背景技术

猫冠状病毒(Feline coronavirus,FCoV)是家猫和野生猫致死性疾病猫传染性腹膜炎(Feline infectious peritonitis,FIP)的病原,通过口鼻传播,主要感染幼年和老龄猫,在世界猫群中广泛流行。

FCoV病毒有4种主要结构蛋白,分别是刺突蛋白(Spike protein,S)、膜蛋白(Membrane protein,M)、小膜蛋白(Small envelope protein,E)和核衣壳蛋白(Nucleocapsid protein,N)。其中S蛋白是排列在包膜突起的糖蛋白,分子量约为180~200kDa,它对于诱导宿主的抗体应答和细胞免疫很重要。冠状病毒可通过S蛋白与特定的细胞受体结合,是决定病毒致病性和组织嗜性的关键因素,S蛋白是诱导机体产生中和抗体的保护性抗原,目前疫苗的相关研究的也集中在S蛋白区域。

FCoV根据S蛋白氨基酸序列和抗体交叉中和差异分为2种血清型:I型和II型。血清I型是主要流行毒株类型,其S蛋白完全来源于FCoV。血清II型FCoV临床不常见,由犬冠状病毒(Canine coronavirus,CCoV)和FCoV双重重组而来。FCoV-І型在世界范围内广泛流行,在美国和欧洲一些国家,FCoV-І型毒株感染率高达80-95%;在亚洲地区主要流行FCoV-ІІ型,感染率高达25%。血清I型和II型 FCoV毒株均存在2种在抗原和形态学上不同的生物型(或病理型):猫肠道冠状病毒(Feline enteric coronavirus,FECV)和猫传染性腹膜炎病毒(Feline infectious peritonitis virus,FIPV)。

其中猫肠道冠状病毒(FECV)的感染仅局限于消化道,会引起猫的体温上升,食欲不振甚至脱水等症状,致死率较低。猫传染性腹膜炎病毒,是猫肠道冠状病毒的高毒力突变株,具有高传染性及高致死率的特点。在临床上,猫传染性腹膜炎又可以分为渗出性和非渗出性两种病理型。由于暂无有效治疗药物,携带FIPV的猫咪一旦发病,难以康复,死亡率高。

近年来我国养猫数量逐年增加,由于国内市场并无相关疫苗,猫感染FIP的风险与日俱增。FECV及其转化的FIPV基因型毒株中存在由不同程度的单核苷酸多态性和插入/缺失现象,同时FIP潜伏期几周至几年不等,这种巨大差异给防疫工作带来严峻挑战。目前针对FIPV感染猫的治疗只能起到延缓疾病的作用,不能达到根治的效果,所以疫苗的预防显得尤为重要。

国内外现有疫苗的大都还处于研究阶段,由于该病毒特殊的感染机制,使得常规疫苗和重组疫苗效果不佳。FECV及其转化的FIPV基因型毒株中存在由不同程度的单核苷酸多态性和插入/缺失现象。FECV感染率达80%-95%,而FIP的致死率高达95%,目前并没有有效的疫苗上市,猫一旦感染FIPV几乎只能靠治疗延缓死亡,因此,FCoV疫苗的市场很有潜力,并且对FCoV预防具有重要意义。前期疫苗的研究FIP减毒活疫苗、灭活疫苗和亚单位疫苗免疫猫后均可观察到ADE,使FIP潜伏期缩短至1-2 d,病程更短,死亡更快。随着基因工程技术和分子生物学技术的发展,人们开始把注意力活载体疫苗及核酸疫苗等新型基因工程疫苗上。

发明内容

针对上述问题,本发明的目的在于提供一种用于猫冠状病毒的RNA疫苗及其构建方法,选取特定的5’UTR、3’UTR、S蛋白、终止密码子、polyA核酸序列,构建得到FCoV的RNA疫苗,将其转染细胞后能够高效表达,免疫动物后能够产生高水平的抗体,该抗体具有较高的中和效价及滴度。

本发明的技术内容如下:

本发明提供了一种用于猫冠状病毒的RNA疫苗,所述RNA疫苗为采用含有5’UTR、S蛋白、终止密码子、3’UTR以及polyA核酸的序列合成;

所述S蛋白选自FCoV-I型或FCoV-II型;

