本申请涉及酶工程及生物制药领域,具体涉及一种转氨酶及其在制备西他列汀中的应用。

背景技术

西他列汀(Sitagliptin)是用于治疗Ⅱ型糖尿病的有效药物,全称为(3R)-3-氨基-1-[3-(三氟甲基)-5,6-二氢-1,2,4-三唑并[4,3-a]吡嗪-7(8H)-基]-4-(2,4,5-三氟苯基)丁-1-酮,其为一种二肽基肽酶-4(DPP-4)抑制剂,主要通过保护内源性肠降血糖素和增强其作用而控制血糖水平,具有较好的安全性和耐受性。

在现有技术中,制备西他列汀的方法主要有化学合成法和生物合成法,其中,化学合成法虽然工艺成熟,但是具有工艺步骤繁多、所用试剂(如金属催化剂等)昂贵、部分试剂(如金属催化剂等)对环境和人体具有毒害作用等缺点。生物合成法是利用酶作为催化剂来制备西他列汀,目前生物合成法所用的酶大多为转氨酶(Transaminase,TA),尤其是ω-转氨酶。转氨酶能够进行将西他列汀前体酮不对称转氨形成西他列汀中间体的催化反应,但是自然界存在的野生型转氨酶大多对上述催化反应无催化活性或催化活性较低,因而有必要对其进行定向改造以提高所述催化反应的转化率和产物收率,如Codexis公司对一种源自节杆菌属(Arthrobacter sp.)的ω-转氨酶进行改造,获得的转氨酶突变体对西他列汀前体酮1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-1,3-丁二酮具有高催化活性。

但是,现有的生物合成法仍存在诸多缺陷,例如:(1)由于现有的转氨酶活性较低,所以反应体系中需加入大量酶液,使得反应结束后大量的游离蛋白质残存于反应体系中,不利于产物的分离纯化;(2)游离转氨酶稳定性较差,对酶的储存环境与运输操作要求高;(3)转氨酶对有机溶剂的耐受性差,在含有有机溶剂的反应液中难以用于长时间反应。因此,有必要研发新型转氨酶以制备西他列汀。

发明内容

本申请提供了一种转氨酶及其在制备西他列汀中的应用,以改善现有技术中生物合成法制备西他列汀存在的问题。

第一方面,本申请提供了一种转氨酶,所述转氨酶的氨基酸序列如SEQ ID NO:1所示,或者为与所述SEQ ID NO:1至少具有80%、至少具有85%、至少具有90%、至少具有95%、至少具有96%、至少具有97%、至少具有98%或至少具有99%相似性的氨基酸序列。

进一步地,所述转氨酶的氨基酸序列是由SEQ ID NO:1所示的氨基酸序列发生一个或多个点突变得到的氨基酸序列。

进一步地,发生所述点突变的氨基酸包括SEQ ID NO:1中的第34位氨基酸T、第77位氨基酸H、第80位氨基酸F、第153位氨基酸Y、第155位氨基酸I、第158位氨基酸L、第163位氨基酸P、第181位氨基酸S、第183位氨基酸A、第185位氨基酸V、第199位氨基酸V、第218位氨基酸S、第237位氨基酸T、第259位氨基酸R、第260位氨基酸D、第276位氨基酸R、第277位氨基酸G、第293位氨基酸V、第309位氨基酸W、第321位氨基酸M和第322位氨基酸D中的一个或多个。

进一步地,发生所述点突变的方式为T34A、H77N、F80A、Y153T、I155V、L158Q、P163F、S181T、A183R、V185R、V199I、S218C、T237V、R259E、D260S、R276Y、G277D、V293F、W309V、M321N或D322V。

在本申请的一些实施例中,所述转氨酶的氨基酸序列为SEQ ID NO:3、SEQ ID NO:5、SEQ ID NO:7、SEQ ID NO:9、SEQ ID NO:11、SEQ ID NO:13、SEQ ID NO:15、SEQ ID NO:17、SEQ ID NO:19、SEQ ID NO:21、SEQ ID NO:23、SEQ ID NO:25、SEQ ID NO:27、SEQ IDNO:29、SEQ ID NO:31、SEQ ID NO:33、SEQ ID NO:35、SEQ ID NO:37、SEQ ID NO:39、SEQ IDNO:41或SEQ ID NO:43所示的氨基酸序列。

