Background

The Chinese medicinal material bupleuri radix is derived from dried root of Bupleurum chinense DC or Bupleurum angustifolium Willd belonging to Umbelliferae, and has been used as traditional Chinese medicine for thousands of years. The research shows that: the bupleuri radix contains saponin, volatile oil, polysaccharide, flavone, etc., and has antiinflammatory, antibacterial, and antiviral effects. Saikosaponin (saikoaponins, SS) is the main chemical and bioactive components of bupleurum, and the chinese pharmacopoeia (first part of 2020 edition) uses the total amount of Saikosaponin a (saikoaponin a, SSa) and Saikosaponin d (saikoaponin d, SSd) as the quality control index of bupleurum medicinal material. Both SSa and SSd belong to the class of native saponins. Clinically, the bupleurum is mainly decocted into a decoction, so that the content of SSa and SSd in the decoction can be influenced, and further the drug effect is influenced.

(3) soaking 50g of radix bupleuri crude decoction pieces in 30mL of sodium bicarbonate aqueous solution containing 3g of sodium bicarbonate for 2 h;

Example 3:

(1) removing impurities and residual stems of radix bupleuri raw medicinal materials, cleaning with clear water, moistening with clear water until the medicinal materials are thoroughly permeated, cutting into 8mm thick pieces, and oven drying at 63 deg.C to obtain radix bupleuri crude decoction pieces;

(2)1g baking soda (NaHCO)3) Adding 15mL of distilled water, and dissolving by ultrasonic to prepare a sodium bicarbonate water solution;

(3) soaking 50g of radix bupleuri crude decoction pieces in 30mL of sodium bicarbonate aqueous solution containing 1g of sodium bicarbonate for 2 h;

(4) parching the soaked radix bupleuri crude decoction pieces with slow fire at 120 deg.C for 8min, taking out, cooling, and making into radix bupleuri processed product.

Comparative example 1

A processing method of radix bupleuri decoction pieces comprises the following steps: removing impurities and residual stems of radix bupleuri raw medicinal materials, cleaning with clear water, moistening with clear water until the medicinal materials are thoroughly permeated, cutting into 8mm thick pieces, and drying in a 60 ℃ oven to obtain radix bupleuri raw medicinal slices.

Contents of one, total saponins, SSa and SSd

The contents of total saponins in bupleuri radix prepared in examples 1-3 and comparative example 1 were respectively detected by UV spectrophotometry.

The content of SSa and SSd in the bupleurum prepared in the examples 1-3 and the comparative example 1 is respectively detected by adopting a high performance liquid chromatography and referring to a method of content determination under the item of bupleurum in the Chinese pharmacopoeia (the first part of 2020 edition), and the specific detection method comprises the following steps: respectively taking the processed radix bupleuri products prepared in examples 1-3 and the raw radix bupleuri decoction pieces prepared in comparative example 1, and crushing to obtain radix bupleuri powder (sieving by a sieve IV); chromatographic conditions and system applicability test: using octadecylsilane chemically bonded silica as a filler, acetonitrile as a mobile phase A, water as a mobile phase B, and performing gradient elution under the following conditions: acetonitrile (A) -water (B), gradient elution (0-30min, 35% A, 30-50min, 35% -90% A, 50-60min, 90%); the detection wavelength is 210nm, and the number of theoretical plates is not less than 10000 calculated according to the peak of saikosaponin a; preparation of control solutions: taking proper amount of saikosaponin a reference substance and saikosaponin d reference substance, precisely weighing, adding methanol to obtain solution containing saikosaponin a0.4mg and saikosaponin d0.5mg per 1mL, and shaking up to obtain; preparation of a test solution: taking about 0.5g of powder of a product to be detected (passing through a No. four sieve), precisely weighing, placing the powder in a conical flask with a plug, adding 25mL of methanol solution containing 5% concentrated ammonia test solution, sealing the plug, carrying out ultrasonic treatment (power 200W and frequency 40kHz) at 30 ℃ for 30 minutes, filtering, washing a container and medicine residues by 20mL of methanol for 2 times, combining the washing solution and the filtrate, recovering the solvent to dryness, dissolving the residues in methanol, transferring the solution to a 5mL measuring flask, adding methanol to scale, shaking up, filtering, and taking a subsequent filtrate to obtain the product; the determination method comprises the following steps: precisely sucking 20 μ L of the reference solution and 10-20 μ L of the sample solution, respectively, injecting into a liquid chromatograph, and measuring; the powder to be detected contains saikosaponin a and saikosaponin d in percentage by mass based on the dry product.

The above detection results are shown in table 1, and it can be seen from table 1 that the total saikosaponin content of the processed product of bupleurum root prepared by the present invention is about 2 times higher than that of the raw bupleurum root, and the content of saikosaponin a and saikosaponin d is more than 2 times higher than that of the raw bupleurum root.

Second, the dissolution rate of saikosaponin obtained by decocting bupleurum root with water

Clinically, the bupleurum roots prepared in the example 1 and the comparative example 1 are respectively decocted by distilled water, the contents of the total saponins, SSa and SSd in the extracting solution are detected by an ultraviolet spectrophotometry and a high performance liquid chromatography, the dissolution rates (the percentages of the dissolved substances to the mass of the processed product) of the total saponins, SSa and SSd are obtained, the detection results are shown in table 2, and the dissolution rates of the saikosaponin a and the saikosaponin d in the processed product of the bupleurum roots prepared by the invention are 2 times higher than that of raw bupleurum decoction pieces when the bupleurum roots are decocted by water.
