3. Use of a compound of general formula (I) as defined in claim 1 for the preparation of a medicament for the treatment or prevention of diseases and conditions which are affected by modulation of TMEM 16A.

4. A method of treatment or prevention of diseases and conditions which are affected by modulation of TMEM16A, which method comprises administering to a patient in need of such treatment an effective amount of a compound of general formula (I) as defined in claim 1.

5. The compound for use, the use or the method according to any one of claims 2 to 4, wherein the disease or disorder which is affected by modulation of TMEM16A is selected from respiratory diseases and disorders, dry mouth (xerostomia), intestinal hyperactivity, cholestasis and ocular symptoms.

6. The compound for use, use or method according to claim 5 wherein the respiratory diseases and disorders are selected from the group consisting of cystic fibrosis, Chronic Obstructive Pulmonary Disease (COPD), chronic bronchitis, emphysema, bronchiectasis including non-cystic fibrosis, asthma and primary ciliary dyskinesia.

7. The compound for use, the use or the method according to claim 5, wherein the dry mouth (xerostomia) is caused by sjogren's syndrome, radiotherapy or a drug causing dry mouth.

8. The compound for use, the use or the method according to claim 5, wherein the intestinal hyperactivity is associated with gastric dyspepsia, gastroparesis, chronic constipation or irritable bowel syndrome.

27. A pharmaceutical composition comprising a compound according to claims 24 and 25 and a pharmaceutically acceptable excipient.

The pharmaceutical composition according to claim 27, which is formulated for oral, rectal, nasal, bronchial (inhalation), topical (including dermal, transdermal, eye drop, buccal and sublingual), vaginal or parenteral (including subcutaneous, intramuscular, intravenous and intradermal) administration.

29. The pharmaceutical composition according to claim 27 or 28, further comprising an additional active agent for the treatment or prevention of a respiratory disorder.

30. A product containing a compound of general formula (I) and a further active agent for the treatment or prophylaxis of a respiratory disorder as a combined preparation for simultaneous, sequential or separate use in the treatment of a disease or disorder which is affected by modulation of TMEM 16A.

31. A compound for use, use or method according to claim 23, a pharmaceutical composition according to claim 29 or a product according to claim 30, wherein the additional active agent for treating or preventing a respiratory disorder is selected from:

beta 2 adrenoceptor agonists such as metaproterenol, isoproterenol, albuterol, salbutamol, salmeterol, indacaterol, terbutaline, metaproterenol, bitolterol mesylate, pirbuterol, metaproteterol, vilanterol, and abediterol;

Antihistaminic agents, e.g. histamine H1Receptor antagonists, for example loratadine, cetirizine, desloratadine, levocetirizine, fexofenadine, astemizole and chlorpheniramine or H4A receptor antagonist;

a streptococcal enzyme alpha;

corticosteroids such as prednisone, adrenocorticosterone, flunisolide, triamcinolone acetonide, beclomethasone dipropionate, budesonide, fluticasone propionate, mometasone furoate, and fluticasone furoate;

leukotriene antagonists such as montelukast and zafirlukast;

anticholinergic compounds, in particular muscarinic antagonists, such as ipratropium, tiotropium, glycopyrrolate, aclidinium and umeclidinium bromide;

32. A process for preparing a pharmaceutical composition according to claim 27 or 28, which process comprises bringing a compound according to claim 24 or 25 into association with an excipient.

Examples

The invention is illustrated by the following examples.

General conditions:

mass spectra were performed on LC-MS systems using electrospray ionization, these were run using a Waters Acquity uPLC system with a Waters PDA and ELS detector or a Shimadzu LCMS-2010EV system. [ M + H ] + means a monoisotopic molecular weight.

NMR spectra were recorded on Bruker Avance III HD 500MHz, Bruker Avance III HD 250MHz with a 5mm Broad Band Inverse probe, or SmartProbe 400MHz Avance III HD Nanobay equipped with a 5mm Broad Band observer SmartProbe, using solvent as internal deuterium lock. Unless otherwise stated, spectra were recorded at room temperature and referenced using solvent peaks.

With reference to the following examples, compounds of the preferred embodiments are synthesized using the methods described herein or other methods known in the art.

The various starting materials, intermediates and compounds of the preferred embodiments can be isolated and purified, where appropriate, using conventional techniques such as precipitation, filtration, crystallization, evaporation, distillation and chromatography. All starting materials were obtained from commercial suppliers and used without further purification unless otherwise indicated. Salts may be prepared from the compounds by known salt-forming methods.

Preparative HPLC using acidic pH, early elution method

Purification was performed on a Gilson LC system using a Waters Sunfire C18 column (30mm x 100mm, 10 μ M; temperature: RT) with a gradient of 10-95% B (a ═ water containing 0.1% formic acid; B ═ acetonitrile containing 0.1% formic acid) over 14.44 minutes, followed by 2.11 minutes at 95% B, an injection volume of 1500 μ L, a flow rate of 40mL/min, and UV spectra were recorded at 215nm using a Gilson detector.

Preparative HPLC using acidic pH, Standard elution method

Purification by preparative HPLC (acidic pH, standard elution method) was carried out on a Gilson LC system using a waters sunfire C18 column (30mm x 100mm, 10 μ M; temperature: RT) with a gradient of 30-95% B (a ═ water containing 0.1% formic acid; B ═ acetonitrile containing 0.1% formic acid) over 11 minutes, then at 95% B for 2.11 minutes with an injection volume of 1500 μ L and a flow rate of 40mL/min, and UV spectra were recorded at 215nm using a Gilson detector.

Preparative HPLC using basic pH, early elution method

Purification by preparative HPLC (basic pH, early elution method) was performed on a Gilson LC system using a Waters Xbridge C18 column (30mm x 100mm, 10 μ M; temperature: RT) with a gradient of 10-95% (a ═ water containing 0.2% ammonium hydroxide; B ═ acetonitrile containing 0.2% ammonium hydroxide) over 14.44 minutes, then 95% B for 2.11 minutes, an injection volume of 1500 μ Ι _ and a flow rate of 40mL/min, with a Gilson detector recording the UV spectrum at 215 nm.

Preparative HPLC using basic pH, Standard elution method

Purification by preparative HPLC (basic pH, standard elution method) was carried out on a Gilson LC system using a waters xbridge C18 column (30mm x 100mm, 10 μ M; temperature: RT) with a gradient of 30-95% (a ═ water containing 0.2% ammonium hydroxide; B ═ acetonitrile containing 0.2% ammonium hydroxide) over 11 minutes, then with 95% B for 2.11 minutes, an injection volume of 1500 μ L and a flow rate of 40mL/min, and UV spectra were recorded at 215nm using a Gilson detector.

All evaporations were carried out under vacuum, if not mentioned otherwise, preferably between about 15 and 100mm Hg (═ 20-133 mbar).