第二方面,本申请提供了一种核酸分子,所述核酸分子包括用于编码第一方面中任一种所述转氨酶的核苷酸序列,所述核苷酸序列为下述任一项:

(a)所述核苷酸序列如SEQ ID NO:2所示;

(b)所述核苷酸序列与所述SEQ ID NO:2至少具有80%、至少具有85%、至少具有90%、至少具有95%、至少具有96%、至少具有97%、至少具有98%或至少具有99%相似性;或

第四方面,本申请提供了一种转化体,所述转化体包括表达系统,以及如第二方面中任意一种所述的核酸分子或如第三方面中所述的重组表达载体。表达系统可以是真核生物,也可以是原核生物。

第五方面,本申请提供了一种酶制剂,所述酶制剂包括第一方面中任一种所述的转氨酶本领域技术人员可以理解的是,所述酶制剂还可以包括食品或药学上可接受的佐剂和/或辅料,以达到赋形、保护酶活性成分、提高酶稳定性等目的。

第六方面,本申请提供了如第一方面中任意一种所述的转氨酶、或如权第二方面中任意一种所述的核酸分子、或如第三方面中所述的重组表达载体、或如第四方面中所述的转化体、或如第五方面中所述的酶制剂在制备西他列汀中的应用,所述转氨酶或所述酶制剂,或者经由所述核酸分子、所述重组表达载体以及所述转化体中的至少一种生成的转氨酶,与底物西他列汀前体酮(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮转氨接触而发生酶催化反应,生成西他列汀中间体(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮。

本申请提供了一种转氨酶及其在制备西他列汀中的应用,该转氨酶或含有该转氨酶的酶制剂能够不对称催化西他列汀前体酮(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮转氨形成西他列汀中间体(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮,转氨酶的酶活可达1100U/g,在酶催化反应进行2h时,50μL的转氨酶酶液可使底物转化率达到55%。相较于催化同一反应的现有转氨酶,本申请的转氨酶具有活性高、稳定性理想、对有机溶剂耐受性佳的优点,将本申请的转氨酶应用于西他列汀的制备工艺中能够有利于产物的分离纯化。

本申请实施例提供了一种转氨酶及其在制备西他列汀中的应用,其中,转氨酶的氨基酸序列如SEQ ID NO:1所示,或者为与所述SEQ ID NO:1至少具有80%、至少具有85%、至少具有90%、至少具有95%、至少具有96%、至少具有97%、至少具有98%或至少具有99%相似性的氨基酸序列。该转氨酶能够不对称催化西他列汀前体酮(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮转氨形成西他列汀中间体(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮,反应式如(1)所示:

如本申请所用,“载体”是指能够运输另一种核酸的核酸分子,例如可以是质粒、病毒、粘粒等,在本申请实施例中,未插入外源基因的pUC-19质粒可以作为载体。

如本申请所用,“重组表达载体”是指一种DNA构建体,其含有与合适的控制序列可操作地连接的核酸分子,所述控制序列能够实现核酸分子在合适的表达系统中表达。在本申请实施例中,重组表达载体是指采用分子生物学技术将外源基因插入至载体上,由此形成的DNA构建体,示例pUC19-WT为重组表达载体。

如本申请所用,“表达系统”是指用于表达外源基因而产生蛋白的一类宿主,表达系统例如可以是真核生物、原核生物、病毒等,其中,作为表达系统的真核生物包括但不限于哺乳动物细胞、酵母、真菌、昆虫细胞和植物细胞,作为表达系统的原核生物包括但不限于芽孢杆菌属、梭菌属、乳酸菌属、链霉菌属、葡萄球菌属、大肠杆菌、假单胞菌属和类芽孢杆菌属。在本申请实施例中,pUC19-WT的表达系统为大肠杆菌。

如本申请所用,“酶制剂”是指从生物(如微生物)中提取的具有生物催化能力的物质,辅以其他成分而制得的功能化产品。在本申请实施例中,酶制剂的活性成分为转氨酶,转氨酶的氨基酸序列如SEQ ID NO:1所示,或者转氨酶的氨基酸序列为与所述SEQ ID NO:1至少具有80%、至少具有85%、至少具有90%、至少具有95%、至少具有96%、至少具有97%、至少具有98%或至少具有99%相似性的氨基酸序列,该酶制剂能够应用于西他列汀的生产工艺中。在一些实施例中,所述酶制剂可包括必要的赋形剂、稳定剂、保护剂等食品或药学上可接受的佐剂和/或辅料。

