Background

Ozone is a broad-spectrum, high-efficiency, quick, safe and secondary-pollution-free sterilizing gas, not only has strong killing capability to various bacteria, but also is effective to microorganisms such as viruses, fungi and the like. In addition to sterilization, ozone also has the effects of activating and repairing cells and improving the secretion of autoimmune substances. However, the ozone is extremely active and difficult to store, the half-life period in the air is 15-30 minutes, the ozone is more easily decomposed in water than in the air, and the ozone is extremely unstable and difficult to store and transport, so that the ozone can be produced at present only by using equipment. Ozone oil is the product of ozone curing application technology. Compared with ozone gas, the ozone oil has more stable property, can be stored for two years, and can be widely applied to the aspects of antibiosis, antiphlogosis, itching relieving, detoxification and the like of skin and mucous membrane.

The treatment of chronic pelvic inflammatory disease mainly adopts Chinese and western medicine treatment and physical therapy. Enema refers to a treatment method in which a medicinal liquid is infused into the intestines through the anus, and the disease is treated by absorption through the intestinal mucosa. It has certain therapeutic effect on many diseases, such as male prostate and female pelvic inflammation, and has certain therapeutic effect on inflammation through local administration of enema; for intestinal diseases, enema has a very remarkable treatment effect, such as ulcerative colitis, acute and chronic bacillary dysentery and chronic inflammation of colorectal, and enema administration has many advantages compared with oral administration. The retention enema is mostly used for enema treatment, and a small amount of liquid (50-100 ml) is mixed with the medicine and injected into the deeper part of the anus (rectum or sigmoid colon) for more than 10-15 minutes. At present, most of the drugs used for clinical enema treatment are antibiotics, but the antibiotics are not used properly and cause great harm, and the following aspects are mainly adopted:

Based on the state of the art, there is a need for an enema drug that has no side effects and reduces the harm caused by the overuse of antibiotics in clinical practice.

Disclosure of Invention

The invention aims to overcome the defect that harm is caused by excessive use of antibiotics in enema medicines in the prior art, and provides application of ozone oil in preparing medicines for enema treatment.

The invention also provides application of the ozone oil in preparing a medicine for treating pelvic inflammation through enema.

The invention further provides application of the ozone oil in preparing a medicine for treating colitis through enema.

The pelvic inflammatory disease can be acute pelvic inflammatory disease or chronic pelvic inflammatory disease.

The colitis may be acute colitis or chronic colitis.

The source of the ozone oil is not particularly required in the invention, and the ozone oil can be obtained by commercial products or the ozone oil prepared by the prior art. Preferably, the ozone oil comprises ozone and vegetable oil, wherein the content of the ozone is 20-120 mu g/mL relative to 1mL of the ozone oil; preferably 45-75. mu.g/ml.

The vegetable oil used in the present invention may be based on vegetable oils known to those skilled in the art, for example, the vegetable oil may be at least one of olive oil, coconut oil, soybean oil, sunflower oil, corn oil, hemp oil, camellia oil, cottonseed oil, palm oil, rapeseed oil, canola oil, walnut oil, sesame oil, peanut oil, or linseed oil; preferably linseed oil and/or camellia oil.

The method for preparing the ozone oil has no special requirements, and can be obtained by a method for preparing the ozone oil by the prior art. For example, ozone oil is produced by contacting ozone with vegetable oil in a reactor as follows.

The conditions for contacting ozone and vegetable oil in the reactor are not particularly limited, and the contacting temperature can be 15-65 ℃; preferably 25 to 60 ℃; the contact time may be 3-25 h; preferably 5-15 h.

The invention has no special requirement on the reactor for preparing the ozone oil, and can be a conventional gas-liquid mixing tank, for example, an ozone oil all-in-one machine.

Compared with the enema medicament in the prior art, the enema medicament has no toxic or side effect on human bodies, no irritation, no residue, no drug resistance, no mutagenicity, safety and reliability. The traditional Chinese medicine composition has the advantages of no pain, obvious pain relief effect and obvious curative effect in the treatment process, and can also obviously reduce the use of antibiotics, shorten the inflammation time, improve the cure rate and reduce the treatment cost of patients. Can be used as a substitute drug of antibiotics, and can overcome the harm caused by excessive use of antibiotics.

