Technical Field
The invention relates to the technical field of biology, in particular to a lytic and high-titer vibrio parahaemolyticus bacteriophage and application thereof in vibrio parahaemolyticus infection in aquaculture.
Background
Vibrio parahaemolyticus, also known as pathogenic halophilic bacteria, is a marine pathogenic bacterium and is a zoonosis pathogen. Isolated in 1950 from an explosive food poisoning in japan. The vibrio parahaemolyticus is extremely wide in distribution and mainly distributed in seawater and aquatic products, and according to statistics, the detection rate of the vibrio parahaemolyticus in coastal seawater in east China is 47.5% -66.5%, the average bacteria carrying rate of marine fishes and shrimps is 45.6% -48.7%, and the detection rate can reach more than 90% in summer. The existence of Vibrio parahaemolyticus is found in livestock and poultry meat, salted vegetables, salted eggs, freshwater fish, and the like, in addition to marine products.
In recent years, the aquaculture industry in China is rapidly developed. With the increasing expansion of the mariculture scale and the continuous popularization of intensive culture modes, the frequent occurrence of fish and shrimp diseases poses serious threats to the healthy development of the large-scale culture of aquatic products. The pathogenic factors of aquatic animals mainly include bacteria, viruses, parasites and the like, wherein bacterial diseases are diseases with serious damage degree to the aquatic animals. At present, the main method for preventing and controlling vibrio parahemolyticus is to use antibiotics, although the antibiotics are convenient to use, have broad-spectrum bacteriostasis and quick response, the abuse of the antibiotics also causes the serious drug resistance of bacteria, and the effect in preventing and treating diseases caused by the vibrio parahemolyticus is increasingly reduced, so that the development of novel vibrio parahemolyticus resistant drugs is urgently needed.
The bacteriophage is a bacterial virus, has higher abundance in nature and strong host specificity, and cannot cause damage to the micro-ecological environment. After the phage infects host bacteria, new phage can be released during cracking, and a small amount of phage can play a role in preventing and treating. The action target of the vibrio parahaemolyticus phage is on the cell wall of prokaryotic microorganisms, and corresponding action sites do not exist on animal and plant cells, so that the use safety of the phage is improved. And the phage and the pathogenic bacteria can evolve together to prevent the occurrence of bacterial drug resistance. Therefore, the utilization of the phage to solve the biological control approach with wide prospect for bacteria.
The invention provides a safe and reliable lytic vibrio parahaemolyticus phage for treating vibrio parahaemolyticus in response to national order of comprehensive banning of resistance, solves the problems of bacterial drug resistance and antibiotic abuse, and lays a foundation for developing antibiotic substitutes for treating vibrio parahaemolyticus by expanding a phage stock library.
Disclosure of Invention
The invention aims to provide a specific lytic vibrio parahaemolyticus bacteriophage RDP-VP-21010 to replace antibiotics in preventing and treating aquaculture and solve the problems that aquatic animals are not treated timely due to acute disease, drug taking is ineffective due to drug resistance of pathogenic bacteria and the like.

The invention also provides application of the vibrio parahaemolyticus phage RDP-VP-21010 in preparation of anti-vibrio parahaemolyticus BV-21002 drugs.
Another purpose of the invention is to provide an application of the vibrio parahaemolyticus bacteriophage RDP-VP-21010 in preparing medicines for preventing and or treating diseases caused by vibrio parahaemolyticus.
Another purpose of the invention is to provide an application of the vibrio parahaemolyticus bacteriophage RDP-VP-21010 in preparing feed for preventing and/or treating diseases caused by vibrio parahaemolyticus.
Another purpose of the invention is to provide a bactericidal composition for preventing and treating Vibrio parahaemolyticus infection, which comprises an effective amount of Vibrio parahaemolyticus RDP-VP-21010.