所述疫苗还包括纳米脂质颗粒,所述纳米脂质颗粒包裹RNA疫苗;

所述纳米脂质颗粒包括BHEM-Chol、PEG5k-b-PLGA11k、PLGA11k聚合物的混合物;

所述5’UTR、3’UTR的序列选取自猫源的猫血红蛋白或猫白蛋白,所选序列的同源性在50%~100%;

所述5’UTR、3’UTR分别选自六组不同的猫血红蛋白或猫血清白蛋白,其核酸序列分别为序列表SEQ ID NO.1和SEQ ID NO.2、SEQ ID NO.3和SEQ ID NO.4、SEQ ID NO.5和SEQ ID NO.6、SEQ ID NO.7和SEQ ID NO.8、SEQ ID NO.9和SEQ ID NO.10、SEQ ID NO.11和SEQ ID NO.12所示的六组;

所述5’UTR、3’UTR的六组序列选自GenBank中NCBI参考序列XM_023246781.1、XM_011290755.3、XM_003992859.2、XM_003992882.4、XM_003998693.4、X84842.1;

所述猫冠状病毒的S蛋白,其氨基酸序列如序列表SEQ ID NO.13所示,其核酸序列如序列表SEQ ID NO.14所示;

所述RNA疫苗的核酸序列包括如序列表SEQ ID NO.15~20所示;

所述终止密码子为TGA、TAA、TAG的自由组合,个数为1-10个;

所述polyA中的碱基A的个数为1-200个。

本发明还提供了一种用于猫冠状病毒的RNA疫苗的构建方法,包括如下步骤:

选取5’UTR、3’UTR、S蛋白、终止密码子、polyA核酸序列合成RNA疫苗的核酸序列构建于载体上,转染到细菌得到重组菌,将重组菌进行质粒提取得到重组质粒,将重组质粒进行酶切、转录、纯化得到RNA疫苗核酸,所述RNA疫苗的核酸序列如序列表SEQ ID NO.15~20所示;

所述S蛋白选自FCoV-I型或FCoV-II型;

所述RNA疫苗包括由脂质纳米颗粒载体包裹,所述脂质纳米颗粒的制备原料包括聚合物PEG5k-b-PLGA11k、PLGA11k和BHEM-Chol;

所述脂质纳米颗粒包裹RNA疫苗的操作为:将RNA疫苗核酸溶液与BHEM-Chol、PEG5k-b-PLGA11k、PLGA11k混合,超声得到油相-水相的初乳液,加入无核酸酶水再超声乳化得到水相-油相-水相的复乳液,将复乳液中的有机溶液去除,浓缩,即得到脂质纳米颗粒包裹的RNA疫苗;

所述RNA疫苗核酸溶液为RNA疫苗核酸使用无核酸酶水溶解得到;

所述BHEM-Chol、PEG5k-b-PLGA11k、PLGA11k使用之前分别溶于色谱级三氯甲烷溶液中。

本发明的有益效果如下:

本发明的用于猫冠状病毒的RNA疫苗,为通过选取特定的5’UTR、3’UTR、S蛋白、终止密码子、polyA核酸序列,构建得到FCoV的RNA疫苗,将其转染细胞后能够高效表达,免疫动物后能够产生高水平的抗体,该抗体具有较高的中和效价及滴度;

本发明的RNA疫苗能够实现同时诱导细胞免疫和体液免疫,可以做到无细胞生产、且生产时无需导入有毒化学品,能够高效迅速地对付病毒变异,相较传统疫苗,本发明的用于猫冠状病毒的RNA疫苗能够有效地缩短疫苗研发和生产周期,为防控FCoV的爆发提供有效的技术手段;

本发明的RNA疫苗的构建,成本低,生产效率高,因制造速度快,受微生物污染的几乎也相对较低,RNA疫苗的安全性相对较高,制备脂质纳米颗粒作为RNA疫苗的运送载体,脂质纳米颗粒具有核酸包封率高,能够有效转染,组织穿透性增强,细胞毒性和免疫原性低等特点,细胞转染后的脂质纳米颗粒FCoV-S-mRNA能够高效表达。

具体实施方式

以下通过具体的实施案例以及附图说明对本发明作进一步详细的描述,应理解这些实施例仅用于说明本发明而不用于限制本发明的保护范围,在阅读了本发明之后,本领域技术人员对本发明的各种等价形式的修改均落于本申请所附权利要求所限定。

