(1) grinding Galla chinensis and radix Sophorae Flavescentis, breaking cell wall, and sieving with 250 mesh sieve; soaking the sieved powder in 8 times of water for 40min, reflux extracting for 2h, and filtering; pumping the filtrate into a concentration device, and concentrating under reduced pressure until the concentration of crude drug is 0.3 g/g; standing the concentrated solution at 4 deg.C for 12 hr, and centrifuging at 6000rpm to obtain Galla chinensis and radix Sophorae Flavescentis extractive solution.

(2) Grinding fructus Cnidii and/or cortex moutan, breaking cell wall, and sieving with 250 mesh sieve; soaking the sieved powder in 27 times of 70% ethanol water solution for 70min, reflux extracting for 3h, and filtering; pumping the filtrate into a concentration device, and concentrating under reduced pressure until the concentration of crude drug is 0.1 g/g; standing the concentrated solution, and centrifuging at 8000rpm to obtain fructus Cnidii and cortex moutan extractive solution.

(3) Inoculating lactobacillus plantarum stored at 4 ℃ into an MRS liquid culture medium for activation, and culturing for 24h at 37 ℃ for 2 times; inoculating the activated strain into an MRS liquid test tube culture medium, and culturing at 37 ℃ for 24h to obtain a seed solution;

(4) inoculating the seed solution into a fermentation tank according to the inoculation amount of 5%, wherein the fermentation temperature is 37 ℃, the dissolved oxygen amount is 10%, the pH value is controlled at 6.4, and the fermentation time is 60 hours, so as to obtain fermentation liquor;

(5) separating the fermentation liquor in a high-speed centrifugation mode, and collecting thallus precipitates; washing the precipitate with pure water for 3 times, dispersing the precipitate with pure water according to the feed-liquid ratio of 1:20, and adding 4% trehalose as protectant. Crushing the liquid at low temperature with a high-pressure crusher at 130MPa and 4 deg.C for 3 times to obtain Lactobacillus plantarum fermented product.

(6) Mixing the extractive solutions of Galla chinensis, fructus Cnidii, cortex moutan and radix Sophorae Flavescentis with Lactobacillus plantarum fermented product.

Example 3

The embodiment provides a composition for restoring microecological balance of female private parts, which comprises the following raw materials in parts by weight: 140 parts of gallnut, 40 parts of common cnidium fruit, 35 parts of cortex moutan, 30 parts of radix sophorae flavescentis and 40 parts of lactobacillus plantarum fermentation product. The preparation method comprises the following steps:

(1) grinding Galla chinensis and radix Sophorae Flavescentis, breaking cell wall, and sieving with 400 mesh sieve; soaking the sieved powder in 12 times of water for 40min, reflux extracting for 4h, and filtering; pumping the filtrate into a concentration device, and concentrating under reduced pressure until the concentration of crude drug is 0.3 g/g; standing the concentrated solution at 4 deg.C for 10 hr, and centrifuging at 6000rpm to obtain Galla chinensis and radix Sophorae Flavescentis extractive solution.

(2) Grinding fructus Cnidii and cortex moutan, breaking cell wall, and sieving with 400 mesh sieve; soaking the sieved powder in 33 times of 70% ethanol water solution for 70min, reflux extracting for 3h, and filtering; pumping the filtrate into a concentration device, and concentrating under reduced pressure until the concentration of crude drug is 0.1 g/g; standing the concentrated solution, and centrifuging at 8000rpm to obtain fructus Cnidii and cortex moutan extractive solution.

