5) inactivating enzyme of the biochemical bionic degradation liquid, filtering, and collecting filter residue and filtrate;

controlling the pH value in the reaction process of the step 4) as follows: (0-15) min, from 8.0 to 7.0, (15-25) min, from 7.0 to 7.5, (25-100) min, from 7.5 to 5.5.

The invention belongs to the technical field of animal fresh waste treatment, and particularly relates to a composite digestion extract and a method for treating animal fresh waste by using the same.

Background

With the increasing environmental protection requirements of China, domestic garbage classification work is actively deployed all over the country, and a simple 4-class garbage or 6-class garbage classification method is primarily seen so as to facilitate subsequent garbage treatment and recycling. But the field of garbage classification and recovery in China just starts at present, and a plurality of places still have the problem that the garbage is mixed and buried again after classification because the garbage cannot be treated; the main problem is in the recovery and disposal of the fine classification back-end.

For the field of animal fresh waste, at the present stage, no good treatment technology is available, or the animal fresh waste is treated together with kitchen waste or mixed and buried. In fact, the garbage cannot be treated optimally, and if the garbage is treated and fermented together with kitchen garbage, a large amount of liquid and putrefactive substances are generated, so that the fermentation process is difficult to control, the influence of mixed bacteria is great, and a large amount of harmful gas can be generated; if mixed landfill is adopted, a large amount of land is occupied like a way of treating biological hazards, and the land after the landfill can not be used for a long time because of containing a large amount of harmful substances such as butyric acid, propionic acid and the like.

The quantity of the garbage such as the fresh animal waste is very large, and the garbage is an important garbage in our daily life: one fifth to one seventh of the weight of one fish is fresh waste (fish intestinal offal), the maximum one fourth of the weight of the chicken is unusable fresh waste (chicken intestinal offal), and the fresh waste such as dead fish and dead shrimps, expired meat and the like which are slightly rotten due to untimely sale; in addition, there are dead livestock and poultry in transit which are not received by slaughter houses, livestock and poultry dead of illness (pigs, cattle, sheep, chickens, ducks, geese, etc.), corpse meat (frozen pork, beef) punished by customs, expired frozen products (frozen pork, frozen beef, frozen chicken, frozen seafood, etc.); and the remains of pets such as accidents or diseases, old dead cats, dogs and the like in cities belong to fresh meat waste. Taking Shenzhen as an example, according to incomplete statistics of 2 garbage classification transport companies in Shenzhen market, the fresh and fresh animal wastes generated in each supermarket and vegetable market exceed 20 tons every day in Shenzhen dragon sentry region; according to incomplete statistics of related departments of city administration in Tianjin, more than 2000 tons of fresh animal wastes (including wastes generated by aquatic product processing and slaughtering enterprises in Tianjin) can be generated every day. This is a large category of garbage.

However, the disposal cost and environmental cost of such wastes as animal fresh wastes are relatively high; at present, no efficient pollution-free method for treating fresh animal waste exists.

Disclosure of Invention

In view of the above, the present invention provides a non-microbial fermentation, composite digestion extract and a method for treating fresh animal waste using the same, the composite digestion extract can be used for treating fresh animal waste, and can rapidly decompose the fresh animal waste into harmless protein solution and a small amount of solid slag, and the protein solution and the solid slag can be used for agricultural production, thereby realizing harmless zero-emission treatment of the fresh animal waste.

The invention provides a method for treating fresh waste of animals by using a composite digestion extract, which comprises the following steps:

1) crushing fresh animal waste, mixing with water, and adjusting the pH value to 8.0-8.5 to obtain mixed feed liquid;

2) heating the mixed material liquid to 90-105 ℃, and keeping the temperature for 8-12 min to obtain cured material liquid;

3) centrifuging to remove grease in the curing feed liquid to obtain an oil-removing feed liquid;

4) mixing the oil removing material liquid with the composite digestion extract, and reacting at 55-60 ℃ for 50-100 min to obtain a biochemical bionic degradation liquid;

5) inactivating enzyme of the biochemical bionic degradation liquid, filtering, and collecting filter residue and filtrate;

controlling the pH value in the reaction process of the step 4) as follows: (0-15) min, from 8.0 to 7.0, (15-25) min, from 7.0 to 7.5, (25-100) min, from 7.5 to 5.5.