若无特殊说明,本发明的所有原料和试剂均为常规市场的原料、试剂。

实施例

一种用于猫冠状病毒的RNA疫苗及其构建:

1.重组菌构建:

选取5’UTR、3’UTR、S蛋白(FCoV-I型中的UU4毒株S蛋白(GenBank:FJ938054.1))、终止密码子、polyA核酸序列进行人工合成RNA疫苗的DNA于pUC57载体上,无缝克隆于pUC57的EcoRV酶切位点,并转化大肠杆菌DH5α,获得FCoV-S-pUC57-DH5α;

本实施例所述S蛋白选用来自FCoV-I型中的UU4毒株S蛋白(GenBank:FJ938054.1),所述S蛋白还可选自FCoV-II型,也可以是所选S蛋白中的一段氨基酸序列或具有50-100%同源性的氨基酸序列,操作与上相同,在此不作重复赘述;

2.质粒的提取

将获得的重组菌进行摇菌,按照常规方法提取质粒,参照质粒抽提试剂盒(E.Z.N.A® Plasmid MiniprEppendorf Kit I)说明书进行:

2.1)取菌液于1.5mL Ep 管中12000r/min离心30s,弃上清;

2.2)加250µL溶液Ⅰ/RNase A,充分重悬菌泥;

2.3)加250µL溶液Ⅱ,颠倒4~6次混匀,室温作用2min至溶液变成澄清;

2.4)加350µL溶液Ⅲ,颠倒4~6次混匀以形成白色沉淀; 12000r/min离心10min,

2.5)转移上清至DNA吸附柱中,室温静置2min。12000r/min离心1min,弃液。

2.6)加入500 μL的Buffer HB,12000r/min离心1min,弃液。

2.7)加入750 μL的DNA Wash Buffer,12000r/min离心1min,弃液。

2.8)重复该步骤一次,再空管12000r/min离心1min。

2.9)移至新的1.5 mLEp管中,加入50~100 μL的TE室温下放置2min,12000r/min离心1min,洗脱的质粒于-20℃保存备用,取少量于1.0%琼脂糖凝胶中低压电泳检测质粒效果。

3.酶切线性化、纯化

对重组质粒进行Quick Cut MluI酶切,酶切反应体系如表1所示:

将上述成分加入0.5mL的EP管中,混匀,37℃酶切15min。纯化试剂盒进行纯化回收线性化重组质粒。

根据OMEGA公司的Cycle-pure纯化试剂盒使用说明书,线性化质粒进行纯化回收,方法如下:

3.1)将线性化质粒收集在一个1.5ml的EP管中;

3.2)加入6倍体积的Buffer CP于装有PCR产物的EP管中,小心混匀;

3.3)将上述混合液全部加至试剂盒配的有滤膜的管HiBind spin-column中,10000 r/min离心1 min,弃去离心出来的溶液;

3.4)加入700 µLSPW Buffer,等待2~3min,10 000 r/min离心1 min,弃去离心出来的溶液;

3.5)重复步骤3.4);

3.6)空管10 000 r/min离心1min;

3.8)弃去下面的离心管,将柱子放入新的干净的1.5mL离心管中,在柱子中央加入50µL DNA Elution Buffer,室温下放置2min,10 000r/min离心1min,收集离心管中的液体即为回收的DNA片段,经琼脂糖凝胶电泳检测后即用或-20℃冰箱保存备用。提高洗脱液的温度至55℃~80℃,有利于提高DNA的洗脱效率。

4.体外转录和纯化

采用体外转录试剂盒(mMESSAGE mMACHINE™ T7 ULTRA Transcription Kit)对线性化的质粒进行体外转录。

采用纯化试剂盒(MEGAclear™ Transcription Clean-Up Kit)对转录的产物进行纯化处理,即得RNA疫苗核酸;

取来自的猫源的六组5’UTR、3’UTR(选自GenBank中NCBI参考序列XM_023246781.1、XM_011290755.3、XM_003992859.2、XM_003992882.4、XM_003998693.4、X84842.1),按照以上步骤制备成六组RNA疫苗核酸,纯化之后的RNA疫苗核酸进行1%琼脂凝胶电泳鉴定,如图1所示,可见其体外转录成功。