一、本申请实施例中涉及培养基的说明

(1)LB培养基

每100mL的LB液体培养基中,包括:1.0g的蛋白胨,0.5g的酵母粉,以及1.0g的NaCl。

对于LB固体培养基,是在LB液体培养基配方的基础上添加20g/L的琼脂。

(2)自诱导培养基

分别称取120g的酵母粉、60g胰蛋白胨、0.75g的硫酸镁(MgSO4)、16.5g的硫酸铵((NH4)2SO4)、32.5g的磷酸二氢钾(KH2PO4)、35.5g的磷酸氢二钠(Na2HPO4)、2.5g的葡萄糖以及10g的α-乳糖,然后将各个称取好的组分全部加入至磨粉机内,充分研磨至粉状,获得粉末状的自诱导培养基。将粉末状的自诱导培养基溶于1L去离子水,充分混匀后调节pH至7.0,然后121℃灭菌30min。

本申请实施例中涉及的基因片段,包括引物、如SEQ ID NO:2所示的核苷酸序列等由生工生物工程(上海)股份有限公司合成。

本申请实施例中涉及的限制性内切酶(如:BamH I、Nde I和Dpn I)、T4连接酶、KOD高保真酶试剂盒、胶回收试剂盒、10×T4连接酶缓冲液(Buffer)、双蒸水(ddH2O)等分子试剂均购自宝生物工程(大连)有限公司。

本申请实施例中涉及的底物化合物(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮,以及(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮标准品商购获得。

下面结合实施例进一步说明本申请的技术方案和有益效果。

实施例1:筛选转氨酶以及构建包含转氨酶编码基因的重组表达载体

1.1筛选转氨酶

在杭州市海宁市长安镇的某稻田中采集5~15cm的土壤样品,以作为转氨酶的筛选样本。称取5g的土壤样品,并置于50mL的离心管中,然后加入5mL的0.1%(w/w)焦磷酸钠溶液,颠倒数次混匀后置于室温条件下20min,其中每隔10min轻柔颠倒混匀;接着,在室温条件下12000×g离心10min,去除上清液以收集沉淀物;称取0.5g的沉淀物,采用FastDNA试剂盒对称取的沉淀物进行基因组抽提操作,获得土壤基因组。

根据转氨酶上下游的保守氨基酸序列设计PCR引物对F1(上游引物)和R1(下游引物),并在引物对的端部添加酶切位点BamH I和Nde I,其中,F1的核苷酸序列如SEQ ID NO:73所示,R1的核苷酸序列如SEQ ID NO:74所示。PCR体系选用KOD体系(商购,购自宝生物工程(大连)有限公司),模板为上述提取的土壤基因组。PCR反应条件为:95℃预变性5min;95℃变性30s;55℃退火30s;72℃延伸1.5min,30个循环;72℃延伸10min;4℃保持。

PCR反应结束后,对PCR产物进行琼脂糖凝胶电泳,琼脂糖凝胶电泳的结果如图1所示,对片段大小为500~2000bp的条带进行切胶回收,然后对切胶回收的多个DNA样品分别进行测序,获得对应的多个核苷酸序列。运用NCBI数据库的Blast对比分析功能,筛选获得一条与现有转氨酶编码基因的核苷酸序列相似度较高的核苷酸序列,该核苷酸序列如SEQID NO:2所示,SEQ ID NO:2所示核苷酸序列所编码的转氨酶的氨基酸序列如SEQ ID NO:1所示,其中,SEQ ID NO:1所示的氨基酸序列与NCBI登录号为RBP85366.1的氨基酸序列相似度为94.61%。

1.2构建重组表达载体pUC19-WT

选择未插入外源基因的pUC-19质粒作为载体,pUC-19质粒上具有酶切位点BamH I和Nde I,pUC19-WT的构建包括如下步骤:

S1.21、人工合成如SEQ ID NO:2所示的核苷酸序列,然后采用BamH I和Nde I限制性内切酶对合成的核苷酸序列进行双酶切,1%琼脂糖凝胶电泳检测酶切完全后,胶回收目的基因片段,其中,双酶切后回收目的基因片段的操作根据胶回收试剂盒操作说明实施;

S1.22、采用BamH I和Nde I限制性内切酶对pUC-19质粒进行双酶切,1%琼脂糖凝胶电泳检测酶切完全后,胶回收载体骨架,其中,双酶切后回收载体骨架的操作根据胶回收试剂盒操作说明实施;