Another object of the present invention is to provide the use of a Vibrio parahaemolyticus bacteriophage which provides a potential therapeutic drug or health product by using the Vibrio parahaemolyticus bacteriophage RDP-VP-21010 as a bacterial infection caused by Vibrio parahaemolyticus.
Another object of the present invention is to provide an additive for preventing and/or treating diseases caused by Vibrio parahaemolyticus, which is achieved by including Vibrio parahaemolyticus bacteriophage RDP-VP-21010 as an active ingredient.
The invention also provides a cleaning agent or a disinfectant, which comprises vibrio parahaemolyticus phage RDP-VP-21010.
The invention has the beneficial effects that:
(1) the invention discovers and separates a pathogenic vibrio parahaemolyticus BV-21002, and separates a vibrio parahaemolyticus bacteriophage RDP-VP-21010 by taking the pathogenic vibrio parahaemolyticus BV-21002 as a host, the bacteriophage RDP-VP-21010 has strong cracking effect on the pathogenic vibrio parahaemolyticus BV-21002 in an aquaculture environment, and provides a bacteriophage source for the industrial production of the bacteriophage for preventing and treating the pathogenic vibrio parahaemolyticus BV-21002 in the aquaculture environment;
(2) the vibrio parahaemolyticus phage RDP-VP-21010 provided by the invention is a virulent phage test phage separated from nature and does not contain virulence genes or undesirable genes; the invention does not carry out any genetic modification on the test phage;
(3) the vibrio parahaemolyticus phage RDP-VP-21010 provided by the invention has an obvious lysis effect within 8 hours after the vibrio parahaemolyticus phage RDP-VP-21010 is added into a water body, has strong lysis activity and shows a good application prospect.
(4) The vibrio parahaemolyticus phage RDP-VP-21010 provided by the invention acts for 6 hours at pH 5-10, the titer is high and can be 2.2 x 1010Above pfu/mL, there is a clear alkalophilicity. The present invention includes, but is not limited to, the prevention of food spoilage caused by infection with Vibrio parahaemolyticus in the form of liquid immersion, spraying, use in combination with synthetic ingredients, and the like, and is particularly suitable for cooked or non-sterilizable foods. The liquid soaking and spraying forms comprise but are not limited to food degerming agents, food disinfectants, food preservatives and the like; synthetic components of the present invention include, but are not limited to, benzoic acid, sodium benzoate, sorbic acid, potassium sorbate, calcium propionate, and the like.
(7) The vibrio parahaemolyticus phage RDP-VP-21010 of the invention can be used for preparing compositions, reagents or kits; the method can be applied to the rapid detection of the vibrio parahaemolyticus, and comprises the steps of detecting the vibrio parahaemolyticus in a target sample in the forms of test paper, a kit and the like, or screening target pathogenic bacteria in a clinical sample, so that the detection sensitivity can be effectively ensured;
(8) the vibrio parahaemolyticus phage RDP-VP-21010 can be used alone or in a mixture, and can be used as a biological bactericide, a feed additive, or a therapeutic drug, a health product and a medical apparatus for bacterial infection caused by vibrio parahaemolyticus; the vibrio parahaemolyticus phage RDP-VP-21010 can be applied to the treatment or prevention of infectious diseases caused by vibrio parahaemolyticus;
(9) the vibrio parahaemolyticus phage RDP-VP-21010 can be applied to industrial production, can be specifically amplified by host bacteria, can be highly purified by applying a standard virus purification method, and has a very good popularization prospect. According to the description of the invention and the common knowledge in the field, the technical personnel can prepare the vibrio parahemolyticus phage RDP-VP-21010 or the composition thereof into various products applied to the aspects of medical treatment, detection, disinfection, food protection and the like for industrial application. The product form can include, but is not limited to, the carrier carrying, the concentrated injection or the medicament soaking and the like, and the product is applied to the controlled host body surface, the oral part, the rectum, the inner pleura and other parts; as one embodiment, the carrier-borne form includes, but is not limited to, oral aqueous carriers, oral anhydrous carriers, cream formulations, and the like; concentrated injection forms include, but are not limited to, vaccine injections, pleural cavity injections, transvenous injections, etc.; dosage soaking forms include, but are not limited to, aerosols, rinses, and the like.

