本发明涉及革兰氏阳性菌的转化方法。
背景技术
以往,作为细菌等的转化方法,已知有如下方法:将重组质粒、DNA片段导入大肠杆菌等宿主中的感受态细胞转化法(参照非专利文献1);原生质体转化法、电穿孔法(Electroporation法)、农杆菌介导法、粒子枪法、磷酸钙法、脂质体转染法等。然而,这样的现有的转化方法中,难以将例如100kb以上的大尺寸的DNA片段导入宿主DNA中。
作为将大尺寸的DNA片段导入宿主DNA中的方法,已知使用基于获得了自然转化能力的枯草芽孢杆菌的接合转移的方法。然而,该方法中存在如下缺陷:需要将枯草芽孢杆菌导入自然转化能力高的细胞这样特殊的培养条件;转化本身需要花费时间和精力。进而,难以将超过数百kb的大尺寸的DNA无损伤地导入宿主DNA中。
另一方面,已知源自纳豆菌的质粒pLS20具有通过接合转移由枯草芽孢杆菌等革兰氏阳性菌细胞复制转移给其它革兰氏阳性菌细胞的能力。进而还明确了:该pLS20在供体中作为辅助质粒发挥作用,能够将供体所具有的其它质粒转移至受体(参照非专利文献2和3)。由于与其它质粒同时作为辅助质粒的pLS20也转移至受体侧的革兰氏阳性细菌中,因此对于其它革兰氏阳性细菌也能够作为供体进一步重复接合转移,从控制自然环境的观点出发也可能成为问题(参照非专利文献4和5)。

发明要解决的问题
在这样的情况下,本发明的目的在于提供,能将大尺寸的DNA无损伤地导入革兰氏阳性菌的宿主DNA中、简便且有效的革兰氏阳性菌的新型转化方法。进而,目的还在于提供,将期望的DNA片段蓄积于受体(受体菌)的染色体中,能够制作经人为设计的长的DNA的方法、转化后的细胞从控制自然环境的观点出发也不会成为问题的方法。
用于解决问题的方案
本发明人等为了解决上述课题而进行了深入研究,结果发现:通过使用使pLS20cat的oriTLS20区域失活的pLS20catΔoriT作为辅助质粒,从而在革兰氏阳性菌的接合转移中,能够制作仅转移了期望的染色体、质粒的转化细胞而辅助质粒本身不被转移至受体(受体菌)侧。根据本发明的方法,得到的转化细胞不具有辅助质粒,因此再次利用同样的方法能够作为受体(受体菌)接受期望的染色体、质粒的接合转移。因此,根据本发明,能将期望的DNA片段蓄积于受体(受体菌)的染色体中,设计并制作长的DNA。进而,由于利用本发明的方法而得到的转化细胞不具有辅助质粒,因此也不会作为供体进一步重复接合转移,因此从控制自然环境的观点出发也不会出现问题。另外,基于本发明的方法的转化仅通过将供体(供体菌)与受体(受体菌)的液体培养液混合就能够迅速且简便地进行,在合成生物学的产业活动中,可以适宜地用于微生物、培养细胞等的制作。即,本发明的主旨如下。
[1]一种通过接合转移(conjugative transfer)进行的革兰氏阳性菌的转化方法,其特征在于,使用使DNA转移起始位点(oriT)区域失活的辅助质粒。
[2]根据[1]所述的转化方法,其中,上述辅助质粒为从pLS20cat使oriT区域失活的质粒。
[3]根据[1]或[2]所述的转化方法,其中,上述接合转移中的供体菌具有:使上述DNA转移起始位点(oriT)区域失活的辅助质粒、及组入了上述DNA转移起始位点(oriT)区域的染色体DNA或质粒。

发明的效果
根据本发明的革兰氏阳性菌的转化方法,能够将大尺寸的DNA无损伤地且在短时间内转移至受体侧。进而,通过使用使pLS20cat的oriTLS20区域失活的pLS20catΔoriT作为辅助质粒,从而在革兰氏阳性菌的接合转移中,能够制作仅转移了期望的染色体、质粒的转化细胞而辅助质粒本身不被转移至受体侧。根据本发明的方法,由于得到的转化细胞不具有辅助质粒,因此再次利用同样的方法,能够作为受体(受体菌)接受期望的染色体、质粒的接合转移。因此,根据本发明,能将期望的DNA片段蓄积于受体(受体菌)的染色体中,设计并制作长的DNA。进而,利用本发明的方法而得到的转化细胞不具有辅助质粒,所以也不会作为供体进一步重复接合转移,因此从控制自然环境的观点出发也不会出现问题。另外,基于本发明的方法的转化仅通过将供体(供体菌)与受体(受体菌)的液体培养液混合就能够迅速且简便地进行,在合成生物学的产业活动中,可以适宜地用于微生物、培养细胞等的制作。