Preferably, the particle size of the crushed fresh animal waste is 40-60 meshes.

The invention has the beneficial effects that: the composite digestive extract provided by the invention comprises the following components in parts by weight: pepsin, trypsin, intestinal juice extract, bromelain, papain, bacillus subtilis culture solution extracellular extract and escherichia coli culture solution extracellular extract; the composite digestion extract obtained by compounding the enzyme and the extracellular extract of the microorganism culture solution simulates animal gastrointestinal digestion liquid and microorganisms, can directly digest and decompose animal fresh wastes, and the obtained filtrate and filter residue can be used as protein liquid and solid fertilizer for agriculture, can realize rapid and harmless treatment of the animal fresh wastes, and provide organic protein liquid and solid residue for agricultural production, thereby being environment-friendly, efficient, harmless and beneficial to the people.

The invention provides a compound digestive extract, which comprises the following components in parts by weight: 2-5 parts of pepsin, 15-18 parts of trypsin, 2-3 parts of intestinal juice extract, 27.5-33 parts of bromelain, 35-41 parts of acid papain and 10-12 parts of bacillus subtilis culture solution extracellular extract; and 3-4 parts of an extracellular extract of the escherichia coli culture solution.

In the present invention, the complex digestive extract preferably comprises the following components in parts by weight: 2.5-4.5 parts of pepsin, 16-17 parts of trypsin, 2.2-2.8 parts of intestinal juice extract, 28-32.5 parts of bromelain, 36-40 parts of papain and 10.5-11.5 parts of bacillus subtilis culture solution extracellular extract; and 3.2-3.8 parts of an extracellular extract of the escherichia coli culture solution.

In the invention, the bacillus subtilis culture solution extracellular extract is preferably an extracellular extract of bacillus subtilis culture solution for 45-50 h, and is more preferably an extracellular extract of bacillus subtilis culture solution for 48 h; the extracellular extract of the escherichia coli culture solution is an extracellular extract of 24-40 h of escherichia coli culture solution. In the present invention, the culture method of the bacillus subtilis or the escherichia coli is not particularly limited, and a culture method conventional in the art may be employed; in the present invention, the extracellular extract of the culture solution is preferably obtained by culturing the Bacillus subtilis or Escherichia coli, centrifuging, and collecting the supernatant. The rotation speed and time of the centrifugation are not particularly limited in the invention, and the rotation speed and time of separating solid cells and culture solution which are conventional in the field can be adopted. In the present invention, the bacillus subtilis culture solution extracellular extract and the escherichia coli culture solution extracellular extract function to decompose and dissolve animal organisms by using substances of digestive chemical organic matters secreted by bacillus subtilis and escherichia coli.

In the invention, the intestinal juice extract is a porcine intestinal juice extract; the main components of the pig intestinal juice extract comprise trypsin, pepsin and enteroprotease; the pig intestinal juice extract is preferably produced and provided by a consignment enzyme preparation manufacturer.

In the present invention, the pepsin (5 ten thousand U/g), trypsin (10 ten thousand U/g), bromelain (2 ten thousand U/g) and papain acidocaldarin (20 ten thousand U/g) are preferably commercially available products which are conventional in the art. The stomach digestive protease, the trypsin, the bromelain and the acid papain simulate the enzyme environment in the digestive juice of the intestines and stomach of the animals, and after the components are compounded according to the proportion, the treatment effect on the fresh waste of the animals is the best. The four biological enzymes have good enzymolysis and digestion effects on animal proteins, and can hydrolyze insoluble macromolecular proteins into soluble proteins. However, the activity and action site of each enzyme are different, so that the efficient digestion and decomposition can be carried out on different protein molecules. Meanwhile, the enzyme and the complex enzyme in the intestinal juice extract have the functions of mutual promotion and supplement and mutual supplement and balance control in different pH value ranges, so that the enzyme can decompose and digest different protein sites in different pH value control intervals, and rapid decomposition is realized.