(3) Inoculating lactobacillus plantarum stored at 4 ℃ into an MRS liquid culture medium for activation, and culturing for 24h at 37 ℃ for 2 times; inoculating the activated strain into an MRS liquid test tube culture medium, and culturing at 37 ℃ for 24h to obtain a seed solution;

(4) inoculating the seed solution into a fermentation tank according to the inoculation amount of 5%, wherein the fermentation temperature is 37 ℃, the dissolved oxygen amount is 20%, the pH value is controlled at 6.6, and the fermentation time is 24 hours, so as to obtain a fermentation liquid;

(5) separating the fermentation liquor in a high-speed centrifugation mode, and collecting thallus precipitates; washing the precipitate with pure water for 3 times, dispersing the precipitate with pure water according to the feed-liquid ratio of 1:30, and adding 5% pullulan as a protective agent. Crushing the liquid at low temperature with a high-pressure crusher at 170MPa and 4 deg.C for 1 time to obtain Lactobacillus plantarum fermented product.

(6) Mixing the extractive solutions of Galla chinensis, fructus Cnidii, cortex moutan and radix Sophorae Flavescentis with Lactobacillus plantarum fermented product.

Example 4

This example provides a composition for restoring microecological balance in female private parts, which is prepared from the same raw materials as in example 1, except that the fructus Cnidii and cortex moutan are extracted with 60% ethanol in step (2) of the preparation method, and other conditions are kept unchanged.

Example 5

This example provides a composition for restoring microecological balance in female private parts, which was prepared from the following raw materials: 90 parts of gallnut, 60 parts of common cnidium fruit, 50 parts of cortex moutan, 55 parts of radix sophorae flavescentis and 30 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Example 6

This example provides a composition for restoring microecological balance in female private parts, which was prepared from the following raw materials: 160 parts of nutgall, 40 parts of common cnidium fruit, 30 parts of cortex moutan, 25 parts of radix sophorae flavescentis and 30 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Example 7

This example provides a composition for restoring microecological balance in female private parts, which was prepared from the following raw materials: 130 parts of gallnut, 60 parts of common cnidium fruit, 45 parts of cortex moutan, 50 parts of radix sophorae flavescentis and 60 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Comparative example 1

This comparative example provides a composition which differs from example 1 in the starting materials for its preparation: 90 parts of fructus cnidii, 80 parts of cortex moutan, 85 parts of radix sophorae flavescentis and 30 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Comparative example 2

This comparative example provides a composition which differs from example 1 in the starting materials for its preparation: 150 parts of Chinese gall, 50 parts of cortex moutan, 55 parts of radix sophorae flavescentis and 30 parts of lactobacillus plantarum fermentation products. The preparation method is the same as that of example 1.

Comparative example 3

This comparative example provides a composition which differs from example 1 in the starting materials for its preparation: 150 parts of Chinese gall, 55 parts of common cnidium fruit, 50 parts of lightyellow sophora root and 30 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Comparative example 4

This comparative example provides a composition which differs from example 1 in the starting materials for its preparation: 150 parts of Chinese gall, 60 parts of common cnidium fruit, 45 parts of cortex moutan and 30 parts of lactobacillus plantarum fermentation product. The preparation method is the same as that of example 1.

Comparative example 5

This comparative example provides a composition which differs from example 1 in the starting materials for its preparation: contains no lactobacillus plantarum fermentation product component, and other raw materials and contents are kept unchanged. The preparation process is as in example 1.

Test example 1

The test examples carried out a 24-hour bacteriostatic test on the compositions prepared in examples 1-7 and comparative examples 1-5, and characterized the inhibitory effect of each composition on pathogenic bacteria in private parts. The test method is specifically referred to "Disinfection Specification" (2002 edition).

The results are shown in table 1:

Escherichia coli, Lactobacillus plantarum and Lactobacillus acidophilus were inoculated into an agar nutrient medium containing 5% of the composition prepared in examples 1 to 7 and comparative examples 1 to 5 (see "Disinfection Specification" (2002) for formulation) at a viable count ratio of 1:1:1, and after 24 hours of culture according to a standard method, the number of colonies of each bacterial population was counted. The pharmacological effects of the composition on up-regulation of resident flora and down-regulation of pathogenic flora are demonstrated by co-culturing the resident flora and pathogenic bacteria causing the destruction of microecological balance in the same environment, simulating the actual situation of the colonies in the private area under the condition of administration of patients and changing the abundance of different floras.