S1.23、将步骤S1.21获得的目的基因片段,与步骤S1.22获得的载体骨架相混合,在T4连接酶的作用下16℃连接过夜,然后将连接产物转化至DH5a感受态细胞内,挑取单克隆子测序验证,提取测序正确的重组质粒,获得包含转氨酶编码基因的重组表达载体,命名pUC19-WT,其中,连接体系为20μL,具体是:2μL的10×T4连接酶缓冲液(Buffer)、5μL的目的基因片段、5μL的载体骨架、2μL的T4连接酶以及6μL的双蒸水(ddH2O)。

实施例2构建含有转氨酶编码基因的基因工程菌

将具有如SEQ ID NO:1所示氨基酸序列的转氨酶进行一个或多个点突变,以获得多个转氨酶突变体,其中,具体突变方式详见表2。

采用定点突变策略,根据待突变的氨基酸位点利用Oligo7软件来设计点突变引物,通过在上下游突变引物的5’端以插入、替换或缺失碱基的方式引入突变。选择实施例1中构建的重组表达载体pUC19-WT为模板,采用KOD高保真酶试剂盒进行反向PCR,从而获得转氨酶突变序列。使用Dpn I限制性内切酶处理获得的转氨酶突变序列,酶切产物经T4连接酶连接后转化大肠杆菌BL21(DE3)感受态,随后涂布含卡那霉素的LB抗性平板,置于37℃倒置培养18h,挑选单菌落转接含卡那霉素的LB液体培养基中,挑选培养液送样测序,将测序正确的克隆子保存备用,从而获得以大肠杆菌为宿主的基因工程菌。

需要说明的是,本领域技术人员根据突变位点引入的方式,结合引物设计的基本原则可以获得突变引物的核苷酸序列。以构建T34A为例,用于反向PCR扩增的上游引物的核苷酸序列如SEQ ID NO:75所示,用于反向PCR扩增的下游引物如SEQ ID NO:76所示。

此外,反向PCR的反应程序为:95℃预变性3min;98℃变性30s,55℃退火30s,68℃延伸3min,28个循环;72℃延伸5min。

实施例3含有转氨酶编码基因的基因工程菌的诱导表达及后处理

将实施例2获得的基因工程菌接种至含50μg/mL卡那霉素的LB液体培养基中,在37℃、180r/min的条件下培养至OD600为0.6~0.8,获得种子菌液。将种子菌液以1%的体积浓度接种至新鲜的含50μg/mL卡那霉素的自诱导培养基,置于30℃培养18h,获得培养液。将培养液在25℃、8000r/min的条件下离心10min,弃上清液以收集沉淀物,将沉淀物用pH为7.0的PB缓冲液清洗数遍,收集湿菌体备用。

通过超纯水重悬制得的湿菌体,以制得菌体浓度为20%的菌液。采用超声波破碎法或高压均质破碎法处理菌液,破碎条件可依据实际需要自行选择。示例超声波破碎法的工作参数为:破碎1s;暂停2s;在180W的功率下,破碎10min。示例高压均质破碎法的工作参数为:在50HZ和800bar的条件下,破碎两次。

菌液破碎处理后,在4℃、12000r/min的条件下离心10~15min,以除去细胞碎片和大分子杂质,收集上清液保存于-20℃和4℃以备用,上清液即为含有转氨酶的酶液。

实施例4含有转氨酶编码基因的基因工程菌所表达的转氨酶性质测定试验

取实施例3制得的酶液50μL置于反应板上,向酶液中加入0.75mL pH为8.5的储备混合物,其中,储备混合物包含0.2mol/L的三乙醇胺、2mmol/L的磷酸吡哆醛(PLP)、2mol/L的异丙胺(IPM)以及无菌水,获得混合体系。随后,向混合体系中加入200μL含有25g/L底物化合物的无水乙醇以进行起始反应,获得反应体系,其中底物化合物为(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮。测定反应体系中各个组分的含量为:5g/L的底物化合物、1.5mol/L的IPM、2mmol/L的PLP以及20%(v/v)的无水乙醇,并且整个反应体系的pH值为8.5。

将含有上述反应体系的反应板热密封,在45℃、200r/min的摇动条件下持续反应2h,待反应结束后,利用高效液相色谱(High Performance Liquid Chromatography,HPLC)法对反应产物进行检测分析。