5.纳米脂质颗粒的制备

5.1)将-80℃冰箱保存的聚合物PEG5k-b-PLGA11k,PLGA11k和BHEM-Chol取出,平衡到室温后在干燥环境下开封,分别以62.5 mg/mL,39.5mg/mL和22.5mg/mL的浓度溶于色谱级三氯甲烷溶液中;

5.2)将-80℃冰箱保存的mRNA溶液在冰上融化,使用无核酸酶水(DEPC水)调整其浓度为4 mg/mL,在50mL无菌离心管中,依次加入25 μL mRNA(0.1 mg),100 μL BHEM-Chol(2 mg),350 μL PEG5k-b-PLGA11k(21.875 mg)和50 μL PLGA11k(1.925 mg);

5.3)将该离心管置于冰水中,超声探头用DEPC水冲洗后放入离心管的溶液中,在80W功率和冰水浴条件下,以超声5s停2s的方式超声乳化溶液60 s,得到油相-水相的初乳液。随后,将5 mLDEPC水沿管壁加入到初乳液中,并在同样条件下,以超声10s停2s的方式再次乳化60 s,形成水相-油相-水相的复乳液;

5.4)将复乳液转移到DEPC处理并高压过的无菌无核酸酶100mL圆底烧瓶中,使用旋转蒸发仪在低温条件下缓慢地减压旋蒸以除去乳液中的三氯甲烷,并浓缩其水溶液体积到1mL左右,制备好的脂质纳米颗粒包裹的mRNA转移到1.5mL EP管中保存于4C冰箱中待用。

6.检测细胞转染后的FCoV-S-mRNA脂质体表达情况

6.1)取24孔板,接种猫肾细胞(CRFK)(200000个细胞/孔),采用含10%血清无抗培养基培养18小时;

6.2)当细胞贴壁融合度约达60 %时,在转染前一小时更换为无血清的Opti-MEM培养基,转染脂质纳米颗粒FCoV-S-mRNA(1μg RNA/孔),培养6小时;

6.3)将培养基替换为新鲜的培养基,培养42小时;

6.4)1000g离心10min,收集上清液;

6.5)取上清液,进行非还原型SDS-PAGE,然后进行Western blot。Western blot采用的一抗为抗E2蛋白的鼠源单克隆抗体,二抗为抗鼠源抗体。

7. FCoV-S-mRNA疫苗免疫小鼠及其抗体的测定

将6-8周龄雄性BALB/c小鼠随机分为5组(每组5只):FCoV-S-mRNA1组、FCoV-S-mRNA2组、FCoV-S-mRNA3组、FCoV-S-mRNA4组、FCoV-S-mRNA5组、FCoV-S-mRNA6组及PBS空白对照组。

采用肌内免疫,试验第1天进行初次免疫,试验第28天进行加强免疫,FCoV-S-mRNA中的各组组每次免疫剂量相同,均为20g,体积为200L,PBS空白注射组注射等体积的PBS(pH7.2,0.01M)。分别在二免14天后进行尾部采血,收集血清,用间接ELISA法测定抗体水平。

如图3所示,OD450测定结果显示FCoV-S-mRNA1组、FCoV-S-mRNA2组、FCoV-S-mRNA3组、FCoV-S-mRNA4组、FCoV-S-mRNA5组、FCoV-S-mRNA6组实验组均能够达到一定的抗体水平。

8.血清中和试验中中和效价及抗体滴度的测定

对第42d的血清进行血清中和试验,如表2所示,结果显示FCoV-S-mRNA1组、FCoV-S-mRNA2组、FCoV-S-mRNA3组、FCoV-S-mRNA4组、FCoV-S-mRNA5组、FCoV-S-mRNA6组抗体效价均显著高于PBS对照组。

如图4所示,抗体滴度测定试验中FCoV-S-mRNA1组、FCoV-S-mRNA2组、FCoV-S-mRNA3组、FCoV-S-mRNA4组、FCoV-S-mRNA5组、FCoV-S-mRNA6组实验组中和抗体滴度均高于24,可见,本发明制备的猫冠状病毒RNA疫苗可以起到很好的保护作用,为FCoV的防控提供了有效的技术手段。