In the invention, the fresh animal waste is firstly crushed. In the present invention, the fresh animal waste refers to the intestinal, digestive gland and other auxiliary parts which are generated in the slaughtering process of chicken, fish and other animals and can not be used for food processing, and the waste of meat, fish and other animals which are partially or completely rotten and deteriorated due to the overlong shelf storage time, and the fresh animal waste is slightly harmful garbage. In the invention, the fresh waste of animals can be collected from places such as supermarkets, vegetable markets and the like; after the fresh waste of the animals is collected, the fresh waste is preferably directly treated or sprayed with a disinfectant for refrigeration and then treated. In the invention, when the animal fresh waste cannot be treated in real time, the animal fresh waste is refrigerated and stored; the refrigeration temperature is preferably 0 ℃, and the freezing preservation time is not longer than 20 h. In the invention, the particle size of the crushed fresh animal waste is preferably 40-60 meshes; the crushing preferably adopts high-speed crushing equipment, particularly a high-energy high-speed meat grinder or a colloid mill; the invention has no special limitation on the parameter setting of the high-speed crushing equipment, and the conventional parameter setting in the field can be adopted.

In the invention, the crushed fresh animal waste is mixed with water, and the pH value is adjusted to 8.0-8.5 to obtain mixed feed liquid. In the invention, the mass ratio of the crushed fresh animal waste to water is preferably (0.8-1.2), and more preferably 1: 1. The reagent for adjusting the pH value is not particularly limited, and the reagent can be an alkaline solution which is conventional in the field, and in the specific implementation process of the invention, the pH value is adjusted by sodium hydroxide. In the invention, the crushed fresh animal waste is preferably put into a reaction tank through a feeding machine and then mixed with water in the reaction tank.

The mixed material liquid is heated to 90-105 ℃ and kept for 10-30 min to obtain cured material liquid. In the invention, the temperature of the mixed liquid is preferably raised to 98-102 ℃, and more preferably 100 ℃; the holding time is preferably 9-11 min, and more preferably 10 min. In the present invention, stirring is preferably accompanied in the temperature raising process of the mixed material liquid, and the rotation speed of the stirring is preferably not more than 65 rpm. The heating of the invention is used for killing harmful germs in the mixed material liquid, so that the mixed material liquid is continuously cured at high temperature until 90 percent of solid materials are basically melted.

After the curing feed liquid is obtained, grease in the curing feed liquid is removed centrifugally, so that the oil-removing feed liquid is obtained. In the invention, the rotation speed of the centrifugation is preferably not less than 5200rpm, and the time of the centrifugation is preferably 5-12 min; the centrifugation is preferably carried out in a high-speed liquid-oil centrifuge; according to the invention, after the centrifugation is finished, the separated grease is preferably collected; the grease can be applied to agricultural production or biodiesel recycling.

After the oil removing liquid is obtained, the oil removing liquid and the composite digestion extract are mixed and react for 50-100 min at 55-60 ℃ to obtain the biochemical bionic degradation liquid. In the present invention, the mass of the composite digestion extract is preferably 0.1% to 0.5%, and more preferably 0.2% to 0.4% of the mass of the degreasing liquid. In the present invention, stirring is preferably accompanied during the reaction; in the invention, the reaction temperature is preferably 56-69 ℃, and more preferably 57-58 ℃; the reaction time is preferably 55-70 min, and more preferably 60 min. The invention controls the pH value in the reaction process as follows: (0-15) min, from 8.0 to 7.0, (15-25) min, from 7.0 to 7.5, (25-100) min, from 7.5 to 5.5, the invention simulates the enzyme environment of animal gastrointestinal tract by using the compound digestion extract, and further simulates the process of digesting food in animal gastrointestinal tract by the limitation of temperature, time and pH change, thereby realizing the high-efficiency harmless treatment of the animal fresh waste.