Detailed Description
The invention is further illustrated with reference to the following figures and examples. The following examples will assist those skilled in the art in further understanding the invention, but are not intended to limit the invention in any way. It should be noted that variations and modifications can be made by persons skilled in the art without departing from the spirit of the invention. All falling within the scope of the present invention.
The invention separates vibrio parahaemolyticus BV-21002 from a diseased shrimp focus of a Qingdao, i.e. a Munich farm, determines pathogenicity and drug resistance of the BV-21002 through experiments, separates a lytic vibrio parahaemolyticus phage from culture sludge by taking the BV-21002 as a host, and determines the genotype of RDP-VP-21010 through a whole genome sequence.
Example 1 isolation and identification of pathogenic bacteria and phages
(1) Isolation and identification of pathogenic vibrio parahaemolyticus BV-21002
Sampling from gills of the ill prawns, coating the ill prawns on a TCBS culture medium by adopting an aseptic operation method, culturing at 37 ℃ for 18-24 hours, and then presenting round green colonies with neat edges on a TCBS agar plate, wherein the diameter of the round green colonies is about 2-3 mm; then selecting a typical colony to streak on a Vibrio coimajiaensis color development culture medium, culturing for 18-24h at 37 ℃, then presenting a purple colony on the Vibrio coimajiaensis color development culture medium, selecting the typical colony to continuously streak and purify for 3 times, then selecting a single colony to inoculate in 5mL of 2216E liquid culture medium, culturing for 6h at 37 ℃ and 200rpm in a shaking way, and obtaining a uniform and turbid bacterial suspension. Then identified as vibrio parahaemolyticus by 16sRNA molecular identification, one of the vibrio parahaemolyticus is named BV-21002 and stored in a refrigerator at minus 80 ℃.
(2) Phage isolation and identification
The sludge sample for the test is collected from the water bottom sludge of Qingdao, i.e. inkfarmer in 2021 years and is used as a sample for phage separation.
Sample treatment: soaking a proper amount of sludge in 20mL of 2216E liquid, centrifuging at 10000rpm for 10 minutes, removing larger impurities and most bacteria, and filtering and sterilizing by using a 0.22-micron microporous filter membrane.
Preparing a mixed bacterial suspension: 2mL of the filtrate and 1mL of the host cell suspension were added together to 27mL of 2216E liquid medium after autoclaving, and then placed in an incubator at 37 ℃ for overnight culture. After culturing, 3mL of the solution is taken, centrifuged at 10000rpm for 5 minutes, filtered and sterilized by a 0.22 mu m microporous filter, and the filtrate is collected for later use, namely the stock solution which is supposed to contain the phage.
Phage separation: separating phage by double plate method, mixing the mixed bacterial suspension filtrate 0.1mL and secondary hemolytic vibrio suspension 0.1mL, adding into 2216E agar heated to 45-54 deg.C 3mL 4%, spreading double plates, culturing in 37 deg.C incubator for 6-8h, observing result, if there is phage, there is transparent plaque on the plate. Soaking plaque in 1mL of 3% saline for 10min, adding 0.1mL of leachate and 0.1mL of bacterial suspension into 3mL of 2216E agar heated to 45-54 ℃, spreading a double plate, culturing in a 37 ℃ incubator for 4-6h for purification, and purifying for 2-3 times according to the steps until the plaque is uniform in size, thereby obtaining a phage, and the phage is named as RDP-VP-21010. The plaque formed by RDP-VP-21010 on the double plate has a diameter of 4mm-5mm, a round shape, transparency and a halo.