具体实施方式
以下对本发明的革兰氏阳性菌的转化方法进行详细地说明。需要说明的是,本说明书中,DNA、载体的制备等分子生物学方法只要没有特别声明,则可以利用本领域技术人员公知的通常记载于实验书的方法或依据其的方法来进行。另外,本说明书中使用的术语只要没有特别声明,则用该技术领域中通常使用的含义来解释。
<革兰氏阳性菌的转化方法>
本发明的革兰氏阳性菌的转化方法的特征在于,使用在接合转移中使DNA转移起始位点(oriT)区域失活的辅助质粒。
本发明中革兰氏阳性菌是指革兰氏染色阳性的细菌及其它菌类。革兰氏阳性菌通常具有相对较厚(15nm~80nm)的细胞壁,大多数菌在外层缺乏脂多糖。另外,对溶菌酶敏感性高的菌较多。作为具体的革兰氏阳性菌,可列举出芽胞杆菌属(Bacillus;枯草芽孢杆菌(Bacillus subtilis)、炭疽杆菌(Bacillus anthracis)、嗜热菌(地芽孢杆菌属(Geobacillus))、蜡样芽孢杆菌(Bacillus cereus)、地衣芽孢杆菌(Bacilluslicheniformis)、巨大芽孢杆菌(Bacillus megaterium)、短小芽孢杆菌(Bacilluspumilus)、苏云金芽孢杆菌(Bacillus thuringiensis)等)、李斯特菌(Listeria)、葡萄球菌属(Staphylococcus;葡萄球菌)、链球菌属(Streptococcus;链球菌)、肠球菌属(Enterococcus;乳酸菌等)、梭菌属(Clostridium)等。本发明中,作为革兰氏阳性菌,优选芽孢杆菌,其中,更优选枯草芽孢杆菌、炭疽杆菌、嗜热芽孢杆菌属细菌,进一步优选嗜热地芽胞杆菌(Geobacillus kaustophilus)。
本发明中转化是指:通过革兰氏阳性菌的基因组DNA的修饰或向革兰氏阳性菌中导入DNA而引起的修饰,包括由染色体DNA的缺失、复制、突变、或自主复制质粒的导入引起的全部性状变化。所导入的DNA可以被组入染色体中进行维持、复制,还可以如质粒等那样独立于染色体地进行维持、复制。将DNA组入革兰氏阳性菌的染色体的特定位点时,还可以使用同源重组技术。
本发明中接合转移是在将具有不同性状的细菌混合培养时所发生的现象,是指某细菌(供体菌)的一部分基因转移至其它细菌(受体菌)中。接合转移能力的强度受到供体的染色体上或者供体所具有的质粒上的与接合转移相关的基因的影响。与接合转移相关的基因有自传递性基因、共轭传递性基因、以及oriT序列。自传递性基因所编码的蛋白质参与供体菌/受体菌之间的相互作用。共轭传递性基因所编码的蛋白质具有:在oriT序列加入缺口的功能、进而稳定地搬运成为单链的DNA的功能。oriT序列由缺口位点和用于进入缺口的识别序列组成。这3种共存而首次发生接合转移。
本发明中辅助质粒是指在预将某基因转移至目标菌中时有助于基因转移至该菌中的质粒,具体而言,可以列举出枯草芽孢杆菌中的pLS20等。若将具有辅助质粒的菌用于转化时,与自然转化相比,能显著提高转化效率、技术的简便性、所需的时间效率。
本发明中DNA转移起始位点(oriT)区域是指包含DNA迁移的启动序列的区域。
以下对本发明的革兰氏阳性菌的转化方法进行具体地说明。即,本发明的革兰氏阳性菌的转化方法具有如下工序:
工序(1):制备供体菌,所述供体菌具有:使DNA转移起始位点(oriT)区域失活的辅助质粒、及组入了上述DNA转移起始位点(oriT)区域的染色体DNA或质粒;
工序(2):由上述供体菌对受体菌进行接合转移。