The experimental procedures used in the following examples are all conventional procedures unless otherwise specified.

Materials, reagents and the like used in the following examples are commercially available unless otherwise specified.

The iodometry in the following examples and comparative examples was tested using the following method:

1mL of the sample was taken out and placed in a 250mL conical flask with a stopper, and 30mL of a chloroform-glacial acetic acid mixture (chloroform: glacial acetic acid 2: 3) was added thereto, and the sample was completely dissolved by gentle shaking. 1.00mL of saturated potassium iodide solution was added accurately, the cap was closed, shaken gently for 0.5min, and left in the dark for 3 min. The calculation results are expressed as the arithmetic mean of two independent measurements obtained under repetitive conditions, with two significant figures retained.

Example 1: preparation of ozone oil

1) 40L of linseed oil is measured and placed in a gas-liquid mixing tank (maximum capacity of 50L) of a JY-CY-50L ozone oil all-in-one machine (Xuzhou Jinyuan ozone equipment Co., Ltd.);

2) electrifying the ozone-oil integrated machine, and opening a circulating cooling system; opening oxygen vent valve to ensure normal oxygen supply, starting ozone generator, and regulating oxygen gas inflow to 2m3H, leading the ozone concentration to be 110 mg/L;

3) keeping aeration for 12.7h, and checking whether aeration is normal or not through an observation hole on the gas-liquid mixing tank; and the temperature of the gas-liquid mixing tank body is not more than 55 ℃;

4) after the preparation is finished, after cooling, opening a liquid discharging port at the bottom of the tank body, and discharging oil liquid in the gas-liquid mixing tank. The ozone concentration was 70. mu.g/ml as determined by iodometry.

Example 2: preparation of ozone oil

1) Measuring 30L of camellia oil, and placing the camellia oil in a gas-liquid mixing tank (maximum capacity of 50L) of a JY-CY-50L ozone oil all-in-one machine (Xuzhou Jinyuan ozone equipment Co., Ltd.);

2) electrifying the ozone-oil integrated machine, and opening a circulating cooling system; opening oxygen vent valve to ensure normal oxygen supply, starting ozone generator, and regulating oxygen gas inflow to 1.5m3H, the ozone concentration is 120 mg/L;

3) keeping aeration for 8.3h, and checking whether aeration is normal or not through an observation hole on the gas-liquid mixing tank; and the temperature of the gas-liquid mixing tank body is not more than 55 ℃;

4) after the preparation is finished, after cooling, opening a liquid discharging port at the bottom of the tank body, and discharging oil liquid in the gas-liquid mixing tank. The ozone concentration was measured by iodometry to be 50. mu.g/ml.

Example 3: preparation of ozone oil

1) Measuring 30L of camellia oil, and placing the camellia oil in a gas-liquid mixing tank (maximum capacity of 50L) of a JY-CY-50L ozone oil all-in-one machine (Xuzhou Jinyuan ozone equipment Co., Ltd.);

2) electrifying the ozone-oil integrated machine, and opening a circulating cooling system; opening oxygen vent valve to ensure normal oxygen supply, starting ozone generator, and regulating oxygen gas inflow to 2m3H, the ozone concentration is 120 mg/L;

3) keeping aeration for 8h, and checking whether aeration is normal or not through an observation hole on the gas-liquid mixing tank; and the temperature of the gas-liquid mixing tank body is not more than 55 ℃;

4) after the preparation is finished, after cooling, opening a liquid discharging port at the bottom of the tank body, and discharging oil liquid in the gas-liquid mixing tank. The ozone concentration was measured by iodometry to be 64. mu.g/ml.

Example 4: application of ozone oil in enema treatment of colitis

1) Clinical data: 30 cases of chronic colitis patients, age 28-70 years, course of disease more than 1 year.

2) The treatment method comprises the following steps: the patient takes the left lateral decubitus position, the enema tube is coated with paraffin oil, the enema tube is inserted into the anus for about 10-15cm, 50ml of ozone oil prepared in the embodiment 1 is slowly pushed into the anus by a 50ml injector, then the enema tube is slowly pulled out, paraffin oil outside the anus and overflowing ozone oil are wiped off, the knee chest decubitus position of the patient is 0.5-1 hour, and the retention time is prolonged. 15 days is a course of treatment.