After the reaction is finished, the biochemical bionic degradation liquid is subjected to enzyme deactivation and filtration, and filter residue and filtrate are collected. In the invention, the temperature of enzyme deactivation is 75-85 ℃, the time of enzyme deactivation is 8-12 min, and more preferably 10 min; in the enzyme deactivation, stirring is preferably accompanied. After the enzyme deactivation step is finished, enzymes in the feed liquid, the bacillus subtilis culture solution extracellular extract and the escherichia coli culture solution extracellular extract lose biological activity and do not react. The method also comprises the step of adding a filter aid after the enzyme deactivation and before the filtration, wherein the mass of the filter aid is preferably 0.4-0.6 percent of that of the biochemical bionic degradation liquid, and more preferably 0.5 percent; in the present invention, the filter aid is preferably perlite powder; the mass of the filter aid is preferably 0.4-0.6% of the mass of the feed liquid, and more preferably 0.5%; after the filter aid is added, stirring is preferably carried out to mix the filter aid with the feed liquid, and the stirring time is preferably 4-6 min, and more preferably 5 min. In the invention, the filtration is preferably plate-and-frame filtration, and the equipment and parameters of the plate-and-frame filtration are not particularly limited, and the plate-and-frame filtration can be performed by adopting the conventional plate-and-frame filtration in the field. After the filtration, filter residue and filtrate are collected; after the filter residue is preferably pressurized and dried, the filter residue is used as a solid fertilizer for agricultural production; the solid soluble content of the filtrate reaches more than 5 percent, and the filtrate can be used as a protein solution for agricultural production and is a very good nutrient source for agricultural production.

The technical solutions provided by the present invention are described in detail below with reference to examples, but they should not be construed as limiting the scope of the present invention.

Example 1

1. Collecting raw materials: collecting fresh waste of animals in supermarket and vegetable market, transporting to factory, spraying disinfectant immediately, sealing (using environment-friendly disinfectant of dibromo hydantoin with concentration of 250ppm, spraying for 10 seconds intermittently at 5 seconds, covering and sealing, and pushing into refrigerator, wherein the temperature of refrigerator is controlled at 0 deg.C, and the storage time is not more than 20 h;

2. pretreatment: physically crushing the cold materials to 40-60 meshes by using ultrahigh-speed crushing equipment, and putting the crushed materials into a reaction tank by using a feeding machine;

3. adjusting: adding 100kg of materials into a reaction tank, and adding water to ensure that the ratio of the materials to the water is 1:1, adding a pH adjusting aid (sodium hydroxide) to adjust the pH value to enable the feed liquid to reach 8.0;

4. and (3) cooking and sterilizing: continuously stirring, heating to 100 deg.C, and maintaining for 10min to kill harmful bacteria in the material, and continuously aging at high temperature until 90% of solid material is basically melted;

6. high-speed centrifugal oil removal: reducing the temperature of the materials, simultaneously pumping the material liquid into a high-speed liquid-oil centrifugal separator, and separating out the grease to obtain about 8.6kg of grease for recycling the biodiesel;

7. biochemical reaction: adding compound digestive extract (raw material mass ratio of 0.25%) into the cooled material liquid, stirring continuously for biochemical reaction, and performing biochemical biomimetic degradation at 55 deg.C for 1h under pH control curve (figure 1) of technical design;

8. high-temperature inactivation: after the bionic biochemical degradation is finished, starting heating circulation to heat the materials in the reaction tank to more than 80 ℃, stirring for 10min, and inactivating the biological activity of the microbial extract and the biochemical preparation;

9. and (3) filtration assistance: after inactivation, adding 0.5% by mass of inorganic filter aid (perlite powder) while the mixture is hot, and stirring uniformly (5 min);

10. deslagging and filtering: pressing the uniformly stirred materials into a plate-and-frame filter to filter solid slag, collecting the slag, pressurizing and drying the slag, wherein the slag can be used for agricultural production;

11. blending: the filtered feed liquid is filled and stored at low temperature and can become nutrition for agricultural production; the solid soluble content of the feed liquid reaches 5.7 percent, 10.8kg of solid slag is obtained, 420kg of concentrated feed liquid is a very good agricultural production nutrient source.