本发明的革兰氏阳性菌的转化方法优选根据需要还包括工序(3):使期望的DNA片段蓄积于受体菌的染色体中。
[工序(1)]
(受体株(受体菌)的构建工序)
作为受体株(受体菌),如上所述优选芽孢杆菌,其中可以更优选使用枯草芽孢杆菌、炭疽杆菌、嗜热菌,进一步优选使用枯草芽孢杆菌。在这些受体株(受体菌)中预先导入抗生素抗性基因以便能够确认在工序(2)中有无基于接合转移的转化。向受体株(受体菌)中导入抗生素抗性基因可以通过本领域技术人员利用现有公知的方法来进行。
例如,作为受体株(受体菌)而使用枯草芽孢杆菌时,可以通过基于奇霉素抗性基因等的置换使枯草芽孢杆菌的comK等基因失活而导入。即,以枯草芽孢杆菌DNA作为模板通过PCR来扩增分别对应于comK等基因的上游和下游区域的2个DNA片段。此时,设计并使用适于上游片段和下游片段的引物。另外,使用适合的引物来扩增包含TMO310株等其它枯草芽孢杆菌株的奇霉素抗性基因等的其它DNA片段。通过重组PCR将3个片段连接,并将奇霉素抗性基因等插入comK等上游与下游的区域之间。通过转化为枯草芽孢杆菌并将重组PCR片段导入受体株中而赋予奇霉素等抗生素抗性,能够得到在本发明中受体株得到修饰的新型的枯草芽孢杆菌株。
(使oriT区域失活的辅助质粒的构建工序)
辅助质粒的oriT区域的失活可以通过本领域技术人员利用现有公知的基因操作方法来进行。需要说明的是,oriT区域的失活包括:一部分oriT区域的缺失、整个oriT区域的缺失、oriT区域的1个或多个碱基的置换。例如,可以利用无痕敲除(marker-freedeletion)等方法使oriT区域缺失而失活。以下对使用pLS20cat作为辅助质粒时的例子进行说明。
将pLS20catDNA作为模板,使用上游区域的引物oriT-uF/oriT-uR和oriT-dF/ori并通过PCR来扩增对应于pLS20cat的oriTLS20的上游(片段1)和下游(片段2)区域的2个DNA片段。片段1的末端和片段2的前端在22bp处是相同的,因此可以利用这些区域通过转化而使oriTLS20区域缺失。使用mazF-F/mazF-R作为引物由TMO311 DNA扩增mazF kan盒的另一个DNA片段(片段3)。3个PCR片段可以通过使用了oriT-uF/oriT-dR作为引物的重组PCR以1-3-2顺序连接的方式进行设计。其次,将重组PCR片段转化为PKS11株并赋予卡那霉素抗性,能够得到新的株YNB022。对于该株,可通过oriTLS20区域的双交叉事件组入PCR片段来修饰pLS20cat。将YNB022在含有卡那霉素的LB液体培养基中以37℃进行一晩左右增殖。将培养液的整分试样(aliquot)移至含有1mM异丙基硫代吡喃半乳糖苷(IPTG)的新鲜的LB液体培养基中,将细胞以37℃进行2小时左右增殖。其次,将培养液的整分试样在包含1mM IPTG的LB孔板上展开,以37℃孵育一晩左右。在IPTG的存在下表达mazF而产生自杀毒素,只有能够通过分子内重组而逃离mazF kan盒的细胞才能存活。需要说明的是,在孔板上出现的菌落中,对卡那霉素敏感性菌落进行测序,能够确认oriTLS20区域被准确删除。上述各引物是下述表2所述的引物。
(供体株的构建工序)
作为供体株(供体菌),如上所述优选芽孢杆菌,其中可以更优选使用枯草芽孢杆菌、炭疽杆菌、嗜热菌,进一步优选使用枯草芽孢杆菌。本工序中,在制作供体株时,使用事先导入使上述中制备的oriT区域失活的辅助质粒而得到的枯草芽孢杆菌,在该染色体DNA中组入源自辅助质粒的oriT区域。另外,还可以代替染色体DNA来转移质粒,在此情况下,将包含源自辅助质粒的oriT区域的质粒导入至事先已导入了使oriT区域失活的辅助质粒的枯草芽孢杆菌中,从而制备能在工序(2)中使用的供体株。