(3) Measurement of cell titer
Laying a double-layer plate on the purified phage, taking a single plaque, adding the plaque into 2216E liquid culture medium, simultaneously adding host bacterial suspension, after the phage is proliferated, centrifuging the proliferation liquid at 10000rpm for 5 minutes, filtering by using a 0.22-micrometer microporous filter, performing 10-fold gradient dilution on the filtrate to 10-8 times, taking 0.1mL of phage diluent and 0.1mL of host bacterial suspension with the last 3 dilutions, laying the double-layer plate, and determining the titer of the phage. Phage titer (pfu/mL) ═ plaque number dilution factor ÷ 0.1. The result shows that the plaque has consistent shape, size and definition and has typical lytic phage characteristics; the titer of the phage can reach more than 10^12 pfu/mL.
(4) Preservation of phages
Phage multiplication fluid was mixed with 60% glycerol according to 1:1, and storing in liquid nitrogen.
EXAMPLE 2 Electron microscopy of phages
Dropping 20 μ L of liquid containing crude phage particles on a copper net, naturally precipitating for 15min, sucking off excessive liquid from the side by using filter paper, adding a drop of 2% phosphotungstic acid (PTA) on the copper net to dye the phage for 10min, sucking off the staining solution from the side by using filter paper, and observing the form of the phage by using an electron microscope after a sample is dried.
As can be seen from FIG. 1, this phage RDP-VP-21010 is a Long-tailed phage with a regular hexagonal head diameter of about 56nm and a tail length of 103 nm.
Example 3 phage genome sequencing
After enrichment culture of a single phage, centrifuging for 15 minutes at 8000g under the condition of 4 ℃, adding 10% PEG8000 and 0.5M NaCl, standing overnight, adding equal amount of chloroform, mixing uniformly, standing for layering, centrifuging for 10 minutes at 5000g, removing a chloroform layer and a PEG layer, adding restriction endonuclease for digestion treatment, suspending the phage under the condition of gradient density cesium chloride, dialyzing for 3 times by TM buffer solution at the later stage, and sending the phage to a Huada gene biological sequencing company for whole genome sequencing after 30 minutes each time. The results are shown in the sequence listing.
Example 4: determination of phage pH stability
Adjusting pH of normal saline with dilute hydrochloric acid and dilute NaOH solution to obtain buffer solution with pH of 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, 12, and diluting phage with prepared buffer solution to 1 × 1010pfu/mL, putting the diluted solution in a water bath at 37 ℃ for 1h, diluting with 10 times of normal saline, and then spreading a double-plate; counting after inverted culture at 37 ℃ for 4-6 h.
As can be seen from FIG. 2, after 1h of incubation under different pH conditions, the phage RDP-VP-21010 can maintain better activity in the pH range of 4.0-11.0, the phage titer does not change significantly, and the phage titer is highest at pH 8.0, reaching 5X 109pfu/ml. The titer of the phage decreased significantly with increasing or decreasing pH. The results show that the bacteriophage RDP-VP-21010 can resist weak acid and strong alkali conditions and has good acid-base tolerance.
Example 5: determination of optimal multiplicity of infection
The phage multiplication solution and the host were added to 2216E in a ratio of multiplicity of infection of 10:1, 1:10, 1:100, 1:1000, 1:10000, and the total volume of the culture system was kept the same. After shaking culture at 37 ℃ and 200rpm for 8h, centrifuging at 12000r/min at normal temperature for 5min, and spreading the supernatant on a double-plate to determine the titer. The results are shown in Table 1.

As can be seen from the data in Table 1, when the infection complex number is 1:1, the proliferation medium is relatively clear and the titer is the highest after 8 hours of culture, indicating that the optimal infection complex number of the Vibrio parahaemolyticus phage RDP-VP-21010 of the present invention is 1: 1.