3) The curative effect standard is as follows: and (3) curing: disappearance or substantial disappearance of symptoms, signs; the effect is shown: symptoms and physical signs are obviously improved; the method has the following advantages: symptoms and signs are improved; and (4) invalidation: symptoms and signs do not improve or even worsen.

4) The treatment results are as follows: 20 of 31 cases with 2 treatment courses are effective, and the effective rate is 64.5%; of 42 cases treated in 5 courses of treatment, 36 cases are effective, and the effective rate is 85.7%.

After the treatment of 5 treatment courses, compared with the treatment before and after the treatment, 9 cases are cured, 30.0 percent of the treatment course, 7 cases are obviously effective, 23.3 percent of the treatment course, 9 cases are effective, 30 percent of the treatment course, 5 cases are ineffective, 16.7 percent of the treatment course, and the total effective rate is 85.3 percent.

Comparative example 1: application of antibiotic enema for treating colitis

1) Clinical data

The same as in example 4.

2) The treatment method comprises the following steps: 100mL of 0.2% metronidazole solution, 2mL of gentamicin: 8 ten thousand U, dexamethasone 1 mL: 1mg of tin powder and 1.5g of tin powder are mixed to prepare a mixed solution. Enema is carried out on the mixed solution 1 time per day, 100mL each time, 15 days later, 1 time every other day, and 3 months are 1 course of treatment.

4) The treatment results are as follows: compared with the patients before and after treatment, 9 cases of the patients are cured, accounting for 30.0 percent, 8 cases of the patients with significant effect accounting for 26.7 percent, 6 cases of the patients with significant effect accounting for 20.0 percent, 7 cases of the patients with no effect accounting for 23.3 percent, and the total effective rate accounting for 76.7 percent.

The results of example 4 and comparative example 1 for enema treatment of colitis are given in the following table:

Example 5: application of ozone oil in enema treatment of pelvic inflammation

1) Clinical data: 60 patients with acute or chronic pelvic inflammatory disease, 20-50 years old.

2) The enema treatment method comprises the following steps: before sleep, the patient can take the treatment after defecating. The patient takes the left lateral decubitus position, the enema tube is coated with paraffin oil, the enema tube is inserted into the anus for about 10-15cm, 50ml of ozone oil prepared in the embodiment 1 is slowly pushed into the anus by a 50ml injector, then the enema tube is slowly pulled out, the paraffin oil outside the anus and the ozone oil overflow are wiped off, the retention time is more than 4 hours, and the menstrual period of the female patient is suspended. The medicine is taken once a day, 10 days are a treatment course, and the treatment course is three.

3) The curative effect standard is as follows: the clinical symptom sign completely disappears to cure; the symptom sign is improved to be better than before; the patients with no improvement in the treated signs were ineffective.

4) The treatment effect is as follows:

40 cases are cured, accounting for 67%; 18 cases are improved, accounting for 30 percent; 2 cases of the traditional Chinese medicine are invalid, the traditional Chinese medicine accounts for 3 percent, and the total effective rate reaches 97 percent.

Comparative example 2: application of traditional Chinese medicine enema for treating pelvic inflammation

1) Clinical data

2) The treatment method comprises the following steps: 30g of patrinia, 30g of coix seeds, 20g of red paeony roots, 120g of frankincense, 120g of myrrh, 20g of Chinese honeylocust spines, 20g of corydalis tuber, 20g of peach kernels and 20g of salvia miltiorrhiza, adding water for decoction, taking 150mL of liquid medicine, and carrying out enema when the temperature of the liquid medicine is proper (39-41 ℃). Enema is started 3d after menstruation, 1 time per day, and the administration is continued for 10 days, and 3 months are 1 course of treatment.

3) The curative effect standard is as follows:

4) The treatment results are as follows: compared with the patients before and after treatment, the traditional Chinese medicine composition has the advantages that 41 cases are cured, accounting for 68.3%, 13 cases are improved, accounting for 22.7%, 6 cases are invalid, accounting for 10%, and the total effective rate is 90%.

The effects of example 5 and comparative example 2 for enema treatment of pelvic inflammatory disease are as follows:

compared with the comparative example 2, the treatment effect is better and the total effective rate is higher in the example 5.