在供体株的染色体DNA中组入源自辅助质粒的oriT区域的方法可以通过本领域技术人员依据现有公知的基因操作的方法来进行。以下,以将使用pLS20cat作为辅助质粒来构建作为供体株的枯草芽孢杆菌YNB060的情况为例,对本工序进行说明。
即,使用引物yhfM-uF/yhfM-uR1(针对上游)和yhfM-dF/yhfM-dR(针对下游),由枯草芽孢杆菌168株DNA来扩增对应于yhfM的上游(片段1)和下游(片段4)区域的2个片段。使用引物oriT-F/oriT-R并以pLS20cat作为模板来扩增包含oriTLS20的片段2。进而,使用引物erm-F1/erm-R并以质粒pMutin2为模板来扩增具有红霉素抗性基因的片段3。通过使用了引物yhfM-uF/yhfM-dR的重组PCR以1-2-3-4的顺序连接片段1~4。可以通过重组PCR片段来转化菌株TMO311(aprE::kan),选择对红霉素和卡那霉素这两者具有抗性的菌落,作为供体株(YNB060)。
[工序(2)]
对于本工序,由上述供体株对受体株进行接合转移。将供体株和受体株在各自含有适合的抗生素的LB液体培养基中边以180rpm进行振荡边以37℃培养一晩左右。将各培养物在不含抗生素的新鲜的LB培养基中稀释至OD600为0.05左右的的细胞密度,边以180rpm进行振荡边以37℃进行孵育。在OD600达到0.5~0.7时,在1.5mL的微型管中混合500μL的供体和受体培养物,在37℃下放置2分钟~2小时、优选放置5分钟~1小时、更优选放置10分钟~30分钟、进一步优选放置15分钟左右。以往需要数小时以上的接合转移的工序根据本发明的方法就能在15分钟左右够实现充分的转移。将混合物连续稀释,在包含各种组合的抗生素的LB孔板上展开,使菌落增殖一晩。在它们各自的孔板上,为了计算出基于接合转移的转化效率,而测定菌落形成单位(CFU),求出经转化的CFU/总受体的CFU×106(ppm)。
[工序(3)]
对于本工序,使期望的DNA片段蓄积于受体菌的染色体中。具体而言,将利用上述工序(2)的接合转移而得到的转化细胞作为受体菌,重复上述工序(1)和(2),由此能够使期望的DNA片段蓄积于受体菌的染色体中。
本发明的转化方法中,在供体株与受体株为异种的情况下,被接合转移的DNA有时在受体株内因限制性内切酶等而降解。在这样的情况下,上述工序(1)中,可认为为了在供体株中进行与受体株内同样的DNA修饰(甲基化修饰等),而进行预先将甲基化酶导入至供体株中的转化。例如,作为受体株而使用嗜热菌(嗜热地芽胞杆菌(Geobacilluskaustophilus;GK))时,可以采用日本特开2011-211968号公报中公开的方法。具体而言,向GK中转移的质粒传输供体通过实施DNA甲基化酶的赋予及用于强制增强rap基因表达的基因操作而能够更进一步提高效率。另外,转移至GK的质粒(pGK1)可以使用:例如向pGR16B中添加了用于在GK中复制的ori、及耐受在GK中的选拔所需的高温的卡那霉素抗性基因KmR(TK101)等的质粒。
实施例
通过以下的实施例对本发明进行具体地说明,但本发明不限定于实施例。
[实施例1]枯草芽孢杆菌的转化方法
1.细菌株和培养条件
下述表1示出用于本研究的细菌株和质粒。另外,下述表2示出作为PCR引物而使用的合成寡核苷酸(序列号1~33)。在LB培养基(Difco公司制)中在37℃的条件下使细菌株增殖而使用。根据需要,培养基中添加了抗生素(氯霉素5μg/mL、红霉素1μg/mL、奇霉素100μg/mL和卡那霉素10μg/mL)。

2.受体株的构建
