技术领域

本发明属于生物医药技术领域,尤其是指TBC1D3及其家族在制备肿瘤诊断药物与预后判断药物中的应用。

背景技术

肾透明细胞癌是肾癌中最常见的恶性肿瘤,占原发性肾脏恶性肿瘤的75-82%。在各 种临床和基因组研究中,肾透明细胞癌被证明是一种高度免疫浸润的肿瘤,是最早的免疫治疗肿瘤之一。由于肾透明细胞癌对常规放疗和化疗不敏感,其治疗主要依赖于靶向 治疗和免疫治疗。靶向治疗包括酪氨酸激酶抑制剂,如舒尼替尼和索拉非尼。然而,肿 瘤异质性、动态变化和对治疗的适应细胞死亡相关信号通路的改变显著限制了靶向药物 在肾透明细胞癌治疗中的应用。

发明内容

本发明通过在线数据库发现肾透明细胞癌组织中TBC1D3及其家族基因的表达显著 高于癌旁正常组织;肾透明细胞癌中TBC1D3及其家族蛋白的表达水平与患者的预后密切相关,肾透明细胞癌组织中TBC1D3及其家族蛋白表达水平高的患者总的生存率低于 表达低的患者。同时我们构建TBC1D3质粒,转染肾透明细胞癌细胞,发现可以促进其 细胞增殖。同时发现TBC1D3家族成员与免疫浸润相关。

RNA干扰(RNA interference,RNAi)是指内源性或外源性双链RNA介导的细胞内mRNA发生特异性降解,导致靶基因的表达沉默,产生相应的功能表型缺失。它可高效、 特异地阻断体内特定基因的表达,导致其降解,从而引起生物体内特异基因的沉默,使 细胞表现出某种基因表型的缺失,是近年来新兴的一种常用的研究基因功能、寻找疾病 治疗方法的实验室技术。目前,RNAi特异性的抑制基因表达,已应用于基因病、病毒 感染性疾病、癌症的基因治疗等方面。

TBC1D3、TBC1D3B、TBC1D3C、TBC1D3D、TBC1D3E、TBC1D3F、TBC1D3G 或TBC1D3H作为肿瘤诊断及预后标记物的应用。

在本发明的一个实施例中,所述TBC1D3、TBC1D3B、TBC1D3C、TBC1D3D、 TBC1D3E、TBC1D3F、TBC1D3G或TBC1D3H的核苷酸序列分别如SEQ ID NO.1-SEQ ID NO.8所示。

在本发明的一个实施例中,所述肿瘤为肾透明细胞癌、肾乳头状细胞癌、肝癌、肺腺癌、肺鳞状细胞癌和甲状腺癌。

TBC1D3、TBC1D3B、TBC1D3C、TBC1D3D、TBC1D3E、TBC1D3F、TBC1D3G 或TBC1D3H的RNA干扰靶点序列分别如SEQ ID NO.9-SEQ ID NO.14所示。

所述的TBC1D3、TBC1D3B、TBC1D3C、TBC1D3F、TBC1D3G或TBC1D3H的 RNA干扰靶点序列在制备治疗肿瘤药物中的应用。

在本发明的一个实施例中,所述肿瘤为肾透明细胞癌、肾乳头状细胞癌、肝癌、肺腺癌、肺鳞状细胞癌或甲状腺癌。

一种抗肿瘤的药物,所述药物中含有所述TBC1D3、TBC1D3B、TBC1D3C、 TBC1D3F、TBC1D3G或TBC1D3H的RNA干扰靶点序列。

在本发明的一个实施例中,所述药物还包括抑制CD4+T细胞或T细胞耗竭的抑制剂。

本发明首次证实TBC1D3及其家族是一个肾透明细胞癌预后标记物,首次发现TBC1D3可作为肾透明细胞癌诊断和预后判断的分子标记及治疗药物的靶标。TBC1D3 家族成员可以作为肾透明细胞癌的免疫治疗靶点。

具体实施方式

下面结合附图和具体实施例对本发明作进一步说明,以使本领域的技术人员可以更 好地理解本发明并能予以实施,但所举实施例不作为对本发明的限定。

实施例1 TBC1D3及其家族在肾功能透明细胞癌中的表达

通过GSCA数据库分析,TBC1D3在肾透明细胞癌中的表达。发现TBC1D3表达在 肾透明细胞癌、肾乳头状细胞癌、肝癌、肺腺癌、肺鳞状细胞癌和甲状腺癌中高表达,在 头颈部鳞状细胞癌中低表达。我们同时通过UALCAN数据库和下载肾透明细胞癌TCGA 数据,通过R语言分析数据,发现TBC1D3家族(TBC1D3,TBC1D3B,TBC1D3C, TBC1D3D,TBC1D3E,TBC1D3F,TBC1D3G,TBC1D3H)在肾透明细胞癌中相比于癌 旁组织都高表达。

