Compared with the classic ATAC-seq which counts a certain number of fresh primary cells or cell strains firstly and then carries out lysis by a lysis solution, the method can carry out preliminary lysis on the nuclear membrane of the cell nucleus while extracting the cell nucleus from the frozen tissue of the gastric cancer, and is convenient for subsequent experiments.