The results are shown in table 2:

phase A: 80 parts of deionized water; 0.2 part of sodium hyaluronate; 0.2 part of acrylate/C10-30 alkanol acrylate cross-linked polymer; 5 parts of glycerol; 2 parts of 1, 3-propylene glycol;

phase B: 2 parts of 1, 2-pentanediol; 0.2 part of ethylhexyl glycerin; 0.1 part of aminomethyl propanol; citric acid monohydrate 0.09 part; example 1 preparation of 10 parts of a microecologically balanced composition;

and adding the sodium hyaluronate in the phase A into glycerol for dispersion, dispersing the acrylate/C10-30 alkanol acrylate cross-linked polymer by using water, adding other components for mixing, mixing the A, B, C phases, and stirring uniformly to obtain the private care gel for human clinical experiments.

(2) Test method

51 patients diagnosed with vaginal pathogenic microorganism infection accompanied with pudendum pruritus and increased vaginal secretion with mucopurulent, dark complexion and withered yellow are divided into A, B, C groups, each group comprises 17 patients, and the group A uses the private microecological nursing liquid twice a day, 5mL each time and lasts for two weeks; b, using a conventional nursing solution sold in the market twice a day, 5mL each time, and lasting for two weeks; group C only performed a simple two week daily nursing rinse. Microbial detection of vaginal secretion is carried out before and after use, and compared with healthy people, the influence of the private part treatment gel on the female private part micro-ecology is analyzed from three aspects of flora concentration, dominant bacteria, fungal infection and the like. The vaginal secretion microorganism detection results of healthy women are shown in the following table:

(3) results of the experiment

The results of the microbial testing of vaginal secretions before and after use of the product by subjects are shown in table 3:

As can be seen from Table 3: after the group A uses the private microecological care solution, the germ flora density of the reproductive system of a patient is reduced, the diversity of the germ flora is reduced, beneficial bacteria dominate, and no fungal infection exists; after the group B uses the conventional nursing liquid sold in the market, the germ density of the reproductive system of a patient is reduced, the diversity of the florae is still high, and a large number of pathogenic bacteria still exist and the florae are abnormal; group C did not use the nursing and repairing product, and the reproductive system microecology was abnormal, and the symptoms were not improved.

The composition for repairing the micro-ecological balance of the female private parts is formed by combining traditional Chinese medicines and lactobacillus plantarum fermentation products, traditional Chinese medicine extracts can effectively reduce the number of pathogenic bacteria, the lactobacillus plantarum fermentation products contained in the formula have a promoting effect on the growth of the lactobacillus in the private ecological system, and the lactobacillus can inhibit the growth of the pathogenic bacteria through competitive inhibition among microbial communities, so that the resistance of the vagina to the pathogenic bacteria is enhanced, and a good long-acting maintenance effect is achieved.

The applicant states that the present invention is illustrated by the above examples to a composition for restoring micro-ecological balance in female private parts, a method for preparing the same and applications thereof, but the present invention is not limited to the above examples, i.e. it does not mean that the present invention must be implemented by means of the above examples. It should be understood by those skilled in the art that any modification of the present invention, equivalent substitutions of the raw materials of the product of the present invention, addition of auxiliary components, selection of specific modes, etc., are within the scope and disclosure of the present invention.

The preferred embodiments of the present invention have been described in detail, however, the present invention is not limited to the specific details of the above embodiments, and various simple modifications may be made to the technical solution of the present invention within the technical idea of the present invention, and these simple modifications are within the protective scope of the present invention.

It should be noted that the various technical features described in the above embodiments can be combined in any suitable manner without contradiction, and the invention is not described in any way for the possible combinations in order to avoid unnecessary repetition.