实施例2 TBC1D3及其家族的基因和蛋白网络的基因改变

我们使用cBioportal数据库来确定TCGA KIRC样本中TBC1D3家族改变的类型和频率。结果显示TBC1D3家族成员很少发生突变(少于5个频率),这是高度保守的(结 果见图2-A)。然后,我们使用cBioportal的“相关分析”对TBC1D3家族的五个成员进行 分析,结果表明,TBC1D3家族成员之间的关联呈正相关,但TBC1D3C与TBC1D3G 呈负相关(结果见图2-D)。利用GeneMANIA和STRING生成的基因-基因和蛋白质-蛋 白质相互作用网络显示,20个潜在靶基因和11个潜在靶蛋白与TBC1D3家族相互作用 (结果见图2-B与图2-C)。

实施例3 TBC1D3家族成员在肾透明细胞癌中预后表达

使用GSCA、TISIDB、LinkedOmics和Kaplan-Meier绘图仪评估TBC1D3家族表达 对生存率的影响。GSCA分析显示,TBC1D3在KIRC多发性肿瘤中的表达与OS和PFS 呈正相关。此外,高表达的TBC1D3存活时间短(结果见图3-1)。TISIDB和LinkedOmics 的总生存率分析表明,TBC1D3家族成员高表达的患者总生存期较短(结果见图3-2与 图3-3)。为了进一步探讨TBC1D3在KIRC临床特征中的作用,我们使用Kaplan-Meier 绘图仪研究了TBC1D3表达与KIRC临床病理特征之间的关系。如表1所示,TBC1D3 和TBC1D3B的高表达与I期、II期、III期、IV期、3级和4级患者的OS恶化相关。 为了探讨TBC1D3表达是否是KIRC患者总生存率的独立预测因素,我们进行了单变量 和多变量Cox回归分析。单因素Cox回归分析显示,年龄、分级、分期、TNM分级均 为OS的独立危险因素(p=0.012、3.61E-08、1.26E-10、2.69E-08、2.76E-10和0.001); 在多变量Cox回归分析中,年龄、年级和TBC1D3表达是OS的独立危险因素(分别 p=0.0003、0.014和0.013)(结果见表2)。

实施例4 TBC1D3在肾透明细胞癌中的功能

为了研究TBC1D3家族在KIRC中的功能,我们使用CancerSEA进行了单细胞分析。结果表明,TBC1D3D正调控KIRC细胞的增殖,负调控炎症(图4A-B)。我们构建了 TBC1D3质粒并转染Caki-1细胞,结果显示TBC1D3能促进肾透明细胞癌细胞的增殖(结 果见图4-C)。TBC1D3的生物学过程通过过表达富集分析(ORA)进行鉴定,结果表明 TBC1D3的表达与免疫应答密切相关(结果见图4-D)。

实施例5 TBC1D3家族表达与免疫抑制剂在肾透明细胞癌中的相关性

选择TISIDB数据库研究TBC1D3家族表达与免疫抑制效应之间的关系。因此,TBC1D3、TBC1D3B、TBC1D3C和TBC1D3G是TBC1D3家族的四个成员,分别与CD160、 CTLA4、CD244、KDR、LAG3、PDCD1、PDCD1LG2和TIGIT相关。此外,TGFBR1 和HAVCR2与TBC1D3、TBC1D3B和TBC1D3G相关。CD274与TBC1D3B、TBC1D3C 和TBC1D3G相关。此外,TBC1D3与LGALS9相关。TBC1D3B与CD274相关。TBC1D3C 与CD96和LGALS9相关。TBC1D3G与IL10RB和PVRL2相关(结果见图5-1,图5-2, 图5-3,图5-4)。

实施例6 TBC1D3家族表达与免疫浸润在肾透明细胞癌中的相关性

采用TIMER数据库探讨TBC1D3家族表达与肾透明细胞癌浸润淋巴细胞的关系。TBC1D3家族表达与CD4+T细胞浸润水平呈正相关。此外,巨噬细胞浸润水平仅与 TBC1D3表达显著相关,树突状细胞浸润水平与TBC1D3B表达呈负相关。中性粒细胞 浸润水平与TBC1D3和TBC1D3H表达呈正相关(结果见图6-1,图6-2,图6-3,图6-4, 图6-5)。为了进一步证实TBC1D3在KIRC中的表达与免疫细胞浸润水平之间的关系, 我们使用TIMER数据库来探讨TBC1D3表达与各种免疫浸润相关标记物之间的关系。 我们的结果显示,TBC1D3的表达与大多数中性粒细胞、Th1、Th2、Treg和T细胞衰竭 的标志物之间存在显著相关性(结果见表3)。特别是T细胞衰竭,结果与DISTIB分析 一致。体细胞拷贝数改变(SCNA)模块显示TBC1D3家族成员的臂水平缺失与肾透明 细胞癌中的免疫细胞浸润水平显著相关(结果见图7-1,图7-2,图7-3)。
