wound healing process comprises a series of coordinated processes including hemostasis, inflammatory reaction, cell and granulation tissue proliferation, extracellular matrix (ECM) reconstruction and the like, chronic wounds such as diabetic foot, pressure sore, venous ulcer and the like (the healing process is more than 8 weeks) all have wound healing process disorder with different degrees, and especially the ECM and new blood vessel reconstruction are blocked, and the wounds bring huge psychological and economic burden to patients. The wound surface moisture is kept, and the skin cell proliferation, the skin cell repair and the formation of new blood vessels are promoted, so that the wound healing is very important. Therefore, it is necessary to develop a safe and non-irritating dressing for healing skin wounds, which has good moisture retention and can promote the proliferation of skin wound cells and angiogenesis, so as to meet the requirements of repairing skin wounds which are difficult to heal, and the technical problem to be solved at present is urgent.

Disclosure of Invention

In view of the above, the present invention provides a dressing for promoting healing of a skin wound and a preparation method thereof, which can form a protective barrier on the skin surface, keep the wound moist, and promote healing of the skin wound to overcome at least one of the defects of the prior art.

In order to solve the technical problems, the invention adopts the following technical scheme:

a dressing for promoting skin wound healing comprises 0.01-0.1 g/L of 3-sodium hydroxybutyrate solution and a medical wound covering layer, and when the dressing is used, the medical wound covering layer and the wound contacting part are at least soaked by the 3-sodium hydroxybutyrate solution.

The dressing for promoting the healing of the skin wound comprises 0.01-0.1 g/L of 3-sodium hydroxybutyrate solution, the solution can be used for soaking a medical wound covering layer to cover the wound and can also be directly dripped on the wound for use, and the effects of promoting cell proliferation and angiogenesis are verified through cell experiments and animal experiments. When the medical wound dressing is used, the sodium 3-hydroxybutyrate solution at least wets the part of the medical wound dressing, which is in contact with the wound, and the wetted part of the medical wound dressing provides a moist environment for the skin wound, so that the water loss on the surface of the skin is reduced, the harm of external adverse environment to the skin can be prevented, and the healing of the skin wound can be promoted.

The medical wound surface covering layer can be medical gauze dressing, also can be medical transparent dressing with gauze, preferably adopts the latter, promptly medical wound surface covering layer is located the medical absorbent cotton gauze of inboard and is located the medical transparent dressing in the outside when using. When the medical absorbent cotton gauze is used, the medical absorbent cotton gauze is soaked by the sodium 3-hydroxybutyrate solution, the medical absorbent cotton gauze can be soaked by the sodium 3-hydroxybutyrate solution to cover a wound surface, and the medical absorbent cotton gauze and the medical transparent dressing can also be directly dripped on the wound surface to be used and then sequentially covered, so that the medical absorbent cotton gauze is soaked by the sodium 3-hydroxybutyrate solution, and the thickness of the medical absorbent cotton gauze is 3-5 layers. More preferably, the medical transparent dressing uses a commercial 3M transparent dressing with viscosity (product standard: YZB/USA5456-2014), and compared with the medical adhesive tape, the cotton yarn is fixed by using the 3M transparent dressing, so that the movement of the cotton yarn is reduced, and meanwhile, the air permeability of the cotton yarn is maintained, and the irritation to the skin is reduced.

The concentration of the sodium 3-hydroxybutyrate solution is most preferably 0.05 g/L.

The preparation method of the dressing for promoting the healing of the skin wound comprises the following steps:

s1, preparing a medical wound covering layer capable of completely covering a wound according to the size of the wound;

s2, soaking at least the part, in contact with the wound, of the medical wound covering layer in the step S1 by using 0.01-0.1 g/L sodium 3-hydroxybutyrate solution to prepare the dressing for promoting the healing of the skin wound.

Preferably, in step S1, according to the size of the wound surface, preparing a medical absorbent cotton gauze capable of completely covering the wound surface and a medical transparent dressing capable of completely covering the medical absorbent cotton gauze and larger than the medical absorbent cotton gauze, and covering the medical transparent dressing outside the medical absorbent cotton gauze to obtain the medical wound surface covering layer.

Preferably, in step S2, the medical absorbent cotton gauze in step S1 is soaked by 0.01-0.1 g/L of 3-hydroxybutyric acid sodium solution.

Preferably, in step S2, sterile PBS or deionized water is used to dissolve the sodium 3-hydroxybutyrate powder with a purity of more than 98%, and the sodium 3-hydroxybutyrate solution is obtained by sufficiently shaking and centrifuging to remove impurities.

Preferably, the concentration of the sodium 3-hydroxybutyrate solution is 0.05 g/L.

Compared with the prior art, the invention has the following beneficial effects: the dressing for promoting the healing of the skin wound comprises 0.01-0.1 g/L (preferably 0.05g/L) of 3-sodium hydroxybutyrate solution, medical cotton yarns can be soaked by the solution to cover the wound and can also be directly dripped on the wound for use, and the 3-sodium hydroxybutyrate solution provided by the invention has been verified to promote the cell proliferation and the angiogenesis through cell experiments and animal experiments. The soaked cotton yarn provides a moist environment for the skin wound surface, reduces the water loss on the skin surface, can prevent the skin from being damaged by external adverse environment, and can promote the healing of the skin wound surface. Compared with the medical adhesive tape, the 3M transparent dressing is used for fixing the cotton yarns, so that the movement of the cotton yarns is reduced, the air permeability of the cotton yarns is kept, and the skin irritation is reduced.