Example 2

1. Collecting raw materials: after being collected in supermarkets and vegetable markets, the fresh animal wastes are transported to factories, and immediately sprayed with disinfectants to be sealed; then pushing into a cold storage, controlling the temperature of the cold storage at 0 ℃, and storing for no more than 20h for later use;

2. pretreatment: physically crushing the cold materials to 40-60 meshes of particle size by using ultrahigh-speed crushing equipment, and putting the materials into a reaction tank by using a feeding machine;

3. adjusting: adding 120kg of materials into a reaction tank, and adding water to ensure that the ratio of the materials to the liquid is 1:1, adding a pH adjusting aid (sodium hydroxide) to adjust the pH value to enable the feed liquid to reach 8.5;

4. and (3) cooking and sterilizing: continuously stirring, heating to 100 deg.C, and maintaining for 10min to kill harmful bacteria in the material, and continuously aging at high temperature until 90% of solid material is basically melted;

6. high-speed centrifugal oil removal: reducing the temperature of the materials, simultaneously pumping the feed liquid into a high-speed liquid-oil centrifugal separator, separating out the grease to obtain 11kg of grease, and delivering the 11kg of grease to farmers for agricultural planting;

7. biochemical reaction: adding the composite digestive extract (raw material mass ratio is 0.5%) into the cooled feed liquid, continuously stirring for biochemical reaction, and performing biochemical biomimetic degradation at a constant temperature of 57 ℃ for 1h under a pH control curve (figure 1) designed by the technology;

8. high-temperature inactivation: after the bionic biochemical degradation is finished, starting heating circulation to heat the materials in the reaction tank to more than 80 ℃, stirring for 10min, and inactivating the biological activity of the microbial extract and the biochemical preparation;

9. and (3) filtration assistance: after inactivation, adding 0.5% by mass of inorganic filter aid (perlite powder) while hot, stirring uniformly (5min),

10. deslagging and filtering: pressing the uniformly stirred materials into a plate-and-frame filter to filter solid slag, collecting the slag, pressurizing and drying the slag, wherein the slag can be used for agricultural production;

11. blending: the filtered feed liquid is filled and stored at low temperature and can become nutrition for agricultural production; the solid soluble content of the feed liquid reaches 7 percent, so that 9.3kg of solid slag and 400kg of concentrated feed liquid are obtained, and the feed liquid is a very good agricultural production nutrient source.

Example 3

1. Raw material income: after being collected in supermarkets and vegetable markets, the fresh animal wastes are transported to factories, and immediately sprayed with disinfectants to be sealed; then pushing into a cold storage, controlling the temperature of the cold storage at 0 ℃, and storing for no more than 20h for later use;

2. pretreatment: physically crushing the cold materials to the particle size of 40-60 meshes by using ultrahigh-speed crushing equipment, and putting the materials into a reaction tank by using a feeding machine;

3. adjusting: adding 150kg of raw materials into a reaction tank, and adding water to ensure that the ratio of the materials to the liquid is 1: 1.1, adding a pH adjusting aid (sodium hydroxide), and adjusting the pH value to enable the feed liquid to reach 8.5;

4. and (3) cooking and sterilizing: continuously stirring, heating to 100 deg.C, and maintaining for 10min to kill harmful bacteria in the material, and continuously aging at high temperature until 90% of solid material is basically melted;

6. high-speed centrifugal oil removal: reducing the temperature of the materials, simultaneously pumping the material liquid into a high-speed liquid-oil centrifugal separator, and separating out grease to obtain 16kg of grease for recycling biodiesel;