Example 6: phage versus host lysis curve
The host bacterial suspension cultured for 6h was inoculated into 100mL of 2216E medium at a ratio of 1:1, and cultured with shaking at 37 ℃ and 200rpm for 2h (OD about 0.45, about 6.0X 10)8cfu/mL) of the bacterial suspension was divided into 4 sterilized Erlenmeyer flasks, and 0.2mL (about 4.0X 10) of a phage-proliferating solution was added to 3 of the Erlenmeyer flasks at the optimum multiplicity of infection11pfu/mL, filtered through a 0.22 μm membrane), while taking the average value with no phage added as a control, the results are shown in FIG. 3.
As can be seen from FIG. 3, the Vibrio parahaemolyticus phage RDP-VP-21010 is added into the suspension of the host bacterium BV-21002, after 1h, a certain lysis effect appears, after about 1h, the OD600 value begins to decrease, the effect lasts until 8.5h, after 9h, the mixed bacterium solution begins to become turbid, the OD value gradually increases, and the balance is reached after 12h, but the OD value is still lower than that of the host bacterium. In the early stage, the phage is in the adsorption and infection stage of the host bacteria, only a small part of the phage starts to crack the host, so the amount of the cracked host is far less than the growth amount of the host, the OD600 still keeps a certain rising trend in the first 1h, the number of the cracked host is larger than the proliferation speed of the host along with the exponential increase of the number of the phage, the OD600 value starts to be gradually reduced, and the mixed bacteria liquid is clearest. After the continuous action for 9 hours, the mixed bacteria liquid turns turbid, the original sensitive bacteria are cracked by the bacteriophage, and the flora replaced by the mutant tolerant bacteria is alternated.
Example 7: determination of the thermostability of phages
The phage stock solution is subpackaged into EP tubes, incubated for 30min at 40 ℃, 50 ℃, 60 ℃ and 70 ℃, diluted by 10 times of normal saline, and spread on double plates to determine the titer. The results are shown in FIG. 4.
As can be seen from FIG. 4, the activity of the phage was kept stable at 40 ℃ to 60 ℃ and the titer was 1012pfu/mL or more; after the phage is acted for 30min at 70 ℃, the titer can still reach 7.4 x 10^4pfu/mL, which indicates that the phage has certain temperature tolerance.
Example 8: genetic test for stability
The RDP-VP-21010 is continuously passaged for 15 times, the same amount of RDP-VP-21010 and BV-21002 is added into each generation of culture medium, the culture time of each generation is 6h, and then double plates are paved to determine the titer. The results are shown in FIG. 5.
As shown in FIG. 5, the titer of RDP-VP-21010 was measured after continuous passage through 15 generations, and the titer stabilized at 1011And pfu/mL or more indicates that the phage has better genetic stability and is suitable for industrial production.
Example 9: cracking rate test
Under aseptic conditions, 1mL of sample and 1mL of host bacterium solution (1X 10)6CFU/mL), incubating for 15 minutes at 37 ℃, mixing uniformly, and diluting to 10 with physiological saline-1-10-3Each gradient was plated at 100. mu.L on 2216E agar plates and incubated at 37 ℃ for 24 hours, with each gradient repeated twice. Simultaneously, 1mL of 3% saline and 1mL of host bacterium solution (1X 10)6CFU/mL) as a blank, repeat the above steps. Plates with 30-300 colonies were picked for enumeration. The test was repeated 3 times and the average was taken. The phage lysis rate was (1-number of treated colonies/number of control colonies) × 100%.
Through calculation, the cracking rate of the RDP-VP-21010 reaches 93.1%, and the RDP-VP-21010 has a good cracking effect on a host and is suitable for being used in a culture process.
Example 10: RTD experiment
The plates were divided into zones, a drop of host bacteria (about 10. mu.L) was added to the center of each zone, air dried, a drop of phage was added at different dilutions to the spot of host bacteria, air dried, and incubated overnight at 37 ℃. The result is shown in FIG. 6, where the host bacteria cannot grow continuously, and is called CL concentration (CL concentration is the concentration of host bacteria in the environment that can be completely lysed and can be used as a dilution scheme for guiding the production practice).
