Technical Field

The invention relates to the field of medical health-care food, in particular to a tinospora root water extract and a new application thereof.

Background

Radix tinosporae, dried root tuber of Tinospora cordifolia or Tinospora cordifolia of Menispermaceae. Wherein, the physiological characteristics of the tinospora: winding vines, wherein the roots are deep and long, the root tubers are yellow, the shapes are different, the small branches are slender, the coarse branches are provided with grooves, and short bristles are arranged on the joints; the physiological characteristics of the tinospora cordifolia are as follows: evergreen wound liana, oval, elliptical, kidney-shaped or round root tuber, often several connected, earthy yellow epidermis.

At present, few researches are made on the pharmacodynamic action of tinospora cordifolia, and the researches are mainly focused on the aspects of anti-inflammation and analgesia. For example, patent document CN110269885A discloses an application of tinospora root extract in preparing drug-resistant acinetobacter baumannii drugs, patent document CN105707305A discloses a tinospora root milk tea for clearing heat and toxic materials and a production method thereof, and patent document CN1939417B discloses that a pharmaceutical composition of asiatic moonseed rhizome and tinospora root or extracts thereof synergistically plays the roles of resisting infection, resisting inflammation, relieving fever, relieving pain and the like. As a traditional Chinese medicine, deep research needs to be carried out on the new efficacy of the tinospora cordifolia to improve the application value of the tinospora cordifolia.

Disclosure of Invention

Based on the above, there is a need for an aqueous extract of tinospora root and a novel use thereof.

The technical scheme of the invention is as follows:

the invention provides an application of tinospora root water extract in preparation of a preparation product for promoting the growth of probiotics. Wherein, the probiotics include but are not limited to bifidobacteria or non-pathogenic escherichia coli.

The invention also provides a preparation method of the tinospora root water extract, which comprises the following steps: adding water into the washed radix tinosporae tuber, uniformly crushing, and performing ultrasonic extraction to obtain a homogenate extraction product; and (3) centrifuging and filtering the homogenate extraction product, collecting an extracting solution, and concentrating to obtain the product.

Preferably, the volume ratio of the washed radix tinosporae tuber to water is 1: (8-30).

Preferably, the density of the concentrated extract of the tinospora root water extract is 1.10-1.15.

The invention also provides a preparation product for promoting the growth of probiotics, and the active ingredient of the preparation product comprises the aqueous extract of tinospora root. Wherein, the preparation products include, but are not limited to, tinospora oral liquid, tinospora pill, tinospora granule and tinospora liposome.

In some embodiments, the tinospora root liposome powder is mainly prepared from tinospora root water extract with the concentration extract density of 1.10-1.15, phospholipid and cholesterol by a film dispersion method.

The invention has the beneficial effects that:

the tinospora root water extract can effectively promote the growth of probiotics, can be used for preparing a preparation product for promoting the growth of the probiotics, improves the gastrointestinal health of a human body or an animal body, and can obviously reduce the blood sugar level.

The terminology used in the description of the invention herein is for the purpose of describing particular embodiments only and is not intended to be limiting of the invention.

The raw material sources are as follows:

radix tinosporae root tuber: shiweir, Heng Biotech, Inc.

The rest of the reagents are conventional and commercially available.

Example 1

The embodiment provides a preparation method of an tinospora root water extract, which comprises the following steps:

s1, cleaning the dried radix tinosporae tuberous roots by ultrasonic waves (the ultrasonic power is 200-300 w).

S2, adding ultrapure water into the cleaned radix tinosporae tuber according to the material liquid volume ratio of 1:10, then smashing and uniformly mixing the mixture by using a high-speed pulverizer, carrying out ultrasonic treatment (with the ultrasonic power of 200-300 w) for 10min, ultracentrifuging or filtering the homogenate to remove residues, and collecting the water extract solution.

And S3, concentrating the water extract solution until the density of the extract is 1.10-1.15, wherein the temperature of the liquid inlet material is 55-65 ℃, the temperature of the air inlet is 170 ℃, the temperature of the air outlet is 80 ℃.