7. biochemical reaction: adding compound digestive extract (raw material mass ratio of 0.4%) into the cooled material liquid, stirring continuously for biochemical reaction, and performing biochemical biomimetic degradation at 60 deg.C for 1h under pH control curve (figure 1) of technical design;

8. high-temperature inactivation: after the bionic biochemical degradation is finished, starting heating circulation to heat the materials in the reaction tank to more than 80 ℃, stirring for 10min, and inactivating the biological activity of the microbial extract and the biochemical preparation;

9. and (3) filtration assistance: after inactivation, adding 0.4% by mass of inorganic filter aid (perlite powder) while the mixture is hot, and stirring uniformly (5 min);

10. deslagging and filtering: pressing the uniformly stirred materials into a plate-and-frame filter to filter solid slag, collecting the slag, pressurizing and drying the slag, wherein the slag can be used for agricultural production;

11. blending: the filtered feed liquid is filled and stored at low temperature and can become nutrition for agricultural production; the solid soluble content of the feed liquid reaches 6.5 percent, 10.2kg of solid slag is obtained, 420kg of concentrated feed liquid is a very good agricultural production nutrient source.

Example 4

1. Collecting raw materials: after being collected in supermarkets and vegetable markets, the fresh animal wastes are transported to factories, and immediately sprayed with disinfectants to be sealed; then pushing into a cold storage, controlling the temperature of the cold storage at 0 ℃, and storing for no more than 20h for later use;

2. pretreatment: physically crushing the cold materials to 40-60 meshes of particle size by using ultrahigh-speed crushing equipment, and putting the materials into a reaction tank by using a feeding machine;

3. adjusting: adding 110kg of raw materials into a reaction tank, and adding water to ensure that the ratio of the materials to the liquid is 0.9: 1, adding a pH adjusting aid (sodium hydroxide) to adjust the pH value to enable the feed liquid to reach 8.0;

4. and (3) cooking and sterilizing: continuously stirring, heating to 100 deg.C, and maintaining for 10min to kill harmful bacteria in the material, and continuously aging at high temperature until 90% of solid material is basically melted;

6. high-speed centrifugal oil removal: reducing the temperature of the materials, and simultaneously pumping the material liquid into a high-speed liquid-oil centrifugal separator to separate out the grease to obtain about 10kg of grease or be used for recycling the biodiesel;

7. biochemical reaction: adding the composite digestion extract (raw material mass ratio is 0.35%) into the cooled feed liquid, continuously stirring for biochemical reaction, and performing biochemical biomimetic degradation at a constant temperature of 58 ℃ for 1h under a pH control curve (shown in figure 1) designed by the technology;

8. high-temperature inactivation: after the bionic biochemical degradation is finished, starting heating circulation to heat the materials in the reaction tank to more than 80 ℃, stirring for 10min, and inactivating the biological activity of the microbial extract and the biochemical preparation;

9. and (3) filtration assistance: after inactivation, adding 0.5% by mass of inorganic filter aid (perlite powder) while the mixture is hot, and stirring uniformly (5 min);

10. deslagging and filtering: pressing the uniformly stirred materials into a plate-and-frame filter to filter solid slag, collecting the slag, pressurizing and drying the slag, wherein the slag can be used for agricultural production;

11. product blending: the filtered feed liquid is filled and stored at low temperature and can become nutrition for agricultural production; the solid soluble content of the feed liquid reaches 6.5 percent, so that 9.8kg of solid slag and 400kg of concentrated feed liquid are obtained, and the feed liquid is a very good agricultural production nutrient source.

The comparison of the effect of the compound digestive extract provided by the invention and the effect of the conventional single enzymolysis is shown in table 1.

Comparison of differences between different enzyme treatments

The embodiment shows that the composite digestion extract and the method for treating the animal fresh waste by using the composite digestion extract can efficiently and harmlessly treat the animal fresh waste, and the obtained grease, filter residues and filtrate can be effectively reused, so that the method is eco-friendly and pollution-free.

The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various modifications and decorations can be made without departing from the principle of the present invention, and these modifications and decorations should also be regarded as the protection scope of the present invention.