通过基于奇霉素抗性基因的置换而使枯草芽孢杆菌168株的comK基因失活。即,通过将枯草芽孢杆菌168株的DNA作为模板利用PCR来扩增分别对应于comK的上游和下游区域的2个DNA片段。作为引物,上游的片段使用comK-uF/comK-uR,下游的片段使用comK-dF/comK-dR。另外,使用引物spc-F/spc-R来扩增包含TMO310株的奇霉素抗性基因的另一个DNA片段。通过使用了comK-uF/comK-dR的重组PCR来连接3个片段,将奇霉素抗性基因插入comK的上游与下游的区域之间。导入重组PCR片段而将枯草芽孢杆菌168株转化为奇霉素抗性,得到了在该研究中用作受体株的新型的枯草芽孢杆菌株YNB001(comK::spc)。
3.pLS20catΔoriT的构建
通过无痕敲除pLS20cat的oriTLS20区域而进行失活。即,将pLS20catDNA作为模板并使用上游区域的引物oriT-uF/oriT-uR和oriT-dF/ori通过PCR来扩增对应于oriTLS20的上游(片段1)和下游(片段2)区域的2个DNA片段。片段1的末端和片段2的前端在30bp处相同,因此利用这些区域,通过转化而将oriTLS20区域删除。作为引物使用mazF-F/mazF-R由TMO311 DNA扩增mazF kan盒的另一个DNA片段(片段3)。3个PCR片段以通过使用oriT-uF/oriT-dR作为引物的重组PCR以1-3-2的顺序连接的方式设计。导入重组PCR片段将PKS11株转化为卡那霉素抗性,得到新的株YNB022。对于该株,通过oriTLS20区域的双交叉事件而组入PCR片段,由此修饰pLS20cat。将YNB022在含有卡那霉素的LB液体培养基中在37℃下增殖一晩。将培养液的整分试样移至含有1mM异丙基硫代吡喃半乳糖苷(IPTG)的新型的LB液体培养基中,将细胞在37℃下增殖2小时。其次,将培养液的整分试样在包含1mM IPTG的LB孔板上展开,在37℃下孵育一晩。在IPTG的存在下表达mazF而产生自杀毒素,只有能够通过分子内重组而逃离mazF kan盒的细胞才能存活。在孔板上出现的菌落中,对卡那霉素敏感性菌落进行测序,确认oriTLS20区域己被准确删除。将得到的质粒命名为pLS20catΔoriT。将该质粒的结构示于图1。另外,将基因序列在序列表中显示为序列号34。
4.供体株的构建
如下那样构建了供体株YNB060(图2)。使用引物yhfM-uF/yhfM-uR1(关于上游)和yhfM-dF/yhfM-dR(关于下游),由168DNA扩增了对应于yhfM的上游(片段1)和下游(片段4)区域的2个片段。使用引物oriT-F/oriT-R并将pLS20cat作为模板扩增了包含oriTLS20的片段2。进而,使用引物erm-F1/erm-R并以质粒pMutin2为模板扩增了具有红霉素抗性基因的片段3。通过使用了引物yhfM-uF/yhfM-dR的重组PCR以1-2-3-4的顺序连接了片段1~4。导入该重组PCR片段以红霉素和卡那霉素这两者具有抗性的方式转化菌株TMO311(aprE::kan),选择了菌落。将得到的菌株命名为YNB060。对于该菌株,在oriTLS20处具有红霉素标记物,在相同染色体上距离彼此6.6kb的位置的yhfM和aprE基因座这两者上具有卡那霉素标记物。进而,oriTLS20的复制方向朝向位于6.6kb下游的卡那霉素标记物。
其它菌株的YNB061也与上述同样地构建。分别使用引物yhfM-uF/yhfM-uR2和yhfM-dF/yhfM-dR扩增了yhfM的2个片段(上游和下游区域)。分别使用引物oriT-F/oriT-R和erm-F2/erm-R,扩增了oriTLS20片段(片段2)和红霉素抗性片段(片段3)。通过使用了引物yhfM-uF/yhfM-dR的重组PCR来连接4个片段。导入该重组PCR片段而将TMN311株转化为红霉素抗性,选择菌落而得到YNB061。与YNB060对照地,在YNB061中,yhfM基因座上的oriTLS20的复制方向朝向aprE基因座上的与卡那霉素标记物的相反方向。