Detailed Description

The drawings are only for purposes of illustration and are not to be construed as limiting the invention. The present invention will be described in further detail with reference to specific examples.

Example 1

The invention provides a dressing for promoting the healing of a skin wound and a preparation method thereof, and as shown in figure 1, the dressing comprises the following components: 0.05g/L of 3-sodium hydroxybutyrate solution, medical absorbent cotton gauze 2 and adhesive medical transparent dressing 1.

The preparation method of the dressing comprises the following steps:

s1, preparing medical absorbent cotton gauze capable of completely covering a wound surface and medical transparent dressing which can completely cover the medical absorbent cotton gauze and is larger than the medical absorbent cotton gauze according to the size of the wound surface, and covering the medical transparent dressing on the outer side of the medical absorbent cotton gauze to obtain a medical wound surface covering layer;

s2, soaking the medical absorbent cotton gauze obtained in the step S1 in 0.05g/L sodium 3-hydroxybutyrate solution to obtain the dressing for promoting the healing of the skin wound.

The dressing for promoting the healing of the skin wound comprises 0.05g/L of 3-sodium hydroxybutyrate solution, medical cotton yarns can be soaked by the solution to cover the wound and can also be directly dripped on the wound for use, and the 3-sodium hydroxybutyrate solution provided by the invention has been verified to have the effects of promoting cell proliferation and angiogenesis through cell experiments and animal experiments. The soaked cotton yarn provides a moist environment for the skin wound surface, reduces the water loss on the skin surface, can prevent the skin from being damaged by external adverse environment, and can promote the healing of the skin wound surface.

Among them, sodium 3-hydroxybutyrate is the sodium salt of 3-hydroxybutyric acid (3-HB), and 3-hydroxybutyric acid is produced in mammals by the metabolism of long-chain fatty acids in the liver, and is abundantly present in plasma and peripheral tissues as a main ketone body. Exogenous 3-HB has been reported in the literature for oral or injection treatment of diseases and injuries such as hemorrhagic shock, myocardial injury, extensive burns, insufficient brain oxygen supply, hypoxia and ischemia. Under these circumstances, 3-HB ameliorates tissue damage, protein metabolism, and metabolic disorders. Although monomer 3-HB may have a broad-spectrum growth promoting effect on various cells, no report on promotion of vascular endothelial cell proliferation, migration and angiogenesis of 3-HB and derivatives thereof has been made so far because 3-HB is an acidic substance. The cell experiment proves that 0.05g/L of 3-HB sodium salt has the effect of promoting the proliferation and migration of vascular endothelial cells and the formation of blood vessels, animal experiments prove that the 3-HB sodium salt has the effect of promoting the healing of skin wounds, and the sodium salt is applied to prepare the dressing, thereby obtaining remarkable effect on promoting the healing of the skin wounds and making up the blank of skin wound repair which is difficult to heal.

Example 2

As a preferable embodiment of example 1, this example provides a method for preparing a sodium 3-hydroxybutyrate solution, which comprises dissolving 1g of a sodium 3-hydroxybutyrate reagent (with a purity of 98% or more) in 10ml of sterile PBS buffer, shaking sufficiently with a shaker, centrifuging at 900r/min for 5min to remove impurities, obtaining 100g/L of a sodium 3-hydroxybutyrate solution, and further diluting to obtain 0.05g/L of a sodium 3-hydroxybutyrate solution. As the solvent, sterile purified water, distilled water or water for injection may be used in addition to PBS buffer. Cell experiments prove that 0.01-0.1 g/L sodium 3-hydroxybutyrate solution has promotion effects on cell proliferation, migration and angiogenesis, and the promotion effects on proliferation, migration and angiogenesis are most obvious when the concentration is 0.05g/L, as shown in figures 2-4.

Example 3

As a preferable scheme of embodiment 1, in this embodiment, the size of the medical cotton yarn is determined by the size of the wound, the medical cotton yarn with a certain size is adopted to ensure that the wound can be completely covered, the medical cotton yarn is soaked in 0.05g/L sodium 3-hydroxybutyrate solution to cover the skin wound, and the skin wound can be moistened on the wound with 0.05g/L sodium 3-hydroxybutyrate solution before covering.

Example 4

As a preferable scheme of the example 1, in the example, the medical transparent dressing uses a commercial 3M transparent dressing (product standard: YZB/USA5456-2014) as a dressing for fixing cotton yarns, and ensures that the dressing completely covers the medical absorbent cotton yarns. Compared with the medical adhesive tape, the 3M transparent dressing is used for fixing the cotton yarns, so that the movement of the cotton yarns is reduced, the evaporation of water can be reduced to a certain degree, the air permeability of the cotton yarns can be kept, and the stimulation to the skin can be reduced.

Example 5

The effect of the skin wound dressing on promoting the healing of the skin wound is investigated through animal experiments in the embodiment:

a full-thickness wound model with the diameter of 1cm is established on the back of the SD rat, an experimental group is covered by the skin wound healing-promoting dressing in the example after the wound is established, a control group is covered by medical absorbent gauze which is not soaked in the sodium 3-hydroxybutyrate solution, and the healing condition and the healing time of the wound are observed. After 2 weeks, the wound surface of the experimental group was completely healed, and the healing area of the control group was about 70%.

It should be understood that the above-described embodiments of the present invention are merely examples for clearly illustrating the present invention, and are not intended to limit the embodiments of the present invention. Other variations and modifications will be apparent to persons skilled in the art in light of the above description. And are neither required nor exhaustive of all embodiments. Any modification, equivalent replacement, and improvement made within the spirit and principle of the present invention should be included in the protection scope of the claims of the present invention.
