Technical Field

Background

The dental caries has high morbidity and wide distribution, is a main common disease of the oral cavity and one of the most common diseases of human beings, can cause secondary pulpitis and periapical periodontitis and even can cause alveolar bone and jaw inflammation. If not treated in time, the lesion continues to develop, forming a cavity, eventually the crown is completely destroyed and disappears, the end result of which is the loss of teeth.

Bacteria are a prerequisite for caries, and the most common of the numerous bacteria that induce caries is streptococcus mutans. Streptococcus mutans living in human body can rapidly ferment various carbohydrates to generate a large amount of acid, can reduce local pH to below 5.5, and can continue to live and produce acid at pH 4.5 due to strong acid resistance of Streptococcus mutans. The local pH drop is maintained for a considerable time, avoiding the buffering action of saliva, thus causing local hard tissue to drop, inducing the onset of caries lesions. Culturing and detecting the quantity of the caries-causing acid-producing bacteria is an important method for judging the susceptibility value of the caries-causing acid-producing bacteria.

Detailed Description

The invention provides a primary screening culture medium for increasing the number of bacteria of acid-producing bacteria causing dental caries, which comprises the following components in parts by weight: 10-20 parts of starch, 0.1-0.3 part of monopotassium phosphate, 4-6 parts of saccharides, 0.4-0.6 part of sodium chloride, 80-120 parts of water and 0.003-0.007 part of indicator.

In the present invention, the weight part of the starch is preferably 12 to 18 parts, more preferably 14 to 16 parts, and still more preferably 15 parts.

In the present invention, the weight part of the monopotassium phosphate is preferably 0.15 to 0.25 part, and more preferably 0.2 part.

In the present invention, the amount of the saccharide is preferably 4.5 to 5.5 parts by weight, and more preferably 5 parts by weight.

In the present invention, the sodium chloride is preferably 0.45 to 0.55 part by weight, and more preferably 0.5 part by weight.

In the present invention, the water is preferably 90 to 110 parts by weight, more preferably 95 to 105 parts by weight, and still more preferably 100 parts by weight.

In the present invention, the indicator is preferably 0.004 to 0.006 part by weight, and more preferably 0.005 part by weight.

(2) heating and boiling for 20-30 min;

(3) supplementing water to the original water level height;

(4) adding sugar, potassium dihydrogen phosphate, sodium chloride and indicator to obtain blue suspension, and obtaining primary screening culture medium for increasing bacteria of the dental caries pathogenic acid-producing bacteria;

(5) filling 3ml of culture medium in each detection tube;

(6) the test tubes were sterilized at 121 ℃ for 15 min.

The invention also provides a use method of the primary screening culture medium for increasing the number of bacteria of the acid-producing bacteria causing dental caries, which preferably comprises the following steps:

(1) the samples were inoculated into the culture medium after 4 samples were rubbed on the teeth;

(2) culturing the inoculated culture medium at 37 ℃ for 24 h;

(3) and (4) measuring the pH value of the culture medium and judging the infection risk of the cariogenic tooth.