进而作为其它菌株与上述同样地构建了YNB069、YNB062及YNB097(图2)。对于YNB069,作为引物使用yhfK-uF/yhfK-uR1和yhfK-dF/yhfK-dR(表2),由168DNA分别扩增了yhfK的上游(片段1)和下游(片段4)区域这2个片段。对于YNB062,作为引物使用yhfC-uF/yhfC-uR1和yhfC-dF/yhfC-dR,分别扩增了yhfC的上游和下游区域这2个片段。对于YNB097,作为引物使用yhcT-uF/yhcT-uR1和yhcT-dF/yhcT-dR,分别扩增了yhcT的上游和下游区域这2个片段。oriTLS20片段(片段2)和红霉素抗性片段(片段3)与关于YNB060构建中上述使用的片段相同。对于各自的情况,使用YNB069的引物yhfK-uF/yhfK-dR、YNB062的引物yhfC-uF/yhfC-dR及YNB097的引物yhcT-uF/yhcT-dR,通过重组PCR连接了4个片段。通过各自的重组PCR片段来转化TMO311(aprE::kan),选择对红霉素和卡那霉素这两者具有抗性的菌落。将得到的菌株命名为YNB069、YNB062和YNB097。所有这些在yhfK、yhfC和yhcT基因座中具有红霉素标记物及oriTLS20。此外在染色体上位于自aprE基因座的卡那霉素标记物起分别距离9.5、16.4和113kb的位置。进而,在全部这些菌株中,oriTLS20的复制方向与卡那霉素标记物为正向。
依据常规方法从上述得到的YNB060、YNB061、YNB069、YNB062、YNB097中提取染色体DNA,并导入具有pLS20cat的168株(PKS11)或具有pLS20catΔoriT的YNB026中,由此得到在以下的实验中使用的供体株。作为具有pLS20cat的株,得到YNB065、YNB066、YNB071、YNB067、YNB099,另外,作为具有pLS20catΔoriT的株,得到YNB091、YNB095、YNB092、YNB094、YNB100。
5.DNA的接合转移
在液体培养基中进行DNA的接合转移。即,将供体和受体株分别在含有适合的抗生素的5mL的LB液体培养基中边以180rpm进行振荡边在37℃下培养一晩。将各培养物在不含抗生素的5mL的新鲜的LB培养基中稀释至OD600为0.05的细胞密度,边以180rpm进行振荡边在37℃下进行孵育。在OD600达到0.5~0.7时,在1.5mL的微型管中混合500μL的供体和受体培养物,在37℃下放置15分钟。连续稀释混合物,在包含各种组合的抗生素的LB孔板上展开,使菌落增殖一晩。在它们各自的孔板上,为了计算出基于接合转移(Transconjugate)的转化效率,测定菌落形成单位(CFU),求出经转化的CFU/总受体的CFU×106(ppm)。
6.结果
上述的实验结果可知,pLS20catΔoriT(oriT缺失的质粒)本身变得无法转移至受体株中,但维持了将具备共存的oriTLS20的质粒、染色体DNA转移至受体株中的能力。以下进行详细地说明。
(质粒的转移实验)
图3示出质粒的转移实验的结果。需要说明的是,图3中,CS(黑色的柱)表示获得了氯霉素抗性的细胞浓度(ppm),ES(灰色的柱)表示获得了红霉素抗性的细胞浓度(ppm)、CES(白色的柱)表示获得了氯霉素抗性和红霉素抗性的细胞浓度(ppm)。另外,ND表示检测限以下(<0.01ppm),*表示未进行实验。
如图3所示,pLS20cat从作为供体株的PKS11(具有pLS20cat的168株)或GR138(具有pLS20cat和pGR16B的168株)转移至受体株,得到了大量获得了氯霉素抗性的受体细胞(2500ppm以上)。另外,具备作为辅助质粒的pLS20cat和作为转移质粒的pGR16B(具有oriTLS20和红霉素抗性基因)的供体株GR138对受体株赋予了红霉素抗性,因此也可知pLS20cat能够介导具有oriTLS20的转移质粒pGR16B的转移(图3)。需要说明的是,在上述的实验中,由于供体株GR138对受体细胞赋予红霉素抗性至约1000ppm,因此可以说与转移质粒pGR16B相比,辅助pLS20cat在受体细胞的转化效率几乎为2倍。进而,约100ppm的受体细胞对红霉素和氯霉素这两者显示出抗性(图3),因此启示出接受了pGR16B的转染接合体的约10%有可能获得pLS20cat。