As can be seen from FIG. 6, RDP-VP-21010 is diluted 105After doubling, the bacillus subtilis still has better lysis effect on host bacteria.
Example 11: method for large-scale production of bacteriophage
The process flow is simple, reliable and effective, and is favorable for industrial production.
The concrete steps
1. Seed preparation
(1) Preparing a host seed: under aseptic condition, selecting single colony, streaking on 2216E agar medium, culturing at 37 deg.C overnight,
then, the cultured single colony is picked up and put into 5mL 2216E liquid culture medium, shake culture is carried out for 6h at 37 ℃ and 180rpm, then the single colony is inoculated into the 2216E liquid culture medium by the inoculation amount of 1%, and shake culture is carried out for 6h at 37 ℃ and 180 rpm. The prepared seed liquid is stored in a refrigerator at 4 ℃ for later use.
(2) Preparing a phage seed: diluting the preserved phage seeds properly, placing 100 μ L and 100 μ L host bacteria in the upper layer culture medium, pouring the double-layer plate, culturing at 37 deg.C for 4-6h, scraping with sterile coating rod, centrifuging at 10000rpm for 5min, sterilizing with 0.22 μm filter membrane, and storing the collected filtrate in 4 deg.C refrigerator.
The phage and the host are respectively inoculated in 2216E liquid culture medium in the proportion of 1 percent and 5 percent, and cultured for 6-8h at 37 ℃ and 180rpm until the proliferation liquid is clear and a large amount of floccules appear.
2. And (3) phage multiplication: inoculating 2% of host bacteria in a seeding tank, culturing for 2-3h, then inoculating 0.6-0.8 OD600, then inoculating 1% of the inoculum size to phage seeds, culturing for 6-8h, until the dissolved oxygen is reversely increased and the pH value tends to be stable.
Inoculating 2% of host bacteria in a seeding tank, culturing for 4-5h until dissolved oxygen begins to slowly decrease.
Inoculating the prepared host bacteria into a large fermentation tank with the inoculation amount of 5%, culturing for 2-3h until dissolved oxygen begins to slowly decrease, then inoculating phage seeds with the inoculation amount of 5%, increasing the ventilation amount to 1vvm, increasing the rotation speed to 150rpm, culturing for 8-10h until the dissolved oxygen reversely increases and the pH value increases, and finishing proliferation.
Note: fermentation tank parameters: the seeding tank has 200-300rpm, 37 deg.c and ventilation amount of 0.6-1 vvm. Fermentation tank: 120-150pm, 37 ℃ and the ventilation volume is 0.6-1 vvm.
The components of the culture medium: 0.5% of yeast extract powder, 1% of peptone, 0.1% of beef extract, 0.5% of glycerol, 0.5% of sodium chloride, 0.1% of monopotassium phosphate, 0.1% of dipotassium phosphate, 20ppm of calcium chloride and 20ppm of magnesium chloride.
3. Phage post-treatment process
(1) And (3) filtering: and (3) respectively adopting 800nm and 500nm ceramic membranes for filtration to remove cell fragments and host bacteria which are not cracked, wherein the filtration pressure is less than 0.4mpa, and the damage of shearing force to the bacteriophage is reduced.
(2) Concentration: hollow fiber membranes are used for concentration to improve the titer of the phage.
(3) A stabilizer: adding 5% of glycerol, 3% of trehalose, 3% of polysorbate, 3% of sodium dihydrogen phosphate and 2% of sodium phosphate, and stirring until the mixture is completely dissolved.
(4) And (3) degerming: three-stage filtration (glass fiber 0.45 μm, glass fiber 0.2 μm, polyethersulfone 0.22 μm) was used for sterilization.
(5) The filter-sterilized phage liquid was stored at 4 ℃.