其中,HPLC的仪器型号为Agilent 1260(安捷伦),HPLC的工作条件如下:

(1)进样液的制备:取50μL的反应产物,向其中加入1mL的含饱和氯化钠的无水异丙醇,然后加入10μL的5mol/L氢氧化钠溶液,充分混匀离心以收集上清液;取500μL的上清液,向其中加入500μL的无水异丙醇,充分混匀制得进样液,每次进样量为10μL;

(2)色谱柱:CHIRALPAK IC,4.6*150mm,5μm。

(3)流动相的制备:将无水正己烷、无水乙醇和无水三乙胺按照600:400:1的体积比混合配制而成。

(4)流速:1mL/min。

(5)检测波长:268nm。

(6)柱温:30℃。

根据底物化合物的减少量计算底物转化率(%),底物转化率(%)的测定结果详见表2,底物转化率(%)的计算公式如下式(2):

在式(2)中,A1为反应后的底物化合物峰面积,A2为反应前的底物化合物峰面积。

根据转氨反应生成(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮产物的质量M(mg)、转氨反应时间以及细胞质量m(g)来计算转氨酶的酶活(U/g),其中,细胞质量m(mg)是指实施例3中所述湿菌体的质量,酶活(U/g)测定结果详见表2,其中,M的计算公式如下式(3),酶活(U/g)的计算公式如下式(4):

在式(3)中,C为(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮标准品的浓度(mg/mL),A3为(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮标准品的峰面积,A4为转氨反应生成的(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮产物的峰面积,V为整个反应体系的体积(mL)。

在式(4)中,M为转氨反应生成(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮产物的质量(mg),T为转氨反应时间(h),m为细胞质量(mg)。

下面详细说明编号1、编号17以及编号36所对应转氨酶的底物转化率和酶活的推导过程。

在本实施例中,C为0.472mg/mL,图2示出了(3R)-3-氨基-1-[3-(三氟甲基)-5,6,7,8-四氢-1,2,4-三唑并[4,3-a]吡嗪-7-基]-4-(2,4,5-三氟苯基)丁-1-酮标准品的HPLC图谱,其中A3为1257276。图3示出了初始反应时反应体系中底物化合物的HPLC图谱,其中A2为1215423。此外,细胞质量m为10mg,转氨反应时间T为2h,整个反应体系的体积V为1mL,稀释倍数为100倍。

图4示出了包含编号1转氨酶的反应体系反应2h时的HPLC图谱,其中,A1为1203269,按式(2)计算得出底物转化率为1%;A4为10650,按式(3)计算得出M为0.004mg;按式(4)计算得出酶活为20U/g。

图5示出了包含编号17转氨酶的反应体系反应2h时的HPLC图谱,其中,A1为948030,按式(2)计算得出底物转化率为22%;A4为234300,按式(3)计算得出M为0.088mg;按式(4)计算得出酶活为440U/g。

图6示出了包含编号36转氨酶的反应体系反应2h时的HPLC图谱,其中,A1为546940,按式(2)计算得出底物转化率为55%;A4为585750,按式(3)计算得出M为0.220mg;按式(4)计算得出酶活为1100U/g。

在表2中,编号2至编号36所示的转氨酶是在编号1所示转氨酶的基础上作出一个或多个位点突变而获得,相较于编号1所示的转氨酶,编号2至编号36所示转氨酶的转化率和酶活均有显著提高。采用T34A、F80A、L158Q、P163F、S181T、A183R、V185S、M321N和D322V组合突变方式获得的底物转化率和酶活最高,酶活可达1100U/g,在酶催化反应进行2h时,50μL的转氨酶酶液可使底物转化率达到55%。

在通过单一位点突变方式获得的转氨酶中,采用R276Y突变方式获得的底物转化率和酶活最高,其次是采用M321N突变方式获得的转氨酶;采用H77N突变方式获得的底物转化率和酶活最低。在通过多位点组合突变方式获得的转氨酶中,采用T34A、F80A、L158Q、P163F、S181T、A183R、V185S、M321N和D322V组合突变方式获得的底物转化率和酶活最高,其次是采用F80A、L158Q、P163F、S181T、R276Y和G277D组合突变方式获得的转氨酶;采用T34A和F80A组合突变方式获得的底物转化率和酶活最低,但其高于部分采用单一位点突变方式获得的转氨酶,例如:采用T34A突变方式获得转氨酶、采用Y153T突变方式获得转氨酶等。