Example 2

Example 3

The embodiment provides a preparation method of an tinospora root beverage, which comprises the following steps: taking 100mL of the tinospora root water extract obtained in the example 1, adding 400mL of water, 75g of cane sugar and 1-2 g of citric acid, heating to 50 ℃, filtering by using a filter after the cane sugar is fully dissolved, degassing by using a vacuum degassing tower, instantly sterilizing at ultrahigh temperature, sealing and filling to obtain the tinospora root water extract.

Example 4

The embodiment provides an tinospora root compound tea, which comprises the following steps:

s1, adding green tea (green tea in Fangxian county) into purified water, and extracting in 80 deg.C water bath oscillator.

S2, mixing the tinospora root water extract and the green tea juice in the embodiment 1 uniformly, adding Vc, homogenizing under high pressure, and fixing the volume to make the tinospora root water extract account for 10% of the volume of the final solution, the mass ratio of the green tea (leaves) to the final solution is 1:250, and the addition amount of Vc in each 100mL of the solution is 0.01-0.015 g.

And S3, sterilizing and filling.

Example 5

The embodiment provides tinospora root granules, which comprise the following steps:

s1, weighing 20g of soybean lecithin and 6.66g of cholesterol, placing the soybean lecithin and the cholesterol into a 1000mL flask, adding 66mL of absolute ethanol, placing the mixture into a water bath at 65-70 ℃, stirring to dissolve the mixture, rotating the mixture on a rotary evaporator, and performing reduced pressure evaporation to prepare the phospholipid membrane.

S2, dissolving the aqueous extract of Tinospora cordifolia obtained in example 1 in phosphate buffer (pH about 5.7) to obtain aqueous extract solution with concentration of 1 mg/mL.

S3, transferring 1000mL of the aqueous phase extract solution into a flask containing a phospholipid membrane, and stirring the aqueous phase extract solution on a magnetic stirrer or shaking the aqueous phase extract solution in a constant-temperature shaker to form a liposome solution.

Comparative example 1

The comparative example provides a method for preparing an alcoholic extract of tinospora cordifolia, comprising the following steps:

s1, the dried radix tinosporae tuber is cleaned by ultrasonic wave.

S2, adding 70% ethanol into the washed radix tinosporae root tuber according to the volume ratio of the material liquid of 1:10, carrying out vacuum reflux extraction, collecting ethanol extract, carrying out reduced pressure distillation to remove organic solvent, and collecting concentrated solution.

Effect test

(1) Probiotic test:

the tinospora capillipes extracts of example 1 and comparative example 1 were added to water, diluted in the ratio of 1:10, 1:100, and 1:1000, added to a culture medium, sterilized, added to a bacterial culture plate, inoculated with probiotic non-pathogenic escherichia coli, cultured at 37 ℃ for 24 hours, photographed, and automatically counted by a computer, and the results are shown in table 1 below:

The tinospora formulations of examples 1 to 6 (in the same amount of extract) were added to the culture medium, added to the bacterial culture plate, inoculated with probiotics (bifidobacteria), cultured at 37 ℃ for 24 hours, photographed, and then automatically counted by a computer, and the results were summarized in table 2 below:

(2) Hypoglycemic test

Selecting 120C 57 mice, randomly dividing the mice into four groups (n is 30, a normal group, a diabetes group, a tinospora capillipes water extract group and a tinospora capillipes alcohol extract group), preparing a diabetes model (60mg/kg) by injecting streptozotocin into the abdominal cavity of the mice of the diabetes, the tinospora capillipes water extract group (which are divided into 3 groups) and the tinospora capillipes alcohol extract group (which are divided into 3 groups), collecting tail venous blood after 3 days of injection, and measuring the blood sugar, wherein the blood sugar is more than 11.1mmol/L, namely the model is successful.

Then, mice in the tinospora capillipes water extract group and the tinospora capillipes alcohol extract group are subjected to intragastric gavage three times a day, 2mL of the extract is obtained each time, the concentrations of the extracts are respectively 1:10, 1:100 and 1:1000, after three days, the blood sugar level is measured again, and the statistical results are shown in the following table 3:
