Background

Ceftiofur has the advantages of strong bactericidal power, wide antibacterial spectrum, small toxic effect, small allergic reaction and the like, and is widely applied to prevention and treatment of bacterial diseases of beef cattle, dairy cows, pigs, sheep, poultry and the like in veterinary clinic. Ceftiofur is absorbed mainly in the intestinal tract after oral administration. Related researches show that the bioavailability of ceftiofur in duck bodies is less than 90%, the ceftiofur belongs to low-permeability medicines, and the low bioavailability of ceftiofur is possibly related to efflux transporters on intestinal mucosa, particularly P-glycoprotein.

Luteolin is a natural compound contained in a plurality of plants, and particularly, a great amount of luteolin is contained in fruits and vegetables which are eaten by people at ordinary times. Because it has pharmacological actions of bacteriostasis, antivirus, immunoregulation, anti-tumor, anti-inflammation, antioxidation, etc., it can be used for preventing and curing diseases. Therefore, people can change the diet to determine the intake of luteolin, so as to reduce the occurrence of diseases. So far, no report about the influence of flavonoid compounds such as luteolin on the absorption of ceftiofur in the duck intestinal tract is available.

In the following embodiment, a duckling in vitro intestine turning method is adopted for research, an in vitro intestine turning bag model is established by taking a section of jejunum of a healthy duckling at the age of 4 weeks, an intestine turning bag is prepared, and the intestinal activity of the intestine turning bag is detected; establishing an HPLC detection method for ceftiofur and luteolin, detecting the concentrations of the luteolin and the ceftiofur by an HPLC method, and calculating the apparent permeability coefficient, the absorptivity and the absorption rate parameters of the two, thereby verifying that the luteolin can promote the permeability of the ceftiofur in the organism. The method has the advantages of strong controllability of test conditions, short test period, simplicity and feasibility, and good in-vivo correlation of test data.

Ceftiofur hydrochloride used in the following examples, purity 92.1%, was purchased from zilu animal health products, ltd; de-furoyl ceftiofur (DFC) standard, 98.0% pure, available from sigma; luteolin standard with purity of 98% is purchased from Shanghai Purui Biotech limited; dichloroerythrol (DTE) with a purity of 99.0%, Biotechnology engineering (Shanghai) Ltd.