对比例

选择来源于烟曲霉(Aspergillus fumigatus)的转氨酶编码基因,所述转氨酶编码基因的核苷酸序列如SEQ ID NO.77所示,人工合成所述转氨酶编码基因,并按照实施例1中1.2所示的方法构建获得重组表达载体pUC19-AF。

将pUC19-AF转化入大肠杆菌BL21(DE3)感受态,随后涂布含卡那霉素的LB抗性平板,置于37℃倒置培养18h,挑选单菌落转接含卡那霉素的LB液体培养基中,挑选培养液送样测序,将测序正确的克隆子保存备用,从而获得基因工程菌E.coli BL21(DE3)/pUC19-AF。

将E.coli BL21(DE3)/pUC19-AF按照实施例3的方法进行诱导表达及后处理,将获得的酶液保存于-20℃和4℃以备用。

试验例1转氨酶稳定性实验

选择编号36的转氨酶和对比例的转氨酶进行稳定性实验,按照酶液的保存时间4h、8h、16h、24h、48h、7d、15d和30d分别取样并检测酶活,其中酶活的检测方法按照实施例4进行。

按照下式(5)计算各个样品的酶活损失率,实验结果详见下表3:

在式(5)中,U0为酶液的初始酶活(U/g),UT为取样的酶活(U/g)。

表3转氨酶稳定性实验结果

由表3可知,随着保存时间的延长,编号36的转氨酶和对比例的转氨酶活性均降低,且4℃保存条件下的酶活损失率高于-20℃保存条件下的酶活损失率。在4℃保存条件下,对比例的转氨酶的酶活损失率在第7d已达100%,而编号36的转氨酶在第7d仍具有300U/g的酶活。在-20℃保存条件下,对比例的转氨酶和编号36的转氨酶保存30d后仍具有一定的酶活,并且编号36的转氨酶酶活是对比例的转氨酶酶活的2.3倍。本试验例充分说明:相较于对比例的转氨酶,编号36的转氨酶初始酶活更高,且稳定性更佳,更有利于运输及储存。

试验例2转氨酶对有机溶剂的耐受性实验

选择编号36的转氨酶和对比例的转氨酶进行有机溶剂耐受性实验,具体实验方法包括如下步骤:

Sa、取实施例3制得的酶液200μL置于反应板上,向酶液中加入1.0mL pH为8.5的储备混合物,其中,储备混合物包含0.2mol/L的三乙醇胺、10mmol/L的磷酸吡哆醛(PLP)、10mol/L的异丙胺(IPM)以及无菌水,获得混合体系;

Sb、向步骤Sa获得的混合体系中加入20mg的底物化合物(2Z)-4-氧代-4-[3-(三氟甲基)-5,6-二氢-[1,2,4]三唑并[4,3-a]吡嗪-7-(8H)-基]-1-(2,4,5-三氟苯基)丁-2-酮,然后加入有机溶剂(无水乙醇或DMSO)直至体系中有机溶剂的总浓度为20%(v/v)、30%(v/v)、35%(v/v)、40%(v/v)或50%(v/v),再加水补齐至总体系为2.5mL以进行起始反应,获得反应体系,整个反应体系的pH值为8.5;

Sc、将含有步骤Sb中反应体系的反应板热密封,在45℃、200r/min的摇动条件下持续反应2h,待反应结束后,利用HPLC对反应产物进行检测分析,并计算底物转化率,其中HPLC的检测条件和底物转化率的计算方法参照实施例4。

有机溶剂耐受性实验结果详见下表4

由表4可知,无论是采用无水乙醇作为有机溶剂,还是采用DMSO作为有机溶剂,编号36的转氨酶对有机溶剂的耐受性能均优于对比例的转氨酶。在酶催化反应条件、反应酶量(200μL)以及反应体系中除转氨酶之外的其他成分均相一致的前提下,编号36中转氨酶和对比例中转氨酶分别在同种浓度的有机溶剂中反应2h后,编号36中转氨酶的底物转化率均高于对比例的转氨酶,例如当有机溶剂为无水乙醇且浓度为50%(v/v)时,编号36中转氨酶的底物转化率是对比例中转氨酶的底物转化率的1.2倍,又如当有机溶剂为DMSO且浓度为50%(v/v)时,编号36中转氨酶的底物转化率是对比例中转氨酶的底物转化率的1.4倍。