已知具有pLS20cat的细菌不接受pLS20cat介导性基因转移,因此接受了pLS20cat的转化体无法使用相同的接合转移系统来再次进行转化。另一方面,如上述实验所示,保有pLS20cat的细胞不仅pLS20cat自身转移,而且pGR16B也进一步转移至其它株中。可认为假如在环境中释放出这些转染接合体,在抗生素抗性基因扩散至其它细菌细胞中,有出现新型的抗生素抗性菌的可能性。
因此我们利用上述方法,为了避免pLS20cat的自转移而敲除了pLS20cat的oriTLS20并进行了pLS20catΔoriT的构建。如预想的那样,作为具备pLS20catΔoriT的供体株的YNB026完全未转移pLS20catΔoriT,但具备pLS20catΔoriT和pGR16B的YNB031能够仅转移pGR16B来对受体株赋予红霉素抗性(图3)。进而,对于pGR16B的转移效率,pLS20cat为辅助质粒的情况及pLS20catΔoriT为辅助质粒的情况均相同。这些结果显示出:pGR16B的pLS20cat依赖性转移无需辅助质粒、pLS20cat的自转移性。进而,pLS20cat中的oriT的基因敲除未对共存的pGR16B的转移效率产生影响。
(染色体DNA的转移实验)
接着我们对pLS20catΔoriT能否依赖于oriTLS20区域的状态而介导染色体DNA的转移进行了确认。将结果示于图4。需要说明的是,图4中,CS(黑色的柱)表示获得了氯霉素抗性的细胞浓度(ppm),KS(灰色的柱)表示获得了卡那霉素抗性的细胞浓度(ppm),CKS(白色的柱)表示获得了氯霉素抗性和卡那霉素抗性的细胞浓度(ppm)。另外,ND表示检测限以下(<0.01ppm)。
在YNB060和YNB061的供体染色体中,在aprE基因座中的卡那霉素抗性基因的6.6kb上游的yhfM基因座导入oriTLS20。在株YNB060和YN0B61中,oriTLS20的复制方向分别与卡那霉素抗性基因为正向和反向。制作了将pLS20cat或pLS20catΔoriT作为辅助质粒导入至供体中而得到的新株:(1)YNB065(将pLS20cat导入至YNB060中)、(2)YNB066(将pLS20cat导入至YNB061中)、(3)YNB091(将pLS20catΔoriT导入至YNB060中)及(4)YNB095(将pLS20catΔoriT导入至YNB061中)。另一方面,对于受体株YNB001,使编码自然感受态所需的转录因子的comK失活,完全删除了自然感受态(数据未示出)。
YNB065和YNB066这两者的pLS20cat对2300ppm以上的受体细胞赋予了氯霉素抗性。另一方面对于卡那霉素抗性的赋予,YNB065仅对1ppm的受体细胞赋予卡那霉素抗性,而YNB066完全未赋予卡那霉素抗性(图4)。这些结果示出:oriTLS20复制的方向与卡那霉素抗性基因为正向时,pLS20cat可以转移位于oriTLS20的6.6kb下游的卡那霉素抗性基因。此处,受体不具有自然感受态,因此可以说卡那霉素抗性的获得仅依赖于接合转移。另一方面,YNB065对受体细胞(约1ppm的受体细胞)不仅赋予卡那霉素抗性,而且赋予氯霉素抗性(图4)。该结果启示出,导入了大的染色体DNA的成为卡那霉素抗性的受体细胞的大部分能够进一步获得辅助质粒pLS20cat。
使用了将pLS20catΔoriT作为辅助质粒导入至YNB060中的YNB091、及导入至YNB061中的YNB095的实验中,确认了受体细胞没有获得氯霉素抗性,pLS20catΔoriT失去自转移性(图4)。另一方面,更重要的是,在使用YNB091作为供体细胞时,pLS20catΔoriT以与pLS20cat同样的效率地依赖于oriTLS20取向的方式对受体细胞赋予卡那霉素抗性。该结果明确了:pLS20catΔoriT不仅在移入转移质粒时,而且在移入染色体DNA时也能够与原本的辅助质粒pLS20cat同样有效地发挥辅助活性。YNB095未对受体赋予卡那霉素抗性,因此确认了DNA移入依赖于正向的oriTLS20。
接着,我们进行了确认pLS20catΔoriT能够介导转移的染色体DNA的长度的实验。将该结果示于图5。需要说明的是,图5中,CS(黑色的柱)表示获得了氯霉素抗性的细胞浓度(ppm),KS(灰色的柱)表示获得了卡那霉素抗性的细胞浓度(ppm)。另外,ND表示检测限以下(<0.01ppm)。
菌株YNB069、YNB062及YNB097中,oriTLS20与卡那霉素标记物之间的距离分别为9.5kb、16.4kb、113kb。在所有这些菌株中,oriTLS20的复制的方向与卡那霉素抗性基因为正向。包括YNB071(将pLS20cat导入至YNB069中)、YNB067(将pLS20cat导入至YNB062中)、YNB099(将pLS20cat导入至YNB097中)、YNB092(将pLS20catΔoriT导入至YNB069中)、YNB094(将pLS20catΔoriT导入至YNB062中)及YNB100(将pLS20catΔoriT导入至YNB097中)。具有pLS20cat的全部供体对超过600ppm的受体细胞赋予氯霉素抗性,但具有pLS20catΔoriT的其它供体完全未赋予氯霉素抗性(图5)。然而,进而重要的是,具有pLS20catΔoriT的全部株能够对0.5-10.0ppm的受体细胞赋予卡那霉素抗性(图5)。效率与YNB091中作为供体而实现的效率几乎等同,作为能够移入的DNA长度,示出能至少为113kb。进而,在转移比染色体DNA长的片段时,pLS20catΔoriT显示出与pLS20cat同样的效率(图5)。这些结果还示出:pLS20cat本身的转移性并不需要用于转移比染色体DNA更长的片段的辅助功能。
[实施例2]GK(嗜热地芽胞杆菌)的转化方法
进行作为受体株使用嗜热菌(嗜热地芽胞杆菌;GK)的实验。实验方法采用日本特开2011-211968号公报中公开的方法。具体而言,对向GK中转移的质粒传输供体实施了DNA甲基化酶的赋予及用于强制增强rap基因表达的基因操作。另外,转移至GK中的质粒pGK1可以使用:向pGR16B中添加了用于在GK中复制的ori、及耐受在GK中的选拔所需的高温的卡那霉素抗性基因KmR(TK101)的质粒。在接合转移时,测定了作为辅助质粒使用pLS20cat和pLS20catΔoriT时的pGK1转移效率。其结果如图6所示,可知使用了pLS20catΔoriT时能够以高于pLS20cat的效率转移pGK1。
产业上的可利用性
根据本发明的革兰氏阳性菌的转化方法,能够将大尺寸的DNA转移至受体侧。进而,通过使用pLS20cat的oriTLS20区域失活的pLS20catΔoriT作为辅助质粒,从而在革兰氏阳性菌的接合转移中,能够制作辅助质粒本身不会转移至受体侧而仅转移了期望的染色体、质粒的转化细胞。根据本发明的方法,得到的转化细胞不具有辅助质粒,因此再次利用同样的方法而能够将期望的染色体、质粒的接合转移用作受体(受体菌)。因此,根据本发明,能够将期望的DNA片段蓄积于受体(受体菌)的染色体中,设计并制作巨大的DNA。另外,基于本发明的方法的转化仅通过将供体(供体菌)和受体(受体菌)的液体培养液混合就能够迅速且简便地进行,在合成生物学的产业活动中,可以适宜地用于微生物、培养细胞等的制作。
